Effects of Radix Curcumae Aromaticae Extract in Rat Cardiac Endothelial Cells (울금 추출물이 배양 심장내피세포에 미치는 영향)

  • To test the protective effect of Radix Curcumae Aromaticae (RCA) on the damage of cardiac endothelial cells by xanthine oxidase (XO)/hypoxanthine (HX)-induced oxygen free radical, Neutral Red (NR), thiobarbituric acid reactive substances (TSARS), and DNA synthesis assay were used in the presence of RCA extract. The results of these experiments were obtained as follows ; Cardiac endothelial cells treated with XO/HX showed the cytotoxicity such as decreases in viability and DNA synthesis, a increase in lipid peroxidation. Cardiac endothelial cells pretreated with RCA extract protected the increase of lipid peroxidation by XO/HX. Cardiac endothelial cells pretreated with RCA extract inhibited the decrease of DNA synthesis by XO/HX. These results show that XO/HX elicits toxic effects in cultured cardiac endothelial cells derived from neonatal rat, and suggest that RCA extract is very effective in the prevention of XO/HX-induced toxicity.

  • To test the protective effect of Sophorae Radix (SR) on the damage of cardiac endothelial cells by xanthine oxidase (XO)/hypoxanthine (HX)-induced oxygen tree radical, Neutral Red (NR), lactate dyhydrogenase (LDH), and c-fos immunopositive cells assay were used in the presence of SR extract. The results of these experiments were obtained as follows ; Cardiac endothelial cells treated with XO/HX showed the cytotoxicity such as a decrease in viability, and increases in LDH activity and c-fos immunopositive cells. Cardiac endothelial cells pretreated with SR extract protected the increase of LDH activity. Alos, cardiac endothelial cells pretreated with SR extract inhibited the increase of c-fos immunopositive cells. These results show that XO/HX induces toxic effects in cultured cardiac endothelial cells derived from neonatal rat, and suggest that SR extract is very effective in the prevention of XO/HX-induced toxicity.

Isolation and Characterization of an Antibacterial Substance from Rheum palmatum for Treatment of Bacterial Vaginosis (대황으로부터 세균성 질염 치료를 위한 항균성 물질의 분리 및 특성)

  • Rheum palmatum has traditionally been used as a preventive agent and medication against fever and infection. The aim of this study was to isolate and characterize an antibacterial substance from R. palmatum that is effective against bacterial vaginosis. A methanol extract from R. palmatum showed antibacterial activity against Lactobacillus vaginalis KC TC 3515, Chryseobacterium gleum KCTC 2904, and Sphingomonas paucimobilis KCTC 2834, which cause bacterial vaginosis. After extraction and pH control of the methanol extract from R. palmatum, we found that acidic and alkaline extracts did not show antibacterial activity. A neutral extract (50 mg/mL) displayed an inhibitory zone of 18 mm on a nutrient agar plate with C. gleum KCTC 2904. Fractions No. 11 and 12 among 41 fractions obtained by silica gel column chromatography produced inhibitory zones of 10 mm on nutrient agar plates with C. gleum KCTC 2904. $R_f0.15$ and $R_f0.17$ spots produced by TLC of fraction No. 11 showed antibacterial activity against C. gleum KCTC 2904. Isolation and purification of the peak at a retention time (Rt) of 9.427 min was achieved by HPLC of $R_f0.29spots$. The peak at Rt 9.427 min showed antibacterial activity against C. gleum KCTC 2904.

  • It was investigated effect of Fructure schizandra chinesis Water extract on the white rat which was induced hypercholesterolemia by fatty diet. The results obtained were summarized as follows; Neutral lipid level in blood lipid was greatly decreased on the group being fed animal fat diet and Fructure schizandra chinesis Water extract, total cholesterol was decreased on the group being fed vegetative fat diet, HDL-cholesterol was shown that the group being fed vegetative fat diet and Fructure schizandra chinesis Water extract at a time more increased HDL-cholesterol in a little than the other group, and normal fat diet group show to highest figure,38.5mg/dl. Animal fat diet group is 20.75 mg/dl. but, the group being fed Fructure schizandra chinesis Water extract was increased to 31.00mg/dl. LDL-cholesterol had no difference between fat diet group and the group being fed Fructure schizandra chinesis Water extract.

  • This study was carried out to evaluate antitoxic effects Taraxaci Herba extract against Cadium by calorimetric methods. The antitoxic activity of Taraxaci Herba ex tract in NIH 3T3 fibroblasts was evaluated by MTT (3-(4,5-Dimethylthiazol-2-yl)-2,5-phenyl-2H-tetrazoliumbromide), NR (Neutral red) and SRB (Sulforhodamine B protein) assay. The light microscopic study was carried out to observe morphological changes of the treated cells. These results were obtained as follows; The concentration of $10^{-2}mg/ml$ of Taraxaci Herba extract was shown significant antitoxic activity. The number of NIH 3T3 fibroblasts were antitoxic and tend to regenerate. These results suggest that Taraxaci Herba extract retains a potential antitoxic activity.

Effect of Proanthocyanidin-rich Extracts from Pinus radiata Bark on Termite Feeding Deterrence

  • Antioxidants are known to affect the feeding habits of termites and a good source is pine bark which contains high levels of antioxidants which can be extracted with neutral solvents. In this study procyanidins (PCs)-rich MeOH-extract and a hot-water extract (HWE) were prepared from Pinus radiata bark. HWE was further separated into crude PCs (CPCs), polymeric PCs (PPCs), and low-molecular-weight PCs (LMWPCs) fractions. The MeOH extract and these fractions were examined for termite (Reticulitermes flavipes Kollar) antifeedant activities using a no-choice test. Catechin was used as a positive control. The LMWPCs was found to be the most effective deterrent to termite feeding. The comparative efficiency of the compounds tested were LMWPCs > PPCs > CPCs > MeOH extract, with the latter being more dependent on molecular weight than on its antioxidant activity.

Effect of Old Antler Extracts on the Galactosamine-induced Hepatotoxicity in Rats (녹각이 Galactosamine에 의해 유도된 간장해에 미치는 영향)

  • This study was conducted to investigate the biochemical efforts of old antler extracts on the enzyme activities in serum of galactosamine (GaIN)-induced rats. Male rats of Sprague-Dawley strain weighing the average of 110$\pm$10g were divided into four groups and each group was fed either AIN-76 diet or modified AIN-76 diet with old antler extracts (water extract, neutral extract, ether extract) for four weeks. CaIN (400mg/ kg of body weight) was injected to CaIN treatment groups intraperitoneally on the same day every week. CaIN treatment decreased growth performance of rats. But dietary supplementation of old antler extracts prevented this decrement. Serum aminotransferase activities were increased by CaIN treatment, but this increment was reduced by feeding the old antler extracts, and the effect of water extract was the most prominent of the old antler extracts. Activities of lactate dehydrogenase (LDH) and alkaline phosphatase (ALP) in serum of CaIN treatment groups were higher than those of control groups and were decreased by old antler extracts supplementation.

The Possibility of Passiflora Caerulea Extract as Cosmetic Material (Passiflora caerulea 추출물의 화장품 소재 활용 가능성)

  • This study attempted to investigate the possibility of Passiflora caerulea extract as a cosmetic material by measuring its antioxidant activities and melanin biosynthesis-inhibiting effects. For this, the substance was extracted with 70% ethanol, and ABTS radical scavenging activities were measured, using the TEAC assay. In addition, cell viability through the neutral red assay, ROS inhibition and inhibition of melanin biosynthesis were analyzed, and the results found the followings: In terms of ABTS radical scavenging activities of the extract, dose-dependent scavenging activities were observed. In trolox used as a positive control group and 0.1 mg/mL, similar radical scavenging activities were found. The production of ROS through CFDA was inhibited in a dose-dependent fashion. Furthermore, inhibition of melanin biosynthesis induced by 𝛼-MSH was confirmed. Therefore, it is reasonable to conclude that Passiflora caerulea extract has a potential as a cosmetic ingredient.

  • To investigate the protective effect of Sophorae Radix (SR) on the damage by pulmonary vascular endothelial cells by xanthine oxidase (XO)/hypoxanthine (HX)-induced oxygen tree radical, Neutral Red (NR) and c-fos immunopositive cell assay were used. The results were obtained as follows ; The viability of vascular endothelial cells treated with XO/HX was decreased. And c-fos immunopositive cells represented a maximal increase in group treated with XO/HX for 2 hour in pulmonary vasvular endothelial cells. But pretreated groups with SR extracts were not inhibited the increase of c-fos immunopositive cells by XO/HX in a dose-dependent manner. These results show that XO/HX elicits toxic effects in cultured pulmonary vascular endothelial cells, and suggest that SR extract is very effective in the prevention of XO/HX-induced increase of c-fos immunopositive cells.

Effects of Bulbus Allii Macrostemi Extract on PKC activity in Pulmonary Vascular Endothelial Cells Damaged by XO/HX (산소자유기에 의해 손상된 혈관내피세포의 PKC 활성도에 미치는 해백 추출물의 효과(I))

  • To investigate the protective effect of Bulbus Allii Macrostemi (BAM) on the damage by pulmonary vascular endothelial cells by xanthine oxidase (XO)/hypoxanthine (HX)-induced oxygen free radical, Neutral Red (NR) and protein kinase c (PKC) activity assay were used. The results were obtained as follows ; The viability of vascular endothelial cells treated with XO/HX was decreased. And activation of PKC represented a maximal increase in group treated with XO/HX for 15 mins in vasvular pulmonary endothelial cells. But pretreated groups with BAM extracts were not inhibited the increase of PKC activation by XO/HX in a dose-dependent fashion. These results show that XO/HX elicits toxic effects in cultured pulmonary vascular endothelial cells, and suggest that BAM extract is very effective in the prevention of XO/HX-induced PKC activation.
