Preference evaluation of stink bugs to leguminous seeds by video tracking system (VTS를 이용한 두류종실에 대한 노린재류의 선호성 평가)

  • Preference of stink bugs to various leguminous seeds was evaluated by using VTS (Video Tracking System) in laboratory. Major soybean stink bugs such as bean bug, Riptortus pedestris (Fabricius), one-banded stink bug, Piezodorus hybneri (Gmelin), eastern green stink bug, Nezara antennata (Scott), and sole bug, Dolycoris baccarum (L.) were significantly most attracted to Cheongjakong, a soybean variety, baited fish-net trap, followed by soybean varieties Ilpumgeomjeongkong and Taekwangkong, a peanut variety Daekwangdangkong, a kidney bean variety Gangnangkong, and a adzuki bean variety Jungwonpat, respectively in a soybean field. VTS observation in laboratory showed that R. pedestris and D. baccarum had significantly higher frequency of visit on Cheongjakong, followed by Ilpumgeomjeongkong. However, P. hybneri, N. antennata and Halyomorpha halys (Stal) had significantly higher number of visits on Cheongjakong, Seonnogkong and Jinpumkong, followed by Ilpumgeomjeongkong. The sojourned time of stink bugs, however, was significantly longer on Cheongjakong regardless of species of the bugs. Accordingly, Cheongjakong was evaluated as the most preferred soybean seed by fish-net trap and VTS. Thus, VTS is found to be an effective means to evaluate the food preference of stink bugs.

Anti-inflammatory Effects of Canavaliae Semen(Canavalia gladiate) Extracts in a Systemic Anaphylaxis Food Allergy Mouse Model (전신과민성 식품 알레르기 마우스 모델에서의 도두(刀豆) 추출물의 항염 효과)

  • Objective : An allergy to peanuts is a major cause of fatal food-induced anaphylaxis, with food allergies becoming an increasingly important health research issue. Food allergy as clinical entity has been recongnized for many years, although there is yet no general concord as to the incidence of this symptom.1) Methods : This study was undertaken to verify the effect of seeds of Canavalia gladiata (Jacq.) DC. extract (CGE) on the inhibition of allergic reactions using a cholera toxin and peanut extract-immunized food allergy mouse model. We determine whether the changes in rectal temperature were related to energy consumption owing to heat production in the body. Mast cell distribution and degranulation in the dermis and epidermis were observed with an optical microscope. Subsequently, Ara h1 levels in serum and interleukin (IL)-4, IL-10, and $IFN-{\gamma}$ levels in cultured supernatants of splenocytes were measured. Results : CGE treatment significantly attenuated the secretion of the Ara h1 antibody in serum and splenocytes. Ara h 1 was undetected in the cholera toxin and peanut extract-immunized food allergy mouse model. Improvement in ear tissue inflammation symptoms was the CGE experimental group. In the control group and peanut extract control group, the expression of mast cells was higher, whereas that in the CGE experimental group was significantly lower. Conclusion : CGE causes suppression in a food allergy mouse model via the inhibition of Ara h1 secretion, and might be useful for developing functional health foods.

Uptake and Accumulation of Soil Strontium-90 by Peanut and Sesame (토양 strontium-90의 땅콩과 참깨에 의한 흡수 및 축적)

  • A greenhouse experiment on the Sr-90 uptake by peanut and sesame was conducted through pot cultures on a sandy loam soil of pH 6.35 treated with Sr-90 in 5.2 and 31.2Bq per gram. The rate of Sr-90 transfer from soil to each plant part, the ratio of Sr-90 concentration in the part to the concentration in soil, and the patterns of their temporal changes were not, on the whole, significantly different between the two treatments. About 0.7 and 0.5 % of Sr-90 in soil transferred to all the mature plants of peanut and sesame, respectively, with the radioactivities in their roots not counted. Only 4% and less than 15% of Sr-90 absorbed by peanut and sesame, respectively, translocated to their seeds. Both crops showed the highest Sr-90 concentration in the leaf and the lowest in the seed. At maturities, the concentration ratio in dry seed was 0.4 in peanut and 3.3 in sesame and that in dry leaf was 12.5 and 10.7, respectively. Sr-90 concentrations in the top 15 cm soil after harvests averaged about 80 % of the concentrations at starting. Sr-90 uptake resulted in neither growth inhibition nor yield decrease.

Study on functional improvement of peanut sprouts by LEDs

  • The research was carried out to investigate a total polyphenol content, antioxidant activity, amino acid and resveratrol content of peanut sprouts (cotyledon, epicotyl, leaf, hypocotyl, root), in different light (white, blue, red, F-red, UV-A, UV-B, UV-C) conditions for 24 hours. Peanut seeds were sown on a $27.5{\times}15.9{\times}13cm$ tray and grown at the $25^{\circ}C$ under the dark condition for 14 days. Total polyphenolic contents of epicotyl and leaf were about 288mg GAE/100g in blue light. The DPPH radical scavenging of cotyledon and hypocotyl were 1.3~1.5 times (63%) and 2 times (40%) compared to control (43%, 19%), respectively. As to ABTS activity, its activity was increased by all LEDs treatment, Especially, the highest ABTS activity of the hypocotyl and leaf was shown as 99.1% in blue light. The essential amino acid content of hypocotyl and leaf was increased 1.9 times in the UV-B, 1.6 times in red, and 1.5 times in F-red, respectively. The non-essential amino acid content was increased by all LEDs treatment in hypocotyl and leaf. The content of resveratrol was increased by 1.3 times in UV-B compared to that of other tissues. Assessing inclusively, this study showed that there was a significantly positive effect between increase of physiological substance activity and LED light treatment, resulting in stably producing peanut sprouts. Therefore, a material treated with LEDs is thought to be useful as a functional food resources.

Development of Selectable Marker of High Oleate Trait in Peanut (Arachis hypogaea L.) (땅콩에서 고 올레인산 형질관련 분자마커의 선발)

  • Peanut(Arachis hypogaea L.) is one of the major oilseed crops. The peanut oil consists of palmitic, oleic and linoleic acids, which are present at levels of 10%, 36-67% and 15-43%, respectively. High oleate mutant of peanut F435 contains 80% oleate and as little as 2% linoleate in seed oil. Previous study indicated that delta 12 fatty acid desaturase is a major enzyme controlling the oleate content in seeds of oilseed crops. F435 sequence alignment of their coding regions disclosed that an extra A(adenine) was inserted at the position +2,823 bp of delta 12 fatty acid desaturase gene. This study was to develop molecular marker (SNP marker) co-segregating with the high oleate trait. Chopyeong ${\times}$ F435 $F_2$ 41 population were investigated using molecular marker and fatty acid assay (NIR and gas chromatography). Finally, this marker segregates Chopyeong type 26 lines, heterotype 9 lines and F435 type 6 lines. These results in our study suggested that SNP marker conform fatty acid assay.

Monitoring Pesticide Residues in Nuts and Seeds in Gyeonggi-do (경기도 내 유통 견과종실류의 잔류농약 실태 모니터링)

  • This study was aimed to investigate pesticides residue levels in nuts and seeds distributed in Gyeonggi-do. We investigated nuts and seeds in 131 products, which is pumpkin seeds (n=17), sunflower (n=12), sesame (n=23), perilla (n=11), peanut (n=18), almond (n=8), chestnut (n=6), walnut (n=14), pine nut (n=3), pecan (n=4), macadamia (n=3), brazil nut (n=5), cashew nut (n=4) and ginkgo nut (n=3). As a result of evaluating 339 pesticide residues, eight out of 131 samples were detected at the range of 0.01-0.07 mg/kg. Additionally, 7 out of 8 detected samples exceeded MRLs (Maximum Residue Limits) and 85% of them were imported. Pyraclostrobin applied on the positive list system (PLS) was detected in five Chinese pumpkin seeds samples, and the range was 0.02-0.04 mg/kg. In Indian and Korean sesame seeds, pendimethalin, boscalid, and bifenthrin were detected at 0.04, 0.05, and 0.06 mg/kg, respectively. The ratio of estimated daily intake (EDI) to acceptable daily intake (ADI) was 0.002-0.372%, indicating that the risk level was safe.

  • trans-Resveratrol(3,5,4'-trihydroxystilbene) is phenolic compound present in grapes, wines, and peanuts, has been reported to have health benefits including anticarcinogenic effects, protection against cardiovascular diseases and reduced cancer risk. A simple method for the quantitative extraction of trans-resveratrol from peanut has been developed. Optimal conditions for extraction were investigated. Type of solvent, time, and temperature assayed influenced trans-resveratrol yield. Adequate extraction condition was decided to ethanol/water (80:20v/v) maintained at $25^{\circ}C$ for 45 min. After extraction, the protocol consists of sample preparation using a $\textrm{C}_{18}$ solid-phase extraction (SPE) cartridge after concentrate with rotary evaporator and quantified by reversed phase HPLC using a $\textrm{C}_{18}$ column at 308 nm. Analytical methods for measuring trans-resveratrol in peanut were adapted to isolate, identify, and quantify trans-resveratrol in 11 peanut varieties by HPLC (high-performance liquid chromatography) with UV detector, The 11 peanut varieties content ranged from 0.018 to 1.125 $\mu\textrm{g}/\textrm{g}$ with an average of 0.289 $\mu\textrm{g}/\textrm{g}$. The contents were higher in the seeds with than without testa, regardless of varieties. The trans-resveratrol content was Higher in 110, 130 days after sowing than that of other period.

Evaluation of the Antioxidant Effects of Extracted Seed Oils by Pressure Method using Domestic Seeds and Nuts (국내 종실류를 이용한 압착 오일의 화장품 소재로서 항산화 활성 평가)

  • This study was performed to evaluate the antioxidant activity capacity of extracted seed oils by pressure method using domestic plant resources as a cosmetics material. Four type of oil were extracted from pumpkin seed, camellia seed, red pepper seed, peanut. The extracted seed oils were analyzed for fatty acid composition by GCMS, The antioxidant activity evaluated by DPPH radical scavenging activity, ABTS radical cation decolorization activity. Pumpkin seed oil(PSO), camellia seed oil(CSO), peanut oil(PO) showed higher ratio of unsaturated fatty acid than saturated fatty acid. After heat treatment, the content of unsaturated fatty acids was higher than that of saturated fatty acids except for red pepper seed oil(RSO). In the result of DPPH, ABTS antioxidant activity, RSO were the highest 87.84%, 76.72% at the $200{\mu}{\ell}/m{\ell}$, PSO, PO and CSO were the highest antioxidant activities at the $1000{\mu}{\ell}/m{\ell}$. Compared with the positive control olive oil, DPPH radical scavenging activity of RSO, PSO and PO showed higher than the control. ABTS radical cation decolorization activity of RSO and PSO is stronger than the control. After heat treatment, the antioxidant activity capacity showed a slight difference, four type of oils is expected as having potential to be useful as a cosmetic material.

Effect of the Mixing Extraction of Perilla Seed and Peanut on Physicochemical Characteristics and Oxidative Stability of Perilla Oil (들께와 땅콩의 혼합 채유가 들기름의 이화학적 특성 및 산화안정성에 미치는 영향)

  • The oils were extracted from the mixture of roasted(for 20 min at 190oC) perilla seeds(RPS) and roasted (commercially) peanuts(RPN) by solvent extraction(SE) and mechanical expression(ME). The effects of mixing ratio on physicochemical characteristics and oxidative stability of their oils were investigated. Yields of both SE and ME oils were increased as the RPN ratio in the mixture increased. In all the SE and ME oils, the major fatty acids were oleic, linoleic and linolenic acid, and total saturated fatty acids increased gradually, but total unsaturated fatty acids decreased gradually as the RPN ratio in the mixture was increased. The specific gravity and refractive index of both SE and ME oils decreased as the RPN ratio in the mixture was increased. Acid value, saponification value and iodine value of SE oils decreased as the RPN ratio in the mixture increased, whereas acid value and iodine value of ME oils decreased and saponification value increased. The colors of ME oils were darker brownish than SE oils. The oxidative stability of SE oils was decreased as the RPN ratio in the mixture increased, whereas that of ME oils was increased. Sensory evaluation of all the oils extracted from the mixture with various mixing ratio showed significant differences in flavor, taste, color and overall acceptance(p<0.01). The oil extracted from the mixture of the mixing ratio of 8:2(RPS:RPN) showed slightly higher preference regardless of extraction method.

Studies on the Composition of Sapindus Mukurossi Seeds (무환자(無患子)나무 종자(種字)의 성분(成分)에 관(關)한 연구(硏究))

  • The composition of fatty acids and sterols from Sapindus Mukurossi oil were analyzed by gas liquid chromatography and amino acids from Sapindus Mukurossi seed oil cake were determined by amino acid autoanalyzer. The results obtained were summarized as follows; 1. Oil content (45.7%) was higher in Sapindus Mukurossi seed than in other plant seeds such as soybean, sesame and peanut. 2. From the analysis of fatty acids, it was found that oleic acid(61.6%) and gadoleic acid(20.4%) were major fatty acid and that high monoenoic acid content(82%) could be a characteristic of sample seed oil. 3. Glutamic acid contained the most amounts among all amino acids determined, and isoleucine, leucine, lysine, phenylalanine, threonine and valine were relative high amounts compared with content of other amino acids. 4. The sterol compositions of 4-desmethyl sterol, 4-monomethyl sterol and 4,4-dimethyl sterol fraction were found to be similar to those of higher plant oil, but 4-monomethyl sterol fraction was characterized by containing the unknown sterol (31.5%) with RRT 1.27.
