Study on antioxidative, antidiabetic and antiobesity activity of solvent fractions of smilax china L. leaf extract (청미래덩굴잎 추출물 용매분획의 항산화, 항당뇨 및 항비만 활성연구)

  • Smilax china L., a native plant found in Asian countries, has several medicinal properties including antioxidant, anti-inflammatory, and anti-cancer effects. Although the root of the plant is commonly used as traditional herbal medicine in Korea and China, the medicinal properties of the leaves have not gained the same attention. In this study, we analyzed the antioxidant activity, ${\alpha}$-glucosidase inhibitory effect and lipid accumulation inhibition effect of Smilax china L. leaf water extract (SCLE) and its solvent fractions. SCLE was fractionated by using a series of organic solvents, including ethylacetate (EA) and n-butanol (BuOH). The EA fraction had the highest total polyphenol content ($440.20{\pm}12.67$ mg GAE/g) and total flavonoid content ($215.14{\pm}24.83$ mg QE/g). The radical scavenging activity $IC_{50}$ values of the EA fraction for 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2-azino-bis-(3-ethylbenzthiazoline)-6-sulfonic acid (ABTS) were 0.022 mg/mL and 0.13 mg/mL, respectively. Further, SOD-like activity and reducing power values of the EA fraction were higher than those of the other fractions. However, both the ${\alpha}$-glucosidase and lipid accumulation inhibition assays showed that the BuOH fraction ($83.35{\pm}4.18%$ at 1 mg/mL) and water extract ($11.27{\pm}2.67%$) were more effective than the EA fraction ($64.13{\pm}6.35%$, and $45.66{\pm}7.20%$). These results provide new insights into the potential anti-diabetic and anti-obesity effects of Smilax china L. leaf.

Comparative Analysis of the Amino Acids and Effective Components from Viola mandshurica (자화지정(Viola mandshurica)의 아미노산과 유용성분 비교 분석)

  • This study compared amino acids and effective components of Viola mandshurica produced in Korea with those of Chinese plants, as part of a study on nutritional andfunctional materials, to aid in the development of a valuable food. The contents of reducing sugar, free sugar, polyphenols, and soluble protein in the flowers of V. mandshurica were higher than in other fractions, being 10,485.11 mg%, 118.85 mg%, 1,259.26 mg%, and 23.67 mg%, respectively. Total flavonoid content of Chinese leaves was the highest at 2,370.75 mg%, followed by 1,844.43 mg% in Korean leaves. Free amino acids were highest in Korean leaves (5,345.21 mg%), followed by roots (4,692.24 mg%), flowers (3,807.87 mg%), and leaves of China plants (3,613.81 mg%). Arginine accounted for 55% of free amino acids in the roots, and all plant fractions contained more valine, aspartic acid, glutamic acid, alanine, and arginine, than other amino acids. The total content of amino acid derivatives was the highest (9,058.80 mg%) in Korean leaves. Especially, $\alpha$-aminoadipic acid constituted over 50% of total amino acid derivatives in all plant parts of V. mandshurica. When mineral contents were analyzed, leaves grown in China showed the highest content of 5,917.57 mg%, and the contents of K, Ca, and Mg were much greater than those of other minerals, together comprising over 95% of total mineral content in leaves and flowers from Korea.

Effects of Polyphenol and Catechin Levels on Antioxidant Activity of Several Edible Flower Extracts (주요 식용꽃 추출물의 폴리페놀과 카테킨류 함량이 항산화 활성에 미치는 영향)

  • Contents of phenolics, flavonoids, and catechins, and antioxidant activity were investigated in the ethanol extracts of three different flower species, Magnolia denudata, Prunus mume, Carthamus tinctorius. Total phenolics were more present in M. denudata than P. mume or C. tinctorius, ranging from 72.6 to 118.0 mg/kg (p < 0.05). Total flavonoids level had same tendency to total phenolics content, showing highest amount (25.1 mg/kg) in M. denudata. The antioxidant activity of the methanol extracts from all the flowers dose-dependently increased. DPPH free radical scavenging activity at 250 mg/kgwas higher in M. denudata and P. mume by 93.0 and 92.6%, respectively, than C. tinctorius by 23.0% (p < 0.05). Total catechins content including 6 compounds was higher in M. denudate (9,425.5 mg/kg) than that in P. mume or C. tinctorius (3,407.8 or 190.6 mg/kg). However, P. mume extracts showed highest amount in caffeine as well as vitamine C. DPPH radical scavenging activity in three different flowers was highly correlated with contents of total phenolics ($r^2=0.7994$), total flavonoids ($r^2=0.9131$), and total catechins ($r^2=0.5929$), and their content and activities were different depending on species.

Antioxidant, Antibacterial and Anti-inflammatory Effects of Stachys sieboldii Extract (초석잠 추출물의 항산화, 항균 및 항염 활성)

  • This study was conducted to evaluate antioxidant, antibacterial, anti-inflammatory, and digestive enzyme activity in water extract (SAW) and 60% ethanol extract (SAE) from Stachys sieboldii. As the treatment concentration of each extract S. sieboldii extract increased, antibacterial and antioxidant activity increased. The total polyphenol and total flavonoid contents of SAW were 106.25 ± 0.94 mgGAE/g, 24.4 ± 0.24 mgQE/g and SAE were 124.61 ± 1.11 mgGAE/g, 45.2 ± 3.52 mgQE/g, respectively. The 400 ㎍/mL of SAW and SAE performed more than 53% protective effects against oxidative stress in HepG2 cell lines. All extracts were not showed cytotoxicity to RAW 264.7 cell line at 100 ㎍/mL. NO production was reduced to 44.3 ± 1.4% for SAW and 45.1 ± 1.0% for SAE at a concentration of 100 ㎍/mL. The production of inflammatory cytokines each TNF-α, IL-1β, and IL-6 was inhibited in a concentration-dependent. S. sieboldii extract did not showed Caco-2 cells cytotoxicity and inhibited NO production in concentration-dependent. As the concentration of the S. sieboldii extract increased of α-amylase and protease enzymes activity, which are digestive enzyme. As a result of the experiment, it is judged that it can be used as basic data for the development of health food using S. sieboldii.

  • 나고야 의정서가 국내에 본격 시행됨에 따라 전남산림자원연구소는 도내 유용 산림 자원의 주권 확립 및 자원 정보의 DB 구축을 위하여 산림생명자원 추출물은행을 운영 중에 있다. 그 일환으로 1차년 난대 수종 4종(동백, 비자, 편백, 황칠)의 부위(잎, 가지) 시기(1차:4월, 2차:7월, 3차:9월, 4차:11월) 용매별(열수, 에탄올) 추출물을 제작하여, 유효성분 및 생리활성의 분석을 실시하였다. 총 폴리페놀(Total Polyphenol) 함량을 150 ug/g 이상을 기준으로 분석 분류한 결과, 동백 잎(4차) 열수, 가지(1, 3차), 잎(4차) 에탄올, 비자 잎(3, 4차) 열수, 가지(2, 3차), 잎(1, 3, 4차) 에탄올, 편백 가지(3차) 열수, 잎(3차), 가지(2, 3차) 에탄올, 황칠 잎(2, 3, 4차) 에탄올 추출물에서 높은 함량의 총 폴리페놀을 확인 할 수 있었다. 다음으로, 프리라디칼(DPPH) 소거능을 측정하여 50 % 이상을 기준으로 분류한 결과, 동백(1~4차), 가지(1~3차) 열수, 잎(1~4차), 가지(1, 3, 4차) 에탄올, 비자 잎(3차) 열수, 잎(2, 4차) 에탄올, 편백 잎(3, 4차), 가지(1차) 열수, 잎(1~3차), 가지(1, 3, 4차) 에탄올, 황칠 잎(2~4차) 열수 추출물에서 높은 프리라디칼(DPPH) 소거능을 보여 주었다. 이와 같이, 수종별로 부위 시기 용매별 추출물에 따른 유효성분과 생리활성에 차이를 확인 할 수 있었다. 이를 바탕으로 산업화 활용 시 채취 시기 및 부위를 특정 하는 자료로 도움 될 것으로 생각되며, 지속적으로 연계 추출 및 분석을 통해 보다 유의성 있는 자료 수집을 계속 진행 중 입니다. 이러한 산림생명자원 추출물은행의 연구 결과를 바탕으로 보다 심도 있게 반복적인 데이터를 수집 구축하고, 연구개발에 활용한다면, 고부가가치 임산 자원을 개발 및 산업화에 큰 도움이 되고 임업임 소득증대에 기여할 것이다.

Increased Antioxidative Activity of Fermented Ligusticum striatum Makino Ethanol Extract by Bioconversion using Lactobacillus plantarum BHN-LAB 129 (Lactobacillus plantarum BHN-LAB 129의 생물전환공정을 통한 천궁 발효 추출물의 항산화 활성 증대)

  • Phytochemical compounds of Ligusticum striatum Makino are used as traditional medicinal herbs in Asia. These compounds are reported to have pain relief and antioxidant activities in gynecological and brain diseases. In this study, we investigated the antioxidant effects of Ligusticum fermented ethanol extract from Lactobacillus plantarum BHN-LAB 129 isolated from Kimchi, a Korean traditional food. The total polyphenol and total flavonoid contents increased by about 116.2% and 281.0% respectively, in the fermented Ligusticum extract as compared with those in the nonfermented Ligusticum ethanol extract. Superoxide dismutase-like (SOD), DPPH radical scavenging, ABTS radical scavenging, and reducing power activities increased by around 139.9%, 199.6%, 301.0%, and 137.1%, respectively, in the fermented Ligusticum ethanol extract as compared with these parameters in the nonfermented Ligusticum ethanol extract, respectively. In conclusion, the fermented Ligusticum ethanol extract with L. plantarum BHN-LAB 129 was effective in increasing the antioxidant effects. The bioconversion process in this study points to the potential of using Ligusticum to produce phytochemical-enriched natural antioxidant agents with high added value. The findings may prove useful in the development of improved foods and cosmetic materials.

Increased Antioxidants of Agastache rugosa by the Night Interruption Time (야파(night interruption)처리에 의한 배초향의 항산화 물질 증가)

  • The objective of this study was to determine the proper night interruption of photoperiods and dark periods for accumulating bioactive compounds of Agastache rugosa without decreasing plant growth. Five-week-old seedlings were transplanted in a DFT system with white LEDs. A. rugosa was treated with night interruption time treatments of 18:1:2:3, 18:2:2:2, 18:3:2:1 (light:dark:light:dark), and 20:4 (control) for 4 weeks. There were no significant differences except for leaf length, leaf width, and the number of flowers. The content of antioxidants in the shoot of A. rugosa was high in tilianin and acacetin, and the content of rosmarinic acid (RA) was significantly higher in the underground part. The RA content per dry weight of A. rugosa was 47.92 and 51.46% higher than that of the control in 18:1:2:3 and 18:2:2:2, and tilianin and acactin per dry weight were significantly higher in 18:3:2:1. There was no significant difference in growth due to the same day light integral, but 18:2:2:2 showed high total polyphenol contents. Therefore, it is thought that the effect of increasing secondary metabolites of A. rugosa without degradation of growth can be expected through night interruption treatment in plant factory cultivation systems using artificial light.

Analysis of Chlorogenic Acid Content and Biological Activities of Aralia elata Ethanol Extract (두릅 에탄올 추출물의 Chlorogenic acid 함량 분석 및 생리활성)

  • This study was conducted to quantify chlorogenic acid content and evaluate biological activity, such as antioxidant, antibacterial, anti-inflammatory, and digestive enzyme activity of Aralia elata ethanol extract (AEE). The SC50 of DPPH and ABTS radical scavenging activities of AEE were 4.79±0.05 mg/mL, 5.79±0.05 mg/mL; total polyphenol and total flavonoid contents were 170.0±1.8 mgGAE/g, 105.5±4.1 mgQE/g, respectively. Nitric oxide (NO) was increased in RAW 264.7 cells and Caco-2 cells with treatment of LPS, and production of NO was inhibited by AEE in a concentration-dependent manner. Production of NO was reduced by 60.0±1.1% in RAW 264.7 cells and 50.7±2.8% in Caco-2 cells at of AEE. Similarly, the production of inflammatory cytokines (TNF-α, IL-1β and IL-6) was inhibited in a concentration dependent manner. Antibacterial activity increased as the dose concentration of AEE increased, and the MIC was 75 mg/mL for L. monocytogenes, and 100 mg/mL for S. typhimurium and H. pylori. In addition, amylase and protease enzyme activity was observed in AEE and increased enzyme activity was observed according to the concentration of the extract. AEE contained 7.06±0.01 mg/g of chlorogenic acid. As a result of the experiment, it is judged that it can be used as basic data for the development of health food using Aralia elata.

Antioxidant and Elastase Inhibitory Effects of Equisetum hyemale Extract (속새(Equisetum hyemale) 추출물의 항산화 및 Elastase 저해 효과)

  • Background: The presence of antioxidants was not confirmed in the medicinal plant Equisetum hyamale grass. Purposes: This study was to determine the antioxidant and elastase inhibition effects of extracts of E. hyemale. Methods: Antioxidant functions of E. hyemale stems and roots were measured and extracted with hot water (HW) and ethyl alcohol (70EOH, 100EOH). Results: The extraction yield of stems was higher in HW extraction than in ethyl alcohol extraction. The polyphenol content was significantly higher in the root extract than in the stem. Total flavonoid content of 70EOH extract was significantly higher in root extract than in stem. The elastase inhibitory function of the extract was 46% in the root and 49% in the stem at 100 ppm of the extract. The ABTS free radical scavenging function was in the order of HW<70EOH<100EOH

Antioxidative and Cytoprotective Effects of Annona muricata (Graviola) Extract for HDF Cell Damage Induced by Hydrogen Peroxide (H2O2에 의해 유도된 HDF 세포 손상에 대한 그라비올라 추출물의 항산화 및 세포 보호 효과)

  • As interest in functionality and environmentally friendly cosmetics is growing in recent years, materials that use safe and effective plant extracts have been developed. Therefore, this study also attempted to check the possibility of the graviola extract, which is known to have various efficacy mainly as a health functional material as a functional cosmetic material. In order to find out the antioxidant activity of graviola, we measured total polyphenol, total flavonoid content and DPPH radical scavenging activity and measured the ROS activity inhibition effect and cytoprotective effect on oxidative stress by treating HDF with hydrogen peroxide cells at an appropriate concentration after checking cytotoxicity in HDF cells. Based on the results of this experiment, the graviola extract was found to contain as high as 26.6 mg(CA)/100g, 14.3 mg(Q)/100g of total polyphenol and flavonoid, which are the antioxidant indexes and to have the high radical scavenging activity. The cell survival rate of the HDF cells was measured, and as a result, no significant cytotoxicity was observed at all concentrations and the experiment was carried out at a concentration of $100{\mu}g/mL$ afterwards. Inhibition of ROS activity in HDF cells induced by hydrogen peroxide was measured and the concentration-dependent inhibition of ROS activity was found and the cell protection effect of graviola was measured after hydrogen peroxide was treated for 4, 24 and 48 hours. As a result, the cell protection effect as high as 89.92% was confirmed at a $25{\mu}g/mL$ concentration up to 24 hours. As these results show that the graviola extract has excellent antioxidant activity, almost no toxicity to HDF cells, an effective activity inhibitory effect on active oxygen generated by hydrogen peroxide and excellent cytoprotective effect, the possibility as various functional materials with antioxidant and cytoprotective effects was confirmed.
