Measurements of simulated periodontal bone defects in inverted digital image and film-based radiograph: an in vitro study

  • Purpose: This study was performed to compare the inverted digital images and film-based images of dry pig mandibles to measure the periodontal bone defect depth. Materials and Methods: Forty 2-wall bone defects were made in the proximal region of the premolar in the dry pig mandibles. The digital and conventional radiographs were taken using a Schick sensor and Kodak F-speed intraoral film. Image manipulation (inversion) was performed using Adobe Photoshop 7.0 software. Four trained examiners made all of the radiographic measurements in millimeters a total of three times from the cementoenamel junction to the most apical extension of the bone loss with both types of images: inverted digital and film. The measurements were also made in dry mandibles using a periodontal probe and digital caliper. The Student's t-test was used to compare the depth measurements obtained from the two types of images and direct visual measurement in the dry mandibles. A significance level of 0.05 for a 95% confidence interval was used for each comparison. Results: There was a significant difference between depth measurements in the inverted digital images and direct visual measurements (p>|t|=0.0039), with means of 6.29 mm ($IC_{95%}$:6.04-6.54) and 6.79 mm ($IC_{95%}$:6.45-7.11), respectively. There was a non-significant difference between the film-based radiographs and direct visual measurements (p>|t|=0.4950), with means of 6.64mm($IC_{95%}$:6.40-6.89) and 6.79mm($IC_{95%}$:6.45-7.11), respectively. Conclusion: The periodontal bone defect measurements in the inverted digital images were inferior to film-based radiographs, underestimating the amount of bone loss.

자기공명영상에서 정형보철 금속이 신호강도와 영상왜곡에 미치는 영향 (The Impact of Signal Intensity and Image Distortion Magnetic Resonance Imaging in the Orthopedic Prosthetic Metal)

  • 병원에서 정형보철용 재료로 사용하는 스테인리스와 티타늄 금속을 같은 크기로 제작하여 자기공명영상에 미치는 영상학적 진단가치를 비교하였다. 영상의중심(Center), 영상이외부분(Background), 고 신호부분(High band)에 대한 신호강도 값을 가중치로 변환하여 금속시험편 없는(Norma)영상과 비교하였다. 또한 시상면영상과 횡단면영상의 왜곡범위를 정량적으로 수치화하여 금속시험편 없는 영상면적과 비교하였다. 그 결과 고 신호영상의 신호강도 값은 스테인리스의 경우 팬텀(Phantom) 2, 돼지 뼈(Bone) 1.8, 티타늄의 경우 팬텀 1.7, 돼지 뼈 1.3 만큼 금속시험편 없는 신호강도가중치 값보다 높은 것을 알 수 있었다. 또한 자기공명영상의 왜곡형태와 왜곡 범위는 금속시험편이 없는 영상의 단면적대비 스테인리스의 경우 팬텀 65.8 %, 돼지 뼈 61.5 %, 티타늄은 팬텀 23.1 %, 돼지 뼈 38.5 % 의 왜곡범위를 나타냈다. 결론적으로 본 실험에서는 신호강도가중치값과 왜곡범위가 낮게 변화를 보인 티타늄이 스테인리스 시험편보다 더 진단적 가치가 높은 자기공명영상임을 알 수 있었다.

Polymorphism of Insulin-like Growth Factor Binding Protein-4 Gene in 17 Pig Breeds and Its Relationship with Growth Traits

  • Insulin-like growth factor binding protein-4 (IGFBP-4) is a member of the IGF super family, and regulates the action of IGFs. The polymorphism of porcine IGFBP-4 gene in 17 pig breeds (total n = 570) was detected by PCR-SSCP, and alleles A and B were detected. In these pig breeds, it was found that exotic pig breeds carried high frequencies of allele A, while Chinese native pig breeds carried high frequencies of allele B. The role of porcine IGFBP-4 was investigated in 172 F2 offspring of a $Lantang{\times}Lantang $ population. Forty eight growth traits were recorded for analyzing the association between IGFBP-4 gene polymorphism and quantitative performance traits. In this resource family, pigs with AA genotype had higher fore-body weight, bone weight of mid-body, bone weight of rear-body, fore-leg weight and rear-leg weight than those pigs with BB genotype (p<0.05); while pigs which carried BB genotype had higher back-fat thickness at C point and lard weight than those pigs with AA genotype (p<0.05); pigs with AA genotype had higher body weight than those with BB genotype; for meat quality traits, pigs with AA genotype had higher meat color than those of BB genotype (p<0.01), and pigs with BB genotype had higher marbling than those of AA and AB genotypes (p<0.01 and p<0.05, respectively).Based on these results, it is necessary to do more studies on IGFBP-4 before using the IGFBP-4 locus for the application of marker-assisted selection programs.

비타민 C 결핍이 guinea pig의 실험적 치아이동속도와 치조골 개조에 미치는 영향 (Effect of vitamin C deficiency on the rate of orthodontic tooth movement and alveolar bone remodeling)

  • 이 연구는 비타민 C 결핍이 guinea pig에서의 실험적 치아이동속도와 치조골 개조에 미치는 영향을 알아보고자 시행되었다 이를 위하여 웅성 guinea pig 30마리를 대상으로 정상량의 비타민 C (5 mg/day) 를 투여한 군(정상군)과 결핍량(0.2mg/day)을 투여한 군(결핍군)으로 나누고 치아이동 실험을 시행하였다. 초기 교정력 75gm으로 상악의 좌우 중절치를 이개시키는 치아이동을 시행하고, 순차적으로 실험 경과기간이 지나면 치아이동양을 계측하고 희생시켜 조직 소견을 관찰하였다. 실험 결과. 결핍군에서 치조골과 치주인대의 교원섬유 함량은 현저한 감소와 불규칙한 배열을 보였으며 인대세포의 수적 감소 및 출혈이 관찰되었고 치조골에서 골형성의 감소와 파골세포의 출현 및 골소강의 크기증가와 골소강내의 다수의 파골세포가 관찰되었다. 정상군의 인장측 치조골은 치아이동 시간경과에 따라 조골세포와 골형성이 지속적인 증가를 보였으나, 결핍군에서는 조골세포가 감소하고 골형성은 미약한 양상을 보였다. 치아 이동양은 실험 초기 1일, 3일, 5일, 7일 결핍군의 치아이동양이 정상군보다 많게 나타났다 (p<0.05) 이상의 결과에서 비타민 C 결핍은 치주조직의 교원질의 합성을 억제하여 치조골의 형성을 감소시키고 흡수를 증가시키는 골개조를 초래하며, 치아이동중 인장측 치조골 형성을 억제하고 압박측 치조골 흡수를 촉진하여 치아이동 초기에는 빠른 치아이동을 가져오는 것을 시사한다.

Detection of Mendelian and Parent-of-origin Quantitative Trait Loci in a Cross between Korean Native Pig and Landrace I. Growth and Body Composition Traits

  • This study was conducted to detect quantitative trait loci (QTL) affecting growth and body composition in an $F_2$ reference population of Korean native pig and Landrace crossbreds. The three-generation mapping population was generated with 411 progeny from 38 $F_2$ full-sib families, and 133 genetic markers were used to produce a sex-average map of the 18 autosomes. The data set was analyzed using least squares Mendelian and parent-of-origin interval-mapping models. Lack-of-fit tests between the models were used to characterize QTL for mode of expressions. A total of 8 (39) QTL were detected at the 5% genome (chromosome)-wise level for the 17 analyzed traits. Of the 47 QTL detected, 21 QTL were classified as Mendelian expressed, 13 QTL as paternally expressed, 6 QTL as maternally expressed, and 7 QTL as partially expressed. Of the detected QTL at 5% genome-wise level, two QTL had Mendelian mode of inheritance on SSC6 and SSC9 for backfat thickness and bone weight, respectively, two QTL were maternally expressed for leather weight and front leg weight on SSC6 and SSC12, respectively, one QTL was paternally expressed for birth weight on SSC4, and three QTL were partially expressed for hot carcass weight and rear leg weight on SSC6, and bone weight on SSC13. Many of the Mendelian QTL had a dominant (complete or overdominant) mode of gene action, and only a few of the QTL were primarily additive, which reflects that heterosis for growth is appreciable in a cross between Korean native pig and Landrace. Our results indicate that alternate breed alleles of growth and body composition QTL are segregating between the two breeds, which could be utilized for genetic improvement of growth via marker-assisted selection.

형질변형성장인자가 손상골 재생능에 미치는 영향에 관한 실험적 연구 (THE EFFECTS OF TGF-$\beta$1 ON THE BONE REGENERATION IN GUINEA PIG MANDIBLE)

  • The purpose of this study was to observe the effect of TGF-βl, which promotes differentiation and proliferation of osteoblasts, on bone regeneration. Experimental bone defects that measured 3 mm in diameter were created on the mandibles of guinea pig by removal of bone with the use of trephine burs. In one side of mandibular body, the experimental groups, bone defects were grafted with Biogran(Orthovita Co., U.S.A) and TGF-β1(R&D System Co., U.SA). In the remaining side of the mandiblar body, the control groups, bone defects were grafted with only Biogran. Guinea pigs in the control and experimental groups were serially terminated by fours on the 3 days, the 1 week, the 2 weeks, the 3 weeks, and the 4 weeks after experiment, and both sides of the mandibular bodies were removed and fixed with 10% neutral formalin. They were decalcified and embedded in paraffin as using the usual method. The specimen sectioned and stained with hematoxylin and eosin. Also, they were radiographed with a soft X -ray apparatus. The obtained results were as follows; 1. Hemorrhagic condition, observed in the granulation tissues, disappeared on the 1 week after experiment in both groups, and more prominent in the experimental group. The granulation tissues of the experimental group had larger number of cells than those of the control group. 2. Osteoblastic differentiation in the margin of grafted material and adjacent bone was observed on the 1 week after experiment in both groups. Also, bone formation was observed in immature form on the 1 week after experiment. and more prominent in the experimental group. 3. In the polarizing microscopic examination, bone matrix was very loose on the 1 week after experiment, but increase in density with time, and more prominent in the experimental group. 4. In the microradiographic examination, newly formed bone was observed in the experimental group on the 2 weeks after experiment, and this was observed earlier than in the control group. Newly formed bone was increased with time and defected area was markedly decreased on the 4 weeks after experiment.

Porcine study on the efficacy of autogenous tooth bone in the maxillary sinus

  • Objectives: This study sought to elucidate the effect of autogenous tooth bone material by experimenting on minipig's maxillary sinus and performing histological and histomorphometric analyses. Materials and Methods: Five 18-24 month-old male minipigs were selected, and right maxillary sinuses were grafted with bone graft material made of their respective autogenous teeth extracted eight weeks earlier. The left sides were grafted with synthetic hydroxyapatite as control groups. All minipigs were sacrificed at 12 weeks after bone graft, which was known to be 1 sigma (${\sigma}$) period for pigs. Specimens were evaluated histologically under a light microscope after haematoxylin-eosin staining followed by semi-quantitative study via histomorphometric analysis. The ratio of new bone to total area was evaluated using digital software for calculation of area. Results: All specimens were available, except one on the right side (experimental group), which was missing during specimen preparation. This study demonstrated new bone at the periphery of the existing bone in both groups, showing evidence of bone remodeling, however, encroachment of new bone on the central part of the graft at the 1 ${\sigma}$ period was observed only in the autogenous tooth bone group (experimental group). Histomorphometric analysis showed more new bone formation in the experimental group compared to the control group. Although the difference was not statistically significant (P>0.05), the mean percentage area for new bone for the experimental and control groups were $57.19%{\pm}11.16%$ and $34.07%{\pm}13.09%$, respectively. Conclusion: The novel bone graft material using autogenous tooth is a good alternative to autogenous bone, comparable to autogenous bone, and outperforming synthetic hydroxyapatite bone graft materials in terms of bone regeneration capacity. Augmentation with autogenous tooth bone materials will reduce donor site morbidity without hampering the safety of the autogenous bone graft.

Production of Transgenic Pigs with an Introduced Missense Mutation of the Bone Morphogenetic Protein Receptor Type IB Gene Related to Prolificacy

  • In the last few decades, transgenic animal technology has witnessed an increasingly wide application in animal breeding. Reproductive traits are economically important to the pig industry. It has been shown that the bone morphogenetic protein receptor type IB (BMPR1B) A746G polymorphism is responsible for the fertility in sheep. However, this causal mutation exits exclusively in sheep and goat. In this study, we attempted to create transgenic pigs by introducing this mutation with the aim to improve reproductive traits in pigs. We successfully constructed a vector containing porcine BMPR1B coding sequence (CDS) with the mutant G allele of A746G mutation. In total, we obtained 24 cloned male piglets using handmade cloning (HMC) technique, and 12 individuals survived till maturation. A set of polymerase chain reactions indicated that 11 of 12 matured boars were transgene-positive individuals, and that the transgenic vector was most likely disrupted during cloning. Of 11 positive pigs, one (No. 11) lost a part of the terminator region but had the intact promoter and the CDS regions. cDNA sequencing showed that the introduced allele (746G) was expressed in multiple tissues of transgene-positive offspring of No.11. Western blot analysis revealed that BMPR1B protein expression in multiple tissues of transgene-positive $F_1$ piglets was 0.5 to 2-fold higher than that in the transgene-negative siblings. The No. 11 boar showed normal litter size performance as normal pigs from the same breed. Transgene-positive $F_1$ boars produced by No. 11 had higher semen volume, sperm concentration and total sperm per ejaculate than the negative siblings, although the differences did not reached statistical significance. Transgene-positive $F_1$ sows had similar litter size performance to the negative siblings, and more data are needed to adequately assess the litter size performance. In conclusion, we obtained 24 cloned transgenic pigs with the modified porcine BMPR1B CDS using HMC. cDNA sequencing and western blot indicated that the exogenous BMPR1B CDS was successfully expressed in host pigs. The transgenic pigs showed normal litter size performance. However, no significant differences in litter size were found between transgene-positive and negative sows. Our study provides new insight into producing cloned transgenic livestock related to reproductive traits.

Effect of bone quality and implant surgical technique on implant stability quotient (ISQ) value

  • PURPOSE. This study investigated the influence of bone quality and surgical technique on the implant stability quotient (ISQ) value. In addition, the influence of interfacial bone quality, directly surrounding the implant fixture, on the resonance frequency of the structure was also evaluated by the finite element analysis. MATERIALS AND METHODS. Two different types of bone (type 1 and type 2) were extracted and trimmed from pig rib bone. In each type of bone, the same implants were installed in three different ways: (1) Compaction, (2) Self-tapping, and (3) Tapping. The ISQ value was measured and analyzed to evaluate the influence of bone quality and surgical technique on the implant primary stability. For finite element analysis, a three dimensional implant fixture-bone structure was designed and the fundamental resonance frequency of the structure was measured with three different density of interfacial bone surrounding the implant fixture. RESULTS. In each group, the ISQ values were higher in type 1 bone than those in type 2 bone. Among three different insertion methods, the Tapping group showed the lowest ISQ value in both type 1 and type 2 bones. In both bone types, the Compaction groups showed slightly higher mean ISQ values than the Self-tapping groups, but the differences were not statistically significant. Increased interfacial bone density raised the resonance frequency value in the finite element analysis. CONCLUSION. Both bone quality and surgical technique have influence on the implant primary stability, and resonance frequency has a positive relation with the density of implant fixture-surrounding bone.

신생자돈의 선천성 전지과골증의 발생예 (Congenital Hyperostosis in Pig Litters)

  • Two cases of congenital hyperostosis at a pig farm were observed and examined pathologically in Landrace piglets. Twenty two piglets from 2 litters: 10 alive piglets in a litter and 2 stillborn and 10 alive piglets in another, were born with thickened forelimbs and some of them with thickened hindlimbs too. These piglets stowed symptoms of staggering and incoordination. Only one of these piglets could be reared normally and all the others died within 3 days of age. The thickened parts of their limbs were felt firm and not well pitted by finger pressing. An apparent ossification was identified on the central regions of the cross sections of the thickened limbs, and some pale, gray and ossiferous signs were found in the peripheral soft areas. On histopathological examination of the thickened limbs, it was found that the bone trabeculae were newly proliferated in a concentric form in central regions and in a radiated form in peripheral regions. The peripheral regions of the periosteum and perichondrium of new bone were proliferated with connective tissue and the edematous muscle bundles were atrophic and degenerative. Both the sows which produced piglets of congenital hyperostosis were mated with a different boar at next breeding and farrowed normal piglets. It was concluded that the swine congenital hyperostosis in the present cases was expressed by autosomal recessive gene of first mating boar.
