Isolation of Monoamine Oxidase B Inhibitory Compound from the Leaves of Eucommia ulmoides Oliv. (두충(Eucommia ulmoides Oliv.)잎으로부터 Monoamine Oxidase B 억제활성물질의 분리)

  • The repeated silica gel colum chromatographies of n-BuOH fraction obtained from MeOH extracts of Eucommia ulmoides leaves led to isolation of a flavonoid-glycoside inhibiting monoamine oxidase B activity. Its chemical structure was determined to be $3-O-[{\beta}-D-glucopyranosyl\;(1{\rightarrow}2)\;{\beta}-D-xylopyranosyl]$ quercetin through interpretation of spectral data and adaptation of acid hydrolysis. The $IC_{50}$ value of this compound in rat brain mitochondrial MAO-B inhibitory activity was evaluated to be $8.05\;{\mu}mol/l$.

Effects of Higenamine and Its Derivatives on the Activity of Rat Brain Mitochondrial Monoamine Oxidase (Higenamine과 그 유도체들이 흰쥐 미토콘드리아 Monoamine Oxidase 활성에 미치는 영향)

  • The effect of higenamine and its derivatives on the activity of rat bran mitochondrial monoamine oxidase(MAO) was studied. Methoxyhigenamine of drugs tested had no effect on isometric contraction of heart and reversibly inhibited MAO towards 5-hydroxytryptamine(5-HT) and phenylethylamine(PEA) in a pure competitive fashion and in a hyperbolic mixed fashion, respectively, but was found to be relatively MAO-A selective inhibitor, with IC50 value for 5-HT lower ten fold than for PEA. The results suggest that methoxyhigenamine is a reversible, relatively MAO-A specific inhibitor in virto.

Effects of Pine Needle Ethyl Acetate Fraction on Acctylcholine(ACh) and Its Related Enzymes in Brain of Rats (뇌 조직의 아세틸콜린 및 그 관련효소에 미치는 솔잎(Pine Needle) 에틸아세테이트획분의 영향)

  • This study was designed to investigate the effects of ethyl acetate(EtOAc) fraction of pine (Pinus densiflora Sieb et Zucc) needle on cholesterol and lipofuscin(LF) accumulations, acetylcholine(ACh) and its related enzyme activities such as choline acetyltransferase(ChAT) and acetylcholinesterase(AChE), and monoamine oxidase-B(MAO-B) activity, which destroyed the catecholamine related neurotransmitters in brain membranes of Sprague- Dawley (SD) rats. Male SD rats were fed basic diets (control group) and experimental diets (EtOAc-25, EtOAc-50 and EtOAc-100) for 45 days. Cholesterol accumulations in mitocholndria and microsomes were significantly inhibited (11.8-12.1% and 9.6-13.0%, respectvely) in EtOAc-50 and EtOAc-100 groups. ACh levels and ChAT activities were significantly increased about 10% in membranes of EtOAc-100 group compared with control group. AChE activities were significantly increased about 8 -12% in membranes of EtOAc-50 and EtOAc-100 groups compared with control group. MAO-B activities were significantly inhibited about 10% in membrane of EtOAc-l00 group compared with control group. These results suggest that ethyl acetate fraction of pine needle may play an effective role in inhibiting cholesterol and improving a membrane fluidity, and learning and memory impairments. Screening on Monoamine Oxidase B Inhibitor and Antioxidant Activity from the Water Extracts of Medicinal Plants (한약재 물 추출물의 Monoamine Oxidase B의 활성 억제 및 항산화 활성 검색)

  • This present study was designed to screen medicinal plants for the treatment of brain diseases such as Parkinson's disease or Alzheimer's disease. The inhibitory activity of monoamine oxidase B (MAO-B) was investigated in the water extracts of 56 species traditional medicines. Among the tested medicinal plants, E. lathyris, R. palmatum, F. rhynchonphylla, E. caryophyllata, E. pekinensis and H. syriacus were showed the strong inhibitory activity against MAO-B. Therefore, MAO-B inhibitory activity of 6 traditional medicine extracts in the different concentration (2.5, 6.5 and 12.5 ㎍/ml) was determined. The inhibitory effect of MAO-B was detected with dose dependently in 6 traditional plants extracts. E. caryophyllata and R. palmatum were showed the highest inhibitory activity, the MAO-B inhibitory activity at 2.5㎍ of herbal extract being 58% and 52%, respectively. The water extracts of 6 species were tested on antioxidant activity using radical scavenging effects against ABTS/sup +/. The water extracts of R. palmatum, E. caryphyllata, E. pekinensis and H. syriacus were showed strong antioxidant capacity at 20 ㎍ concentration. Among the 56 medicinal plants investigated, the water extracts of R. palmatum and E. caryphyllata were showed significant antioxidant capacity and MAO-B inhibiory activity. Therefore, R. palmatum and E. caryphyllata are expected to ameliorate the clinical symptoms in Parkinson's disease due to significant MAO-B inhibition and radical scavenging effect.

Effect of Selenium Yeast on MPTP (1-methyl-4-phenyl-propion-oxypiperidine)-Induced Neurotoxicity in Mice (Selenium이 MPTP(1-methy-4-phenyl-1,2,3,6-tetrahydropyridine)에 의해 유도된 생쥐의 신경독성에 미치는 영향)

  • This study is investigated the effect of selenium against neurotoxicity induced by MPTP(1-methy-4-phenyl-propion-oxypiperidine) in mice. In order to demonstrate neuroprotective activity of selenium, mice were administrated orally with selenium(25, 50, 100 ${\mu}g/kg$, once/day) for 10 days, and MPTP(10 mg/kg) was injected subcutaneously into the mice for 6 days from the beginning 1hr before selenium treatment. Test of rota road activity was inhibited by treatment with selenium in MPTP-induced neurotoxicity group when compared to MPTP treatment group in normal mice. Monoamine oxidase(MAO)-B activity and cerebral lipid peroxide content were significantly decreased in the treatment of selenium in MPTP-induced neurotoxicity group when compared to MPTP treatment group in normal mice and MAO-A was not affected. Activities of cerebral superoxide dismutase, catalase and glutathione peroxidase were significantly increased in the treatment of selenium in MPTP-induced neurotoxicity group when compared to MPTP treatment group in normal mice. These results suggest that selenium might be estimated the result from the cooperative action of its inhibitory effect on monoamine oxidase-B with that of the enhancement of antioxidant(SOD, catalase, GSH-Px) defence ability.

Effects of Acorn (Quercus acutissima CARR.) Supplementation on the Level of Acetylcholine and Its Related Enzyme Activities in the Brain of Dementia Mouse Model (도토리 급여가 치매모델 마우스 뇌조직의 아세틸콜린 및 관련효소 활성에 미치는 영향)

  • This study was designed to investigate the anti-dementia effects of acorn (Quercus acutissima CARR.) in brain of the mouse. Dementia model was induced by administration of scopolamin (30 mg/kg BW) Male ICR mouse $(30{\pm}2g\;BW)$ were fed basal diet (control group), and experimental diets (AP-5 and AP-10 groups) added $5\%\;and\;10\%$ of dried acorn powder to basal diet for 8 months. Acetylcholine content significantly increase in AP-5 and AP-10 groups ($4.2\%\;and\;11.3\%$, respectively) compared with control group. Acetylcholinesterase activities were significantly inhibited ($13.5\%\;and\;17.6\%$, respectively) in brain of AP-5 and AP-10 groups. Monoamine oxidase-B (MAO-B) activities were significantly inhibited ($10.0\%\;and\;12.7\%$, respectively) in brain of AP-S and AP-10 groups. These results suggest that acorn (Q. acutissima CARR.) may play an effective role in an attenuating various age-related changes such as dementia including learning and memory impairments in brain.

Supplementary Effects of Lentinus edodes with Different Harvest Period and Part on Neurotransmitters and Lipid Peroxide Levels in the Brain of Diabetic Mice (채취 시기 및 부위가 다른 표고버섯의 급여가 당뇨 마우스 뇌조직의 신경전달물질 및 지질과산화물 수준에 미치는 영향)

  • This study was designed to investigate the supplementary effects of Lentinus edodes which were harvested at different time period and part on acetylcholine content and its related enzyme activities in the brain of diabetic mouse model (KK mouse). We fed mice with standard diet (Control diet; CON) or 4 different kinds of experimental diets (DGC: on time harvested, cap of Dong Go; DGS: on time harvested, stipe of Dong Go; HSC: late harvested, cap of Hyang Sin: HSS: late harvested, stipe of Hyang Sin) to KK mouse for 8 weeks. Neurotransmitter such as acetylcholine contents, acetylcholinesterase activities, monoamine oxidase-B ac-tivities and lipid peroxide contents in the brain were measured. The results showed that acetylcholine content was significantly higher in DGC and HSC groups than CON group. The activities of acetylcholinesterase and monoamine oxidase-B enzyme were significantly inhibited in the brain of DGC and HSC groups compared with CON group. Lipid peroxide content was lower in DGC group than CON group. These results suggested that the cap of Lentinus edodes which were harvested on time and late time contain increased acetylcholine content and decreased acetylcholinesterase activities, monoamine oxidase-B activities and lipid peroxide contents. Thus the cap of Lentinus edodes which were harvested at different time periods may play an effective role in enhancing cognitive function.

Effects of Pine Needle Butanol Fraction on Acetylcholine (ACh) and Its Related Enzymes in Brain of Rats (뇌 조직의 아세틸콜린 및 그 관련효소에 미치는 솔잎(Pine Needle) 부탄올획분의 영향)

  • This study was designed to investigate the effects of buthanol (BuOH) fraction of pine (Pinus densiflora Sieb et Zucc) needle on cholesterol and lipofuscin (LF) accumulations, acetylcholine (ACh) and its related enzyme activities such as choline acetyltransferase (CAhT) and acetylcholinesterase (AChE), and monoamone oxidase-B (MAO-B) activity, which destroyed the catecholamine-related neurotransmitters in brain membranes of Sprague-Dawley (SD) rats. Male SD rats were fed basic diets (control group) or experimental diets (BuOH-25, BuOH-50 and BuOH-100) for 45 days. Cholesterol accumulations in mitochondria and microsomes were significantly inhibited (about 14 - 17% and 23 - 34%, respectvely) in BuOH-50 and BuOH-100 groups, whereas LF levels were significantly inhibited (about 10 - 14%) in BuOH-50 and BuOH-100 groups compared with control group. ACh levels and ChAT activities were significantly increased (about 11 - 17% and 11 - 23%, respectively) in membranes of BuOH-50 and BuOH-100 groups compared with control group. AChE activities were significantly increased (about 14 - 17%) in membranes of BuOH-50 and BuOH-100 groups. There was no significant difference in MAO-B activities between control and experimental diet groups. The results suggest that butanol fraction of pine needle may play an effective role in an antiaging effect and improving a learning and memory impairments.

Effect of Korean Traditional Tea Materials Water Extract on Hepatic Oxygen Free Radical Generating and Scavenging Enzyme Activities in Lead Administered Rats (한국전통차 재료의 열수추출물이 납투여 흰주의 간조직 중 유해 활성산소 생성과 제거효소 활성에 미치는 영향)

  • This study was designed to test the effect of Korean traditional tea materials on oxygen-free radical metabolism in lead (Pb) -administered rats. Male rats were divided into normal, Pb-control (Pb-Con) and Pb-water extract of green tea (Camellia sinensis; GT) , persimmon leaf (Diospyros kaki; PL) , safflower seed (Carhamus tinctorius: SS) , Du-Zhong (Eucommia ulmoides; EU) groups, respectively. Pb intoxication was induced by administration of lead acetate (25 mg/kg. B.W., oral) weekly. The extract was administered based on 1.26 g of raw material/kg B.W./day for 4 weeks. When the GT, PL, SS and EU were supplemented to the Pb-administered rats, hepatic lipid peroxide levels were significantly lower compared to the Pb-Con group. Hepatic cytochrom P-450 content and aminopyrine N-demethylase activity was lower in the Pb-Con group than in the normal group, whereas xanthine oxidase activity was significantly elevated in Pb-administered rats. The water extract of GT, PL, SS and EU supplementation attenuated changes in enzyme activities generating reactive oxygen species in the liver. Hepatic superoxide dismutase, catalase and glucose 6-phosphate dehydrogenase activities were significantly higher in the Pb-Con group than in the normal group, while monoamine oxidase activity also tended to increase in the Pb-administered rats. However, glutathione peroxidase and glutathione S-transferase activities, and glutathione content significantly decreased through Pb intoxication. The supplementation of GT, PL, SS and EU induced alleviation changes of hepatic antioxidant enzyme activity.

Effects of the Deer Antler Extract on Scopolamine-induced Memory Impairment and Its Related Enzyme Activities (녹용 추출물이 치매 동물모델의 기억력 개선과 관련효소 활성에 미치는 효과)

  • The aim of this study was to investigate the ameliorating effects of deer antler extract on the learning and memory impairments induced by the administration of scopolamine (2 mg/kg, i.p.) in rats. Tacrine was used as a positive control agent for evaluating the cognition enhancing activity of deer antler extract in scopolamine-induced amnesia models. The results showed that the deer antler extract-treated group (200 mg/kg, p.o.) and the tacrine-treated group (10 mg/kg, p.o.) significantly ameliorated scopolamine-induced amnesia based on the Morris water maze test. Although there was no statistical significance of brain ACh contents among the experimental groups, the brain ACh contents of the deer antler extract-treated group was slightly higher than that of the scopolamine-treated group. The inhibitory effect of deer antler extract on the acetylcholinesterase activity in the brain was significantly lower than that of scopolamine-treated group. The tacrine- and the deer antler-treated groups reduced the MAO-B activity compared to the scopolamine-treated group, but not significantly. These results suggest that the deer antler extract could be an effective agent for the prevention of the cognitive impairment induced by cholinergic dysfunction.
