Circulating miR-195 as a Therapeutic Biomarker in Turkish Breast Cancer Patients

  • Background: Dysregulation of miRNA expression may be used as a biomarker for specific tumours because it may contribute to development of cancer. Circulating miRNA profiles have been highlighted for their potential as predictive markers in heterogeneous diseases such as breast cancer. In the literature, there is evidence that miR-195 levels are differentially expressed pre- and post-operative periods in breast cancer patients. At the same time, miRNA expression levels may vary because of ethnic origins. This study aimed to determine expression levels and potential roles of miR-195 in Turkish breast cancer patients. Materials and Methods: The expression patterns of miR-195 were initially examined in breast cancer tissues (luminal A and B type) (n=96). Subsequently, blood samples were prospectively collected from preoperative and postoperative Turkish breast cancer patients and disease free controls. Total RNA was isolated, and the expression level of miR-195 was quantified by real-time PCR. Results: We found that miR-195 level was altered in Turkish breast cancer patients, with down-regulation evident in breast cancer tissues compared to normal adjacent specimens. Furthermore, circulating levels of miR-195 was significantly decreased in post-operative blood samples compared with pre-operative levels (p=0.01 and <0.05). However, miR-195 was significantly increased in pre-operative blood samples of the luminal B type (p=0.04 and <0.05). Conclusions: This study represents the first report of a miR-195 expression profile in Turkish breast cancer patients. Our data suggests that miR-195 levels might be a clinically useful biomarker in the earliest stage of Turkish breast cancer patients.

Expression Analyses of MicroRNAs in Hamster Lung Tissues Infected by SARS-CoV-2

  • Coronavirus disease 2019 (COVID-19), caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), is an infectious disease with multiple severe symptoms, such as fever over 37.5℃, cough, dyspnea, and pneumonia. In our research, microRNAs (miRNAs) binding to the genome sequences of severe acute respiratory syndrome coronavirus (SARS-CoV), Middle East respiratory-related coronavirus (MERS-CoV), and SARS-CoV-2 were identified by bioinformatic tools. Five miRNAs (hsa-miR-15a-5p, hsa-miR-15b-5p, hsa-miR-195-5p, hsa-miR-16-5p, and hsa-miR-196a-1-3p) were found to commonly bind to SARS-CoV, MERS-CoV, and SARS-CoV-2. We also identified miRNAs that bind to receptor proteins, such as ACE2, ADAM17, and TMPRSS2, which are important for understanding the infection mechanism of SARS-CoV-2. The expression patterns of those miRNAs were examined in hamster lung samples infected by SARS-CoV-2. Five miRNAs (hsa-miR-15b-5p, hsa-miR-195-5p, hsa-miR-221-3p, hsa-miR-140-3p, and hsa-miR-422a) showed differential expression patterns in lung tissues before and after infection. Especially, hsa-miR-15b-5p and hsa-miR-195-5p showed a large difference in expression, indicating that they may potentially be diagnostic biomarkers for SARS-CoV-2 infection.

miR-195/miR-497 Regulate CD274 Expression of Immune Regulatory Ligands in Triple-Negative Breast Cancer

  • Purpose: Immune suppression is common in patients with advanced breast cancer but the mechanisms underlying this phenomenon have not been sufficiently studied. In this study, we aimed to identify B7 family members that were able to predict the immune status of patients, and which may serve as potential targets for the treatment of breast cancer. We also aimed to identify microRNAs that may regulate the expression of B7 family members. Methods: The Cancer Genome Atlas data from 1,092 patients with breast cancer, including gene expression, microRNA expression and survival data, were used for statistical and survival analyses. Polymerase chain reaction and Western blot were used to measure messenger RNA and protein expression, respectively. Luciferase assay was used to investigate direct microRNA target. Results: Bioinformatic analysis predicted that microRNA (miR)-93, miR-195, miR-497, and miR-340 are potential regulators of the immune evasion of breast cancer cells, and that they exert this function by targeting CD274, PDCD1LG2, and NCR3LG1. We chose CD274 for further investigations. We found that miR-195, miR-497, and CD274 expression levels were inversely correlated in MDA-MB-231 cells, and miR-195 and miR-497 expressions mimic inhibited CD274 expression in vitro. Mechanistic investigations demonstrated that miR-195 and miR-497 directly target CD274 3' untranslated region. Conclusion: Our data indicated that the level of B7 family members can predict the prognosis of breast cancer patients, and miR-195/miR-497 regulate CD274 expression in triple negative breast cancer. This regulation may further influence tumor progression and the immune tolerance mechanism in breast cancer and may be able to predict the effect of immunotherapy on patients.

MiR-186 Inhibited Migration of NSCLC via Targeting cdc42 and Effecting EMT Process

  • In this study, qRT-PCR was employed to identify that miR-186 expression level in NSCLC tissues are highly associated with lymph node metastasis. In addition, through the application of western blotting, luciferase assay and qRT-PCR, it was found that miR-186 targeted 3'UTR of cdc42 mRNA and down-regulated cdc42 protein level in a post-transcriptional manner. Transwell assay indicated that cdc42 partially reversed the effect of miR-186 mimics. Besides, miR-186 was proved to regulate EMT by influencing biomarkers of this process and cell adhesion ability. Thus, miR-186 is a potential target for NSCLC therapy. miR-186 is proposed to be one of tumor-suppressors and may serve as a therapeutic target in NSCLC treatment.

  • Gastric cancer is generally associated with poor survival rates and accounts for a remarkable proportion of global cancer mortality. The prevalence of gastric carcinoma varies in different regions of world and across teh various ethnic groups. On the basis of pathological assessment, gastric cancer can be categorized as intestinal and diffuse carcinomas. The etiology is diverse, including chemical carcinogen exposure, and high salt intake Helicobacter pylori also plays a vital role in the pathogenesis of certain gastric carcinomas. The development of gastric cancer involves various alterations in mRNAs, genes (GOLPH3, MTA2) and proteins (Coronins). miRNAs, Hsa-mir-135b, MiR-21, miR-106b, miR-17, miR-18a, MiR-21, miR-106b, miR-17, miR-18a and MiRNA-375, miRNA-195-5p are the latest diagnostic biomarkers which can facilitate the early diagnosis of gastric carcinomas. Recent development in the treatment strategies for gastric carcinoma include the introduction of monoclonal antibodies, TKI inhibitors, inhibitors of PDGFR ${\beta}$, VEGFR-1, VEGFR-2, Anti-EGFR and anti-HER2 agents which can be applied along with conventional therapies.

Alteration in miRNA Expression Profiling with Response to Nonylphenol in Human Cell Lines

  • Exposures to environmental chemicals that mimic endogenous hormones are proposed for a number of adverse health effects, including infertility, abnormal prenatal and childhood development and above all cancers. In addition, recently miRNA (micro RNA) has been recognized to play an important role in various diseases and in cellular and molecular responses to toxicants. In this study, endocrine disrupting environmental toxicant, nonylphenol (NP) was treated to MCF-7 (Human breast cancer cell) and HepG2 (Human hepatocellular liver carcinoma) cell line at 3 hrs and 48 hrs time point and miRNA analysis using $mirVana^{TM}$ miRNA bioarray was performed and compared with total mRNA microarray data for the same cell line and treatment. Robust data quality was achieved through the use of dye-swap. Analysis of microarray data identifies a total of 20 and 11 miRNA expressions at 3 hrs and 48 hrs exposure to NP in MCF-7 cell line and a total of 14 and 47 miRNA expression at 3 hrs and 48 hrs exposure respectively to NP in HepG2 cell line. Expression profiling of the selected miRNA (let-7c, miR-16, miR-195, miR-200b, miR200c, miR-205, and miR-589) reveals changes in the expression of target genes related to metabolism, immune response, apoptosis, and cell differentiation. The present study can be informative and helpful to understand the role of miRNA in molecular mechanism of chemical toxicity and their influence on hormone dependent disease. Also this study may prove to be a valuable tool for screening potential estrogen mimicking pollutants in the environment.

  • Slim813은 2-사이클로펜텐-1-온 옥심 유도체로서 다양한 염증성 자극원에 대한 TNF 생성을 억제하는 항 자극 및 UV에 의한 MMP1 억제를 통한 항 노화 효능을 갖는 물질이다. 본 연구를 통하여 이와 같은 효능 외에도 완전히 분화된 지방 세포에서 cAMP (cyclic adenosine monophosphate) 생성 촉진을 통한 HSL (hormone sensitive lipase) 활성을 증대시킴으로써 지방분해를 증가시킴을 확인하였다. 또한 Slim813을 사람의 팔과 허벅지 부위에 2주간 도포하였을 때 지방의 두께 감소를 유발하는 것으로 나타나 체내 피하 지방을 분해하는 효능을 갖는 것으로 사료된다.

스마트폰 과다사용이 사이버불링에 미치는 영향에 대한 융합 연구 : 공격성의 매개효과를 중심으로 (The convergence study of smartphone overuse on cyberbullying : Focusing on mediating effects of aggression)

  • 본 연구는 고등학생의 스마트폰 과다사용과 사이버불링의 관계에서 공격성의 매개효과를 확인하기 위해 진행되었다. K시에 거주하는 고등학생 328명을 대상으로 자가보고식 설문지를 이용하여 자료를 수집하였으며, 수집된 자료는 SPSS 22.0과 AMOS 22.0을 이용하여 분석하였다. 분석결과 사이버불링은 스마트폰 과다사용(r=.321, p<.001), 공격성(r=.397, p<.001)과 정적 상관관계가 있는 것으로 확인되었다. 스마트폰 과다사용은 공격성(${\beta}=.195$, p=.014)에 직접적인 영향을 미치고, 공격성은 사이버불링(${\beta}=.260$, p=.007)에 직접적인 영향을 미치는 것으로 나타났다. 또한 스마트폰 과다사용은 공격성(${\beta}=.051$, p=.007)을 매개로 하여 사이버불링에 간접적인 영향을 미쳤다. 고등학생의 사이버불링을 감소시키기 위해 스마트폰 과다사용에 대한 규제와 함께 공격성 감소를 위한 사이버 윤리의식 교육 프로그램 개발 등이 필요할 것이라 생각된다.

남자간호사의 대인관계능력과 직무 스트레스가 간호업무성과에 미치는 영향 (Influence of Interpersonal relation and Job Stress on Nursing Performance of Male Nurses)

  • Purpose: The purpose of this study was to identify the effect of interpersonal relation and job stress on nursing performance among male nurses. Methods: This study was cross-sectional descriptive one. The data were collected from self-administered questionnaires by 110 male nurses members affiliated with the Korean Man Nurses Association. The data were analyzed using descriptive statistics, independent t-test, one way ANOVA with Duncan test, Pearson correlation, and stepwise multiple regression with the SPSS/WIN 21.0. Results: The average scores of interpersonal relation, job stress, and nursing performance were respectively 3.64, 3.10, 3.63. Nursing performance was high in married, good health condition, manager, and high salary among male nurses. Nursing performance showed significant positive correlation with interpersonal relation (p<.001), and a significant negative correlation with job stress (p=.010). Influencing factors of nursing performance among male nurses were interpersonal relation, health status, total duration of career, position, and the adjusted $R^2$ of the regression model was 42%. Conclusion: The results suggest that we need to develop a training program to improve the nursing performance of the male nurses.

Effects of Oral Rutaecarpine on the Pharmacokinetics of Intravenous Chlorzoxazone in Rats

  • It has been reported that hepatic microsomal cytochrome P450(CYP) 2E1 is responsible for the metabolism of chlorzoxazone(CZX) to 6-hydroxychlorzoxazone. The present study was undertaken to assess the possible interaction of rutaecarpine, an alkaloid originally isolated from the unripe fruit of Evodia rutaecarpa, with CZX. Male Spraque-Dawley rats were administered with 80 mg/kg/day of oral rutaecarpine for three consecutive days. Twenty four hr after the pre-treatment with rutaecarpine, the rats were treated with 20 mg/kg of intravenous CZX. Rat hepatic microsomes isolated from rutaecarpine-treated rats showed greater(50% increase) activity of p-nitrophenol hydroxylase(a marker of CYP2E1) when compared with the control rats. Compared with control rats, the AUC of CZX was significantly smaller(84% decrease) possibly due to significantly faster CL(646% increase) in rats pretreated with rutaecarpine. This could be, at least partially, due to induction of CYP2E1 by rutaecarpine.
