Transdermal Permeation Behavior of 5-FU using Microneedle (마이크로니들을 이용한 5-FU의 경피투과 거동)

  • 5-Fluorouracil (5-FU) is an antimetabolic of the pyrimidine derivatives that is used in chemotherapy for the treatment of several types of cancer. 5-FU have poor oral absorption and short biological half-time and strong side effects. Microneedle introduced to find a solution of problems. Microneedle device with roll was manufactured for transdermal delivery of various drugs. 5-FU was mixed in non-ionic surfactant such as tween 20 and tween 80. Camscope was used to analysis the permeation magnitude of treated skin by microneedle and trypan blue staining. The 5-FU solution with surfactant measured by ZETA-potential analysis system for stability of solution. The skin permeation rate of 5-FU determined by HPLC. We confirmed that cross treated skin was dyed more deeply than parallel treated skin through trypan blue staining. The results indicate that skin permeation rate of 5-FU was increased with the treatment types and treatment times.

A Seven Cases of Treating Atrophic Acne Scar with Subcision, Microneedle Therapy System and Herbal Peeling Therapy (절개침과 미세피부침, 약초필링을 이용한 위축성 여드름 흉터 치료 7례 임상보고)

  • Objectives : The purpose of this study is to present the effect of subcision, microneedle therapy system and herbal peeling therapy which is treatment of Korean medicine to atrophic acne scar patients. Methods : We administered subcision, microneedle therapy system and herbal peeling therapy to atrophic acne scar patients and tested the results by Qualitative Global Acne Scarring Grading System and Korean Acne Grading System as well. Results : After the treatment, all of the patients showed improvements in Qualitative Global Acne Scarring Grading System and Korean Acne Grading System as well. Qualitative Global Acne Scarring Grading System of patients changed from grade 2.71±0.71 to grade 1.14±0.86 along with the change of Korean Acne Grading System from grade 1.57±0.57 to grade 1.00±0.00. Conclusions : Subcision, microneedle therapy system and herbal peeling therapy may be effective in the treatment of moderate to severe acne scars, and could be applied in clinical practice.

  • Microstereolithography technology has potential capability for fabrication of 3D microstructures. It evolved from conventional SLA which is one of the RP processes. In a microstereolithography process, 3D microstructures can be easily fabricated by continuously stacking 2D layer which is photopolymerized using a liquid prepolymer. Combination between biocompatible/biodegradable photocurable prepolymer and 3D complex fabrication in microstereolithography makes broad application areas such as medical, pharmaceutic, and bio devices. In particular, a 3D microneedle for transdermal drug delivery and a scaffold for tissue engineering are fabricated using this technology. In this paper, the authors address development of microstereolithography system adapted to large surface and fabrication of various microstructures. In addition, to apply human body we suggest a biodegradable 3D microneedle and a scaffold using biodegradable photocurable prepolymer.

  • Background & Objective : Microneedle therapy system(MTS) is popular these days. However, there are few reports about it in oriental medicine academia. This study is performed to evaluate the effect of MTS on acne patients. Methods : This study was conducted to evaluate the effect of MTS. Seven patients were treated for 7 weeks. Follow-up was performed after the treatment with Janus facial analysis system, skindex-29 and Digital Infrared Thermographic Imaging machine. Satisfaction degree and inconvenience were asked from the patients. Results and Conclusions : Microneedle therapy system(MTS) was good for acne like pore, wrinkle, spot(polarised), sebum, porphyrin and skin tone. Especially it had excellent effects on sebum and porphyrin.

  • Transdermal drug delivery device is a method of drug delivery through the skin. Skin has a very large area, so it is attractive route to drug delivery. When drug delivery through the skin, microneedle has a advantage that painless, constant drug deliver and penetration efficient; nevertheless the cost is expensive because fabrication process need a particular equipment and not suitable in mass production. This study shows microneedle fabrication process using convergence of general MEMS process and dicing process that can make 3-D sharp microneedle tip and this fabrication process suitable for mass production.

  • Conventional drug delivery methods mainly include subcutaneous injection and oral administration. Subcutaneous injection has the advantages of delivering the correct concentration, but it might cause pain and trauma to patients. Although oral administrations do not accompany pain as the subcutaneous injection, unexpected side effects may occur because they undergo digestion process and it is not easy for many drugs to be exposed to targeted sites with proper concentration. While dissolving microneedles have been extensively studied to overcome the limitations of conventional subcutaneous injection, the effects on microneedle shapes for drug release have not been fully explored. In this study, the characteristics of hyaluronic acid microneedles for cone and quadrangular pyramidal shapes were examined by the size, volume, contact surface area, skin permeability, and dissolution rate. As a result, the cone shapes of hyaluronic acid microneedles had high skin permeability, and the quadrangular pyramid of shapes hyaluronic acid microneedles showed a fast dissolution rate, given by the large contact area on the skin. Based on the results, we could confirm that the shape of a dissolving microneedles can affect skin permeability and the drug delivery rate.

  • The feasibility of lecithin as a material for dissolving microneedles to improve skin permeability of drugs and the effect of the composition of lecithin on the mechanical strength, solubility, and skin permeability of rhodamine B (RhB) of the dissolving microneedles were investigated. Dissolving microneedles with needles of uniform shape and size were fabricated with the mold made using the laser-writing technique, simpler and more efficient method compared to the photolithography method, the conventional method to fabricate the microneedle mold. The composition of lecithin in the microneedle affected the mechanical strength and solubility of the needle thus, the mechanical strength of the needle increased as the composition of lecithin in the needle increased, resulting in improving the skin permeability of RhB contained in the microneedles. When comparing the microneedles containing same composition of amylopectin (AP), the skin permeability of RhB of the microneedles containing lecithin was higher. These results indicate that lecithin can be used as a material for dissolving microneedles and the skin permeability of the microneedle could be controlled by changing the composition of lecithin.

  • Objectives This clinical study is aimed to examine the effects of herbal medication and pharmacopunture on child with different types of alopecia. Methods 13 year old girl was treated with Hominis placenta pharmacopuncture microneedle therapy system and Ganoderma lucidum pharmacopuncture mesotherapy on the alopecia lesion for 20 months. Herbal medication, meridian pharmacopuncture therapy and physiotherapy were also used to reduce physical symptoms. Results The patient had a relapse of hair loss with changing alopecia types, but eventually alopecia has been resolved and maintained throughout the Korean medicine treatment. Conclusions This study shows the effect of the Korean medical treatments for a pediatric patient with alopecia. And more research is needed to study about patients with different types of alopecia.

  • Background & Objective : Rejuvenate treatment with oriental medicine is popular these days. However, there are few reports about it. This study was performed to evaluate the effect of derma stamp treatment. Derma stamp is a new clinical equipment for Microneedle System. Methods : Restrospective study was conducted. Ten patients were treated for 5 weeks. Follow-up was performed after the treatment with Janus facial analysis system. Satisfaction degree was asked from the patients. Results and Conclusions : Derma stamp treatment was good for skin aging problems like pore, wrinkle, spot, sebum and porphyrin. Especially oily type had better effect than dry type.
