Antithrombotic Effect of Galla Rhois (오배자의 항혈전 효과)

  • The possibility of Galla Rhois(GR) water extract as an antithrombotic agent was investigated. The effect of GR on platelet aggregation in human platelet-rich plasma(PRP) induced by collagen and ADP in vitro and coagulation parameters in a pathological model induced by endotoxin and hydrocortisone acetate(HA) in vivo were examined. In platelet aggregation assay, GR extract significantly inhibited platelet aggregation induced by collagen and ADP in a dose-dependent manner. GR extract significantly increased the number of platelet and shortened prothrombin time(PT) and activated thromboplastin time(APTT) as compared with the control in pathological model induced by endotoxin and HA. Also, GR extract significantly increased fibrinogen level as compared with the control in a pathological model induced HA. These results suggest that GR may be a promising antithrombotic agent.

A Study of Anticoagulation Activity from Perillae Folium Extract (자소엽(紫蘇葉) 추출물의 항응혈(抗凝血) 활성에 관한 연구(硏究))

  • Objectives: This research was investigated to find out the effect of the anticoagulant Perillae folium extract. Methods: To examine an active effect of anticoagulation in Perillae folium extract, the study measured Prothrombin time(PT) and activated partial thromboplastin time(APTT) of human plasma in vitro and measured bleeding time and arterio-venous shunt model in rats in vivo. Results: Bleeding time of Perillae folium extract in vivo had a significant increase 1.6 times and thrombus weight of Perillae folium extract had a significant reduction of thrombus weight as 68%. Perillae folium extract had an effect of anticoagulation by operating on extrinsic pathway factor II, V, VII, X and intrinsic pathway factor VIII, IX, X, VI, VII in the coagulation system. Conclusions: Considering the above mentioned results, it is judged that a Perillae folium extract has a control effect of thrombus creation.

Effect of Terminalia chebula on Immediate Hypersensitivity Reaction in Mice and Rats

  • We investigated the effect of aqueous extract of Terminalia chebula (Combretaceae)(TCAE) on the immediate hypersensitivity reaction in vivo and in vivo. TCAE (0.01 to 1 g/kg) dose-dependently inhibited compound 48/80 induced systemic anaphylaxis in mice. When TCAE was pretreated at concentrations ranging from 0.01 to 1 g/kg, the plasma histamine levels were reduced in a dose-dependent manner. TCAE (0.1 and 1 g/kg) significantly inhibited local immunoglobulin E (IgE)-mediated passive cutaneous anaphylactic reaction. TCAE (0.001 to 1 mg/ml) also dose-dependently inhibited the histamine release from rat peritoneal mast cells (RPMC) activated by compound 48/80 or anti-dinitrophenyl (DNP) IgE. TCAE (0.01 to 1 mg/ml) had a significant inhibitory effect on anti-DNP IgE-induced tumor necrosis $factor-{\alpha}$ production from RPMC. These results indicate that TCAE inhibits immediate hypersensitivity reaction in vivo and in vitro.

  • Polyacrylonitrile diagnostic membranes were prepared to measure blood glucose level of diabetics. Final absorbances at 680 nm through activated polyacrylonitrile membranes were measured at various concentration of glucose in plasma. The end-point results of K/S values obtained from the absorbances had a linear relationship toward the glucose concentration. The effect of temperature on the measurements of glucose concentration was studied. The stability of polyacrlonitrile diagnostic membranes was examined at RH 80%.

  • The covalent conjugation of human recombinant superoxide dismutase (hrSOD) with trichloros-triazine activated polyethylene glycol (PEG) 5000 formed soluble conjugates with molecular weight of 92KD, which retained $90{\sim}98%$ of original activity with a markedly prolonged plasma half-life of enzyme activity. The effect of hrSOD-PEG conjugates on acetaminophen (ACP)-induced hepatotoxicity was tested in male rats which were pretreated with 3-methylcholanthrene. HrSOD-PEG conjugates inhibited the hepatotoxicity produced by ACP, on the other hand, native hrSOD had no protective effect. The above results indicated that oxygen radicals might participate in the mechanism of the ACP-induced hepatotoxicity and that polymer conjugated-protein drugs with prolonged half-lives could be employed as an effective therapeutic agent.

A Novel Anticoagulant Protein from Scapharca broughtonii

  • An anticoagulant protein was purified from the edible portion of a blood ark shell, Scapharca broughtonii, by ammonium sulfate precipitation and column chromatography on DEAE-Sephadex A-50, Sephadex G-75, DEAE-Sephacel, and Biogel P-l00. In vitro assays with human plasma, the anticoagulant from 'S. broughtonii, prolonged the activated partial thromboplastin time (APTT) and inhibited the factor LX in the intrinsic pathway of the blood coagulation cascade. But, the fibrin plate assay did not show that the anticoagulant is a fibrinolytic protease. The molecular mass of the purified S. broughtonii anticoagulant was measured to be about 26.0kDa by gel filtration on a Sephadex G-75 column and SDS-PAGE under denaturing conditions. The optimum activity in the APTT assay was exhibited at pH 7.0-7.5 and $40-45^{\circ}C$ in the presence of $Ca^{2+}$.

Effect of AC-264, a Novel Indole Derivative, on Apoptosis in HL-60 Cells

  • The anticancer effect and apoptotic mechanism of a novel indole derivative AC-264, a lead derived from a chemical library, were investigated in human promyelocytic leukemia HL-60 cells. HL-60 cells treated with AC-264 at various concentrations showed the morphological features of apoptosis, such as plasma membrane blebbing and cell shrinkage. AC-264 exhibited cytotoxic effect in various cancer cell lines with different degrees of potency. Especially, AC-264 was effective on increasing the population of apoptotic cells in HL-60 cells, as detected by the number of cells stained with Annexin V and PI. Furthermore, AC-264 activated caspase-3 enzyme activity and induced internucleosomal DNA fragmentation. These results indicated that AC-264 produces anti-cancer effect via apoptotic cell death by activating caspase-3 and inducing internucleosomal DNA fragmentation in HL-60 cells.

  • Polyethylene glycol (PEG) is a water soluble, biocompatible, non-toxic polymer and PEGylation is a well established technique for the modification of therapeutic proteins and peptides. PEG-protein drugs have been extensively studies in relation to therapies for various diseases: cancer, inflammation and others. The covalent attachment of PEG to proteins and peptides prolonged plasma half-life, reduced antigenicity and immunogenicity, increased thermal and mechanical stability, and prevented degradation by enzymes. Several chemical groups for general and site specific conjugation have been exploited to activate PEG for amino group, carboxyl group, and cysteine groups. PEGylation of many proteins and peptides have been studied to enhance their properties for the potential uses. Also, the different positional isomers in several PEG-proteins have shown the difference in vivo stability and biological indicating that the site of PEG molecule attachment is one of the important factor to develop PEG-proteins as potential therapeutic agents.

  • To lower the fabrication cost of silicon solar cells, a surface treatment using a dielectric barrier discharge instead of a wet cleaning technique was examined on electrode surfaces on silicon solar cells. The fill factor obtained through measuring current-voltage characteristics was evaluated, and the treated surface state was characterized by energy-dispersive X-ray. It was found that the dielectric barrier discharge effectively activated the electrode surface and the surface treatment on finger electrodes contributed greatly to improve the fill factor.

Effects of High Voltage pulse on Seed Germination and Plant Growth

  • It is generally known that electron beam has sterilization effects and can activate plant germination and growth. Compared to electron beam, electrical pulse has not been frequently studied with respect to the biological application. In this study, we have analyzed the effects of high voltage pulse on seed germination and growth using various plant species. We have used the high voltage generator for examining seed's responses to the high voltage pulse. The operating voltage and currents of the generator are about 300 kV and 30 kA, respectively. Pulse width is 60 ns. High voltage pulse has slightly activated germination and growth of radish during early stage. Various levels of germination and growth are observed in different plant species after treated with high voltage pulse.
