Flavonoids of Gomphocarpus sinaicus and Evaluation of Some Pharmacological Activities

  • The aerial parts of Gomphocarpus sinaicus Boiss. yielded four flavonoids that were identified as isorhamnetin 3-O-rhamnoglucoside (1), luteolin-7-O-glucoside-3-O-rhamnoside (2), rutin (3) and rutin-7-O-rhamnoside (4). All of the isolated flavonoids were identified by spectroscopic methods (UV, FAB-MS, $^1H-NMR\;&\;^{13}C-NMR$) and in comparison with the literature data. The isolated flavonoids 1, 2 and 4 are reported here for the first time from Gomphocarpus sinaicus Boiss. Three sets of experiments were carried out using the defatted alcoholic extract of Gomphocarpus sinaicus Boiss: the $1^{st}$ experiment indicated that the $LD_{50}$ was 49.82 mg/100 g b.wt. of intraperitoneally (i.p.) injected mice. The toxic signs were recorded within the first 24 hr post-injection. The $2^{nd}$ experiment revealed that the extract of the plant exhibited significant anti-inflammatory effects in normal rats. The $3^{rd}$ experiment was found that the tested doses of the extract in diabetic rats induced a significant decrease in serum glucose, AST, ALT, triglycerides, cholesterol and LDL, while HDL caused a significant increase.

  • The flavonoids are one of the most numberous and widespread groups of natural consituents. The low molecular weight of benzo-${\gamma}$-pyrone derivative are ubiquitous in plants and are vegetables nuts, seeds, leaves, flowers, and bark. The flavonoids constitute of a large class of compounds ubiquitous in plants containing a number of phenolic hydroxyl groups attached to ring structures conferring the antioxidant activity. Epidemiologic studies suggest that the dietary intake of antioxidants constitutes a risk factor for vasclar disease indicating that oxidation may be important in the pathogenesis of human athero-sclerosis. Elevated plasma low density lipoprotein (LDL) cholesterol concentration are associated with accelerated atherosclerosis, LDL is oxidized by smooth muscle cells resulting in several chemicals and physical changes of LDL. Oxidized LDL is responsible for cholesterol loading of macrophages foam cells formation and atherogenesis. There have been insulficient tests of the protective effects of flavonoids against LDL oxidation to make definitive statements about their structure activity relationships. How-ever hydroxylation of the flavone uncleas can appears to be advantageous because polyhydroxylated aglycone flavonoids are potent inhibitor of LDL modification. This identification may lead to new and more effective antioxidant strategies for abrogating the atherosclerotic process the leading cause of death and disability in industrialized societies.

Development of an Analytical Approach for the Utilization of Edible Tree Sprouts

  • This study evaluated the general nutritional ingredients such as crude fats, crude ashes, crude proteins, total polyphenols, and total flavonoids in 18 kinds of edible tree sprouts. The tree sprouts of Philadelphus schrenckii, Lycium chinense, and Morus alba had the highest crude fat, crude ash, and crude protein content, respectively. The tree sprouts of Cedrela sinensis (CSS) with high ABTS+ radical scavenging activities had a high content of total polyphenols (175.65 mg/g ext.) and total flavonoids (75.18 mg/g ext.). The simultaneous determination of flavonoids such as rutin, isoquercitrin, quercitrin, afzelin, and quercetin in CSS was conducted using high-performance liquid chromatography with a wavelength of 270 nm. Among the flavonoids, the content of quercitrin in CSS was the highest at 59.28 mg/g ext. This study also aids the quality control of many edible tree sprouts by analyzing the general components, total polyphenols, and total flavonoids.

Antiinflammatory Constituents from the Roots of Smilax bockii warb.

  • From $70\%$ ethanol extract of the roots of Smilax bockii warb., seven flavonoids, kaempferol (1), $kaempferol-7-O-\beta-D-glucopyranoside$ (2), quercetin (3), isorhamnetin (4), (+)-dihydro­kaempferol (5), engeletin (6), isoengeletin (7), and $n-butyl-\beta-D-fructopyranoside$ (8), caffeic acid n-butyl ester (9) were isolated and identified by means of chemical and spectroscopic. Compounds 2, 4, and 6-9 were isolated for the first time from the roots of S. bockii and compounds 2, 8, and 9 were firstly isolated from the genus Smilax. In addition, using the SEAP (Secreted alkaline phosphatase) assay system, we investigated the in vitro anti-inflammatory activity of the $70\%$ ethanol extract of the roots of S. bockii, which showed moderate activity in inhibiting $TNF-\alpha-induced NF-{\kappa}B$ activation with an $IC_{50}$ value of $166.6 {\mu}g/mL$.

Flavonoid Glycosides Inhibit Sortase A and Sortase A-Mediated Aggregation of Streptococcus mutans, an Oral Bacterium Responsible for Human Dental Caries

  • Three flavonoids were isolated from dried flowers of Sophora japonica using repetitive column chromatography and high-performance liquid chromatography. The flavonoids were identified as rutin (1), quercetin-3'-O-methyl-3-O-α-ʟ-rhamnopyranosyl(1 → 6)-β-ᴅ-glucopyranoside (2), and quercetin (3) on the basis of spectroscopic analysis and comparison of values reported in the literature. These compounds inhibited the action of sortase A (SrtA) from Streptococcus mutans, a primary etiologic agent of human dental caries. The onset and magnitude of inhibition of saliva-induced aggregation of S. mutans treated with compound 1 was comparable to that of untreated S. mutans with a deletion of the srtA gene.

Effects of Sophoraflavanone G, a Prenylated Flavonoid from Sophora Flavescens, on Cyclooxygenase-2 and In Vivo Inflammatory Response

  • Previously, several prenylated flavonoids having a C-8 lavandulyl moiety were found to inhibit cyclooxygenase-1 (COX-1) as well as 5-lipoxygenase (5-LOX), and sophoraflavanone G was the most potent inhibitor against these eicosanoid generating enzymes among 19 prenylated flavonoids tested. In this investigation, effects of sophoraflavanone G on COX-2 induction from RAW 264.7 cells and in vivo inflammatory response were studied. Sophoraflavanone G inhibited prostaglandin $E_2{\;}(PGE_2)$ production from lipopolysaccharide (LPS)-treated RAW cells by COX-2 down-regulation at 1-50 uM. Other prenylated flavonoids including kuraridin and sanggenon D also down-regulated COX-2 induction at 10-25 uM, while kurarinone and echinoisoflavanone did not. In addition, sophoraflavanone G showed in vivo anti-inflammatory activity against mouse croton oil-induced ear edema and rat carrageenan paw edema via oral (2-250 mg/kg) or topical administration (10-250 ug/ear). Although the potencies of inhibition were far less than that of a reference drug, prednisolone, this compound showed higher anti-inflammatory activity when applied topically, suggesting a potential use for several eicosanoidrelated skin inflammation such as atopic dermatitis.

  • Five flavonoids and one aromatic acid were isolated from the leaves of Zanthoxylum piperitum. The structures of compounds were elucidated as quercetin, afzelin, quercitrin, hyperoside, hesperidin and protocatechuic acid on the basis of spectral evidence.

Inhibitory Activity of IL-6 Production by Flavonoids and Phenolic Compounds from Geranium thunbergii

  • Three flavonoids (1 - 3) and three phenolic compounds (4 - 6) were isolated from the whole plant of Geranium thunbergii Sieb. et Zucc (Geraniaceae). Their structures were determined by chemical and spectral analysis. These compounds were examined for the inhibitory activity of IL-6 production in $TNF-{\alpha}$ stimulated MG-63 cell. Among the isolated compounds, gallic acid (4) and gallic acid methyl ester (6) showed potent inhibitory activity.
