No short-term effects of calorie-controlled Mediterranean or fast food dietary interventions on established biomarkers of vascular or metabolic risk in healthy individuals

  • BACKGROUND/OBJECTIVES: This study addressed the question whether the composition of supposedly 'healthy' or 'unhealthy' dietary regimes has a calorie-independent short-term effect on biomarkers of metabolic stress and vascular risk in healthy individuals. SUBJECTS/METHODS: Healthy male volunteers (age $29.5{\pm}5.9years$, n = 39) were given a standardized baseline diet for two weeks before randomization into three groups of different dietary regimes: fast food, Mediterranean and German cooking style. Importantly, the amount of calories consumed per day was identical in all three groups. Blood samples were analyzed for biomarkers of cardiovascular risk and metabolic stress after two weeks of the baseline diet and after two weeks of the assigned dietary regime. RESULTS: No dietary intervention affected the metabolic or cardiovascular risk profile when compared in-between groups or compared to baseline. Subjects applied to the Mediterranean diet showed a statistically significant increase of uric acid compared to baseline and compared to the German diet group. Plasma concentrations of urea were significantly higher in both the fast food group and the Mediterranean group, when compared to baseline and compared to the German diet group. No significant differences were detected for the levels of vitamins, trace elements or metabolic stress markers (8-hydroxy-2-deoxyguanosine, malondialdehyde and methylglyoxal, a potent glycating agent). Established parameters of vascular risk (e.g. LDL-cholesterol, lipoprotein(a), homocysteine) were not significantly changed in-between groups or compared to baseline during the intervention period. CONCLUSIONS: The calorie-controlled dietary intervention caused neither protective nor harmful short-term effects regarding established biomarkers of vascular or metabolic risk. When avoiding the noxious effects of overfeeding, healthy individuals can possess the metabolic capacity to compensate for a potentially disadvantageous composition of a certain diet.

좀개구리밥(Lemna gibba G3)의 개화에 미치는 Polyamine의 영향 (Effects of Polyamine on Flowering in Lemna gibba G3)

  • 장일식물인 Lemna gibba G3의 개화유도과정에 있어서 polyamine의 관련성을 조사하였다. 이 식물의 개화는 연속광하에서 agmatine, putrescine, spermidine 및 spermine에 의해 촉진되었으며 이들의 체내 합성억제물질인 methylglyoxal-bis(guanylhydrazone)(MGBG)과 cyclohexylamine(CHA)에 의해서 억제되었다. Polyamine은 영양생장률을 거의 일정한 수준으로 유지하면서 개화를 촉진시켰으며, 억제물질에 의한 개화억제는 오히려 영양생장률의 증가를 수반하였다. 이러한 현상은 이 물질들의 개화촉진 또는 억제효과가 생장률의 일반적인 촉진 또는 억제효과에 의한 것이 아니라 선택적으로 개화과정에 작용한 결과일 것이라는 추측을 뒷받침해 준다. 배양액의 첨가된 spermidine 또는 spermine의 개화촉진 폭은 배양액에 이들의 억제물질의 함유여부와 관계없이 거의 일정한 수준으로 나타났다. 이렇듯, 외부에서 공급된 후 체내에 흡수된 spermidine과 spermine의 개화에 대한 기여도가 한정되어 있는 것으로 보아 L. gibba G3의 개화유도과정이 식물체내에서 합성되는 spermidine과 spermine에 크게 의존하고 있을 것으로 판단된다. 한편, 개화유도과정이 시작되어 점차적으로 증가한 체내의 spermidine 함량은 개화가 유도되지 않은 경우에 비하여 24시간만에 약 2배의 수준에 도달하였다. 이렇듯 체내의 spermidine량이 급격히 증가된 사실은 체내의 polyamine의 질적 및 양적 변화가 L. gibba G3의 개화유도초기과정에서 매우 중요한 역할을 하고 있을 것이라는 가정을 뒷받침하는 또 하나의 예비적 증거라 할 수 있다.

Chemical Characterization of Water-Soluble Organic Acids in Size-Segregated Particles at a Suburban Site in Saitama, Japan

  • Saturated n-dicarboxylic acids ($C_2-C_7$, $C_9$), unsaturated dicarboxylic acids (maleic, fumaric, phthalic acid), ketocarboxylic acids (pyruvic, glyoxylic acid), and dicarbonyls (glyoxal, methylglyoxal) were determined in size-segregated samples with a high-volume Andersen air sampler at a suburban site in Saitama, Japan, May 12-17 and July 24-27, 2007 and January 22-31, 2008. The seasonal average concentrations of these detected organic acids were 670 $ng/m^3$, accounting for about 4.4-5.7% (C/C) of water-soluble organic carbon (WSOC) and 2.3-3.6% (C/C) of organic carbon (OC). The most abundant species of dicarboxylic acids was oxalic acid, followed by malonic, phthalic, or succinic acids. Glyoxylic acid and methyglyoxal were most abundant ketocarboxylic acid and dicarbonyl, respectively. Seasonal differences, size-segregated concentrations, and the correlations of these acids with ambient temperatures, oxidants, elemental carbon (EC), OC, WSOC, and ionic components were also discussed in terms of their corresponding sources and possible secondary formation pathways. The results suggested that photochemical reactions contributed more to the formation of particulate organic acids in Saitama suburban areas than did direct emissions from anthropogenic and natural sources. However, direct emissions of vehicles were also important sources of several organic acids in particles, such as phthalic and adipic acids, especially in winter.

Production of 1,2-Propanediol from Glycerol in Saccharomyces cerevisiae

  • Glycerol has become an attractive carbon source in the biotechnology industry owing to its low price and reduced state. However, glycerol is rarely used as a carbon source in Saccharomyces cerevisiae because of its low utilization rate. In this study, we used glycerol as a main carbon source in S. cerevisiae to produce 1,2-propanediol. Metabolically engineered S. cerevisiae strains with overexpression of glycerol dissimilation pathway genes, including glycerol kinase (GUT1), glycerol 3-phosphate dehydrogenase (GUT2), glycerol dehydrogenase (gdh), and a glycerol transporter gene (GUP1), showed increased glycerol utilization and growth rate. More significant improvement of glycerol utilization and growth rate was accomplished by introducing 1,2-propanediol pathway genes, mgs (methylglyoxal synthase) and gldA (glycerol dehydrogenase) from Escherichia coli. By engineering both glycerol dissimilation and 1,2-propanediol pathways, the glycerol utilization and growth rate were improved 141% and 77%, respectively, and a 2.19 g 1,2- propanediol/l titer was achieved in 1% (v/v) glycerolcontaining YEPD medium in engineered S. cerevisiae.

In silico Analysis of Natural Compounds as Modulators of Type I Collagen

  • Collagen plays a vital role in the maintenance of structure and function of a human body. It has been widely applied in various fields including biomedical, cosmeceutical, food, pharmaceutical and tissue engineering. In the present study, the docking behaviour of type I collagen with 15 different ligands namely hydroxymethylfurfural, methylglyoxal, methylsyringate, O-methoxyacetophenone, 3-phenyllactic acid, 4-hydroxybenzoic acid, kojic acid, lumichrome, galangin, artoindonesianin F, caffeic acid, 4-coumaric acid, origanol A, thymoquinone and quercetin was evaluated along with their putative binding sites using Discovery Studio Version 3.1. Docking studies and binding free energy calculations revealed that origanol A has maximum interaction energy (-40.48 kcal/mol) and quercetin with the least interaction energy (-15.44 kcal/mol) as compared to the other investigated ligands. Three ligands which are galangin, methylsyringate and origanol A were shown to interact with Asp21 amino acid residue of chain B (type I collagen). Therefore, it is strongly suggested that the outcomes from the present study might provide new insight in understanding these 15 ligands as potential type I collagen modulators for the prevention of collagen associate disorders.

Chlamydomonas의 Polyamine 함량변화와 엽록체 DNA Methylation (Changes in Polyamine Level and Chloroplast DNA Methylation in Chlamydomonas reinhardtii)

  • Chlamydomonas reinhardtii 배우체 유도기 동안에 polyamine 생합성 억제제인 MGBG[methylglyoxal bis-(guanylhydrazone); SAMDC 억제제] 1mM 처리구와 정상 대조구에서의 polyamine 함량 변화와 Chlamydomonas의 ct-DNA methylation과의 상호 연관성을 조사한 결과, 전반적으로 137C(+)와 137C(-)의 정상 대조구에서 polyamine 수준은 배우체 유도기 전반에 걸쳐서 감소추세로 나타났으나, 특히 137C(+)가 137C(-)보다 polyamine 함량이 많았으며 동시에 감소량도 컸다. 1mM MGBG 처리구에서는 137C(+)의 spermidine 함량이 정상 대조구보다 증가하였다. In vitro에서 ctDNA methylation에 대한 MGBG의 영향은 정상 대조구에서보다도 1mM MGBG 처리구에서 약 20-30%의 ctDNA methylation 감소를 나타내었다. 또한, MGBG는 in vitro에서 DNA methylase에 대하여 억제효과를 나타내어TEk.

Saccharomyces cerevisiae M3G3를 이용한 1,2-Propanediol의 생산 최적화 (1,2-Propanediol Production by Using Saccharomyces cerevisiae M3G3)

  • 1,2-propanediol (1,2-PD) is a commodity chemical that is currently produced from petrochemical derivatives. Saccharomyces cerevisiae is well characterized and a successful industrial microorganism to enable the improvement of the 1,2-propanediol production by metabolic engineering. A recombinant S. cerevisiae M3G3 was used to produce 1,2-propanediol. S. cerevisiae M3G3 is the diploid strain that contains 3 copies of mgs (methylglyoxal synthase) and gldA (glycerol dehydrogenase). S. cerevisiae M3G3 was cultivated at various culture conditions by changing culture temperature, glucose concentration, and inducer concentration. Also the effect of induction time was studied to optimize the production of 1,2-propanediol. Batch and fed-batch cultivation of S. cerevisiae M3G3 was performed by using a 5 L jar fermenter. The highest concentration of 1,2-propanediol in batch cultivation was 0.86 g/L and it was further improved to 1.33 g/L in fed-batch cultivation.

γ-Aminobutyric acid (GABA) confers chromium stress tolerance in mustard (Brassica juncea L.) seedlings by modulating the antioxidant defense and glyoxalase systems

  • Chromium (Cr) toxicity is hazardous to the seed germination, growth, and development of plants. ${\gamma}$-Aminobutyric acid (GABA) is a non-protein amino acid and is involved in stress tolerance in plants. To investigate the effects of GABA in alleviating Cr toxicity, we treated eight-d-old mustard (Brassica juncea L.) seedlings with Cr (0.15 mM and 0.3 mM $K_2CrO_4$, 5 days) alone and in combination with GABA ($125{\mu}M$) in a semi-hydroponic medium. The roots and shoots of the seedlings accumulated Cr in a dose-dependent manner, which led to an increase in oxidative damage [lipid peroxidation; hydrogen peroxide ($H_2O_2$) content; superoxide ($O{_2}^{{\cdot}-}$) generation; lipoxygenase (LOX) activity], MG content, and disrupted antioxidant defense and glyoxalase systems. Chromium stress also reduced growth, leaf relative water content (RWC), and chlorophyll (chl) content but increased phytochelatin (PC) and proline (Pro) content. Furthermore, supplementing the Cr-treated seedlings with GABA reduced Cr uptake and upregulated the non-enzymatic antioxidants (ascorbate, AsA; glutathione, GSH) and the activities of the enzymatic antioxidants including ascorbate peroxidase (APX), monodehydroascorbate reductase (MDHAR), dehydroascorbate reductase (DHAR), glutathione reductase (GR), glutathione peroxidase (GPX), superoxide dismutase (SOD), catalase (CAT), glyoxalase I (Gly I), and glyoxalase II (Gly II), and finally reduced oxidative damage. Adding GABA also increased leaf RWC and chl content, decreased Pro and PC content, and restored plant growth. These findings shed light on the effect of GABA in improving the physiological mechanisms of mustard seedlings in response to Cr stress.

화학적 산화법에 의한 부식산의 분해처리 기술에 관한 연구(II) - 오존처리에 따른 분해특성 분석 - (Characterization of Humic Acid in the Chemical Oxidation Technology(II) - Characteristics by Ozonation -)

  • 본 연구는 수질계의 용존 유기 탄소화합물의 주성분인 부식산을 화학적 산화방법 중 오존처리를 사용하여 $UV_{254}$ 흡광도, TOC, $^{13}C-NMR$, 한외여과법 등을 이용하여 오존 처리후의 특성 변화를 조사하였으며, 오존 산화물의 GC/MS 분석을 통하여 포름알데히드, 아세트알테히드, 아세톤, 글리옥살 및 메틸글리옥살 등의 생성을 확인하였다. $UV_{254}$ 흡광도와 TOC의 측정결과 부식산 농도 20, 100ppm 모두 오존처리시간 20분 이내에서 80%의 색도 감소율과 30분 이내에서 40-50%의 TOC 제거율을 나타냈고, 그 이후의 처리시간에서는 일정한 색도 감소율 및 제거율을 나타냈다. $^{13}C-NMR$, 한외여과법을 통한 실험결과, 오존처리에 의해 고분자량의 부식산이 산화, 분해됨으로서 알데히드, 카르복실산 등이 풍부한 저분자량의 단위체로 나뉘어지며 생물학적 분해가 용이한 지방족 탄소가 풍부한 구조로 전환됨를 확인하였다.

불레기말(Colpomenia sinuosa)의 최종당화산물 저해 효능 및 신장 보호 효과 (Anti-glycation effect and renal protective activity of Colpomenia sinuosa extracts against advanced glycation end-products (AGEs))

  • Here, we evaluated the anti-glycation effects and renal protective properties of 70% (v/v) ethanolic extract of Colpomenia sinuosa (CSE) against AGEs -induced oxidative stress and apoptosis at different concentrations (1, 5, and 20 ㎍/mL). At 20 ㎍/mL, CSE showed that anti-glycation activities via the inhibition of AGE formation (51.1%), inhibition of AGEs-protein cross-linking (61.7%), and breaking of AGEs-protein cross-links (33.3%), were significantly (###p < 0.001 vs. non-treated group) lower than the nontreated group. Methylglyoxal (MGO) significantly (***p < 0.001) reduced cell viability (24.4%) and increased reactive oxygen species (ROS) level (642.3%), MGO accumulation (119.4 ㎍/mL), and apoptosis (55.0%) in mesangial cells compared to the nontreated group. Pretreatment with CSE significantly (###p < 0.001) increased cell viability (57.8%) and decreased intracellular ROS (96.5%), MGO accumulation (80.0 ㎍/mL), and apoptosis (22.6%) at 20 ㎍/mL. Additionally, we confirmed intracellular AGEs reduction by CSE pretreatment. Consequently, our results suggest that CSE is a good source of natural therapeutics for managing diabetic complications by the antiglycation effect and renal protective activity against MGO-induced oxidative stress.
