Development of High Meat Quality Using Microsatellite Markers in Berkshire Pigs (Microsatellite Marker를 이용한 육질 우수 버크셔 계통 조성에 관한 연구)

  • In this study, the efficiency of microsatellite (MS) markers for pork quality was examined and further, their suitability to domestic pork industry also was verified, by measuring meat quality parameters of Berkshire breeds. A total of 323 pigs of Berkshire breeds were slaughtered and subjected to meat quality evaluation. In addition, the genomic DNAs from blood samples of slaughtered pigs were used for genotyping analysis of 50 MS markers. The results revealed that Berkshire breeds have excellent meat quality, compared with the popular domestic breeds such as Duroc, Yorkshire, and Landrace. Noticeably, the Berkshire breeds exhibited a significant post-mortem pH24hr ($5.88{\pm}0.01$) and fat content ($2.878{\pm}0.06$). Through the linkage analysis between MS markers, 14 MS markers showed significant association with meat quality traits (p<0.05). Maximum significant differences of 0.55 pH24hr value and 2.04% fat content were observed between the highest and lowest allele populations. If these 14 MS markers are applied to the pork quality diagnosis kit, the synergistic effect can be expected in meat quality parameters such as meat color, fat content, pH 24 hr, cooking loss, drip loss and water-holding capacity.

Identification of Antioxidant Compound Derived from Methanolic Extract of Houttuynia Cordata (어성초 메탄올 추출물로부터 항산화 효능을 가진 활성물질의 확인)

  • This study was carried out to evaluate the antioxidant effect of methanolic extract of Houttuynia cordata (HCME) and to identify a compound having antioxidant effect. The ethyl acetate fraction of HCME showed the highest antioxidant effect in organic solvent fractions. The fraction was then separated into 12 fractions by open column chromatography. Among these fractions, the fraction 10 (Fr. 10) with the highest antioxidant activity was isolated, and its antioxidant effect was evaluated by DPPH radical scavenging activity, reducing power, TBARS, cell viability, DNA oxidation and DCF fluorescence. The Fr. 10 at a $64{\mu}g/ml$ showed 60% of inhibitory effect similar to that of vitamin C at $10{\mu}g/ml$, compared with blank group. The Fr. 10 at $64{\mu}g/ml$ showed 264% of reducing power, compared with blank group. TBARS assay showed that the Fr. 10 at $64{\mu}g/ml$ had 35.5% of inhibitory effect similar to that of vitamin E at $1,000{\mu}g/ml$, compared with blank group. The Fr. 10 above $32{\mu}g/ml$ displayed cytotoxicity. However, it was observed that the Fr. 10, above $1{\mu}g/ml$ reduced DNA damage. DCF fluorescence assay showed that the Fr. 10 inhibited oxidative stress by $H_2O_2$ in a dose dependent manner. The compound of Fr. 10 was identified to be rutin whose molecular weight is 610 by the IR and LC-MS analyses. Therefore, these results suggest that the rutin of Fr. 10 could use as a natural antioxidant for development of cosmetics and functional foods.

  • Multiplex ligation dependent probe amplification (MLPA) is a PCR-based method to detect gene dosage. Since its introduction, MLPA has been used to test a large number of genes for major deletions or duplications. Genetic testing, as a diagnostic tool for genetic disease, has been used primarily to identify point mutations, including base substitutions and small insertions/deletions, using PCR and sequence analysis. However, it is difficult to identify large deletions or duplications using routine PCR- gel based assays, especially in heterozygotes. The MLPA is a more feasible method for identification of gene dosage than another routine PCR-based methods, and better able to detect deleterious deletions or duplications. In addition to detection of gene dosage, MLPA can be applied to identify methylation patterns of target genes, aneuploidy during prenatal diagnoses, and large deletions or duplications that may be associated with various cancers. The MLPA method offers numerous advantages, as it requires only a small amount of template DNA, is applicable to a wide variety of applications, and is high-throughput. On the other hand, this method suffers from disadvantages including the possibility of false positive results affected by template DNA quality, difficulties identifying SNPs located in probe sequences, and analytical complications in quantitative aspects.

  • Callus formation and plant regeneration from the seeds of zoysiagrass cv. Zenith was tested on MS basal medium supplemented with various concentrations of 2,4-dichlorophenoxyacetic acid(2,4-D) and of several cytokinins. A concentration of 1mg/L 2,4-D on medium stimulated callus formation. In the presence of 5mg/L 2,4-D, addition of 1mg/L kinetin significantly enhanced callus formation and plant regeneration over 2,4-D alone. To transfer foreign DNA into turfgrass, parameters for the bombardment of embryogenic callus with the particle bombardment were partially optimized using transient expression assay of a $chimeric \beta$-glucuronidase(GUS) gene driven by the CaMV 35S promoter. GUS gene was strongly expressed at helium pressure 1,100 psi and 6~9cm target distance.

  • Physiological changes of Escherichia coli during the fed-batch fermentation process were characterized in this study. Overall cellular protein samples prepared at the different stage of fermentation were separated by 2-dimensional gel electrophoresis (2-DE), and differently expressed 15 proteins, Phosphotransferase enzyme I, GroEL, Trigger factor, ${\beta}$ subunit of ATP synthase, Transcriptional regulator KDGR, Phosphoglycerate mutase 1, Inorganic pyrophosphatase, Serine Hydroxymethyl-transferase, ${\alpha}$ subunit of RNA polymerase, Elongation factor Tu, Elongation factor Ts, Tyrosine-tRNA ligase, DnaK suppressor protein, Transcriptional elongation factor, 30S ribosomal protein S6 were identified using matrix-assisted laser desorption / ionization time-of-flight mass spectrometry (MALDI-TOF MS). When bacterial cells grow to high cell density, and IPTG-inducible heterologous protein is produced, expression level of overall cellular proteins was decreased. According to their functions in the cell, identified proteins were classified into three groups, proteins involved in transport process, small-molecule metabolism, and synthesis and modification of macromolecules.

  • Progress in commercialization of the irradiation process, greater international trade in irradiated food, differing regulations relating to use of the technology in many countries, and consumer demand for clear labeling of irradiated food highlighted the need for tests that could be applied to the food itself. Scientists have had to focus on identifying and isolating the minute changes caused in the component food molecules by the process. A number of investigators have reviewed the changes occurring in food after irradiation, detecting and measuring the effects of irradiation. The Joint FAO/IAEA Division of Nuclear Techniques in Food and Agriculture organised a coordinated program on analytical detection methods in irradiation treatment of food (ADMIT) which promoted cooperation in this area and sponsored collaborative testing of some of the most promising methods.

Construction of a Transformed Yeast Strain Secreting Both $\alpha$-Amylase and Glucoamylase for Direct Starch-Fermentation

  • A yeast strain secreting glucoamylase was transformed with an expression vector (pMS12) containing the promoter of yeast alcohol dehydrogenase I gene ADC1, mouse salivary $\alpha$-amylase cDNA, and a segment of yeast $21\mu m$ plasmid. The transformed strain could produce ethanol from starch (4%, w/v) through a direct one-step process with the conversion efficiency of 93.2%, during 5 days of fermentation, while the original, untransformed strain exhibited a conversion efficiency of 38.1% under the same condition. When the regulatory site of the ADC1 promoter region was removed, the production of ethanol increased to 29~37% in the presence of exogenous 3%(v/v) ethanol in the fermentation medium.

Regeneration and Agrobacterium - Mediated Transient Transformation of Button Daisy (Leucanthemum vulgare)

  • Explants of button daisy were screened for their regeneration potential and transient GUS gene expression. Medium containing MS salts minerals and $B_5$ vitamins supplemented with $0.1\;\cal{mg/L}$ BA and $0.1\;\cal{mg/L}$ TDZ showed the best regeneration. Disc florets and receptacles were the most responsive explants in regeneration and transient gene expression respectively. Regenerated plants were successfully rooted and established in the green-house conditions. Infection and co-cultivation of explants with Agrobacterium tumefaciens containing pCAMBIA 1301 resulted in transient GUS foci. Among the different explants, receptacles showed the highest percentage of transient GUS gene expression. Enzymatic and molecular analyses of transformed calli confirmed the integration of GUS gene.

Characterization of Organochlorine Insecticide Endosulfan-Degrading Bacterium Isolated from Seaside Sediment (갯벌에서 분리한 유기염소계 살충제 Endosulfan 분해 세균의 특성)

  • An endosulfan degrading bacterial strain, K-1321, was isolated by endosulfan-enrichment culture from a seaside sediment collected at Dadaepo Beach, Busan, Korea. The strain was identified as a Serratia sp. based on the results of morphological, biochemical and 16S rDNA homology analyses. Serratia sp. K-1321 was able to completely degrade 50 ppm endosulfan in culture media and soil within 6 weeks at $25^{\circ}C$. GC/MS analysis revealed that endosulfan diol was an intermediate of the bacterial endosulfan degradation. Considering the above results, we concluded that Serratia sp. K-1321 utilized endosulfan as a carbon source and metabolized endosulfan via a less toxic pathway, such as the formation of endosulfan diol as an intermediate.

Yeast Cloning Vectors and their Application to the Development of Starch-fermenting Yeast (효모 Cloning Vector와 전분발효 효모의 개발)

  • Transformed, hybrid strains of the yeast Saccharomyces capable of simultaneous secretion of both glucoamylase and ${\alpha}-amylase$ have been produced. These strains can carry out direct, one-step assimilation of starch with conversion efficiency greater than 93% during a 5 day growth period. One of the transformants converts 92.8% of available starch into reducing sugars in only 2 days. Glucoamylase secretion by these strains results from expression of one or more chromosomal STA genes derived from Saccharomyces diastaticus. The strains were transformed by a plasmid(pMS12) containing mouse salivary ${\alpha}-amylase$ cDNA in an expression vector containing yeast alcohol dehydrogenase promoter and a segment of yeast $2{\mu}$ plasmid. The major starch hydrolysis product produced by crude amylases found in culture broths is glucose, indicating that ${\alpha}-amylase$ and glucoamylase act cooperatively.
