Plant Defence Responses: Current Status and Future Exploitation

  • Plants have developed a sophisticated battery of defence responses to protect themselves against attempted pathogen ingress. Manipulation of these defence mechanisms may provide significant opportunities for crop improvement. While plant resistance genes have had a long service history in plant breeding, they possess significant limitations. Recent advances are now providing significant insights into strategies designed to increase the field durability of this class of genes. Hypersensitive cell death is a common feature underlying the deployment of plant defence responses against biographic pathogens. In contrast, necrotrophic pathogens actively kill plant cells. Recently, transgenic plants have been developed that either promote or suppress cell death, providing resistance against either biotrophic or necrotrophic pathogens respectively. Methyl-jasmonate is a key signalling molecule in the establishment of resistance against some fungal pathogens. Increasing the concentration of this molecule in plant cells has been shown to increase resistance against Botrytis cineria, without significantly imparting plant growth or development. Due to the multifarious infection strategies employed by plant pathogens, how-ever, it is unlikely a single commercial product will prove a panacea for global disease control. Future stategies will more likely entail an integrated disease management approach.

Fusarium oxysporum Protects Douglas-fir (Pseudotsuga menziesii) Seedlings from Root Disease Caused by Fusarium commune

  • Fusarium root disease can be a serious problem in forest and conservation nurseries in the western United States. Fusarium inoculum is commonly found in most container and bareroot nurseries on healthy and diseased seedlings, in nursery soils, and on conifer seeds. Fusarium spp. within the F. oxysporum species complex have been recognized as pathogens for more than a century, but attempts to distinguish virulence by correlating morphological characteristics with results of pathogenicity tests were unsuccessful. Recent molecular characterization and pathogenicity tests, however, revealed that selected isolates of F. oxysporum are benign on Douglas-fir (Pseudotsuga menziesii) seedlings. Other morphologically indistinguishable isolates, which can be virulent, were identified as F. commune, a recently described species. In a replicated greenhouse study, inoculating Douglas-fir seedlings with one isolate of F. oxysporum prevented expression of disease caused by a virulent isolate of F. commune. Moreover, seedling survival and growth was unaffected by the presence of the F. oxysporum isolate, and this isolate yielded better biological control than a commercial formulation of Bacillus subtilis. These results demonstrate that an isolate of nonpathogenic F. oxysporum can effectively reduce Fusarium root disease of Douglas-fir caused by F. commune under nursery settings, and this biological control approach has potential for further development.

  • Since 1966 tissue culture has been used as a tool for the production of disease indexed stocks from selected plants and their rapid (clonal) mass propagation through the procedure now referred to as micropropagation. The major advantages have been the rapid introduction of new plant cultivars, created within conventional and mutation breeding programmes, as healthy stock for beneficial distribution and the expansion of the world wide horticultural industry. 비파나무 및 홍가시나무 점무늬병의 살균제 방제효과 (Efficacy of Fungicides for the Control of Leaf Spots on Eriobotrya japonica and Photinia glabra)

  • 2000년대 후반부터 고흥지역의 비파나무(Eriobotrya japonica)와 제주지역의 홍가시나무(Photinia glabra)에 Entomosporium mespili에 의한 극심한 점무늬병이 발생하기 시작하였다. 따라서 방제효과가 높은 살균제를 선발하기 위하여 이 연구를 수행하였다. 효과를 검정한 3개의 농약 중 비파나무 점무늬병에는 propiconazole 처리가 방제가 64.1%로 가장 높게 나타났고, 홍가시나무 점무늬병에는 chlorothalonil 처리가 방제가 87.6%로 가장 높게 나타났다. Thiopanate-methyl 처리는 방제가가 비교적 낮게 나타났고, propiconazole은 홍가시나무에 약해를 나타내었다.

Application of Bacterial Endophytes to Control Bacterial Leaf Blight Disease and Promote Rice Growth

  • Xanthomonas oryzae pv. oryzae (Xoo) causes bacterial leaf blight (BLB) disease in rice (Oryza sativa L.) and it is among the most destructive pathogen responsible for severe yield losses. Potential bacterial biocontrol agents (BCAs) with plant growth promotion (PGP) abilities can be applied to better manage the BLB disease and increase crop yield, compared to current conventional practices. Thus, this study aimed to isolate, screen, and identify potential BCAs with PGP abilities. Isolation of the BCAs was performed from internal plant tissues and rhizosphere soil of healthy and Xoo-infected rice. A total of 18 bacterial strains were successfully screened for in vitro antagonistic ability against Xoo, siderophore production and PGP potentials. Among the bacterial strains, 3 endophytes, Bacillus sp. strain USML8, Bacillus sp. strain USML9, and Bacillus sp. strain USMR1 which were isolated from diseased plants harbored the BCA traits and significantly reduced leaf blight severity of rice. Simultaneously, the endophytic BCAs also possessed plant growth promoting traits and were able to enhance rice growth. Application of the selected endophytes (BCAs-PGP) at the early growth stage of rice exhibited potential in suppressing BLB disease and promoting rice growth.

Characterization of Bacillus luciferensis Strain KJ2C12 from Pepper Root, a Biocontrol Agent of Phytophthora Blight of Pepper

  • In this study, we characterized the bacterial strain KJ2C12 in relation with its biocontrol activity against Phytophthora capsici on pepper, and identified this strain using morphological, physiological, biochemical, fatty acid methyl ester, and 16S rRNA gene sequence analyses. Strain KJ2C12 significantly (P=0.05) reduced both final disease severity and areas under the disease progress curves of 5-week-old pepper plants inoculated with P. capsici compared to buffer-treated controls. As for the production of antibiotics, biofilms, biosurfactant, extracellular enzyme, HCN, and swarming activity, strain KJ2C12 produced an extracellular enzyme with protease activity, but no other productions or swarming activity. However, Escherichia coli produced weak biofilm only. Strain KJ2C12 could colonize pepper roots more effectively in a gnotobiotic system using sterile quartz sand compared to E. coli over 4 weeks after treatments. However, no bacterial populations were detected in 10 mM $MgSO_4$ buffer-treated controls. Strain KJ2C12 produced significantly higher microbial activity than the $MgSO_4$-treated control or E. coli over 4 weeks after treatments. Bacterial strain KJ2C12 was identified as Bacillus luciferensis based on morphological, physiological, and biochemical characteristics as well as FAME and 16S rRNA gene sequence analyses. In addition, these results suggested that B. luciferensis strain KJ2C12 could reduce Phytophthora blight of pepper by protecting infection courts through enhanced effective root colonization with protease production and an increase of soil microbial activity.

미생물을 활용한 친환경적인 벼 병해 방제법 (Biocontrol of Rice Diseases by Microorganisms)

  • 벼는 전 세계 30억 인구의 식량 공급원으로 아시아 인구의 약 절반이 쌀을 식량자원으로 의존하며 100개 이상의 국가에서 재배되고 있다. 따라서 벼의 병해충에 따른 경제적 손실과 수확량 감소는 식량 수요와 공급을 위협할 수 있다. 벼에 발생하는 병해충을 방제하기 위해 가장 일반적으로 사용되는 방법은 농약을 통한 화학적 방제이다. 그러나 이러한 농약을 통한 환경 오염, 잔류 독성, 내성 병원균 출현, 토양 품질 저하 및 생물 다양성 파괴 등의 문제를 야기할 수 있다. 벼의 병해를 방제하기 위한 최근의 대안으로 미생물을 포함한 환경친화적인 생물학적 방제에 대한 연구가 활발히 진행되고 있다. 미생물 작용제는 식물 병원균과의 경쟁, 항생제 효과 및 기생을 통해 식물 병을 방제 할 수 있다. 벼 근권에서 분리한 미생물들, 예를들어 Bacillus spp., Pseudomonas spp., Trichoderma sp. 등은 벼에 발생하는 다양한 곰팡이 및 세균 병들에 대한 생물 방제제로 사용가능성이 보고되었는데, 특히 벼도열병, 벼 잎집무늬마름병, 벼 흰잎마름병, 벼 깨씨무늬병 및 벼 키다리병을 방제하는 것으로 보고되었다. 이 리뷰에서는 벼에 발생하는 다양한 병에 대한 생물 방제제로서 미생물들이 적용된 연구들에 대하여 논의하였다.

Colonizing Ability of Pseudomonas fluorescens 2112, Among Collections of 2,4-Diacetylphloroglucinol-Producing Pseudomonas fluorescens spp. in Pea Rhizosphere

  • Pseudomonas fluorescens 2112, isolated in Korea as an indigenous antagonistic bacteria, can produce 2,4-diacetylphloroglucinol (2,4-DAPG) and the siderophore pyoveridin2112 for the control of phytophthora blight of red-pepper. P. fluorescens 2112 was classified into a new genotype C among the 17 genotypes of 2,4-DAPG producers, by phlD restriction fragment length polymorphism (RFLP). The colonizing ability of P. fluorescens 2112 in pea rhizosphere was equal to the well-known pea colonizers, P. fluorescens Q8r1 (genotype D) and MVP1-4 (genotype P), after 6 cycling cultivations for 18 weeks. Four tested 2,4-DAPG-producing Pseudomonas spp. could colonize with about a 96% dominance ratio against total bacteria in pea rhizosphere. The strain P. fluorescens 2112 was as good a colonizer as other Pseudomonas spp. genotypes in pea plant growth-promoting rhizobacteria.

호장근(Reynoutria elliptica)으로부터 항균활성 물질의 분리 및 구조결정 (Isolation and identification of antifungal compounds from Reynoutria elliptica)

  • In the continued research on natural fungicides for the control of plant diseases by using plant-derived products, we found that Reynoutria elliptica. had a strong fungicidal activity against several plant pathogens. R. elliptica (3.2 kg) were extracted with 80% aq. MeOH and the concentrated extracted was partitioned with n-hexane, EtOAc, n-BuOH and $H_2O$ successively. The four layers were tested their disease control efficacies against 4 plant disease such as rise blast (RCB), tomato grey mold (TGM), tomato late blight (TLB), and barly powdery mildew (BPM). The n-hexane fraction was highly active showing over 95% control against TLB and BPM. and the EtOAc fraction was highly active showing over 95% control against RCB, TLB, and BPM. By using silica gel chromatography, MPLC, and HPLC, three compounds that were expected to have antifungal activity were isolated. Their chemical structures were elucidated as physcion, emodin, and emodie-8-O-glucoside by EI-MS and NMR spectroscopic analyses.
