복분자가 생쥐의 특이적 면역반응에 미치는 영향 (Effect of Rubus coreanus Miquel on the Specific Immune Response in Mice)

  • The specific immune response of unripened fruits and ripened fruits of Rubus coreanus Miquel was examined in BALB/C mice. The 70% ethyl alcohol extracts (20 or 100 mg/kg) of unripened fruits (RCE-I) and of ripened fruits (RCE-II) were administered p.o. once a day for 7 days. RCE-I and RCE-II decreased the viability of thymocytes, but increased the viability of splenocytes. Also, RCE-I enhanced the population of helper T and cytotoxic T cells in thymocytes and RCE-II enhanced the population of helper T cells. Furthermore, RCE-I and RCE-II decreased the population of B220/sup +/ cells and Thy1/sup +/ cells and helper T cells in splenocytes. In a general way, the immunosuppressive action of RCE-I was more potent than those of RCE-II. These results suggest that RCE-I and RCE-II have a regulative action of immune response via decrease the viability of thymocytes and increase the viability of splenocytes.

Genome-wide Identification, Classification, and Expression Analysis of the Receptor-Like Protein Family in Tomato

  • Receptor-like proteins (RLPs) are involved in plant development and disease resistance. Only some of the RLPs in tomato (Solanum lycopersicum L.) have been functionally characterized though 176 genes encoding RLPs, which have been identified in the tomato genome. To further understand the role of RLPs in tomato, we performed genome-guided classification and transcriptome analysis of these genes. Phylogenic comparisons revealed that the tomato RLP members could be divided into eight subgroups and that the genes evolved independently compared to similar genes in Arabidopsis. Based on location and physical clustering analyses, we conclude that tomato RLPs likely expanded primarily through tandem duplication events. According to tissue specific RNA-seq data, 71 RLPs were expressed in at least one of the following tissues: root, leaf, bud, flower, or fruit. Several genes had expression patterns that were tissue specific. In addition, tomato RLP expression profiles after infection with different pathogens showed distinguish gene regulations according to disease induction and resistance response as well as infection by bacteria and virus. Notably, Some RLPs were highly and/or unique expressed in susceptible tomato to pathogen, suggesting that the RLP could be involved in disease response, possibly as a host-susceptibility factor. Our study could provide an important clues for further investigations into the function of tomato RLPs involved in developmental and response to pathogens.

Functional Characterization of PR-1 Protein, β-1,3-Glucanase and Chitinase Genes During Defense Response to Biotic and Abiotic Stresses in Capsicum annuum

  • Spatial and temporal expression of pathogenesis-related (PR) gene and proteins has been recognized as inducible defense response in pepper plants. Gene expression and/or protein accumulation of PR-1, $\beta-1,3-glucanase$ and chitinase was predominantly found in pepper plants during the inoculations by Xanthomonas campestris pv. vesicatoria, Phytophthora capsici and Colletotrichum coccodes. PR-1 and chitinase genes were also induced in pepper plants in response to environmental stresses, such as high salinity and drought. PR-1 and chitinase gene expressions by biotic and abiotic stresses were regulated by their own promoter regions containing several stress-related cis-acting elements. Overexpression of pepper PR-1 or chitinase genes in heterogeneous transgenic plants showed enhanced disease resistance as well as environmental stress tolerances. In this review, we focused on the putative function of pepper PR-1, $\beta-1,3-glucanase$ and chitinase proteins and/or genes at the biochemical, molecular and cytological aspects.

Functional Analysis of Pepper Cys2/His-Type Zinc-Finger Protein Promoter Region in Response to Bacterial Infection and Abiotic Stresses in Tobacco Using Agrobacterium-Mediated Transient Assay

  • The promoter region flanking the 5’ CAZFP1 coding region was isolated from the genomic DNA of Capsicum annuum. To identify the upstream region of the CAZFP1 gene required for promoter activity, a series of CAZFP1 promoter deletion derivatives was created. Each deletion construct was analyzed by Agrobacterium-mediated transient transformation in tobacco leaves after infection by Pseudomonas syringae pv. tabaci, or treatment with methyl jasmonate (MeJA), ethylene, abscisic acid (ABA), salicylic acid (SA), cold and wounding. Promoter fragments of 685 bp or longer showed 7-fold or greater induction after P. s. pv. tabaci infection and MeJA treatment. The CAZFP1 full-length promoter (-999 bp) also showed 6-fold induction in response to ethylene. The transiently transformed tobacco leaves with the CAZFP1 full length promoter fused-GUS gene showed more than 5-fold induction in response to SA, ABA and cold. These results suggest that the CAZFP1 promoter contains responsive elements for pathogen, MeJA, ethylene, SA, ABA and cold.

귀비탕이 C57BL/6 Mouse에 Stress 부하 후 특이적 면역반응에 미치는 영향 (Effects of Kwibitang on the Specific Immune Response after Immobilization Stress in C57BL/6 Mice)

  • Stress is known to influence the immune function via an effect on the central nervous system. To investigated the effects of Kwibitang water extract (KBT) on the specific immune response in C57BL/6 mice stressed by immobilization, we evaluated the changes in the cell viability, DNA fragmentation and subpopulation of thymocytes and splenocytes. KBT enhanced the cell viability of thymocytes and splenocytes decreased by immobilization stress. Also, KBT decreased DNA fragmentation of thymocytes and splenocytes increased by immobilization stress. KBT decreased the population of CD4/sup +/ cells and CD8/sup +/ cells in thymocytes and Thy1/sup +/ cells in splenocytes increased by immobolization stress, but increased the population of B220/sup +/ cells decreased by immobilization stress. In addition, KBT enhanced the production of ν-interferon and IL-2 decreased by immobilization stress. These results indicate that KBT may be useful for the prevention and treatment of stress via enhancement of the specific immune response

  • Experimental studies were done to research the clinical effects of Juglandis Semen aqua-acupuncture and acupuncture(BL13, BL17, BL13ㆍ17 and free points) on the anti-allergic inflammation response. The following results have been obtained ; JSAA was increased in the lymphocyte proliferation in cultured murine thymocytes and splenocytes. The production of IL-1β was decreased in Juglandis Semen aqua-acupuncture and acupuncture(BL13, BL17 and BL13ㆍ17) group. The production of IL-4 was decreased in Juglandis Semen aqua-acupuncture and acupuncture(BL13, BL17 and BL13ㆍ17) group. The production of total IgG was decreased in Juglandis Semen aqua-acupuncture(BL13 and BL13ㆍ17) and acupuncture(BL13, BL17 and BL13ㆍ17) group. According to the above results, Semen aqua-acupuncture and acupuncture(BL13, BL17 and BL13ㆍ17) both enhances the specific immune response in T and B cells and depress the allergic inflammation reaction.

  • To investigated the effects of Kwibi-tang water extract (KBT) on the non-specific immune response in C57BL/6 mice stressed by immobilization, we evaluated the changes in the contents of serum histamine and corticosterone and the phagocytic activity of macrophages. The level of serum histamine and corticosterone was determined with spectrofluorometer. The cell viability was determined by a MTT assay method. The subpolpulation of lymphocytes was determined by a flow cytometry. The phagocytic activity was determined with luminometer. KBT decreased the serum level of histamine and corticosterone increased by immobilization stress. Also, KBT enhanced the phagocytic activity and decreased the level of nitric oxde in murine peritoneal macrophages decreased by immobilization stress. These results indicate that KBT may be useful for the prevention and treatment of stress via suppression of serum histamine and corticosterone level and enhancement of the non-specific immune response.

Overcoming Encouragement of Dragon Fruit Plant (Hylocereus undatus) against Stem Brown Spot Disease Caused by Neoscytalidium dimidiatum Using Bacillus subtilis Combined with Sodium Bicarbonate

  • The use of the supernatant from a Bacillus subtilis culture mixed with sodium bicarbonate was explored as a means of controlling stem brown spot disease in dragon fruit plants. In in vitro experiments, the B. subtilis supernatant used with sodium bicarbonate showed a strong inhibition effect on the growth of the fungus, Neoscytalidium dimidiatum, the agent causing stem brown spot disease and was notably effective in preventing fungal invasion of dragon fruit plant. This combination not only directly suppressed the growth of N. dimidiatum, but also indirectly affected the development of the disease by eliciting the dragon-fruit plant's defense response. Substantial levels of the pathogenesis-related proteins, chitinase and glucanase, and the phenylpropanoid biosynthetic pathway enzymes, peroxidase and phenyl alanine ammonia-lyase, were triggered. Significant lignin deposition was also detected in treated cladodes of injured dragon fruit plants in in vivo experiments. In summary, B. subtilis supernatant combined with sodium bicarbonate protected dragon fruit plant loss through stem brown spot disease during plant development in the field through pathogenic fungal inhibition and the induction of defense response mechanisms.

A Marine Bacterium with Animal-Pathogen-Like Type III Secretion Elicits the Nonhost Hypersensitive Response in a Land Plant

  • Active plant immune response involving programmed cell death called the hypersensitive response (HR) is elicited by microbial effectors delivered through the type III secretion system (T3SS). The marine bacterium Hahella chejuensis contains two T3SSs that are similar to those of animal pathogens, but it was able to elicit HR-like cell death in the land plant Nicotiana benthamiana. The cell death was comparable with the transcriptional patterns of H. chejuensis T3SS-1 genes, was mediated by SGT1, a general regulator of plant resistance, and was suppressed by AvrPto1, a type III-secreted effector of a plant pathogen that inhibits HR. Thus, type III-secreted effectors of a marine bacterium are capable of inducing the nonhost HR in a land plant it has never encountered before. This suggests that plants may have evolved to cope with a potential threat posed by alien pathogen effectors. Our work documents an exceptional case of nonhost HR and provides an expanded perspective for studying plant nonhost resistance.

Role of immunoreactive patterns of lymph nodes in neck dissection cases of oral squamous cell carcinoma: a clinical and histopathological study

  • Objectives: Metastasis in oral squamous cell carcinoma (OSCC) can occur in a variety of ways, and draining lymphatics and lymph nodes serve as a common route. Prior to metastasis, lymph nodes elicit an immune response to either wall off or create a favorable environment for homing of tumor cells. This immune response to tumor stimuli is visualized by recognizing various immunoreactive patterns exhibited by the lymph node. The present study aims to evaluate the role of immuno-morphologic patterns of the lymph node in neck dissection for cases of OSCC. Materials and Methods: Our retrospective study included 50 neck dissection cases of OSCC and a total of 1,078 lymph nodes. The grades of primary tumors with eight different immunoreactive patterns were compared. Vascularity and metastasis in lymph nodes were also evaluated. Results: The lymphocyte predominant pattern was the most common immunoreactive pattern found in 396 of 1,078 lymph nodes. Patterns of lymphocyte predominant (P=0.0005), sinus histiocytosis (P=0.0500), paracortical hyperplasia (P=0.0001), cortical hyperplasia (P=0.0001), and increased vascularity (P=0.0190) were significantly associated with tumor grade. Conclusion: The present study adds to the understanding of lymph node immunoreactivity patterns and their correlation with tumor grade. We recommend further study of lymph node patterns for all sentinel lymph node biopsies and routine neck dissections for OSCCs.
