A Point Mutation at the C-Terminal Half of the Repressor of Temperate Mycobacteriophage L1 Affects Its Binding to the Operator DNA

  • The wild-type repressor CI of temperate mycobacteriophage L1 and the temperature-sensitive (ts) repressor CIts391 of a mutant L1 phage, L1cIts391, have been separately overexpressed in E. coli. Both these repressors were observed to specifically bind with the same cognate operator DNA. The operator-binding activity of CIts391 was shown to differ significantly than that of the CI at 32 to $42^{\circ}C$. While 40-95% operator-binding activity was shown to be retained at 35 to $42^{\circ}C$ in CI, more than 75% operator-binding activity was lost in CIts391 at 35 to $38^{\circ}C$, although the latter showed only 10% less binding compared to that of the former at $32^{\circ}C$. The CIts391 showed almost no binding at $42^{\circ}C$. An in vivo study showed that the CI repressor inhibited the growth of a clear plaque former mutant of the L1 phage more strongly than that of the CIts391 repressor at both 32 and $42^{\circ}C$. The half-life of the CIts391-operator complex was found to be about 8 times less than that of the CI-operator complex at $32^{\circ}C$. Interestingly, the repressor-operator complexes preformed at $0^{\circ}C$ have shown varying degrees of resistance to dissociation at the temperatures which inhibit the formation of these complexes are inhibited. The CI repressor, but not that of CIts391, regains most of the DNA-binding activity on cooling to $32^{\circ}C$ after preincubation at 42 to $52^{\circ}C$. All these data suggest that the 131st proline residue at the C-terminal half of CI, which changed to leucine in the CIts391, plays a crucial role in binding the L1 repressor to the cognate operator DNA, although the helix-turn-helix DNA-binding motif of the L1 repressor is located at its N-terminal end.

Eigenfunctions for Liouville Operators, Classical Collision Operators, and Collision Bracket Integrals in Kinetic Theory Made Amenable to Computer Simulations

  • In the kinetic theory of dense fluids the many-particle collision bracket integral is given in terms of a classical collision operator defined in the phase space. To find an algorithm to compute the collision bracket integrals, we revisit the eigenvalue problem of the Liouville operator and re-examine the method previously reported [Chem. Phys. 1977, 20, 93]. Then we apply the notion and concept of the eigenfunctions of the Liouville operator and knowledge acquired in the study of the eigenfunctions to cast collision bracket integrals into more convenient and suitable forms for numerical simulations. One of the alternative forms is given in the form of time correlation function. This form, on a further manipulation, assumes a form reminiscent of the Chapman- Enskog collision bracket integrals, but for dense gases and liquids as well as solids. In the dilute gas limit it would give rise precisely to the Chapman-Enskog collision bracket integrals for two-particle collision. The alternative forms obtained are more readily amenable to numerical simulation methods than the collision bracket integrals expressed in terms of a classical collision operator, which requires solution of classical Lippmann-Schwinger integral equations. This way, the aforementioned kinetic theory of dense fluids is made fully accessible by numerical computation/simulation methods, and the transport coefficients thereof are made computationally as accessible as those in the linear response theory.

노이즈에 둔감한 밴드패스 이산 코사인 초점 값 연산자 (Mid frequency - DCT focus measure operator for the robust autofocus)

  • 본 논문에서는 노이즈에 둔감한 4*4 밴드패스 이산 코사인 (MF-DCT) 초점 값 연산자를 제안하였다. 제안된 연산자는 DCT 결과 중 중간 주파수 성분을 사용하는 8*8 MDCT 연산자를 노이즈에 둔감하도록 4 형태로 개선한 것으로써 연산자를 180도 회전하여도 같은 구조를 같도록 하였다. 이 연산자는 샘플링 주파수의 절반 부분지 주파수를 통과시키는 밴드패스 필터와 같이 동작하여 초점 정보를 가지고 있지 않는 저주파 신호와 노이즈에 의해 많은 영향을 받는 고주파 신호를 억제함으로써 노이즈에 둔감한 특성을 가진다. 또한 밴드패스 필터를 통과한 성분의 에너지를 초점 값으로 사용함으로써 초점 합의 선형성을 보장받게 된다. 실험 결과에서는 MF-DCT 연산자의 가우시안 노이즈 및 임펄시브 노이즈 특성을 살펴보기 위해 기존의 초점 값 연산자들과의 Autofocusing Uncertain Measure (AUM)비교를 통해 우수성을 검증하였다.

Coiflet Wavelet과 LoG 연산자를 이용한 자연이미지에서의 텍스트 검출 알고리즘 (Text Extraction Algorithm in Natural Image using LoG Operator and Coiflet Wavelet)

  • This paper is to be pre-processing that decides the text recognizability and quality contained in natural image. Differentiated with the existing studies, In this paper, it suggests the application of partially unified color models, Coiflet Wavelet and text extraction algorithm that uses the closed curve edge features of LoG (laplacian of gaussian)operator. The text image included in natural image such as signboard has the same hue, saturation and value, and there is a certain thickness as for their feature. Each color element is restructured into closed area by LoG operator, the 2nd differential operator. The text area is contracted by Hough Transform, logical AND-OR operator of each color model and Minimum-Distance classifier. This paper targets natural image into which text area is added regardless of the size and resolution of the image, and it is confirmed to have more excellent performance than other algorithms with many restrictions.

Importance of Nucleotides Adjacent to the Core Region of Diphtheria tox Promoter/Operator

  • Diphtheria toxin repressor (DtxR) binds to approximately 30 to 35-bp regions containing an interrupted 9-bp inverted repeat within a 19-bp core sequence. The core sequence is fairly conserved and critical for DtxR binding. The flanking regions that are consisted of 5 to 8 more of nucleotides from the core are also required for DtxR binding. The nucleotides in both flanking regions are A-T rich. To examine whether the A-T nucleotides in both flanking regions from the core have significant roles for DtxR binding, a DNA fragment was constructed based on the diphtheria tox promoter/operator, and DNA fragments with substitution of A and T nucleotides In the flanking regions to G and C were also constructed. To assess the effect of these substitutions on binding of DtxR and repressibility by DtxR, $\beta$-galactosidase activity from lacZ fused to the region was assessed. Gel mobility shift of the region by purified DtxR was also examined. The DNA fragments containing the mutations in the flanking regions still exhibited repression and mobility shift with DtxR. The core segment with the mutation is still, therefore, recognized by DtxR. Nonetheless, the results from the assays indicated that the substitution significantly decreased repression of the operator by DtxR in vivo under high-iron condition and decreased binding of DtxR to the operator. These results suggest that A and T nucleotides fur both flanking regions are preferred for the binding of DtxR.

리니어 엔코더를 이용한 선박 엔진용 무선 크랭크 샤프트 디플렉션 검사 장치 개발 (Development of Wireless Device for Inspecting Crankshaft Deflection Using Linear Encoder)

  • A ship engine operator should compensate the crankshaft assembly of ship engine after inspecting crankshaft deflection error in the crank throw regularly to avoid engine vibration and abrasions. In the previous method, the operator enters the bed plate and measures crankshaft deflection using dial gauge on rotating crankshaft manually. However, this method can cause dangerous situation to the operator as well as uncomfortable in an inferior environment. In order to solve the problems, this paper studies the method which makes the operator measure the error outside of the bed plate. In this paper, it is suggested that BlueTooth wireless communication transfers the error data to the outer standing operator with digitalized crankshaft deflection inspection device developed in this paper. So, the wireless measurement system is developed and applied to a medium-speed marine engine through size-miniaturization. After applying test, the developed inspection device showed that it provides much safe and ease inspection method. Furthermore, in the result, the measuring accuracy is more improved.

We define $f_{n+p-1}^{(-1)}(z)$ by using convolution * as $f_{n+p-1}\;*\;f_{n+p-1}^{-1}=\frac{z^p}{(1-z)^{n+p}$. A function p, analytic in E with p(0) = 1, is in the class $P_k(\rho)$ if ${\int}_0^{2\pi}\|\frac{Re\;p(z)-\rho}{p-\rho}\|\;d\theta\;\leq\;k{\pi}$, where $z=re^{i\theta}$, $k\;\geq\;2$ and $0\;{\leq}\;\rho\;{\leq}\;p$. We use the class $P_k(\rho)$ to introduce a new class of multivalent analytic functions and define an integral operator $L_{n+p-1}(f)\;\;=\;f_{n+p-1}^{-1}\;*\;f$ for f(z) belonging to this class. We derive some interesting properties of this generalized integral operator which include inclusion results and radius problems.

NORMAL, COHYPONORMAL AND NORMALOID WEIGHTED COMPOSITION OPERATORS ON THE HARDY AND WEIGHTED BERGMAN SPACES

  • If ${\psi}$ is analytic on the open unit disk $\mathbb{D}$ and ${\varphi}$ is an analytic self-map of $\mathbb{D}$, the weighted composition operator $C_{{\psi},{\varphi}}$ is defined by $C_{{\psi},{\varphi}}f(z)={\psi}(z)f({\varphi}(z))$, when f is analytic on $\mathbb{D}$. In this paper, we study normal, cohyponormal, hyponormal and normaloid weighted composition operators on the Hardy and weighted Bergman spaces. First, for some weighted Hardy spaces $H^2({\beta})$, we prove that if $C_{{\psi},{\varphi}}$ is cohyponormal on $H^2({\beta})$, then ${\psi}$ never vanishes on $\mathbb{D}$ and ${\varphi}$ is univalent, when ${\psi}{\not\equiv}0$ and ${\varphi}$ is not a constant function. Moreover, for ${\psi}=K_a$, where |a| < 1, we investigate normal, cohyponormal and hyponormal weighted composition operators $C_{{\psi},{\varphi}}$. After that, for ${\varphi}$ which is a hyperbolic or parabolic automorphism, we characterize all normal weighted composition operators $C_{{\psi},{\varphi}}$, when ${\psi}{\not\equiv}0$ and ${\psi}$ is analytic on $\bar{\mathbb{D}}$. Finally, we find all normal weighted composition operators which are bounded below.

GENERALIZED BROWDER, WEYL SPECTRA AND THE POLAROID PROPERTY UNDER COMPACT PERTURBATIONS

  • For a Banach space operator $A{\in}B(\mathcal{X})$, let ${\sigma}(A)$, ${\sigma}_a(A)$, ${\sigma}_w(A)$ and ${\sigma}_{aw}(A)$ denote, respectively, its spectrum, approximate point spectrum, Weyl spectrum and approximate Weyl spectrum. The operator A is polaroid (resp., left polaroid), if the points $iso{\sigma}(A)$ (resp., $iso{\sigma}_a(A)$) are poles (resp., left poles) of the resolvent of A. Perturbation by compact operators preserves neither SVEP, the single-valued extension property, nor the polaroid or left polaroid properties. Given an $A{\in}B(\mathcal{X})$, we prove that a sufficient condition for: (i) A+K to have SVEP on the complement of ${\sigma}_w(A)$ (resp., ${\sigma}_{aw}(A)$) for every compact operator $K{\in}B(\mathcal{X})$ is that ${\sigma}_w(A)$ (resp., ${\sigma}_{aw}(A)$) has no holes; (ii) A + K to be polaroid (resp., left polaroid) for every compact operator $K{\in}B(\mathcal{X})$ is that iso${\sigma}_w(A)$ = ∅ (resp., $iso{\sigma}_{aw}(A)$ = ∅). It is seen that these conditions are also necessary in the case in which the Banach space $\mathcal{X}$ is a Hilbert space.
