Groundwater Contamination by Cation, Anion and Pesticides (지하수중 음이온, 양이온, 및 금속의 함량)

  • According to the increase of population and industrialization, the quality of our drinking water are becoming worse by the contamination of resources, production of THM and other halogenated hydrocarbons during the purifying process, the problem of corroded water supplying pipeline, and the water reservoir tanks, Many people choose groundwater to drink instead of city tap water, but sometimes we get report about groundwater contamination by wastes, swage, septic tank, etc. It is reported that in U. S. over 20% of population are drinking groundwater, but U. S EPA reported the groundwater contamination by pesticides, herbicides, fungicides, fertilizer, and various chemical substances. Craun, et at announced the groundwater contamination by bacteria which are related with poor installation of septic tank. Johnson and Kross mentioned aboutmethemoglobinemia by NO3-N originated from human and animal feces, organic chemicals, and fertilizer, and as the results the infant mortality could be risen. Some scientist also reported the high concentration of metals in groundwaters and some cation and anions, and volatile organic compou nds. Authors investigated 80 groundwaters in urban, agricultural, and industrial area during last 3 month(June - August) to check any drinking water quality parameters are exceeding the standards. The results were as follow. 1, The average value of ammonia nitrate were within the standard, but 11.76% of urban area were exceeded the 10 rpm standard, in agricultural area 42.3175 were exceeded, and in industrial area 20.2% were exceeded the drinking water standard of 10 ppm. the highest concentration was 29.37 ppd in industrial area. 2. The mean value of metals is not exceeded the standard, but there were some groundwater whose Mn value was 0.424 ppm(standard is 0,3 ppm) in urban area, 0.737 rpm in agricultural area, and 5.188 ppm in industrial area. The highest Zn value was 1.221 ppm (standard is 1.0 ppm)was found in industrial area. 3. The percentage of contamination by general bacteria was 8.82% in urban area, 15.38% in agricultural area, and 15.00% in industrial area. Escherichia coil group was also contaminated by 35.29% in urban area, 30.76% in agricultural area, and 30.00% in industrial area. 4, The pH value was within the standard which means there was no influence by acid or alkali chemicals, nor acid rain Through the above results, all the groundwater should be tested to check the safety for drinking water and should make some alternative methods suitable for drink.

Anti-oxidant and Anti-inflammatory Effects of Ethanol Extracts from Aerial Part of Coriandrum sativum L. (고수(Coriandrum sativum L.) 지상부 추출물의 항산화 및 항염증 활성 효과)

  • Coriandrum sativum L., an annual herbaceous plant of Apiaceae family. The present study evaluated the anti-oxidant activities and anti-inflammatory effects of ethanol extracts of C. sativum. The anti-oxidant activities of C. sativum were measured by total contents of polyphenol, flavonoid, DPPH and ABTS radical scavenging and reducing power activity. And anti-inflammatory effects of C. sativum were measured by LPS-induced RAW 264.7 cells. The results showed that the contents of total polyphenol and flavonoid were 76.03 ± 1.36 mg of gallic acid equivalents/g and 182.23 ± 4.32 mg of rutin equivalents/g at concentration 1 mg/mL of C. sativum. The DPPH radical scavenging activity was found to be 52.8% at 500 ㎍/mL. The ABTS radical scavenging activity was shown in 58.3% after exposure to 1,000 ㎍/mL. Reducing power activity was found to be 66.8% at 2,000 ㎍/mL. The inhibitory effect of NO production was found to be 65% concentration 500 ㎍/mL. In the generation quantity of inflammatory cytokines such as TNF-α and IL-1β in cell culture medium, the expression levels of inflammatory proteins in cells were showed decrease with the increase of concentration. Therefore, we suggest that the C. sativum should be a potential source of alternative anti-inflammatory drug with good anti-inflammatory effects.

Carbon Metabolism and Its Global Regulation in Corynebacterium glutamicum (Corynebacterium glutamicum의 탄소대사 및 총체적 탄소대사 조절)

  • In this review, the current knowledge of the carbon metabolism and global carbon regulation in Corynebacterium glutamicum are summarized. C. gluamicum has phosphotransferase system (PTS) for the utilization of sucrose, glucose, and fructose. C. glutamicum does not show any preference for glucose when various sugars or organic acids are present with glucose, and thus cometabolizes glucose with other sugars or organic acids. The molecular mechanism of global carbon regulation such as carbon catabolite repression (CCR) in C. glutamicum is quite different to that in Gram-negative or low-GC Gram-positive bacteria. GlxR (glyoxylate bypass regulator) in C. glutamicum is the cyclic AMP receptor protein (CRP) homologue of E. coli. GlxR has been reported to regulate genes involved in not only glyoxylate bypass, but also central carbon metabolism and CCR including glycolysis, gluconeogenesis, and tricarboxylic acid (TCA) cycle. Therefore, GlxR has been suggested as a global transcriptional regulator for the regulation of diverse physiological processes as well as carbon metabolism. Adenylate cyclase of C. glutamicum is a membrane protein belonging to class III adenylate cyclases, thus it could possibly be a sensor for some external signal, thereby modulating cAMP level in response to environmental stimuli. In addition to GlxR, three additional transcriptional regulators like RamB, RamA, and SugR are also involved in regulating the expression of many genes of carbon metabolism. Finally, recent approaches for constructing new pathways for the utilization of new carbon sources, and strategies for enhancing amino acid production through genetic modification of carbon metabolism or regulatory network are described.

Entrainer Enhanced Reactive Distillation for Production of Butyl Acetate: Experimental Investigation in Pilot-Scale (부틸 아세테이트 생산을 위한 외부 공비제 첨가 반응증류: 파일럿 규모 실험을 통한 연구)

  • Butyl acetate is produced from acetic acid and butanol via an esterification reaction in reactive distillation (RD). The product, butyl acetate, has been used as an internal entrainer until now. In this case, butyl acetate and water are removed at the top of column and separated into two different phases (organic and aqueous phases) after condensation, and butyl acetate rich organic phase is refluxed into the RD. This method makes butyl acetate remain high at the reactive zone, leading to lower equilibrium conversion and product yield. We introduced an extraneous entrainer to solve the problem. The extraneous entrainer forms a new azeotrope with water. The proposed process provides lower concentration of butyl acetate in the reactive zone than conventional RD processes using an internal entrainer. We compared the yield and production rate of butyl acetate between the proposed and conventional processes through pilot-scale experiments. Experimental and simulation results showed that the proposed process was more efficient than conventional process using internal entrainer.

Analysis of Antibacterial, Antioxidant, and In Vitro Methane Mitigation Activities of Fermented Scutellaria baicalensis Georgi Extract (발효 황금 뿌리 추출물의 항균, 항산화 효과 및 메탄가스 저감 효과 In Vitro)

  • This study was conducted to investigate the antibacterial, antioxidant, and in vitro greenhouse gas mitigation activities of fermented Scutellaria baicalensis Georgi extract. Seven starter cultures were used, comprising four of lactic acid bacteria and three of Saccharomyces cerevisiae. Ten grams of S. baicalensis Georgi powder was diluted in 90 mL autoclaved MRS broth. Each seed culture was inoculated with 3-10% (v/v) S. baicalensis Georgi MRS broth and incubated at $30^{\circ}C$ for 48 h. Among the starter cultures used, only Lactobacillus plantarum EJ43 could withstand the fermentation conditions. This fermentation broth was dried and extracted with ethanol to assess its antibacterial, antioxidant, and in vitro methane mitigation activities. The extract of S. baicalensis Georgi fermented by L. plantarum EJ43 (SBLp) showed higher antibacterial activity (bigger clear zone) compared to the unfermented S. baicalensis Georgi extract (SB0). SBLp also presented 1.2 folds higher antioxidant activity than SB0. During in vitro rumen fermentation, SBLp showed reduction in methane production compared to SB0 or the control. In conclusion, fermentation by L. plantarum EJ43 may enhance antibacterial and antioxidant activities of S. baicalensis Georgi and decrease enteric methane production.

Electricity Generation and De-contamination Effect for Characteristic Electrode Material in a Microbial Fuel Cell System Using Bay Sediment (MFC의 금속 및 탄소전극에 의한 전기생산 특성과 오염저감 효과)

  • Sediment works as a resource for electric cells. This paper was designed in order to verify how sediment cells work with anodic material such as metal and carbon fiber. As known quite well, sediment under sea, rivers or streams provides a furbished environment for generating electrons via some electron transfer mechanism within specific microbial population or corrosive oxidation on the metal surfaces in the presence of oxygen or water molecules. We experimented with one type of sediment cell using different anodic material so as to attain prolonged, maximum electric power. Iron, Zinc, aluminum, copper, zinc/copper, and graphite felt were tested for anodes. Also, combined type of anodes-metal embedded in the graphite fiber matrix-was experimented for better performances. The results show that the combined type of anodes exhibited sustainable electricity production for ca. 600 h with max. $0.57\;W/m^2$ Al/Graphite. Meanwhile, graphite-only electrodes produced max. $0.11\;W/m^2$ along with quite stationary electric output, and for a zinc electrode, in which the electricity generated was not stable with time, therefore resulting in relatively sharp drop in that after 100 h or so, the maximum power density was $0.64\;W/m^2$. It was observed that the corrosive reaction rates in the metal electrodes might be varied, so that strength and stability in the electric performances(voltage and current density) could be affected by them. In addition to that, COD(chemical oxygen demand) of the sediment of the cell system was reduced by 17.5~36.7% in 600 h, which implied that the organic matter in the sediment would be partially converted into non-COD substances, that is, would suggest a way for decontamination of the aged, anaerobic sediment as well. The pH reduction for all electrodes could be a sign of organic acid production due to complicated chemical changes in the sediment.

Efficiency of methane production from pig manure slurry using anaerobic digestor combined with compost filtration bed (퇴비단 여과상이 부착된 혐기소화조를 이용한 돈분뇨 슬러리 메탄생산 효율분석)

  • The characteristics of methane production from pig manure slurry was investigated using anaerobic digestor combined with compost filtration bed. In this study, raw pig manure slurry was digested in mesophilic rectangular digester (effective volume $250m^3$) for 25 days and anaerobic digestion wastewater was filtered through compost filtration bed, which is composed of double layer, sawdust and chaff. The characteristics of anaerobic digestion wastewater were BOD 1,800 mg/L, COD 3,500 mg/L, SS 11,800 mg/L, T-N 1,200 mg/L and T-P 350 mg/L. After the filtration process, the contents of BOD, COD, SS, T-N and T-P of the anaerobic digestion wastewater were reduced by 97%, 62%, 89%, 39% and 57%, respectively. The concentrations of N, $P_2O_5$, and $K_2O$ of the leachate were 1,024, 111 and 407 mg/L, respectively. However, there was no odor emitted from the leachate.

Effects of Dried Leftover Food and Green Tea By-Product on Performance and Egg Quality in Laying Hens (남은 음식물과 녹차 부산물이 산란계의 산란성적과 계란품질에 미치는 영향)

  • This study was designed to determinate the effects of dried leftover food and green tea by-product on laying hens performance and egg quality in hens. A total of 210 "Tetran Brown" layers 50-weeks of age were assigned to 7 treatments in a completely randomized design. Each treatment had five replicates per treatment with six layers per replication. Seven dietary treatments were a control diet (formula diet) and dried leftover food (DLF) mixed in 10, 20, 30 and 40% to the control diet substituting the corn grain and soybean meal, control diet containing 1.0% GTB without DLF supplementation and control diet containing 30% DLF plus 1.0% GTB supplementation. The trial period was for 8 weeks. The egg production rate of layers was significantly increased in 10, 20 and 40% DLF treatments compared to that of the control treatment (P<0.05). The egg weight was significantly decreased in 10% DLF treatment compared to that of the control (P<0.05). The feed intake of layers was higher in 20% DLF and 30% DLF plus 1.0% GTB treatment than that of the control (P<0.05). The feed conversion ratio significantly decreased in 10% DLF and control plus 1.0% GTB treatments compared to that of the control (P<0.05). The egg yolk cholesterol not varied among the DLF and control treatments (P>0.05). However, the linolenic acid content of egg yolk was significantly increased in DLF and control treatments both containing 1.0% GTB supplementation.

Comparison of Single-stage Thermophilic and Mesophilic Anaerobic Sewage Sludge Digestion (단상 고온 및 중온 혐기성 하수 슬러지의 소화 공정 비교)

  • In this study, single-stage continuous anaerobic reactors to treat sewage sludge were operated under different temperature (55 and $35^{\circ}C$; $R_{TAD}$ and $R_{MAD}$) to evaluate the reactor stability and performance of the thermophilic and mesophilic anaerobic digestion. During the overall digestion, both anaerobic reactors maintained quite stable and constant pH and total alkalinity (TA) values in the range of 6.5-8.0 and 3-4 g $CaCO_3/L$, respectively. After the start-up period, $R_{TAD}$ showed 10% higher VS removal efficiency than that of $R_{MAD}$ ($R_{TAD}$; 43.3%; $R_{MAD}$ : 33.6%). Although organic acids such as acetic and propionic acid were detected in both anaerobic reactors at the start-up period, all organic acids in $R_{TAD}$ and $R_{MAD}$ were consumed at the steady state condition. Also $R_{TAD}$ showed 31.4 % higher methane production rate (MPR) than that of $R_{MAD}$ at the steady state condition ($R_{TAD}$; 243 mL $CH_4/L/d$; $R_{MAD}$ : 185 mL $CH_4/L/d$). Meanwhile, the experimental results indicated similar methane yield between $R_{TAD}$ and $R_{MAD}$.

Mass Cultivation and Characterization of Multifunctional Bacillus velezensis GH1-13 (복합기능성 Bacillus velezensis GH1-13 균주의 대량배양 최적화 및 특성)

  • Bacillus genus are found abundantly in various sites and their secondary metabolites were used as potential agents in agriculture, notably plant growth promoting and bio-control. The objective of this study was to develop the culture conditions of GH1-13 strain including higher cell growth, stable endospore-forming and enhancement of potential agents which are related with plant growth promoting and phytopathogen suppression. The optimal carbon and nitrogen sources were determined by glucose and soy bean flour, respectively, then resulted in $7.5{\times}10^9cells/mL$, $6.8{\times}10^9\;endospore\;cells/mL$ and sporulation yield of 90% after 30 h cultivation in 500 L submerged fermenter at $37^{\circ}C$, pH 7.0. Cells and cell-free supernatant of GH1-13 strains showed the potent antifungal activity against phytopathogenic fungi of Colletotrichum gloeosporioides. It was also confirmed that indole-3-acetic acid (IAA) production of GH1-13 strain was greatly increased by addition of 0.3% tryptophan.
