Callus Induction and Differentiation from Rice (Oryza Sativa L.) Anthers (벼 약(葯)으로부터 callus 형성(形成)과 분화(分化)에 대(對)하여)

  • This study was conducted to obtain basic information on the rice anther culture. Materials used were (Inabawase X YR 2404-14-2-1) $F_1$ hybrid. Callus growth rate on various media, induction frequency of callus in spikelet and panicle, and the effect of treatment on anther and callus were evaluated. The results obtained were summarized as follows ; The growth rate of callus on N-6, M-S, P.E.agar media was 19.8, 13.1, 4.1 times respectively after 30 days inoculated, and on liquid media was 3.8, 5.1, 1.4 times, respectively. Organ differentiation on N-6, M-S, P.E.agar media was 37.5%, 12.5%, 17.5% respectively. The difference of induction frequency of callus per panicle was 0.14%-6.25% and per spikelet was 0-19.05%. Almost callus was induced 30-35 days after inoculation. Organ differentiation of induced callus was decreased by culture. Callus cultured for 13 days after induction did not make shoot. Anthers cold shocked at $8^{\circ}C$ for 5 days obtained 3.32% efficiencys of callus induction per number of anthers plated, and compared with 2.41% of no treated anthers. But anther treated at $8^{\circ}C$ for 7 days decreased 2.24%. Callus induction periods were shortened by cold treatment for about 5 days. Callus cultured on medium containing 2 mg/l of 2, 4-D showed 5% on root formation but medium containing 5 mg/l of 2, 4-D showed 30% of root formation after transfered on the medium without 2, 4-D. Callus cold shocked at $15-18^{\circ}C$ revealed poor efficiency for root formation, but 5 days treatment was good for shoot formation.

Effects of Feeding Softwood By-products or Extract on Growth Performance, Intestinal Organs and Cecal Microbes in Organic Broiler Chicks (유기 육계에 침엽수 부산물과 추출물의 급여가 성장, 장기 및 맹장내 미생물에 미치는 영향)

  • This study was conducted to evaluate the effects of dietary softwood by-products extract on growth performance, organ weight and intestinal microbes in organic broiler chicks. Five treatments were assigned to 200 broiler chicks with four replication, allocating 10 broiler chicks in each replication. Dietary treatments included control(Organic diet), +C(Organic diet + antibiotics), T1(Organic diet + softwood by-product powder 3.0%), T2(Organic diet + softwood by-product extract 0.05%) and T3(Organic diet + softwood by-product extract 0.1%). Body weight gain was higher in +C and T3 for overall period than control and T1. Feed efficiency of were slightly improved in +C and T3 groups compared with that of control and T1. The small intestine weight tended to be higher in control and +C, but it's length showed high in only T1 group. Gizzard weight was significantly higher in T2 compared with other treatments(P<0.05). In contrast, spleen weight was significantly lower in control group(P<0.05), whereas the weight of cecum and liver were not significantly different by dietary treatment. Intestinal microflora was significantly lower or/and seemed to be low in softwood by-products treatment groups than control. Digestibility of energy and protein were statistically lower in control and T1 than other treatments(P<0.05). The IgG concentration of birds fed antibiotics, softwood by-product extract was higher than control and T1, but was no significance. The results of this experiments indicated that softwood by-product extract would be possible to apply for the feed additive in organic broiler diets.

Effects of Scutellariae Barbatae Herba·Alli bulbus·Oldenlandiae Herba Complex Herbal Acupuncture on Tumor and Immune Response (반지련(半枝蓮)·대산(大蒜)·백화사설초(白花蛇舌草) 복합약침(複合藥鍼)이 종양(腫瘍) 및 면역반응(免疫反應)에 미치는 영향)

  • Objective : We need to develop a new treatment method which can curve cancer growth and enhance immunity of patients with various kinds of cancer more safely and effectively, for conventional anticancer treatment has lots of problems to be overcomed, in other words, Its efficacy can be recognizible but it doesn't actually give aid to patients due to its side effects. This study was taken up to evaluate the anticancer and immune-enhancing effect of Scutellariae Barbatae Herba, Alli bulbus, Oldenlandiae Herba(SAO) Herbal acupuncture. Methods : SAO Herbal acupuncture solution was made from Scutellariae Barbatae Herba, Alli bulbus, Oldenlandiae Herba by decoction. Experimental group was divided into normal(N), control(TC, cancer group induced by S 180), high and low concentration SAO complex Herbal acupuncture group. In the high and low concentration SAO complex Herbal acupuncture group, SAO Herbal acupuncture solution was injected, on the left and right Chok-samni(足三里, ST36) of ICR-male S 180 rats alternatively, by 200mg/kg and 100mg/kg respectively. In vitro, S 180 was cultured with $200{\mu}g$ and $500{\mu}g$ of SAO Herbal acupuncture solution. In each experimental group, we examined the effect of SAO complex Herbal acupuncture on body weight, antitumor, organ weight, activity of macrophage, activity of B cell, spleen cell division, IL-2 production and population of lymphocytes. Results : 1. In Body weight, no significant change was shown, but In solid cancer weight, the high concentration SAO complex Herbal acupuncture group showed signigicant(P<0.05) decrease and significant(P<0.05) increase in the weight of kidney, compared with control group. 2. In activity of macrophage, low concentration SAO complex Herbal acupuncture group showed significant(P<0.01) increase, but in vitro, there was no significant increase, compared with control group. 3. In activity of B cell, high and low concentration SAO complex Herbal acupuncture group showed no significant decrease, but in vitro, low concentration SAO complex Herbal acupuncture group showed significant(P<0.01) increase, compared with control group. 4. In spleen cell division, high and low concentration SAO complex Herbal acupuncture group had no significant influence on spleen cell division induced by Co A, meanwhile, it was found that macrophge promote spleen cell division in low concentration SAO complex Herbal acupuncture group(P<0.05), compared with control group. 5. In IL-2 production, high concentration SAO complex Herbal acupuncture group showed significant((P<0.05) increase, compared with control group. 6. In population of lymphocytes, high concentration SAO complex Herbal acupuncture group showed significant increase of CD3+(P<0.05), CD4+(P<0.05), CD3+ and CD4+ T cell(P<0.01) and B cell(P<0.05), while low concentration SAO complex Herbal acupuncture group showed significant increase of CD4+(P<0.05), CD8+ T cell(P<0.05) and B cell(P<0.01), compared with control group. Conclusion : SAO Herbal acupuncture inhibited cancer growth and enhanced immunity.

Classification of Polygonatum spp. Collections Based on Multivariate Analysis (다변량 분석에 의한 둥굴레속 식물의 분류)

  • This study was conducted to obtain the basic data for practical use of the Polygonatum genetic resources. The 20 collections were analyzed by principal component analysis of 8 characters and cluster analysis. In the principal analysis, the first, the second and the third components contributed 54.10%, 18.95% and 11.62% of the variations, respectively. The cumulative contribution from the first to the third principal components was 84.68%. The first principal component was related to shape and size of plant, and assimilatory, reserve and reproductive organs. The second principal component was related to growth and development of plant, and reserve organ. And the third principal component was related to growth and development of plant. Based on cluster analysis, the 20 collections were classified into 4 distinct groups with the average distance greater than 0.7 between groups. Group I was Polygonatum sibiricum $D_{ELAR}$ and Group II included P. odoratum var. pluriflorum $O_{HWI}$, P. odoratum var. pluriflorum $O_{HWI}$ for 'Variegatum' Y. Lee, for. nov., P. odoratum var. thunbergii $H_{ARA}$ and P. odoratum var. maximowiczii $K_{OIDZ}$. GroupIII was P. involucratum $M_{AXIM}$, P. desoulavyi $K_{OMAROV}$ and P. humile $F_{ISHER}$ ex. $M_{AXIM}$. And GroupIV included P. lasianthum var. coreanum $N_{AKAI}$ and P. inflatum $K_{OMAROV}$.

Effects of Dietary Probiotic Mixture on Growth Performance, Caecal Microorganism and Immune Response in Broiler Chickens under Heat Stress (혼합 생균제가 열 스트레스에 노출된 브로일러의 면역반응, 맹장 미생물과 성장능력에 미치는 효과)

  • This study was investigated the effect of dietary probiotic mixture on blood biomarkers, immune responses, caecal microorganism and productivity in broiler chickens under heat stress (HS). A total of 400 broiler chick males (Ross 308) were divided into four groups of 100 heads each, group C (control, room temperature $25^{\circ}C$), HS (heat stress $33^{\circ}C$) and HSP (HS plus probiotic mixture 500, 750 mg/kg of diet), respectively. Broiler body weight gain, feed intake, feed conversion ratio and immune organ weight increased in the HSP group compared to the HS group. The concentrations of blood IgG and lymphocyte were increased in the HSP group compared to the HS group, and the heterophil:lymphocyte (H:L) ratio, corticosterone concentration and mortality were low. Lactobacillus in the cecum increased in the HSP group compared to the HS group, but was lower in Escherichiacoli (E. coli), coliform bacteria and aerobic bacteria. These results indicate that feeding probitic mixture including three strains such as B. subtilis, S. galilaeus and Sphingobacteriaceae to broiler exposed to heat stress can improve production with lowering mortality by improving immune response and microbial balance.

Effects of Brewer's Yeast Slurry on the Growth and Blood Composition of Broiler Chicks (맥주효모 슬러리의 급여가 육계의 성장과 혈액 성상에 미치는 영향)

  • This study was conducted for a period for 5 weeks in order to evaluate effects of brewer's yeast slurry(BYS) on the growth performance, organ development and blood composition of broiler chicks. A total number of 84 one day old broiler chicks were allocated to one of three supplemented diets: 0%(control), 2% and 4% BYS. Body weight gain and feed efficiency of chicks fed the diet supplemented with 4% BYS significantly was increased by 9% and 8%, respectively, compared to the control diet(P<0.05). The BYS supplements did not affect weights of liver, heart and abdominal fat, and their ratios per body weight. However, in 4% BYS supplement group, ratio of liver to body weight significantly decreased(P<0.05) by 12%, compared to the control. The supplement of 4% BYS decreased the concentration of triglyceride by 31%, and increased the concentration of high density lipoprotein(HDL) by 26%, compared to the control(P<0.05). These results indicated that the supplementation of 4% BYS in diet can improve body weight gain as well as feed efficiency, and increased the concentration of blood HDL, and ratio of HDL to total-cholesterol of broiler chicks.

  • Development of modern agricultural machinery and accompanying agricultural development cause soil compaction and reduce growth by stressing roots. Kalanchoe pinnata was used to investigate the impact of stress on rooting and changes in plant growth and reproduction. K. pinnata forms somatic embryos capable of asexual reproduction at the edge of leaves. Impact of root pressurization of K. pinnata on somatic embryogenesis and organ differentiation according to external stress factors was investigated by using a high concentration of agar and this phenomenon was studied histologically. Agar concentration in culture media ranged from 0.5%-1.5% to induce a compression effect on roots. The stem and leaf of K. pinnata were subjected to a microtechnique process to study changes in tissue. In vivo, K. pinnata produced 2nd and 3rd plantlets at edges of leaves from lack of water and excessive lighting conditions. In in vitro culture studies, the lower the concentration of agar, the higher the population and the higher the biomass, but plantlet did not occur in leaf bends. Conversely, as concentration of agar increased, increase in the number of individuals was low. Plantlet development occurred only in agar 1.5% medium. The difference in agar concentration was a stressor in the root of K. pinnata, and thus the pattern of asexual reproduction changed from the division method in root to a plantlet generation in leaf. This suggests root pressurization may act as stress and change in the plant reproduction pattern.

Flower bud development and the influence of sex expression of summer pruning in Schizandra chinensis Baillon (오미자의 화아(花芽) 발달과 성 발현에 미치는 하계전정의 영향)

  • The purpose of this experiment is to improve the ratio of female flower in Schizandra chinensis Baillon. The experiment was carried out in two purposes. One was to understand the development stages on bud formation and determining sex after bud formation by stage. The other was to investigate characteristics of flowers forming on spring when summer pruning was taken place in different cutting part and timing in S. chinensis Baillon. The outcome was that on 7th of June the flower bud was found next to leaf primordium in branch of this year and no changes were found in shape for two weeks by 26th July. On 26th of June, an ovule that forms organ was observed and then, an ovule was found after sex was determined on 6th of July. Second experiment was carried out to find out when the sex was determined by adapting different pruning methods; cutting the branch at 6th node on June 1, June 15, and July 1; cutting the branch at 10th node on June 15, and July 1. The result was that the re-growth ratio on the branch 6th node was cut on June 1 and June 15. July 1 was 34-56% and, however, the number of setting flowers and mail flowers were fewer than the normal branches. More over, even though the re-growth ratio of branches were 34-25% when the cutting was taken place at 10th node on June 15 and July 1, the number of setting flowers and mail flowers were the same as the non treated branch. As a result, it is assumed that cutting controlling in this plant to increase the number of female flowers has no effect.

Effect of Long-day and Night-break Treatment on Growth and Anthesis of Orostachys japonicus A. Berger (장일과 암기중단 처리가 바위솔의 생장과 개화에 미치는 영향)

  • Orostachys japonicus, called Wasong and used as anti-tumor medicinal plant, was cultivated in plastic house. The experiment was done to clarify the effect of long-day and night-break treatment at the timing of bolting on its morphological characters, organ dry weight and flowering of florets. After grown in 15cm plastic boxes containing 2:1 soil:peat moss mixture for about 4 months, long-day of 16 hours and night-break of 2 hours around midnight were treated from Sept. 9. The plants were sampled 5 times at 2-week interval after the treatments. Long-day and night-break treatment delayed the growth of inflorescence and showed greater stem diameter on the last sampling and no. of leaves and bracts than the natural daylength. The treatments also had greater leaf and bract dry weight since 2 weeks, and the other fraction and total dry weights since 4 weeks but less floret dry weight from 4 to 6 weeks after the treatments than the natural daylength. The treatments, however, decreased no. of flowered florets and ratio of flowering plants although all the treatments showed nearly the same no. of total florets per plant until 6 weeks after the treatments, late October, which resulted in the modification of source to sink or vice versa. In the natural daylength, the florets were functioned as sink, while root, leaf and bract as source, but in the long-day and night-break treatments stem and florets were done as sink.
