The Performance of Anion Exchange Expanded Bed Adsorption Chromatography on the Recovery of G6PDH from Unclarified Feedstock with High Biomass Concentration

  • The bed stability of Streamline DEAE (p = 1.2 g/mL) in a 20mm (i.d.) glass expanded bed contactor, and its performance on the recovery of glucose 6-phosphate dehydrogenase (G6PDH) from unclarified yeast homogenate were investigated. A residence time distribution study showed that a stable expanded bed was achieved. The theoretical plate and Bodenstein numbers determined were 25 and 53, respectively. A recovery yield of 87% and purification factor of 4.1 were achieved in the operation using 5% (w/v) biomass concentration feedstock. The performance of the anion exchange EBAC was still considerable good at a biomass concentration as high as 15% (w/v).

  • The surface electronic state of rutile $TiO_2$, which is an oxide semiconductor and has a wide band gap of 3.1 $\sim$ 3.5 eV, was calculated by DV-$X_{\alpha}$ method, which is a sort of the first principle molecular orbital method and uses Hartre-Fock-Slater approximation. The $[Ti_{15}O_{56}]^{-52}$ cluster model was used for the calculation of bulk state and the $[OTi_{11}O_{34}]^{-24}$ model for the surface state calculation. After calculations, the energy level diagrams and the deformation electron density distribution map were compared in both models. As results, it was identified that the surface energy levels are found between the valence and conduction band of bulk $TiO_2$ on the surface area. The energy values of these surface-induced levels are lower than conduction band of bulk $TiO_2$ by 0.1 $\sim$ 1 eV. From this fact, it is expected that the surface energy levels act as donar levels in n-type semiconductor.

  • In this study, the TSC spectroscopy has been applied to investigate the influence of structural change due to a process of curing reaction on the electrical properties of epoxy composites cured with acid-anhydride. Five TSC peaks appeared in -160-250[.deg.C]: in the low temperature region below glass transition temperature(T$\_$g/), three relaxation mode peaks due to action of side chains, substitution group or terminal groups have been observed, a peak associated with T$\_$g/, appeared in 110[.deg. C] and p peak due to ionic space charges located in 150[.deg.C]. Each peak was separated into elementary peaks by the partial polarization procedure, and the distribution of activation energy and relaxation time were analized to clearify the origin of each peak. Also, overaboundantly added hardener separated a .betha. peak near 10[.deg. C] into two peaks of .betha.$\_$1/(10.deg. C) and .betha.$\_$2/(20.deg. C) according to increasement of forming field, and the separated hardener was oxidated thermally with increasing surrounding temperatures. The expansion of the free volume need in molecular motion and the reduction of the structural packing density through thermal oxidation process increased TSC between .alpha. peak and .betha. peak and decreased T$\_$g/.

Investigation of Surface Degradation in Silicone Rubber Due to Corona Exposure (코로나 방전에 노출된 고분자 애자용 실리콘 고무의 표면열화)

  • In this paper we investigated the characteristics of surface degradation in silicone rubber due to corona exposure and recovery mechanism. It was shown that surface free energy was 22.42 mJ/$m^2$ on initial sample but surface free energy was approximately increased to 71.14 mJ/$m^2$ after 45 minutes. However, surface free energy on silicone rubber after corona discharge treatment was completely recovered within a short time due to diffusion of low molecular weight(LMW) silicone fluid. It was shown that corona discharge insured the increase of diffusible LMW chains, which could lead to recover the surface hydrophobicity. 200~370 g/mol distribution of LMW silicone fluid which was extracted by solvent-extraction with gel permeation chromatography (GPC) was contributed to recovery. The surface degradation characteristics on silicone rubbers and the recovery mechanism based on our results were discussed.

  • Small GTPases are well known as one of the signal transduction factors of immune systems. The ADP-ribosylation factors (ARFs) can be classified into three groups based on the peptide sequence, protein molecular weight, gene structure, and phylogenetic analysis. ARF1 recruits coat proteins to the Golgi membranes when it is bound to GTP. The class I duplicated ARF gene was cloned and characterized from the olive flounder (Paralichthys olivaceus) for this study. PoARF1b contains the GTP-binding motif and the switch 1 and 2 regions. PoARF1b and PoARF1b mutants were transfected into a Hirame natural embryo cell to determine the distribution of its GDP/GTP-bound state; consequently, it was confirmed that PoARF1b associates with the Golgi body when it is in a GTP-binding form. The results of the qPCR-described PoARF1b were expressed for all of the P. olivaceus tissues. The authors plan to study the gene expression patterns of PoARF1b in terms of immunity challenges.

Ultrafiltration Characteristics of Hot-Water Extracts from Siberian Larch Barks for Tannin-based Adhesives (타닌접착제를 위한 시베리아산 낙엽송 수피 온수추출물의 한외여과 특성)

  • Hot-water soluble extracts were prepared from medium-sized barks of Siberian larch (Larix gmelineii). The extracts were fractionated by ultrafiltration method for producing uniform quality of adhesives. Molecular weight distribution of the extracts was ranged of 100 to 300.000. pH of the extracts affected amounts of flux, and the range of pH 6~pH 8 was proper for wood adhesives, because of easy concentration and fast gelation time. Removal of particles greater than $0.45{\mu}m$ from the extracts increased both filtration speed (flux) and yields of solids in the filtrates. In ultrafiltration process. operating pressure, filtrate temperature, and flow rate significantly increased with the increase of individual condition. Ultrafiltration using PM10 membrane was very effective to fractionate and concentrate the extracts. Removal of large particles greater than 0.45 m from the extracts increased filtration speed(flux) and yields of solids in the filtrates. A gelation time was accelerated with the increasing pH of the extracts and its concentration. The Stiasny precipitate(26%) from the filtrate obtained by PM 10 membrane was very lower than that(78%) of the retentates. This ultrafiltration method was efficient for obtaining high yield purified phenolic compounds.

  • The aim of this paper is to investigate removal of salts in hydrolyzed sericin solution by a prepared nanofiltration membrane. The nanofiltration membrane was prepared by the interfacial polymerization method, and then compared with commercialized nanofiltration membrane in terms of salts removal. The sericin solution was hydrolyzed by a protease. The molecular weight distribution of the hydrolyzed sericin was determined by electrophoresis method. Relative flux measurements were carried out to investigate membrane fouling.

Molecular Identification of Endophytic Fungi Isolated from Needle Leaves of Pinus thunbergii (곰솔(Pinus thunbergii)의 침엽에서 분리한 내생균의 분자적 동정)

  • Endophytic fungi were isolated from needle leaves of Pinus thunbergii distributed in Boryeong of Korea. Total 7 species of fungi were identified using partial sequences of ITS and LSU regions of nuclear DNA Anthostomella sepelibilis, Beltrania sp., Cladosporium sp., Coniochaeta velutina, Entonaema pallida, Lophodermium sp. and Nigrospora sphaerica sphaerica. The endophytic fungi, Beltrania sp. was the most dominant species isolated from P. thunbergii in this study. Results indicate that the distribution of endophytic fungi in the leaves of P. thunbergii could be influenced by environments of the fungal habitat.

Hyperthermal Collision-induced Dissociation of Bromotoluene Radical Cations at Self-Assembled Monolayer Surfaces

  • Hyperthermal ion/surface collisions of bromotoluene radical cations were studied using perfluorinated (F-SAM) and hydroxyl-terminated (OH-SAM) self-assembled monolayer surfaces in a tandem mass spectrometer with BEEQ geometry. The isomers were differentiated by ion abundance ratios taken from surface-induced dissociation (SID). The dissociation rate followed the order of ortho > meta > para isomers. The peak abundance ratio of m/z 51 to m/z 65 showed the best result to discern the isomers. A dissociation channel leading to tolylium ion was suggested to be responsible for the pronounced isomeric differences. The capability of SID to provide high-energy activation with narrow internal energy distribution may have channeled the reaction into the specific dissociation pathway, also facilitating small differences in reaction rates to be effective in the spectral time window of this experiment. All of the molecular ions experiencing reactive collisions with the F-SAM surface undergo transhalogenation, in which a fluorine atom on the surface replaces the bromine in the incoming ions. This reactive collision was dependent on the laboratory collision energy occurring in ca. 40.75 eV range.

Molecular Characterization of a Bombyx mori Protein Disulfide Isomerase(bPDI) (누에 배양세포로부터 분리한 Protein Disulfide Isomerase 유전자의 발현 특성)

  • Many secreted proteins have disulfide bonds that are important for their structure and function. Protein disulfide isomerase (PDI, EC 5.3.1.4.), an enzyme that catalyzes the formation and rearrangement of thiol/disulfide exchange reactions, is a resident of the endoplasmic reticulum (ER). The subcellular localization and its function as catalyst of disulfide bond formation in the biosynthesis of secretory and cell membrane proteins suggest that PDI plays a key role in the secretory pathway. We have isolated a cDNA encoding protein disulfide isomerase from Bombyx mori(bPDI). It has been characterized under ER stress conditions (dominantly induced by calcium ionophore A23187, tunicamycin and DTT), which is known to cause an accumulation of unfolded proteins in the ER. Furthermore, It has also been examined for tissue distribution(pronounced at the fat body), hormonal regulation (juvenile hormone, insulin and juvenile +transferrin; however, it is not effected by transferrin alone), and the effect of exogenous bacteria (peak at 16 h after infection) on the bPDI mRNA expression. The results suggest that bPDI is a member of the ER stress protein group, and it may play an important role in exogenous bacterial infection in fat body, and that homones regulate its expression.
