Antioxidative and Inhibition Activities on Helicobacter pylori of Spice Extracts (향신료 추출물의 항산화활성 및 Helicobacter pylori 저해효과)

  • For the purpose of developing natural antioxidant, the antioxidative and antimicrobial activities of phenolics isolated from spices were determined. The total phenolics contents of spices were more than 20 mg/g in water and 60% ethanol extracts of all spice, oregano and sage. Electron donating ability assay showed high inhibition rate in water extracts of all spice, nutmeg, white pepper, oregano and sage and 60% ethanol extracts of oregano and nutmeg. Antioxidant protection factor (PF) was higher than 1.2 in 60% ethanol extracts of sage, all spice and oregano and water extracts of sage. The 2,2'-azinobis-3-ethylbenzothiazoline-6-sulfonic acid radical decolorization (ABTS) was inhibited by more than 90% by water and 60% ethanol extracts of all spice and oregano. TBARS (thiobarbituric acid reactive substances) were $0.7{\mu}M$ in the control and $0.2{\mu}M$ in water and 60% ethanol extracts the each spices. The water extracts of each spices did not have antimicrobial activity against H. pylori; however, the 60% ethanol extracts from oregano revealed the high antimicrobial activity as clear zone of 10 mm and inhibition rate of 77.2% with $200{\mu}g/mL$ of phenolics content. The result suggests that spices extract may be useful as potential sources of anti-Helicobacter pylori, antioxidant.

Comparative Analysis of Catechins and Antioxidant Capacity in Various Grades of Organic Green Teas Grown in Boseong, Korea (보성산 유기농 녹차의 품질에 따른 카테킨 함량과 항산화능 비교 분석)

  • The objective of this study was to evaluate the effect of various solvents on extraction of bioactive phenolics and to analyze the antioxidant capacity and contents of individual catechins in various grades of green teas organically grown in Boseong, Korea. The organic green teas, based on their harvest seasons, were categorized into five grades such as Woo-Jeon, Se-Jak, Jung-Jak, Dae-Jak, and coarse tea. Solvents used to extract phenolics from these teas included water at $23^{\circ}C$ and $70^{\circ}C$ as well as 80% (v/v) aqueous methanol and ethanol. In general, aqueous organic solvents of methanol and ethanol led to higher extraction yields of phenolics than water at $23^{\circ}C$ and $70^{\circ}C$. Total phenolics and antioxidant capacity of the teas extracted with the aqueous organic solvents were approximately 1.5 to 3.2 and 1.8 to 3.8 times higher than those with water at $23^{\circ}C$ and $70^{\circ}C$, respectively. Coarse tea, the lowest grade of green tea, showed approximately 30-60% lower total phenolics and antioxidant capacity compared with the higher grade ones. Reversed-phase HPLC analysis was performed quantitatively to identify individual catechins, gallic acid, and caffeine in teas extracted with 80% (v/v) aqueous methanol. Based on their dry weights, the organic green teas contained about 1.7 to 2.9% of caffeine. Content (mg/g dry weight) of tea catechins decreased in the following order: Woo-Jeon (155.4) > Se-Jak (147.7) > Jung-Jak (143.2) > coarse tea (135.1) > Dae-Jak (130.5). (-)-Epigallocatechin gallate was the most abundant among the catechins analyzed. The highest grade of green tea, Woo-Jeon, had the highest amount of (-)-epigallocatechin gallate at 77.4 mg/g dry weight. Overall, the higher grade of organic green teas tended to have the higher level of antioxidant capacity and catechins.

Functional and sensory characteristics of kiwifruit jangachi cured with traditional Korean sauces, doenjang and kochujang (된장과 고추장 키위장아찌의 기능성 및 관능 특성)

  • This study aimed to compare total phenolic, total flavonoid, and vitamin C contents, antioxidant capacities, and sensory characteristics of jangachi made from green kiwifruit (Actinidia deliciosa cv. Hayward) and golden kiwifruit (A. chinensis cv. Hort16A), which were cured with doenjang and kochujang. Golden kiwifruit had higher total phenolic, total flavonoid, and vitamin C contents, and antioxidant capacities than green kiwifruit. Jangachi of both cultivars of kiwifruit had generally higher total phenolic and flavonoid contents, and antioxidant capacities than fresh kiwifruit. Doenjang jangachi of kiwifruit had higher total phenolic contents and antioxidant capacities than kochujang jangachi. Jangachi made from golden kiwifruit cured with kochujang, following pretreatment with sugar solution, had the highest overall quality on sensory evaluation among jangachies tested herein. Together, our findings suggest that kiwifruit jangachi may serve as a good dietary source of phenolics and antioxidants.

Comparative Study of Electron Donating Ability, Reducing Power, Antimicrobial Activity and Inhibition of ${\alpha}$-glucosidase by Sorghum bicolor Extracts (국내산 수수(Sorghum bicolor) 품종에 따른 전자공여능, 환원력, 항균활성 및 ${\alpha}$-glucosidase 저해활성의 비교)

  • Electron donating ability (EDA), reducing power, total phenolic contents, total flavonoid contents, antimicrobial activity, and ${\alpha}$-glucosidase inhibitory effects of twenty-two sorghum varieties were examined using ethanol extracts (70%, v/v). The electron donationg abilities (EDA) of Mesusu and Bulgeunjangmoksusu (94% at 10 ${\mu}g$/mL) indicated higher antioxidant activity compared to those of other varieties and standard antioxidants such as BHT (13%) and BHA (74%). The reducing power of Mesusu ($OD_{700}$=0.71) suggested higher antioxidant activity, which was dependent on sample concentration. Bitjarususu showed the highest content of total phenolics (22.9 mg GAE/g); however, extracts from Heuinsusu exhibited the lowest content of total phenolics (16.4 mg GAE/g). Jangmoksusu showed the highest total flavonolic contents (3.5 mg QE/g), and Sigyeongsusu and Chal (GS) susu displayed the most antibacterial activity (MIC=8 ${\mu}g$/mL) against Escherichia coli. Extracts of Bulgeunjangmoksusu, Moktaksusu, and Ginjangmoksusu showed the highest ${\alpha}$-glucosidase inhibitory effect (98%) at the concentration of 5 ${\mu}g$/mL.

Chemical Composition of Several Herb Plants (서양 허브식물의 화학성분)

  • Chemical compositions of several herbs (rosemary, mint, thyme, sage, and lavender) cultivated in Korea were analyzed. Approximate compositions were as follows: moisture $69.92{\sim}82.10%$, crude ash $2.48{\sim}6.15%$, crude fat $0.40{\sim}2.46%$, crude protein $0.84{\sim}1.57%$, and crude fiber $2.48{\sim}6.15%$. Total contents of phenolics determined by Folin-Dennis's method were in the range of $73.24{\sim}197.79mg%$. Contents of minerals, Na, Ca, Mn, P, Mg, Zn, and Fe determined by ICP-AES were $43.0{\sim}112.5,\;177.5{\sim}304.0,\;0.5{\sim}1.5,\;74.0{\sim}218.5,\;57.0{\sim}116.0,\;1.0{\sim}2.0$ and $3.0{\sim}5.0mg%$, respectively. Free sugar contents determined by HPLC were: sucrose $0{\sim}7.61$, glucose $0.94{\sim}15.92$, and rhamnose $0.64{\sim}7.99mg%$. Fatty acids including palmitic, stearic, oleic, linoleic, linolenic, and arachidonic acids were identified by GC. Linoleic and linolenic acid contents were higher than those of palmitic and stearic acids. Aroma components identified by GC-MS were 1,8-cineole, ${\alpha}-phellandrene,\;{\alpha}-terpinene,\;{\beta}-pinene,\;{\beta}-thujone$, borneol, butan-1-ol, cis-sabinene hydrate, ${\delta}-carene,\;{\gamma}-terpinene$, and verbenone.

Changes of Phytochemicals and Antioxidant Activity during Fermentation of Brown Soymilk (갈색콩 두유의 젖산 발효 중 phytochemicals 및 항산화 활성 변화)

  • The changes of total phenolic and isoflavone contents and antioxidant activities in of brown soymilk fermented by Lactobacillus plantarum P1201 were investigated. The brown soymilk proliferated the cell growth and reached about 11.55log cfu/g after fermentation for 60hr, while pH and titratable acidity ranged from 6.25 to 4.03 and 0.18% to 1.03%, respectively. The total phenolic contents of brown soymilk slightly increased from 2.87mg/g to 2.98mg/g after fermentation for 60hr. The levels of isoflavone-glycosides and -malonylglycosides decreased, while the isoflavone-aglycone contents increased during fermentation of brown soymilk. In particular, the isoflavone contents was 38.30㎍/g, but increased the highest value of 84.31㎍/g after fermentation for 60hr. After then, it was slightly decreased after 60hr of fermentation. In addition, the levels of daidzein, glycitein and genistein among isoflavone aglycones were 24.12㎍/g, 25.25㎍/g and 24.71 ㎍/g, after fermentation for 36hr. The DPPH and ABTS radical scavenging activities and FRAP assay showed to be slightly increased during lactic acid fermentation of brown soymilk.

Comparison of Antioxidant Capacity and Nutritional Composition of three Cultivars of Actinidia arguta

  • Actinidia arguta extracts obtained from three cultivars ('Sae-Han', 'Dae-Sung', and 'Chil-Bo') were assayed for their antioxidant properties and nutritional composition. Antioxidant activity of the extracts was evaluated using 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging method. Total phenolic contents of the extracts were determined by the Folin-Ciocalteu method. Vitamin C (L-ascorbic acid) content measured by a colorimetric method and reducing sugar content estimated by dinitrosalicylic acid (DNSA) method. Crude extracts from A. arguta 'Sea-Han' showed the most potent radical-scavenging activity showing 86.55% at 10 mg/ml. The DPPH radical scavenging activity of extracts and solvent fractions from Sea-Han cultivar was in decreasing order of EtOAc fraction>BuOH fraction>$CH_2Cl_2$ fraction>hexane fraction, among which EtOAc fraction showed the highest antioxidant activity (87.51 % at 5 mg/ml). Total phenolic contents in A. arguta 'Sae-Han', 'Dae-Sung' and 'Chil-Bo', were 32.93, 28.23, and 25.60 mg/g, respectively. Vitamin C contents of them were 840.57, 578.81 and 730.10 ug/g, respectively.

  • Current study investigated phytochemical compositions and antioxidant capacity of Korean leafy vegetables including chajogi, gomchwi, meowi and sseumbagwi. ${\beta}$-carotene, total soluble polyphenol, total flavonoids contents were determined, and antioxidant capacity were evaluated by various methods. ${\beta}$-carotene, total soluble polyphenol, total flavonoids contents in gomchwi were significantly higher (as much as 19 folds) compare to other vegetables (p<0.05). Sseumbagwi extract ($100{\mu}g/mL$) was removed 78.6% of superoxide radicals in xanthin-xanthin oxidase system measured by EPR. Gomchwi showed the highest nitrate scavenging activity as 94.3% at pH 1.2. In SOD-like activity, chajogi, gomchwi, and meowi were evaluated in the range of $12.6{\sim}24.5%$. All samples were revealed to prevent the reaction of ferrous chloride during 9 days, which were comparable ability with 125 mg/100 g of ${\alpha}$-tocopherol. The rich phytochemical contents of gomchwi and sseumbagwi influenced to high antioxidant capacity.

Effect of Potato Polyphenolics on Lipid Peroxidation in Rats (감나 폴리페놀이 흰쥐의 생체내 지질과산화에 미치는 영향)

  • The total contents of polyphenolics in potatoes measured by Folin Denis method were 42~76mg/100g in fresh weight. A major phenolic component contained in potato polyphenolics was identified as chlorogenic acid(3.6~15mg/100g in fresh weight). The antioxidative effects of potato polyphenolics and chlorogenic acid on the lipid peroxidation of liver microsome were studied in vivo and in vitro systems by measuring the formation of thiobarbituric acid reactive substances(TBARS) and the content of urine 8 hydroxy deoxyguanosine(OHdG). The TBARS contents of liver showed an increase in the cholesterol diet compared to those in the normal diet. This trend, however, was minimized when potato polyphenolics and chlorogenic acid were supplemented in the cholesterol diet. On the other hands, urinary 8 OHdG contents showed a marked increase with the supplementation of potato polyphenolics in the cholesterol diet. However, there was a trend of marked decrease by the supplementation of chlorogenic acid. In vitro study, potato poly phenolics and chlorogenic acid effectively inhibited the formation of TBARS in liver microsomal system in a dose dependent manners. These results suggest that potato polyphenolics exerts an antioxidative activity in cholesterol fed rat liver.

Total Polyphenol, Flavonoid Contents and Antioxidant Activities of Dolsan Leaf Mustard Pickle during Storage (저장 기간 중 돌산갓 피클의 폴리페놀, 플라보노이드 및 항산화 활성에 대한 변화)

  • Dolsan leaf mustard has various biological functions, including those in the immune system and cancer prevention. It contains high amounts of nutritional and medicinal compounds, which are important for maintaining optimum health. The objective of this study was to determine the total phenolics contents and antioxidant activity in Dolsan leaf mustard pickle (DLMP) during storage. DLMP methanol extracts had the highest total polyphenol and flavonoid contents of $35.56{\pm}0.01GAEmg/g$ and $4.54{\pm}0.03QEmg/g$, respectively. The DPPH and ABTS radical scavenging activities in the DLMP methanol extracts showed the highest activities of 79.4 and 85.36%, respectively. The ferric reducing antioxidant power (FRAP) assay showed 2.24-3.82 mM $FeSO_4$ eq. (p<0.05) in DLMP extracts. Overall, storage at day 14 showed the highest antioxidant activity.
