Evaluation of Bacterial Blight Resistance Using SNP and STS Marker-assisted Selection in Aromatic Rice Germplasm

  • A molecular survey was conducted to identify the presence of the bacterial blight resistance genes (Xa1, Xa4, xa5, xa13 and Xa21) in 86 accessions of aromatic rice obtained from germplasm. The results revealed that the resistance gene Xa4 (32.5%), Xa21 (17%), and xa5 (16%) were widely observed in tested rice germplasm. Among tested rice germplasm, 49 accessions showed the presence of more than one of five R genes, and 37 accessions possessed none of the R gene. TALLi and 05-IRRi-M-46 showed the presence of Xa4, xa5, xa13 and Xa21. Rice race $415{\times}Ir352$ exhibited positive amplicon for the Xa1, Xa4, xa5 and Xa21. Hyangmibyeo1hos, Ir841-85-1-1-2 and Jasmine85 showed the positive amplicon for the Xa1, Xa4 and xa5 genes. Yekywin Yinkya Hmwe and Khao Dawk Mali105 showed the presence of Xa1, Xa4 and Xa21 gene. Masino Basmati showed the presence of xa5, xa13, Xa21 genes. Xa1 and Xa21 genes were noticed in Mihayngbyeo, Tarana Deshi, Mayataung and AZUCENA. Hyangmibyeo2ho, Basmati 6311 and Basmati405 possessed only two R genes such as Xa4 and xa5, and xa5 and xa13, respectively. The evaluation results of bacterial blight resistance genes in aromatic rice germplasm will help in breeding of multi disease resistant varieties.

  • 미곡종합처리장에서 수집한 2011년산 쌀 시료 중 Fusarium 곰팡이독소의 자연발생을 조사하기 위해서, 벼, 왕겨, 현미, 청치미, 싸라기, 미강, 색채미, 백미 등의 시료를 2012년에 전국적으로 수집하였다. 371점의 시료에 대해 DON, NIV과 ZEA의 자연발생을 LC/MS로 분석하였다. 이중 색채미가 오염율과 오염도가 가장 높았고 다음이 싸라기였다. 백미는 3점이 DON, NIV에 안전한 수준으로 오염되었었지만 나머지는 곰팡이독소가 검출되지 않았다. 부산물은 백미보다 오염도나 오염율이 높았다.

The Complete Nucleotide Sequence of a Korean Isolate Bean yellow mosaic virus from Freesia sp. and Comparison to Other Potyviruses

  • Bean yellow mosaic virus (BYMV; genus Potyvirus, family Potyviridae) causes severe losses to various legume species and a number of non-legume species, particularly freesia plants. In a survey of virus diseases in Gyeonggi province, Korea, BYMV isolates were identified from many cultivated freesia species. Here, we determined the complete nucleotide sequences of a BYMV freesia isolate (BYMV-Fr; accession number FJ492961). BYMV-Fr genome consists of 9,545 nucleotides (nt) excluding the poly (A) tail and encodes 3,057 amino acid (aa), with an AUG start and UAG stop codon, containing one open reading frame typical of a potyvirus polyprotein. The polyprotein of BYMV-Fr was divided to ten proteins and the cleavage sites of each protein were determined. The coat protein (CP) and polyprotein of BYMV-Fr were compared at the aa level with those of the previously reported 4 BYMV isolates. BYMV-Fr shared 90.1 to 97.1 and 91.0 to 92.5% at the CP and polyprotein homology. Interestingly, BYMV-Fr showed identities of a lower level at the nt level of 5' noncoding region (61.4 to 67.6%) and at the aa level of P1 (71.4 to 72.8%), comparing with four BYMV isolates. Based on the aa sequence diversity of CP and polyprotein, phylogenetic analysis with the four BYMV isolates showed two distinct groups and BYMV-Fr and most BYMV isolates were most closely related to the clover yellow vein virus among 52 potyviruses. To our knowledge, this is the first report of the complete genome sequence of BYMV freesia strain.

조사 계절에 따른 식품섭취빈도 조사의 평균 섭취 횟수와 섭취량 변화 (Seasonal Variation of Food Intake in Food Frequency Questionnaire among Workers in a Nuclear Power Plant)

  • Objectives : This study was conducted to investigate the systematic error, such as seasonal change or inadequate food items, in a food frequency questionnaire administered to workers in a Nuclear Power Plant, Korea. Methods : We performed three repeat-tests with 28 subjects on May 13, July 8 and Dec 16, 1992. Our food frequency questionnaire (FFQ) comprised 84 foods organized into 7 food-groups, and was composed of the items of usual intake frequency (8 categories) and the amount per intake (3 or 4 categories) over the previous year. We compared the means of intake frequency and the frequency of the portion-size according to each season using Repeated Measures ANOVA and Pearson's chisquare test with Fisher's exact test. Results : We found the significant seasonal changes of several food items in intake frequency measurement. These items were typical seasonal foods such as mandarin orange, plum and green vegetables, while the single questions consisted of inadequate food items such as thick beef or similar soup and various kimchi products. Significant seasonal changes in portion-size were found in only two items: cooked rice-brown and fresh frozen fishes. Conclusions : The systematic errors observed could caused loss of validity in the FFQ. Consideration should be given for seasonal variation in FFQ survey and methodological concerns are needed to improve the quality for measuring usual diet pattern.

Biological and Molecular Characterization of a Korean Isolate of Cucurbit aphidborne yellows virus Infecting Cucumis Species in Korea

  • Surveys of yellowing viruses in plastic tunnels and in open field crops of melon (Cucumis melo cultivar catalupo), oriental melon (C. melo cultivar oriental melon), and cucumber (C. sativus) were carried out in two melon-growing areas in 2014, Korea. Severe yellowing symptoms on older leaves of melon and chlorotic spots on younger leaves of melon were observed in the plastic tunnels. The symptoms were widespread and included initial chlorotic lesions followed by yellowing of whole leaves and thickening of older leaves. RT-PCR analysis using total RNA extracted from diseased leaves did not show any synthesized products for four cucurbit-infecting viruses; Beet pseudo-yellows virus, Cucumber mosaic virus, Cucurbit yellows stunting disorder virus, and Melon necrotic spot virus. Virus identification using RT-PCR showed Cucurbit aphid-borne yellows Virus (CABYV) was largely distributed in melon, oriental melon and cucumber. This result was verified by aphid (Aphis gossypii) transmission of CABYV. The complete coat protein (CP) gene amplified from melon was cloned and sequenced. The CP gene nucleotide and the deduced amino acid sequence comparisons as well as phylogenetic tree analysis of CABYV CPs showed that the CABYV isolates were undivided into subgroups. Although the low incidence of CABYV in infections to cucurbit crops in this survey, CABYV may become an important treat for cucurbit crops in many different regions in Korea, suggesting that CABYV should be taken into account in disease control of cucurbit crops in Korea.

Podosphaera tridactyla에 의한 자두나무 흰가루병 발생 (Occurrence of Powdery Mildew of Japanese Plum Caused by Podosphaera tridactyla in Korea)

  • 2011년에 양구, 홍천, 횡성, 가평 등 중부지방에서 가족 소비용 또는 관상용으로 식재된 자두나무에 흰가루병이 발생하였다. 상당한 조사에도 불구하고 우리나라 자두나무 과수원에서는 흰가루병이 발견되지 않았다. 초기 병징은 잎과 어린 줄기에 균사층이 흰색으로 얇고 불규칙하게 나타났다. 나중에는 잎이 부분적으로 기형으로 뒤틀렸고 적자색으로 변색되었다. 무성세대 및 유성세대의 형태적 검경과 함께 ITS 영역의 염기서열 분석을 통하여 이 병원균을 Podosphaera tridactyla로 동정하였다. 이는 우리나라에서 자두나무 흰가루병을 처음으로 보고하는 것이다.

Genetic Diversity and Population Structure of the Xanthomonas campestris pv. campestris Strains Affecting Cabbages in China Revealed by MLST and Rep-PCR Based Genotyping

  • Xanthomonas campestris pv. campestris (Xcc) is the causal agent of black rot for cruciferous vegetables worldwide, especially for the cole crops such as cabbage and cauliflower. Due to the lack of resistant cabbage cultivars, black rot has brought about considerable yield losses in recent years in China. Understanding of the pathogen features is a key step for disease prevention, however, the pathogen diversity, population structure, and virulence are largely unknown. In this study, we studied 50 Xcc strains including 39 Xcc isolates collected from cabbage in 20 regions across China, using multilocus sequence genotyping (MLST), repetitive DNA sequence-based PCR (rep-PCR), and pathogenicity tests. For MLST analysis, a total of 12 allelic profiles (AP) were generated, among which the largest AP was AP1 containing 32 strains. Further cluster analysis of rep-PCR divided all strains into 14 DNA groups, with the largest group DNA I comprising of 34 strains, most of which also belonged to AP1. Inoculation tests showed that the representative Xcc strains collected from diverse regions performed differential virulence against three brassica hosts compared with races 1 and 4. Interestingly, these results indicated that AP1/DNA I was not only the main pathotype in China, but also a novel group that differed from the previously reported type races in both genotype and virulence. To our knowledge, this is the first extensive genetic diversity survey for Xcc strains in China, which provides evidence for cabbage resistance breeding and opens the gate for further cabbage-Xcc interaction studies.

건설업 직종별 노출 가능 유해인자 및 노출강도에 관한 평가 (Evaluation on Probability and Intensity of Hazards Exposure by Construction Occupations)

  • Objectives: Construction workers are exposed to various hazardous substances simultaneously. However, little is known about the exposure hazards in construction industry. This study was aimed at identifying the risk of exposure hazards among construction workers. Methods: The expert survey (n=29) was conducted, including construction industry health managers (n=11) and work environment monitoring experts (n=18), on exposure probability, intensity and risk of hazardous substances by construction occupations Results: The exposure hazards of 30 construction occupations were identified and summarized through a literature review and expert survey. The most prevalent hazards were in order of noise, awkward posture, heat/cold, crystalline silica, cement/concrete dust, metal fumes, and volatile organic compounds. The hazards with highest risk score(over seven points) at construction occupations were noise(formwork carpenter, concrete finisher, rebar worker, demolition worker, driller/rock blaster), hazardous rays(welder), heat/cold (earthworks, formwork carpenter, rebar worker, concrete placer, scaffolder), awkward posture(bricklayer, caulker/tile setter, rebar worker) and heavy lifting(bricklayer, rebar worker). Among construction workers, the job types with the highest risk of exposure to carcinogens, and in which occupational cancer has been reported, were in order of stonemason, concrete finisher, rock blaster, welder, insulation installer, painter, scaffolder, plant worker and earthworks in order Conclusions: Systematic research and discussion on occupational disease among construction workers and its various hazardous factors are needed to establish job exposure matrix for facilitating standard for promptly processing the workers' compensation.

유기재배 복숭아 과원의 관리현황 및 병해충 발생 실태 (Survey on Occurrence and Management of Disease and Pests in Organic Peach Orchards)

  • 본 연구에서는 2015년 3월부터 2017년 3월까지 친환경 인증 복숭아 재배현황, 유기농산물인증 복숭아 과원의 관리현황 및 병해충 발생실태를 조사하였다. 조사 기간 동안 복숭아 유기인증 농가는 65.5%, 무농약인증 농가는 31.7% 증가하여 매우 높은 증가 추세를 나타냈다. 복숭아 유기농산물인증 농가 중 대표적인 6 농가를 선발하여 재배현황을 살펴보았다. 조사한 모든 농가에서 녹비작물을 재배하거나 초생재배를 실시하고 예초를 최소한(평균 2.5회)으로 실시하였다. 토양 양분관리를 위해 가축분퇴비, 유박 및 자가제조 액비를 사용하는 농가가 많았으며, 병해충 방제를 위해 보르도액, 석회유황합제, 교미교란제 및 자가제조 식물 추출물을 사용하고 있는 것을 확인할 수 있었다. 병해충 피해현황을 살펴보았을 때 충해로 인한 피해는 31.6%, 병해로 인한 피해는 24.1%로 충해로 인한 피해가 다소 높은 것을 알 수 있었다. 주요 발생 병해충으로는 복숭아순나방이 13.5%로 가장 많은 피해를 유발하는 것을 확인할 수 있었으며, 잿빛무늬병(13.0%), 복숭아심식나방(7.3%) 및 세균성구멍병(7.3%)이 문제 되고 있는 것을 확인할 수 있었다.

구제역 관리를 위한 혈청학적 예찰계획 평가 (Evaluation of Serological Surveillance System for Improving Foot-and-Mouth Disease Control)

  • The primary goal of this study was to compute sample sizes required to achieve the each aim of a variety of foot-and-mouth disease (FMD) surveillance programs, using a statistically valid technique that takes the following factors into account: sensitivity (Se) and specificity (Sp) of diagnostic test system, desired minimum detectable prevalence, precision, population size, and desired power of the survey. In addition, sample sizes to detect FMD if the disease is present and also as proof of freedom were computed. The current FMD active surveillance programs consist of clinical, virological, and serological surveillance. For the 2012 serological surveillance, annual sample sizes (n = 265,065) are planned at four separate levels: statistical (n = 60,884) and targeted (n = 115,232) at breeding pig farms and slaughter house, in together with the detection of structural proteins (SP) antibodies against FMD (n = 88,949). Overall, the sample size was not designed taking the specific aims of each surveillance stream into account. The sample sizes for statistical surveillance, assuming stratified two-stage sampling technique, was based to detect at least one FMD-infected case in the general population. The resulting sample size can be used to obtain evidence of freedom from FMD infection, not for detecting animals that have antibodies against FMD virus non-structural proteins (NSP). Additionally, sample sizes for targeted surveillance were not aimed for the population at risk, and also without consideration of statistical point of view. To at least the author's knowledge, sampling plan for targeted, breeding pig farms and slaughter house is not necessary and need to be included in the part of statistical surveillance. Assuming design prevalence of 10% in an infinite population, a total of 29 animals are required to detect at least one positive with probability of 95%, using perfect diagnostic test system (Se = Sp = 100%). A total of 57,211 animals needed to be sampled to give 95% confidence of estimating SP prevalence of 80% at the individual animal-level with a precision of ${\pm}5%$, assuming 800 herds with an average 200 heads per farm, within-farm variance of 0.2, between-farm variance of 0.05, cost ratio of 100:1 of farm against animals. Furthermore, 779,736 animals were required to demonstrate FMD freedom, and the sample size can further be reduced depending on the parameters assumed.
