Objectives: To investigate the protective effect of hesperidin and hesperetin against oxidative stress in 2,2'-azobis (2-aminopropane) dihydrochloride (AAPH)-induced liver toxicity in rats. Methods: Hesperidin or hesperetin (200 mg/kg/day, respectively) was orally administered for 7 days once daily in rats. Subsequently, AAPH (50 mg/kg/day) was administered intraperitoneally. Lipid peroxidation, nitric oxide production, catalase activity, and protein expressions of nuclear factor-kappa B (NF-κB) and inducible nitric oxide synthase (iNOS) in the liver tissues were measured. Results: Administration of hesperidin and hesperetin significantly decreased serum aspartate transaminase and alanine transaminase levels in AAPH-induced oxidative stress liver tissues compared with control group. Lipid peroxidation and nitric oxide (NO) production were also significantly reduced by hesperidin and hesperetin in AAPH-induced oxidative stress liver tissues. In particular, lipid peroxidation levels of hesperetin-administered group significantly decreased to 5.02 nmole/mg protein in oxidative stress rats. Hesperidin and hesperetin significantly increased antioxidant activity, such as that of catalase. Furthermore, administration of hesperidin and hesperetin substantially down-regulated the expression of NF-κB and iNOS in liver tissues. Administration of hesperidin reduced NO levels and iNOS expression more than in the hesperetin-administered group. Conclusions: Administration of hesperidin and hesperetin led to a reduction in AAPH-induced liver toxicity by regulating oxidative stress.
Objectives Depletion of ovarian function after menopause in women induces estrogen deficiency leading to increased fat and decreased muscle mass. In this study, we examined the effect of herbal medicines by measuring hormone expression in muscle tissue of estrogen-deficient rats induced by ovariectomy. Methods Ovariectomy was performed to induce estrogen deficiency, and mice were given herbal prescription (HP) for 6 weeks. Estrogen-deficient rats were divided into two groups: one group (HPH) which were orally administered HP 200 mg/kg and the other group (HPL) administered HP 40 mg/kg. Weight changes in both groups were measured using polymerase chain reaction (PCR). After extraction of the femoral muscles in mice, the expression of the leptin, lipoprotein lipase (LPL), diacyl glycerol acyltransferase (DGAT)1, peroxisome proliferator-activated receptor-γ coactivator (PGC)-1α, NADH dehydrogenase (NDH), farnesyl diphosphate farnesyltransferase (FDFT)1, lanosterol synthase (LSS), phosphatidylethanolamine N-methyltransferase (PEMT), and peroxiredoxin (Prdx6) were measured using PCR. Results HP increased the expression of leptin, LPL, DGAT1, PGC-1α, NDH, FDFT1, LSS, PEMT, and Prdx6. HP affects body fat metabolism and is effective in improving menopausal obesity and obesity complications caused by estrogen deficiency. However, HP does not affect the expression of tumor necrosis factor-α and 3-hydroxy-3-methylglutaryl-CoA reductase, and thus will not be effective in obesity-related metabolic diseases. Conclusions HP is thought to inhibit weight gain by regulating hormone expression related to glucose metabolism and lipid metabolism in muscle tissue of estrogen-deficient rats.
The purpose of this study is to examine the effects of Gamimahaenggamseok-tang and Typhae Pollen on Body Weight and Lipid Metabolism of Rats with Obesity induced by high fat diet. Sixty four male Sprague-Dawley rats, around 250g of B.W., were divided into normal, control, sample A and sample B groups. Normal groups were orally administrated with saline and normal animal feed, and control groups with saline and high fat diet, sample A with liquid extract of Gamimahaenggamseok-tang and high fat diet, and sample B with liquid extract of Typhae Pollen and high fat diet. After 4 and 8 weeks administration, body weight, epididymal fat pad weight, weight of liverㆍspleenㆍkidney, abdominal skin fold thickness, feed intake, and lipid level in liver tissue on morphological examination; total cholesterol, HDL-cholesterol, LDL-cholesterol, triglyceride, free fatty acid, and phospholipid on serum examination were measured in all of the groups.
Objectives: Alcoholic fatty liver is a potentially pathologic condition which can progress to steatohepatitis, fibrosis, and cirrhosis. The objective of this study is to investigate the effects of Scutellaria Radix (SR) extract on the alcoholic fatty liver induced by long-term EtOH administration. Results: Male Sprague Dawley rats were used in this study. All animals were randomly divided into Normal group, treated with saline (n=10); EtOH group, treated with ethanol (n=10); EtOH+SR group, treated with ethanol+Scutellaria Radix extract (n=10). For oral administration of ethanol in EtOH and EtOH+SR group, the ethanol was dissolved in distilled water in concentrations of 25% (v/v). Throughout the experiment of 8 week, the rats were allowed free access to water and standard chow. Sample group were administrated by Scutellaria Radix extract daily for 8 weeks. Results: The levels of hepatic marker such as aspartate aminotransferase and alanine aminotransferase were altered. Histopathological changes such as ballooning, fatty and hydropic degeneration were reduced and the expression of tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$) was significantly attenuated by Scutellaria Radix extract. Conclusions: These data suggest that Scutellaria Radix extract attenuated the alcoholic simple fatty liver by improving hepatic lipid metabolism via suppression of $TNF-{\alpha}$ protein. Scutellaria Radix could be effective in protecting the liver from alcoholic fatty liver.
The influence of (-0-Hydroxycitrate(HCA), shown to be a competitive inhibitor of adenosine 5-triphosphate(ATP) citrate lyase, on food intake and body weight, serum triglyceride and cholosterol level, in vivo rates of fatty acid and cholesterol synthesis, and fat cell number and size was investigated. 3 groups of female, 5 weeks old Sprague Dawley rats, 8 animals each, were ad libitum meal-fed or pair-fed(3 hours from 10 : 00 to 13 : 00) AIN based high glucose diet for a total period of 8 weeks. Providing normolipidemic rats orally with 400mg of HCA formula containing approximately 20mg of HCA 1 hour prior to daily feeding schedule significantly depressed in vivo hepatic rates of fatty acid and cholesterol synthesis in the liver and adipose tissue. Serum triglyceride and cholesterol levels were significantly reduced by HCA. At the end of treatment period, the rats administered with HCA resulted in a significantly reduction in body weight gain. The reduction in weights was attributable to a significant decrease in fat cell size with a smaller extent, but not significant, reduction in fat cell number. Rats receiving HCA demonstrated less food intake than the controls ; however, this decreased caloric intake was not fully responsible for the HCA induced depression of hepatic and adipocytic lipogenesis, since experiment using pair-fed cojntrol rats showed, less magnitude but similar results. Both a anorectic and an antilipogenic properties of HCA seem to be responsible for this weight reduction activity of HCA. The outcome of this study suggest that metabolic regulation may be a feasible approach to the control of obesity and hyperlipidemia.
Obesity is associated with a number of pathological disorders such as non-insulin-dependent diabetes, hypertension, hyperlipidemia, and cardiovascular diseases. Bangpoongtongsungsankamibang (BTSK) has been widely used in the oriental medicine for the treatment of several diseases associated with inflammatory abnormalities in cardiovascular and nervous system. The BTSK is the modified prescription of Bangpoongtongsungsankamibang in which sea tangle (Laminaria japonica) were added. This study was carried out to detemine the anti-obestic effects of BTSK. Pretreatment with the BTSK at daily dose of 100 or 200 mg/kg (p.o.) far 4 weeks reduced serum triglyceride, total cholesterol contents in rat induced by Poloxamer-407 or Triton WR-1339, respectively. Furthermore, post-treatment with BTSK far four weeks also inhibited body weight gain, adipose tissue mass and hyperlipidemia induced by the high fat diet for six weeks. The BTSK shifted serum total-, HDL- and LDL-cholesterol levels toward the values of normal group, suggesting that BTSK has hypolipldemlc effects. The rats fed BTSK reduced lipid peroxide and hydroxy radical in the rat blood and increased superoxide dismutase (SOD) activity compared to the control group. Taken together, these results superoxide that BTSK improve hyperlidemia and obesity via the upregulation of anti-oxidative mechanism.
This study was conducted to investigate the anti-obesity effect of Discorea Japonica Thunb. fermented with Monascus After inducing obesity by feeding hight fat diet (diet induced obesity model: DIO) for four weeks, each 8 rats were assigned to normal (Nor), high fat diet (HF), high fat diet containing orlistat (PC), high fat diet containing different concentration of Discorea Japonica Thunb. fermented with Monascus (UPDM_L, UPDM_H) and Discorea Japonica Thunb. (UPD) extract. Although the UPD, UPDM_L (ultrafine pulverized Discorea Japonica Thunb. fermented with Monascus: 400 mg/kg) and UPDM_H (DIO oral administration ultrafine pulverized Discorea Japonica Thunb. fermented with Monascus: 800 mg/kg) showed weight gain inhibition effects, the results of poor obesity inhibition rather than PC were confirmed. However, it showed a more effective weight loss effect in UPDM_H than UPD, and significantly reduced the weight of epididymal fat and subcutaneous fat. Furthermore, the possibility of anti-obesity effects of Discorea Japonica Thunb. fermented with Monascus can be confirmed by observing the effects of reducing serum total cholesterol, triglyceride and LDL concentrations, reducing ALT and AST levels, and inhibiting fat build-up in liver tissue. It is believed that Discorea Japonica Thunb. fermented with Monascus can be expected to utilize as a functional material that is important to improve anti-obesity and metabolic syndrome.
The aim of the study was to investigate the anti-obesity effect of Coprinus comatus (CC) in high-fat diet-fed Zucker rat (fa/fa). Obesity was induced by feeding on high-fat diet (HFD) containing 60% kcal fat for 10 weeks, in which CC extracts were administrated through the gastrointestinal tract at a concentration of 200 mg/kg BW/day for 10 weeks. The total polyphenol and flavonoid contents of CC extracts were found to be $18.5{\pm}1.1mg$ of catechin equivalent/g, and $5.24{\pm}0.54mg$ of quercetin equivalent/g extract, respectively. The DPPH, ABTS, and hydroxyl radical scavenging activities of CC extracts were 15.34 %, 17.25%, and 16.18%, respectively. In animal study, CC administration significantly reduced the body weight, while there were no significant differences in the daily food intake between the HFD-fed Zucker rats and HFD plus CC-fed rats. CC treatment decreased epididymal and perirenal fat weights in HFD-fed Zucker rats. Significant decreases in the levels of triglyceride and total cholesterol in the serum and liver were observed in the CC-treated group compared with HFD-fed Zucker rats. Serum HDL-cholesterol levels in the CC-treated group were increased compared with the HFD-fed groups. Serum AST and ALT activities in the CC group were significantly lower than those of the HFD-fed group. Taken together, these data demonstrated that CC has potential in preventing high-fat diet induced obesity and is a good candidate for an anti-obesity agent.
Supplement of dietary fibers (DF) is regarded as one of the most effective way to prevent and relieve chronic diseases caused by long-term intake of a high-fat diet in the current society. The health benefits of soluble dietary fibers (SDF) have been widely researched and applied, whereas the insoluble dietary fibers (IDF), which represent a higher proportion in plant food, were mistakenly thought to have effects only in fecal bulking. In this article, we proved the anti-obesity and glucose homeostasis improvement effects of IDF from pear pomace at first, and then the mechanisms responsible for these effects were analyzed. The preliminary study by real-time PCR and ELISA showed that this kind of IDF caused more changes in the gut microbiota compared with in satiety hormone or in hepatic metabolism. Further analysis of the gut microbiota by high-throughput amplicon sequencing showed IDF from pear pomace obviously improved the structure of the gut microbiota. Specifically, it promoted the growth of Bacteroidetes and inhibited the growth of Firmicutes. These results are coincident with previous hypothesis that the ratio of Bacteroidetes/Firmicutes is negatively related with obesity. In conclusion, our results demonstrated IDF from pear pomace could prevent high-fat diet-induced obesity in rats mainly by improving the structure of the gut microbiota.
100% 글루코만난으로 제조된 국수의 체중감소 효과를 측정하기 위해 고지방식이에 의해 유도된 비만 흰쥐에게 35일간 급여 수준을 달리하여 공급하면서 비만지수, 혈청 및 혈액 영양생화학적 분석을 실시하였다. 그 결과, 고지방식이군인 NO군이 높은 체중증가량을 보였고, N25군, N50군 순으로 체중 감소정도가 컸다. 비만지수와 체지방 함량의 변화도 같은 경향이었으며, 혈액 및 혈청 생화학적 분석으로 영양상태를 측정 한 결과, 대체적으로 국수를 25% 섭취한 N25군이 국수를 50%섭취한 N50군에 비해 영양상태가 양호하게 나타났다. 본 연구 결과, 섬유성분이 대부분인 국수는 적은 양, 적은 칼로리로도 포만감을 주어 체중감량에 효과가 크나, 섭취 비율이 다른식이와 적절할 때 영양상에도 문제가 없음을 알 수 있었다. 따라서, 글루코만난으로 제조된 식품인 ‘국수’는 다이어트 식품소재로 좋은 효과가 있는 것으로 시사되었다.
