Modification and Validation of Analytical Method for Oxypaeoniflorin and Paeoniflorin in Moutan Cortex Radicis Extract (목단피 추출물의 Oxypaeoniflorin 및 Paeoniflorin의 분석법 개선 및 검증)

  • The aim of this study was the validation of a modified analytical method for determination of oxypaeoniflorin and paeoniflorin in Moutan Cortex Radicis extract. For validation of the analytical method, we modified established analytical methods and validated improvement. For validation, the specificity, linearity, precision, accuracy, limit of detection (LOD), and limit of quantification of oxypaeoniflorin and paeoniflorin were measured by high performance liquid chromatography. The results show that the correlation coefficients of the calibration curve for oxypaeoniflorin and paeoniflorin were 1.0000 and 0.9998, respectively. The LOD for oxypaeoniflorin and paeoniflorin were $0.23{\mu}g/mL$ and $0.25{\mu}g/mL$, respectively. The inter-day and intra-day precision values of oxypaeoniflorin and paeoniflorin were 0.70~3.19% and 1.74~2.43%, and 0.32~0.92% and 0.62~2.28%, respectively. The inter-day and intra-day accuracies of oxypaeoniflorin and paeoniflorin were 98.33~102.11% and 97.72~118.12%, and 98.44~101.56% and 97.10~112.00%, respectively. Therefore, the analytical method was validated for the detection of oxypaeoniflorin and paeoniflorin in Moutan Cortex Radicis.

  • From the 60% aqueous methanolic fraction of Moutan Cortex Radicis two hyaluronidase inhibitors were isolated and their structures were elucidated by spectroscopic methods. Their structures were identified as paeoniflorin (com-pound I) and oxypaeoniflorin (compound II). Compound I and II exhibited hyaluronidase inhibitory activities with $IC_{50}$ of 1.71 and 1.73mM, respectively.

Anti-inflammatory and Anti-Oxidant Effects of Oxypaeoniflorin, Paeoniflorin and Paeonia lactiflora cv. 'Red Charm' Flower Petal Extracts in Macrophage Cells

  • The root extracts of Paeonia lactiflora cv. 'Red Charm' has been studied by many groups, however, little attention has been paid to its flower petal. Paeonia is the genus in the Paeoniaceae family. 'Red Charm' Paeonia is a soft-stemmed herbaceous peony hybrid of P. officinalis and P. lactiflora. We previously showed the flower petal extract of Red Charm might have anti-oxidant and anti-inflammatory activities, however, it was not clear which components might be involved in this activity. Bioinformatics analysis previously indicated these extracts have potential anti-oxidant materials. One of them is identified as paeoniflorin, which is major component in root extract of Red Charm. In this study, we compared paeoniflorin and oxypaeoniflorin using DPPH assays to measure its anti-oxidant activities. Oxypaeoniflorin showed higher levels of radical scavenging activity, similar to ascorbic acid control, whereas paeoniflorin did not. Furthermore, nitric oxide assay showed they have similar anti-inflammatory effects. Taken together, these results suggest oxypaeoniflorin may play a more important role in the anti-oxidant activity of the flower petal and root extracts of Red Charm, compared to paeoniflorin. Further studies may be able to provide a platform to develop potential dual effects therapeutics for oxidant-mediated and inflammation-mediated disease in the near future.

Analysis of the Content of Paeoniflorin in Peony Roots Cultivated on Kyeongbuk Area (경북지역(慶北地域)에서 재배중(栽培中)인 작약(芍藥)의 Paeoniflorin 함량분석(含量分析))

  • The effective components of the peony cultivar, Euseongjagyag and Youngcheonjagyag, which were the major cultivars of peony in Kyeongbuk area, were determined with HPLC and TLC. The paeoniflorin content in the root of Euseongjagyag were more than that of Youngcheonjagyag. The root of Euseongjagyag contained much albiflorin and Youngcheonjagyag contained much oxypaeoniflorin in comparision with albiflorin and oxypaeoniflorin contents in both cultivars of peony. Paeoniflorin contents in accordance with peony prodution regions were ranged from 2.15% to 4.08%, and paeoniflorin content of local cultivar of Euseong and that of Geochang were approximatly the same but that of Youngcheon was the lowest. Paeoniflorin content in the 18 accessions of peony cultivar which were collected from Kyeongbuk area and harvested on November 1993, were ranged from 1.41% to 5.30%. The 18 accessions of peony were classified with the HPLC chromatogram pattern of peony root extract into the three groups which composed with Euseong peony group(9 accessions), Youngcheon peony group (4 accessions) and Punggi standard peony group(5 accessions). High content of paeoniflorin WaS contained in peony root harvested in May and November. but low content of paeoniflorin was contained in peony root harvested in March and September.

  • Antibacterial activity-guided fractionation of the $CHCl_3-MeOH$ (1:1) extract of Paeonia suffruticosa root bark furnished three monoterpene glycosides, 6-O-vanillyoxypaeoniflorin (1), mudanpioside-H (2), and galloyl-oxypaeoniflorin (3). Of the isolated compounds, compound 1 is a new compound. All isolated compounds showed broad, but moderate, antibacterial activity with minimum inhibitory concentration (MIC) values in the range of 100 to $500\;{\mu}g/mL$ against eighteen pathogenic microorganisms of concern for public health or zoonosis.

Protective Constituents Against Sepsis in Mice from the Root Cortex of Paeonia suffruticosa

  • The bioassay-guided fractionation of protective agents against sepsis-induced lethality from the root cortex of Paeonia suffruticosa ANDREWS (Ranunculaceae) led to the isolation of eight known compounds: paeonol (1), 2,5-dihydroxy-4-methoxyacetophenone (2), acetovanillone (3), paeonoside (4), paeoniflorin (5), oxypaeoniflorin (6), apiopaeonoside (7), and methyl 3-hydroxy-4-methoxybenzoate (8). Among them, 3 showed the highest survival rate (100% with a dose of 30 mg/kg versus 17% for the control experiment) and reduced alanine aminotransferase level to be a half of the control value on the sepsis model induced by lipopolysaccharide/D-galactosamine.

  • From the polar fractions of 70% EtOH extract of Paeonia lactiflora roots (Paeoniaceae), ten monoterpene glucosides were isolated and identified as lactiflorin (1), benzoylpaeoniflorin (2), mudanpioside C (3), $1-O-{\beta}-D-glucosylpaeonisuffrone$ (4), paeonidanin (5), $1-O-{\beta}-D-glucosyl-8-O-benzoylpaeonisuffrone$ (6), paeoniflorin (7), albiflorin (8), oxypaeoniflorin (9) and mudanpioside E (10) by spectroscopic methods. Among these glucosides, 3-6 and 10 were isolated for the first time from this plant.

Among them, 4 exhibited the highest survival rate in a dose-dependent manner (100% with a dose of 30 mg/kg versus 16.7% for the control experiment) and showed reduction of plasma alanine aminotransferase (ALT) value on the in vivo assay model of sepsis induced by LPS/GalN.

Effect of Fertilizer Applications on the Morphology and the Pharmaceutical Components of Paeonia albiflora Palls (시비관리(施肥管理)에 따른 작약(芍藥)(Paeonia albiflora Palls) 생육특성(生育特性)과 유효성분(有效成分) 연구(硏究))

  • This study was intended to find out the effect of the fertilizer applications on the changes of the morphology, yield and effective components -Paeonol, Benzoic acid, Oxypaeoniflorin, Albiflorin and Paeoniflorin- of Paeonia albiflora Palls. The treatments of the field experiment were designed as follows ; conventional fertilization(R), organic fertilization(OF), chemcial fertiliazation(CF), combination of organic and chemical fertilization(OCF), and 50% and 100% increase of the chemcial fertilizers in combination treatments (OCF-50, OCF-100). The results obtained were summarized as follows ; 1. Depending on the treatments, the changes of the growth characteristics of the aboveground part were not significant, but the yield of the root had a significance on OCF-100. 2. The large quantities of effective components in Paeonia albiflora Palls were increased from the first year to the second year. 3. The excess phosphate application in the conventional fertilization affected to decrease the root yield and the effective components in its root. 4. The organic fertilizer was more favorable to the yield and the contents of the effective components of Paeonia albiflora Palls than the chemical fertilizer. OCF-50 and OCF-100 had higher values of yield and the amounts of the effective components than the treatments of the organic fertilizers.

Quantification of the 25 Components in Onkyung-Tang by Ultra Performance Liquid Chromatography-Electrospray Ionization-Tandem Mass Spectrometry (UPLC-ESI-MS/MS를 이용한 온경탕 중 25종 성분의 함량분석)

  • In this study, an ultra-performance liquid chromatography-electrospray ionization-mass spectrometry (UPLC-ESI-MS/MS) method was established for simultaneous determination of the 25 marker components, including chlorogenic acid, gallic acid, oxypaeoniflorin, homogentisic acid, methyl gallate, caffeic acid, 3,4-dihydroxybenzaldehyde, paeoniflorin, albiflorin, liquiritin, nodakenin, ferulic acid, ginsenoside Rg1, liquiritigenin, coumarin, cinnamic acid, benzoylpaeoniflorin, ginsenoside Rb1, cinnamaldehyde, paeonol, glycyrrhizin, 6-gingerol, evodiamine, rutecarpine, and spicatoside A in traditional Korean formula, Onkyung-tang. All analytes were separated on a Waters Acquity UPLC BEH $C_{18}$ analytical column ($2.1{\times}100mm$, $1.7{\mu}m$) at $45^{\circ}C$ using a mobile phase of 0.1% (v/v) formic acid in water and acetonitrile with gradient elution. The MS analysis was carried out using a Waters ACQUITY TQD LC-MS/MS coupled with an electrospray ionization (ESI) source in the positive and negative modes. The flow rate and injection volume were 0.3 mL/min and $2.0{\mu}L$, respectively. The correlation coefficient of all analytes in the test ranges was greater than 0.98. The limits of detection and quantification values of the 25 marker compounds were in the ranges 0.03-19.43 and 0.09-58.29 ng/mL, respectively. As a result, methyl gallate, 3,4-dihydroxybenzaldehyde, evodiamine, and rutecarpine were not detected in this sample and the concentrations of the 21 compounds except for the above 4 compounds were $33.09-3,496.32{\mu}g/g$ in Onkyung-tang decoction. Among these compounds, paeonol was detected at the highest amount as a $3,496.32{\mu}g/g$.
