Effluent and Aerobic Stability of Cellulase and LAB-Treated Silage of Napier Grass (Pennisetum purpureum Schum)

  • The effects of acremonium cellulase (AC) additive and lactic acid bacteria (LAB) inoculant on effluent production and aerobic stability of silage were investigated. Napier grass (Pennisetum purpureum Schum) was treated with AC at the rates of 0.05 ($AC_1$) and 0.1 g/kg $(AC_2)$ and/or with LAB at the rate of $1.0{\times}10^8cfu/kg$ fresh grass at ensiling. The treatments of LAB, $AC_1$, $AC_2$, $LAB+AC_1$ and $LAB+AC_2$ significantly (p<0.01) decreased pH and contents of volatile basic nitrogen and butyric acid, and significantly (p<0.01) increased lactic acid content compared with the control. All treated silages were well preserved with pH of lower than 4.2. There were no significant differences in fermentation quality between the application rates of AC ($AC_1$ and $AC_2$) and between the mixtures ($AC_1+LAB$ and $AC_2+LAB$). AC ($AC_1$ and $AC_2$) and AC plus LAB ($AC_1+LAB$ and $AC_2+LAB$) resulted in more silage effluent than the control and LAB inoculant alone. When the experimental silos were opened, the silages treated with AC and/or LAB were not as stable as the control silage, as shown by pH increase and lactic acid decomposition.

  • In this paper, some new generalized discrete Halanay inequalities are established. On the basis of these new established inequalities, we obtain the attracting set and the global asymptotic stability of the nonlinear discrete systems. Our results established here extend the main results in [R. P. Agarwal, Y. H. Kim, and S. K. Sen, New discrete Halanay inequalities: stability of difference equations, Commun. Appl. Anal. 12 (2008), no. 1, 83-90] and [S. Udpin and P. Niamsup, New discrete type inequalities and global stability of nonlinear difference equations, Appl. Math. Lett. 22 (2009), no. 6, 856-859].

Effect of Casein Dephosphorylation on Stability of Casein Micelles towards Ethanol (카제인의 탈인산화가 카제인 미셀의 에탄올 안정성에 미치는 영향)

  • Various artificial casein micelle systems were prepared from dephosphorylated whole casein, ${\beta}$- or ${\kappa}$-casein and their stabilities towards ethanol were assessed. Ethanol stability was lower in the micelle systems with dephesphorylated whole casein as compared to the artificial micelles prepared from native whole casein, and the stability decreased with the extent of dephosphorylation. The casein micelles with partially dephosphorylated ${\kappa}$-casein had a lower ethanol stability than those with native ${\kappa}$-casein. Ethanol stability of the micelle system with dephosphorylated ${\beta}$-casein decreased as the degree of dephosphorylation increased. Progressive dephosphorylation of caseins in skim milk system resulted in a decrease of the stability towards ethanol. The decrease was less than that in the system with dephosphorylated individual caseins. Increase in pH of the artificial casein micelle systems in the range of $6.3{\sim}7.2$ led to an increased ethanol stability manifesting that the presence of serine phosphates contributes significantly to the stability towards ethanol.

  • Enzymatic activity of isolated Lysozyme from egg white by cation ion-exchange chromatography was detected with various methods and stability of lysozyme in solution was studied by heat and pH treatments. Lysozyme activity refered to mg pure lysozyme/mg sample was more accurate although it needed standard lysozyme. But lysozyme activity refered to units/mg sample could be detected easily and reducted total detection time. Enzymatic activity of isolated lysozyme which dissolved in 0.066M phosphate buffer(pH 6.3) and then incubated at $37^{\circ}C$ for 2hr was increased remarkably on the lysis of Micrococcus lysodeikticus. The activity of isolated lysozyme by CM Sephadex C-25 was higher in eluting solution of above O. D. 1.0 at 640nm and attained 36, 000 units/mg solid. The stability of isolated lysozyme was decreased by various heat treatment. Activity began to decrease above 6$0^{\circ}C$ and dropped rapidly at $100^{\circ}C$. Especially, 35% loss of activity occured in 0.066M phosphate buffer at $100^{\circ}C$. for 15min. The stability of lysozyme was also affected by pH. lysozyme was very stable in acidic solution but in alkaline solution. Enzymatic activity showed maximum value at pH 3.0 solution while decreased rapidly above pH 6.0 solution.

Micro pH Sensor Using Patterned Hydrogel with pH Indicators

  • This paper presents a study into pH Indicator-Embedded hydrogel micro-particles which are encoded various shapes according to the captured indicator. We incorporate various pH indicators into a photo-curable hydrogel, PEG-DA(Poly(ethylene glycol) diacrylate). Using the latest fluidic lithography techniques, we can easily fabricate variously patterned hydrogel particles based on in-situ photopolymerization of the PEG-DA in a micro-fluidic channel. The shape of the particle is related to the pH indicator inside the particle. We demonstrate that the micro pH sensors change their colors according to the pH levels. The micro pH sensors have various characteristics that are related to the curing time, particle size, etc. By changing these conditions, we can adjust the long term stability and reaction time of the hybrid micro pH sensors.

  • One of the practical limitations with the use of liposomes for delivery of the pharmaceutical substances such antigens is that liposomes are relatively unstable in storage. In order to extend the stability of liposome in storage without affecting their functional activity, solution-type liposomes were dehydrated to form a structurally intact dry liposomes. Comparative immunological evaluation was carried out for both dry and solution-type liposomes containing gag-V3 chimera, consequently it was found that dry liposomes elicited both humoral and cellular response as efficiently as solution-type liposemes did against the same gag-V3 antigen. Especially, long-term stability of the liposomes was remarkably enhanced by the dehydration made to loposomes without a significant change in its ability to elicit immune response in vivo. These results indicate that dry pH-sensitive liposome may become an effective delivery and adjuvant system for general vaccine development.

  • In this study, the stability of concentrated colloidal liquid aphrons (CLAs) containing Aliquat 336 was measured. CLAs in the continuous phase are stabilized by water-soluble surfactant and oil-soluble surfactant. To compare the stability of CLAs, half-life was introduced. According to the change of state of solution, the rate of break-up of concentrated CLAs changed rigorously at critical coagulation concentration. Critical coagulation concentration was measured with changing pH. The effects of ionic strength and concentration of Sodium Dodecyl Benzene Sulfonate (SDBS) on the stability of concentrated CLAs in the continuous phase were also investigated.

  • As for the purpose, we first introduce an random mutation into wild-type gene to expand a mutation space, and then further recombine the mutant genes by staggered extension process PCR. As a result, we obtained the best clones 6-52 that showed a high activity and stability, from a round of error prone and staggered extension process PCR. The purified enzyme showed a similar pH stability to the wild-type enzyme and reveal a slightly high optimum pH at 12. In the optimum temperature, an identical dependency was also showed and a quite high stability in the thermal stability was obtained. Along with this, the enzyme was also stable at a reaction that supplement with a 15 % of ethanol as an additive. The addition of other solvents and surfactants did not improve the reaction and thus resulted in a similar profile to those of wild-type enzyme. The specific activity on the target compound rac-ketoprofen ethyl ester was calculated to be about 85, 000 unit, and the kinetic constants Km and Vmax were determined to be 0.2 mM and 90 mM/mg-protein/min respectively. The deduced amino acid alignment with the wild type enzyme revealed five mutations at L120P, I208V, T249A, D287H and T357A. Based on these observations, the site directed mutagenesis to delineate the mutagenic effect is under progress.

Stability of Mono- and Bis-pyridinium Oximes in Aqueous Systems (Monopyridinium Oxime과 Bispyridinium Oxime 화합물의 수용액 중 안정성 연구)

  • The stability of three oximes, Hl-6 [(4-carbamoyl-2'[(hydroxyimino)-methyl]- 1,1'-oxydimethylenedi-(pyridinium chloride)], Hl-CN [(4 cyano-2'-[(hydroxyimino)-methyl] -1,1'-oxydimethylene-di-(pyridinium chloride)], and 2-PAM [pralidoxime chloride] in aqueous solutions was evaluated by HPLC assay. The rate of degradation is dependent on the pH as well as the temperature at which the solution is stored. The optimum pH for the stability of these oximes was pH 2 to 3. The degradation rate constant for 2-PAM ($k\;at\;70^{\circ}C$, $2.07{\times}10^{-4}/hr;\;E_a\;value$, 27.2 kcal/mol) was smaller than those for bis-pyridiniumoximes, Hl-6 ($k\;at\;70^{\circ}C$, $3.38{\times}10^{-3}/hr$) and Hl-CN ($k\;at\;70^{\circ}C$, $8.66{\times}10^{-3}/hr;\;Ea\;value$, 20.7 kcal/mol). In mechanistic analyses, it was found that Hl-CN was decomposed through not only the hydrolysis of nitrile group but also the cleavage of methylene ether bridge, in contrast to Hl-6 which was degraded mainly through the cleavage of methylene ether bridge.

Effects of Synthetic Temperature and Suspension Stability of CeO2 Abrasive on CMP Characteristics (CeO2 연마입자의 합성온도와 수계안정성이 CMP 특성에 미치는 영향)

  • CMP(Chemical Mechanical Planarization) slurry for STI process is made by mechanically synthesized$CeO_2$as abrasive. The abrasive can be stabilized by electrostatic or steric stabilization in aqueous slurry and steric stabilization is more effective for long-term stability. Blanket-type$SiO_2$and $Si_3N_4$ wafers are polished with CMP slurry containing$CeO_2$synthesized in 50$0^{\circ}C$ or $700^{\circ}C$. Removal rate and surface uniformity of$SiO_2$and$Si_3N_4$wafer and selectivity are influenced by synthetic condition of abrasive, suspension stability and pH of slurries.
