Two experiments were conducted to evaluate developmental gene expression of antimicrobial peptide PR-39 and effect of zinc oxide on gene regulation of PR-39 in piglets using semi-quantitative RT-PCR analysis. In experiment 1, fifteen female Tai-Hu pigs (a local breed in China) in five groups, each of three pigs at 1, 14, 28, 42 and 56 days of age were used to determine effect of age and weaning on mRNA expression of PR-39. In experiment 2, nine groups of pigs (total seventy-two female 36 days-age weanling Tai-Hu piglets) were assigned to three treatments (${ZnO}_0$, ${ZnO}_{100}$ and ${ZnO}_{3000}$). The feeding experimental period lasted 15 days. After feeding experiment, nine pigs with three animals in each treatment were chosen to determine the effect of ZnO on PR-39 mRNA expression of pigs. The results showed that PR-39 mRNA levels increased steadily in postnatal day 1-28 (preweaning), and weaning significantly decreased PR-39 mRNA expression of piglets (p<0.05). ${ZnO}_{3000}$ (3,000 mg zinc/kg diet) significantly increased PR-39 mRNA expression (p<0.05) when piglets were feed ${ZnO}_{3000}$ diet for 15 days. ${ZnO}_{100}$ (100 mg zinc/kg diet) also increased PR-39 gene expression, but the result was not statistically significant (p>0.05). The result was in accordance with the effect of ${ZnO}_{3000}$ and ${ZnO}_{100}$ on weight gain of piglets and prevention of diarrhea.
The goal of this study was to investigate the behavior of sows and their piglets reared using the non-all-in all-out system and the all-in all-out system during a 3-week lactation period. The 24 Landrace${\times}$Yorkshire sows (range of parity: 2 to 4) and their litters (range of litter size: 8 to 13 piglets) for each treatment (8 sows/replication) were used in this experiment. The sows were housed in farrowing crates ($0.6{\times}2.1m$) located in pens ($2.4{\times}1.8m$) with totally perforated metal flooring. The crates were kept in an experimental room that was thermostatically controlled to approximately $23^{\circ}C$. A 250-W infrared heat lamp was turned on above the creep area during lactation. The sows and piglets were conventionally managed. The animals were recorded during a 24-h period at 1, 4, 7, 14 and 21 days postpartum. The videotapes were scanned every minute to obtain an instantaneous behavioral sample. In the behaviors of sows, lateral recumbency decreased and ventral recumbency increased from 1 day postpartum to 21 days postpartum. In comparison to the AIAOS, sows in the NAIAOS spent much less time on lateral recumbency (p<0.01), whereas they spent much more time on ventral recumbency and sitting at 4 days postpartum (p<0.05 and p<0.05). In the behaviors of piglets, lying increased and unsuccessful suckling decreased until 21 days postpartum. Compared to the AIAOS at 4 days postpartum, piglets in the NAIAOS spent less time lying (p<0.01), whereas they spent much more time walking and unsuccessful suckling (p<0.01 and p<0.05). These results suggest that the all-in all-out system may be preferable to the non-all-in all-out system for promoting welfare.
본 연구는 분만시 동복 신생자돈의 제대혈에서 추출한 genomic DNA를 PCR-RFLP 기법을 이용하여 농가수입증대를 위하여 육질이 불량한 PSS 돼지를 판별하는 방법을 개발하기 위한 기초실험으로써 그 결과를 요약하면 다음과 같다. 자돈 제대에서 혈액 genomic DNA를 추출하여 PCR에 의하여 증폭된 ryanodine receptor gene 영역의 산물은 자돈의 제대혈에서 1.8kb의 길이로 증폭되었음을 확인하였다. 제대혈에서 추출된 DNA 의 PCR 증폭 단편을 가지고 Hha I 제한효소로 digest 하여준 결과에서 PSS 돼지는 Yorkshire 종에서 출현하지 않았으나, Landrace 종과 Crossbred 종에서 각각 4.76%와 7.14%로 교잡종(LYD 또는 YLD)에서 더욱 많이 출현되었다. 이상의 결과로 볼 때 분만시 신생자돈의 제대혈을 채취하여 PSS 돼지를 조기에 선발하면 스트레스 감소는 물론 혈액채취의 간편성을 기대할 수 있을 것으로 판단된다.
Piglets' diarrhea is one of the major causes of economic loss in Korean swine industry. To prevent this serious and persistant problem, we have developed systemic farm evaluation and analysis program. The program consists of the checklist and the computerized program. The items of the checklist are selected on the basis of various causative factors related with the piglets' diarrhea including sanitational status, nutritional management, or farm environment. The checklist is composed of 30 items including 6 factors on facility, 8 factors on environment, 6 factors on nutrition and 10 factors on sanitation, respectively. Minimum 1 to maximum 10 points was assigned to each item depending on level or status of farm. The scores calculated by sum of points obtained from each item could find out the problems which the farm was confronted with. To investigate the applicability of a program 150 swine farms raising over 500 heads were selected from 7 provinces throughout the country and surveyed. There were significant correlations between the piglets' diarrhea and the scores of the checklist. Based on above results we revised the checklist which consisted of 22 items including 5 factors on facility, 7 factors on environment, 3 factors on nutrition and 7 factors on sanitation. After that we composed a computerized diagram out of each point of an item which showed and indicated weak factors to be improved promptly. Our present study suggested that this newly established swine farm evaluation program would be a very efficient way to find out the problems and support the systemic control program of the piglets' diarrhea for swine farms.
The purpose of this study was to investigate the protective effects against porcine epidemic diarrhea virus (PEDV) and transmissible gastroenteritis virus (TGEV) in suckling piglets by oral administration of the IgY. The piglets were divided into two groups: test and control group. The former (n=10) were administered orally with IgY for three days from one-day-old and experimentally challenged with PEDV and TGEV at four-day-old. The latter (n=10) were administered with saline solution and challenged with same methods. Several tests were studied and summarized as follows; In clinical signs, the piglets of the control group showed the typical signs such as severe watery diarrhea, depression and anorexia but those of the test group recovered progressively. Control group showed 20% in mortality, but there were no death in the other. The gross lesions in the test were milder than those in the control, and there were typical findings as like congestion and distension of lumen in the control group. In histopathological study, the piglets of the control group had shortened and fused intestinal villi and a marked loss of epithelium, whereas the others showed milder changes. It could be concluded that oral administration of IgY, specific yolk-antibody against PEDV and TGEV is effective to prevent PEDV and TGEV infection in suckling piglets.
Objective: Weaning is an important stage in the life of young mammals, which is associated with intestinal inflammation, gut microbiota disorders, and even death. β-Carotene displays anti-inflammatory and antioxidant activities, which can prevent the development of inflammatory diseases. However, whether β-carotene can affect intestinal microbiota remains unclear. Methods: Twenty-four piglets were distributed into four groups: the normal suckling group (Con), the weaning group (WG), the weaning+β-carotene (40 mg/kg) group (LCBC), and the weaning+β-carotene (80 mg/kg) group (HCBC). The serum, jejunum, colon, and faeces were collected separately from each group. The effects of β-carotene on the phenotype, overall structure, and composition of gut microbiota were assessed in weaning piglets. Results: The results showed that β-carotene improved the growth performance, intestinal morphology and relieved inflammation. Furthermore, β-carotene significantly decreased the species from phyla Bacteroidetes and the genus Prevotella, and Blautia, and increased the species from the phyla Firmicutes and the genera p-75-a5, and Parabacteroides compared to the WG group. Spearman's correlation analysis showed that Prevotella and Blautia were positively correlated, and Parabacteroides and Synergistes were negatively correlated with the levels of interleukin-1β (IL-1β), IL-6, and tumour necrosis factor-α (TNF-α), while p-75-a5 showed negative correlation with IL-6 in serum samples from piglets. Conclusion: These findings indicate that β-carotene could alleviate weaning-induced intestinal inflammation by modulating gut microbiota in piglets. Prevotella may be a potential target of β-carotene in alleviating the weaning-induced intestinal inflammation in piglets.
Edema disease (ED) in pigs is enterotoxemia caused by Shiga toxin type 2e (Stx2e)-producing Escherichia coli (STEC) and frequently occurs in young piglets. Therefore, ED causes enormous economic losses in pig farms. In this study, a modified Stx2e (mStx2e) antigen was expressed and purified using commercial E. coli expression system. Monoclonal antibody was serviced by Ynto Ab Inc., using Phage Display Technique. Anti-Stx2e antibodies in piglets were measured by indirect ELISA using mStx2e antigens. Naive Stx2e in piglets were detected by Sandwich ELISA using Stx2e-monoclonal antibodies and commercial Stx2e-polyclonal antibodies. Among 3,480 piglets, anti-Stx2e antibodies were observed in 2,573 piglets. The 49.4% among 830 piglet serum samples possessed 0.625 ㎍/mL or more of Stx2e proteins. The 18.3% of 830 sera had 0.313 ㎍/mL of Stx2e proteins. The 32.3% of 830 samples held 0.156 ㎍/mL or less of Stx2e proteins. These results show that indirect ELISA using mStx2e antigen and Sandwich ELISA using Stx2e-monoclonal and polyclonal antibodies can be useful to detect ED in piglets.

A goal for swine farming is the improvement in the number of live-born and weaned piglets per sow. Hence, the effect of parities should consider the correlation between the component traits of reproductive performance, weaning, and duration. Sows were housed in farrowing pens (W 2.2 ${\times}$ D 1.8 ${\times}$ H 1.2 m) on a partially slatted plastic floor. Twenty sows used in this study were between the first and sixth parity in gilts (P1), parity 2-5 (P3), and parity 6-9 (P6). Data collection by parity was classified into three categories: (1) reproduction performance (gestation length, total number of piglets born, number of piglets live born, number of piglets stillborn, total piglet birth weight; (2) weaning traits (weaning period, number of piglets weaning, total piglets weaning weight); (3) duration traits (farrowing duration, placenta expulsion duration, time from last piglet to first placenta, average birth interval, and tail wagging behavior). Gestation length was higher in P6 than P1 and P3 of different parity sows. The maximum value in P1 and P3 was 117 days, but the median value in P6 was 117 days (p < 0.05). The total number of piglets born (p = 0.113), number of piglets live born (p = 0.118), number of still piglets born (p = 0.151), and total piglet birth weight (p = 0.117) were not affected by parity. The number of live piglets was higher than the other parities by an average of $15.6{\pm}2.1$ in P1. The duration of farrowing was the lowest at 22.2 min in P6, but the maximum value was 42.2 min more than other parities (p = 0.355). Weaning traits of sows also did not differ significantly (p > 0.05), but the weaning period from P1 was lower than that of the other parities (p = 0.170). The number of piglets weaned was 10 heads on average in P1, P3, and P6 (p < 0.05). However, the mean values of the total piglet weaning weight (p = 0.377) of P6 (62.0 10.4 kg) were higher than those of P1 (54.9 10.2 kg) and P3 (58.4 13.6 kg). The placenta expulsion duration was higher in P6 than that in P1 and P3 (p = 0.447). The time from the last piglet to first placenta was be lower in P3 than that of the other parities (p = 0.206). The average birth interval was higher in P3 than that of the other parities (p = 0.156). Tail wagging (count) behavior was higher in P6 than in the other parities (p = 0.065). The data showed that the reproduction performance, total piglets born, and weight were higher in the gilts group, and higher in the weaning trait than that in the P6 group. This study examined the relationship between reproductive performance, birth interval and tail motion according to sow parity. Regarding duration, farrowing duration was lower in P6 than that in the other parities, but placenta expulsion and tail wagging were higher in P6 than in the other parities. Therefore, it is possible that the results from these sows could be used as basic data for effective farm management.
This study was conducted to evaluate effects of dietary multi-carbohydrases (MCS) in a lactating sow diet on productive performance and immune responses of sows and their piglets. A total of 12 sows (218.37 ± 5.5 kg BW; 2 parity) were randomly assigned to 2 dietary treatments: a diet based on corn-soybean meal (CON) and CON with 0.01% MCS. The MCS contained xylanase (2,700 units/g), β-glucanase (700 units/g), and cellulase (800 units/g). Sows were fed the dietary treatments for 28 days (weaning) after farrowing. Blood samples were collected from sows on d 0, 3, and 7 after farrowing and randomly selected 2 nursing piglets in each sow on d 3, 7, and 14 after birth. Measurements were productive performance of sows, frequency of diarrhea of piglets, and immune responses of sows and their piglets. Sows fed MCS had lower (p < 0.05) their body weight change than those fed CON. Piglets from sows fed MCS had higher (p < 0.05) average weight gain and body weight at weaning day and lower (p < 0.10) frequency of diarrhea than those from sows fed CON. Sows fed MCS had lower number of white blood cells (WBC) on d 3 (p < 0.05) and TGF-β1 on d 7 (p < 0.10) during lactation than those fed CON. Similarly, piglets from sows fed MCS had also lower (p < 0.05) number of WBC on d 3 and d 7 and TGF-β1 and C-reactive protein on d 7 during lactation than those from sows fed CON. In addition, piglets from sows fed MCS had higher (p < 0.10) immunoglobulin G and M on d 7 during lactation those from sows fed CON. In conclusion, addition of dietary MCS in the lactating sow diet based on corn and soybean meal improved productive performance of sows and their litters and modulated their immune responses.
Objective: The objective of the present study was to investigate the effects of farrowing month (FM), parity and sex on the growth performance of Berkshire swine raised in alternative production systems. Methods: A total of 40 farrowing records from 27 sows and 1,258 body weight (BW) records from 274 piglets collected over a two-year period were used for the analysis. The BWs were recorded at birth, weaning (28 d), 56, 84, 112, and 140 days. Any BW not recorded on schedule was recalculated to conform the days of age among corresponding BW records, using growth curves drawn with polynomial functions whose power was determined by the number of existing observations for each individual. Results: The mean parity (${\pm}$standard deviation) of the sows was $3.42{\pm}2.14$. The sows that farrowed in June had the lowest number of total born with an average of $6.25{\pm}2.22$ piglets per sow. However, the lowest average number of piglets weaned at day 28 was found in sows that farrowed in May, as well as the highest number recorded for the stillborn piglets with an average of 2.67 piglets per sow. Moreover, the smallest increase in weight from birth to weaning occurred in piglets that were farrowed in May, which also corresponds with the average daily gain (ADG) of 0.29 kg and the last recorded weight measurement on day 140 of $41.69{\pm}1.45kg$. Contrastingly, the highest growth rate was found among pigs farrowed in June, with the largest increase in weight of 7.55 kg from birth to weaning, the highest ADG of 0.51 kg from birth to 140 day of age and the highest BW of $74.70{\pm}1.86kg$ recorded on day 140. Conclusion: Pigs farrowed in June also had the least number of piglets that died between birth and weaning. The zone of thermal comfort found in sows reared in indoor confinement systems did not improve the reproductive performance of the sows reared in an outdoor, alternative production system, while the growth performance of the piglets was improved when the ambient temperature was consistently hot or consistently cold.
