Biological Activities of Hominis Placenta Herbal Acupuncture prepared by Hydrochloric Acid Hydrolysis (염산 가수분해 추출법으로 조제된 자하거 약침액의 생리활성 효과)

  • Reactive Oxygen Species(ROS) are continuously produced at a high rate as a by-product of aerobic metabolism. Since tissue damage by free radical increases with age, the reactive oxygen species(ROS) such as hydrogen peroxide($H_2O_2$), nitric oxide(NO). Several lines of evidence provided that ROS appears to cause to develop aging-related various diseases such as cancer, arthritis, cardiovascular disease. In this study, we have conducted to investigate the biological activities of Hominis Placenta Herbal Acupuncture by measuring total polyphenol content, DPPH radical scavenging, ABTS radical scavenging, Superoxide dismutase(SOD)-like activity, Nitrite scavenging ability in vitro. The total polyphenol contents of Hominis Placenta Herbal Acupuncture was $24.6m{\ell}/m{\ell}$. Elctron donation ability on DPPH was 49.4%. The 2,2'-azinobis-3-ehtlbezothiazoline-6-sulfonic acid radical decolorization (ABTS) was 50.01%, similar 10 the DPPH free radical scavenging. The superoxide dismutase (SOD)-like activities of hominis placenta herbal acupuncture was 50.876%. The nitrite scavenging abilities at pH 1.5, pH 3.0, pH 6.0 were 52.8%, 29.4%, 15.4%, respectively; these abilities decreased as pH increased. We conclude that Hominis Placenta Herbal Acupuncture may be useful as potential sources of antioxidant.

Nitrite Scavenging and Alcohol Metabolizing Activities of Hot Water Extract from Makgeoly and Its Angiotensin Converting Enzyme Inhibitory Effect (막걸리 열수 추출물의 아질산염 소거능, 알코올 분해능 및 angiotensin converting enzyme 저해 효과)

  • In this study, we investigated the antioxidant activities, alcohol metabolizing activities, nitrite scavenging ability, angiotensin converting enzyme (ACE), and elastase inhibitory effects of hot water extract from Makgeoly (HWM). Antioxidant activities were measured by using 2,2.diphenyl.1.picryl.hydrazyl (DPPH) free radical scavenging activity and SOD (superoxide dismutase).like activity. The DPPH radical scavenging activity and SOD.like activity of HWM were remarkably increased in a dose.dependent manner and were 48.0% and 98.7% at 10 mg/ml, respectively. To determine the influence of HWM on alcohol metabolizing activity, the generating activities of reduced.nicotinamide adenine dinucleotide (NADH) by alcohol dehydrogenase (ADH) and aldehyde dehydrogenase (ALDH) were measured. The facilitating rates of ADH and ALDH activity by HWM were remarkably increased in a dose.dependent manner and were 70.2% and 64.1% at 10 mg/ml, respectively. The inhibitory activity against angiotensin converting enzyme (ACE) of HWM was increased in a dose.dependent manner and was 74.2% at 10 mg/ml. The nitrite scavenging ability of HWM showed the most remarkable effect at pH 1.2 and 2 mg/ml. These results indicated that HWM may have valuable biological properties owing to their antioxidant activities, ADH and ALDH activity, nitrite scavenging ability, and ACE inhibitory activity.

Anti-oxidant Activity of Ethanol Extracts from Fruits of Elaeagnus multiflora THUNB. during Maturation (성숙에 따른 뜰보리수(Elaeagnus multiflora THUNB.) 과실 에탄올 추출물의 항산화 효과)

  • This study was designed to analyze the election donating ability(EDA), superoxide dismutase(SOD)-like activity, nitrite scavenging ability and xanthine oxidase for food nutritional evaluation and excavation of functional materials in fruits of Elaeagnus multiflora THUNB. The electron donating ability of ethanol extracts in the 1.0 mg/ml concentration of extract was higher in extract of unripe fruits than others. The superoxide dismutase (SOD)-like activity was increased according to increasing of the extract concentration. The SOD-like activity was highest in the extract of unripe fruits among the extracts. The nitrite scavenging ability of unripe fruits ethanol extracts in 1.0 mg/ml of extraction solution in pH 1.2 was higher than extract of ripe and Ovenipe fruits. The nitrite scavenging ability was decreased according to increasing of pH. Also, inhibition of xanthine oxidase activity were higher in extracts of unripe fruits than those in extract of ripe and over-ripe fruits.

  • Antioxidant properties and antioxidant activities were analyzed for water extracts and 50% and 70% ethanol extracts of the leaf of Angelica gigas Nakai. The polyphenol and flavonoid contents in water, 50%, and 70% ethanol extract of the leaf of Angelica gigas Nakai, it was found that the polyphenol contents were 18.75 mg GAE/g, 28.95 mg GAE/g, and 34.73 mg GAE/g, respectively, and flavonoid contents were respectively. The DPPH IC50 scavenging activity was 45.84 mg/mL, 36.44 mg/mL, 19.11 mg/mL, respectively, and theABTS+ radical scavenging ability (1 mg/mL) was 28.73%, 22.79%, and 12.70%, respectively. Tyrosinase inhibitory activity, 70% ethanol extract, 50% ethanol extract, and water extract 33.14%, and 4.53%, respectively. Nitrite scavenging activity, 70% ethanol extract, 50% ethanol extract, and water extract were in the order of 1 mg/mL scavenging activity, 36.43%, 34.80%, and 18.85%, respectively.

Biological Activities of Maca (Lepidium meyenii) Extracts (마카 추출액의 생리활성 효과)

  • This study was conducted to determine the optimal extraction conditions for maca by comparing the yields, total polyphenol contents, superoxide dismutase (SOD)-like activity and the nitrite scavenging ability. The proximate composition analysis showed 6.57% moisture, 12.83% crude protein, 1.05% crude fat, 4.80% ash and 74.75% carbohydrate. Maca was extracted with 7 different solvents (water, methanol, ethanol, acetone, ethyl acetate, chloroform and hexane) and the extracts were tested for biological activities. The extraction yields of water, methanol and ethanol extracts were 46.2%, 21.4% and 16.8%, respectively. Acetone, ethyl acetate, chloroform and hexane exhibited very low extraction yield, ranging from 0.2 to 1.0%. Total polyphenol contents and the nitrite scavenging ability were the highest in water extract. Electron donating ability and the SOD-like activity were the highest in methanol extract. When water extract was drawn out at different extraction temperatures (30, 70, $100^{\circ}C$) and time (1, 3, 5 hr), the improved biological activities (total polyphenol contents, electron donating ability, SOD-like activity and nitrite scavenging ability) were found in extracts treated at $100^{\circ}C$ for 3 or 5 hrs.

The Antioxidative and Nitrite Scavenging Ability of Solvent Extracts from Wild Grape (Vitis Coignetiea) Skin (머루 과피 용매추출물의 항산화성 및 아질산염 소거작용)

  • This study was worked out to investigate antioxidant activity of solvent extracts from wild grape skin by measuring electron donating ability (EDA), reducing power, superoxide dismutase (SOD)-like activity, thiobarbituric acid reactive substances (TBARS) and nitrite scavenging ability. Total phenolic compound and flavonoids contents were the highest in ethyl acetate extract, $54.4{\pm}1.18\;mg/100\;g$ and $645.1{\pm}5.05\;mg/100\;g$, respectively. The EDA and reducing power of solvent extracts from wild grape skin were proportionally increased with concentration and ethyl acetate extract $(79.2{\pm}0.06%)$ showed the stronger than BHT $(74.1{\pm}0.15%)$ at concentration of $100\;{\mu}g/mL$, especially. SOD-like ability of ethyl acetate $(25.1{\pm}0.41%)$ and butanol $(20.2{\pm}0.13%)$ extracts were stronger than other extracts at concentration of $100\;{\mu}g/mL$. TBARS of ethyl acetate extract was higher than ascorbic acid. Nitrite scavenging ability of solvent extracts from wild grape skin (pH 2.5, $1000\;{\mu}g/mL$) were in order of ethyl acetate $(90.5{\pm}0.75%)$>butanol $(65.9{\pm}2.16%)$>hexane $(58.1{\pm}1.74%)$>chloroform $(55.4{\pm}1.02%)$>water $(40.9{\pm}0.35%)$. Antioxidant activity of solvent extracts from wild grape skin was the highest in ethyl acetate extract from the results of our experiments.

A Study on the Physicochemical Properties and the Nitrite Scavenging Ability of Japanese Sumac Extracts (붉나무 추출물의 이화학적 특성과 아질산염 소거능에 관한 연구)

  • Japanese sumac (Rhus javanica) is one of the common herbaceous plants growing over the country. This study was conducted to investigate the extraction characteristics and physiological activities inluding nitrite scavenging ability of the water extracts from leaf, fruit and bark of Japanease sumac. Extraction yield was 6.62~13.84%, free amino acids were detected as 24 kinds with 37.9 mg/100g in leaf extract, 23 kinds with 27.0 mg/100g in fruit extract and 27 kinds with 39.0 mg/100g in bark extract, respectively, and seven kinds essential amino acids were detected. Total contents of flavonoids equivalent to naringin were 587.2 mg/100g in bark extract, 557.3 mg/100g in fruit extract and 379.9 mg/100g in leaf extract, respectively. Total contents of phenolics equivalent to gallic acid were 111.2 mg/100g in leaf extract, 108.4 mg/100g in fruit extract and 80.4 mg/100g in bark extract, respectively. The nitrite scavenging ability of extracts was order of 61.93% in bark extract>57.38% in fruit extract>55.49% in leaf extract, and was 78.1~100% of those of BHT (butylated hydroxytoluene) equivalents at pH 1.2. The electron donating ability was order of 47.38% in fruit extract>43.06% in leaf extract>38.55% in bark extract, and was compared to 65.6%, 58.8% and 53.6% of those of BHT equivalents, respectively. The reduction power was evaluated to 37% higher in leaf extract, 43% higher in fruit extract and 46% higher in bark extract than those of BHT equivalents. The metal chelating ability of extracts was considerably low and was order of 27.3% in bark extract>20.6% in leaf extract>11.2% in fruit extract.

Antioxidant Activities of Extracts from Different Parts of Sasa borealis (조릿대의 부위별 추출물의 항산화 활성)

  • Objectives : This study was performed to investigate the antioxidant activity of water and ethanol extracts from Sasa borealis leaves, stems and roots. Methods : Sasa borealis leaves, stems and roots extract were prepared using water and ethanol. The total polyphenol and total flavonoid contents were analyzed. 2,2-diphenyl-1-picrylhydrazyl(DPPH) radical scavenging activity, SOD like activity, 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid)(ABTS), hydroxyl radical scavenging activity, and Nitrite scavenging activity assays were carried out to determine the antioxidant activities. Results : The antioxidant activities of the Sasa borealis appeared higher in ethanol extract than water extracts. Total polyphenol and total flavonoids contents in ethanol extracts of leaves were $24.6mg/m{\ell}$ and $14.3mg/m{\ell}$, respectively, which were much higher than those of any other parts. SOD like activity was 70% ethanol extract of the leaves was highest with 15.68%. Electron donating ability was 70% ethanol extract of the leaves had the highest 59.07%. It exhibited high electron donating ability than BHT(45.68%). Nitrite scavenging activity of 70% ethanol extract was higher than the water extract at pH 2.5 and pH 4.2. Nitrite scavenging activity was 70% ethanol extract of the leaves was the highest 75.2%. Hydroxyl radical scavenging activity was 70% ethanol extract of the leaves was highest with 16.16%, showed very low activity than BHT(61.56%). Conclusions : These results suggest that 70% ethanol extracts from Sasa borealis leaves, exhibited higher antioxidant activities than those of root and stem, and can be potentially used as proper natural antioxidants.

Inhibitory Effect on Tyrosinase, ACE and Xanthine Oxidase, and Nitrite Scavenging Activities of Jubak (Alcohol filter cake) Extracts (주박 추출물의 아질산염 소거와 Tyrosinase, Xanthine Oxidase, ACE 저해 효과)

  • The purpose of this study was to investigate the tyrosinase, xanthine oxidase, angiotensin converting enzyme (ACE) inhibitory effects, and nitrite scavenging abilities of Jubak (alcohol filter cake, AFC) hot water and ethanol extracts. Tyrosinase inhibitory effects of Jubak hot water and ethanol extracts were above 85%. Nitrite scavenging ability was higher at pH 1.2 than at pH 4.2 and 6.0. Nitrite scavenging abilities of water and ethanol extracts were above 90% at pH 1.2. Xanthine oxidase inhibitory effect of Jubak hot water and ethanol extracts showed a lower, and the effect did not different by hot water and ethanol. ACE inhibitory effects of Jubak hot water and ethanol extracts were approximately 43~53% and 36~47%, respectively. Overall, our results indicate that Jubak hot water and ethanol extracts may have tyrosinase and ACE inhibitory effects and nitrite scavenging ability, but no xanthine oxidase inhibitory effect. Therefore, Jubak has potential as a cosmetic raw material. Additionally, Jubak could be used as a food additive.

Antioxidant Activity of Pyrus serotina Fruit in Different Cultivars and Parts (배 품종 및 부위별 항산화 활성)

  • The aim of this study is to compare the total polyphenol, total flavonoid, and antioxidative activity among 5 cultivars of Korean pears cultivated in Korea. As a result of the analysis for the phenolic substance of 5 cultivars, such as Wonwhang, Sunwhang, Whangkeumbae, Chuwhang, and Shingo, the content of total polyphenol was higher in Shingo and Chuwhang, and the content of total flavonoid showed higher levels in Wonwhang and Sunwhang. In addition, this study found that DPPH radical-scavenging ability was the highest in Shingo and that the part of fruit skin showed more favorable reaction to radical ability than fruit flesh. It was also found that much better antioxidative activity was shown for the methanol solvent extraction than for the ethanol solvent extraction. The nitrite-scavenging ability showed the best for Wonwhang and Chuwhang of the cultivars and much better reaction for the methanol solvent extraction than for the ethanol solvent extraction just like the case of DPPH radical-scavenging ability. Further, nitrite-scavenging ability appeared much better reaction for the fruit skin than for the fruit flesh, and antioxidative activity dropped for the higher the range on the pH scale. From the results of this study, Korean pears are worthy of developing as a natural functional food and substance for beauty treatment through the research on bioactivity.
