Stem Rot of Sweet Potato (Ipomoea batatas) Caused by Sclerotium rolfsii in Korea (Sclerotium rolfsii에 의한 고구마 흰비단병)

  • During the early spring of 2007 to 2009, stem rot of sweet potato (Ipomoea batatas L.) caused by Sclerotium rolfsii occurred in seedling stage grown in plastic film house at Iksan. In seedling stage of sweet potato, symptoms were initially appeared in yellowing and then the seedlings were eventually wilted. The fungus produced abundant white silky mycelium on infected tissues and soil line. Seedlings were very susceptible and died quickly once they were infected. The whole area of a petridish was rapidly covered with white mycelium on agar medium. Sclerotia began to produce after 7 days of mycelial growth and white sclerotia quickly melanized to a dark brown coloration. The causal agent isolated from the diseased plants was identified as Sclerotium rolfsii Saccardo on the basis of the morphological and cultural characteristics. All isolates of S. rolfsii caused similar symptoms on the host petioles by artificial inoculation.

Breeding of a New Light Pink Spray-mum 'Brown ND' for Cut Flower (연분홍색의 절화용 스프레이국화 '브라운엔디' 육성)

  • A new light pink spray-mum, 'Brown ND' was bred by crossing between 'Noeul', a yellow anemone type flower, and 'Dalma', a pink anemone type flower in October 2003. The growth and flowering characteristics were investigated in forcing, natural, and retarding cultures from 2005 to 2007. The 'Brown ND' was a medium-flowering fall-mum with light pink anemone type flower and had 10.6 flowers per stem, 4.1 cm diameter of flower in natural culture. The number of days to flowering and flowers per plant was 59 and 9.1, respectively, in forcing culture of 'Brown ND' whereas the number of days to flowering was 49 in retarding culture.

  • A new white spray-mum, 'White Ball' was bred by crossing between 'Stallion', a white anemone type flower, and 'Hyangno', an anemone type with pink petals in October 2003. The growth and flowering characteristics were investigated in forcing, natural, and retarding cultures from 2005 to 2007. The 'White Ball' was a medium-flowering fall-mum with white anemone type flower and had 16.2 flowers per stem, 3.6 cm diameter of flower in natural culture. The number of days to flowering and flowers per plant was 58.5 and 11.9, respectively, in forcing culture of 'White Ball' whereas the number of days to flowering was 54 in retarding culture.

Sclerotinia Rot of Peucedanum japonicum Thunb. Caused by Sclerotinia sclerotiorum (Sclerotinia sclerotiorum에 의한 갯기름나물 균핵병 발생)

  • Sclerotinia rot was occurred on the leaf and stem of Peucedanum japonicum Thunb. in greenhouse field of Pohang city of Gyeongbuk province in Korea. The typical symptom of the disease was light brown spot and tipburn on infected leaves. The colony of the isolated fungus was white to light gray in color. Asci were cylindrical shape and 75-240×5.9-17.3 ㎛ in size. Apothecia were cup-shaped with numerous asci and 0.5-0.9 cm in size. Ascospores were aseptate and ellipsoid in shape, and 8.4-10.7×4.8-5.8 ㎛ in size. Sclerotia formed on the plants and potato dextrose agar medium were globose to irregular in shape and black in color. Partial sequencing of rDNA of this isolate showed that it was 100% consistent with that of Sclerotinia sclerotiorum. It was confirmed that the same lesion was formed by reinoculating this pathogen on a healthy P. japonicum Thunb. and the same strain was isolated. This is the first report on the Sclerotinia rot of P. japonicum Thunb. caused by S. sclerotiorum in Korea.

Mycelial growth and fruit body cultural characteristics of a new Pleurotus ostreatus variety 'Daeseon' (느타리 신품종 '대선'의 균사배양 및 자실체 생육 특성)

  • The oyster mushrooms have known to be a major product in Gyeonggi-do, with production accounting for 69% of the entire country. The 'Daeseon' cultivar, which has white and straight stem, was developed. This cultivar was developed by mating monokaryons isolated from the 'Heuktari' and 'Hwaseong-2ho' varieties. The optimum temperature for the mycelial growth was 26~32℃ on PDA medium and that for the primordia formation and the growth of fruit body of 'Daeseon' was 16~20℃ on sawdust media. It took 35 days to complete spawn running, 3 days for finish primordia formation, and 4 days for finish fruit body growth in the bottle culture. It has shallow funnel-shaped pileus and a white straight stipe. The yield per bottle was 201 g/1,100 ml and was 16% higher than that of control cultivar 'Suhan-1ho'. Based on above results, we expect this cultivar to be suitable for small packaging.

  • A plant regeneration system using shoot basal callus of in vitro cultured black locust(Robinia pseudoacacia L.) was established. Shoot basal callus was induced on MS medium supplemented with BA, or NAA, and mere more proliferated on BA containing medium than NAA containing medium at both light and dark conditions. Shoot basal callus was induced during shoot multiplication procedure. Two types of callus, green colored callus and whitish-yellow colored callus, were cultured on mMS medium containing 2.0 mg/l BA and 0.5 mg/l NAA. Green colored callus showed the shoot regeneration ability while whitish-yellow callus failed to regenerate shoot and died. Regenerated shoot were rooted on hormone-free ${\frac{1}{2}}MS$ medium within 2 weeks.

Effects of Elastic Band-Resistive Exercise using Audio-visual Medium on Pain, Proprioceptive Sense, and Motor Function in Adult Females with Chronic Neck and Shoulder Pain (만성 목-어깨 통증이 있는 여성 성인에게 시청각 매체를 활용한 탄력밴드 저항운동이 통증, 고유수용성 감각과 운동기능에 미치는 영향)

  • Background: This study aimed to investigate the effect of elastic band-resistive exercise using audio-visual medium on pain, proprioception, and motor function in adults with chronic neck and shoulder pain. Design: One group pretest-posttest follow-up experimental design. Method: Twenty adult women with neck and shoulder pain voluntarily participated in this study. Elastic band-resistive exercise using audio-visual medium including cervical flexion and extension, shoulder external rotation, and scapular retraction-protraction motions was conducted 5 times a week for 3 weeks. The Numerical Rating Scale, pressure threshold tool, CROM goniometer, and Image J software were used to assess subjective pain level, tenderness threshold (pain), joint position sense error (proprioception), joint range of motion, and postural alignment (motor function), respectively. Result:: The pain intensity and threshold and joint position sense error showed significant decreases after the intervention, whereas the joint range of motion angle revealed significant increases. The postural alignment including forward head posture and rounded shoulder revealed significant improvements after the intervention. Conclusions: Therefore, we suggest that elastic band-resistive exercise through audio-visual medium would be helpful in preventing and managing pain and physical dysfunction in individuals with chronic neck and shoulder pain, and then it would support the development of health management-related online education content.

Study on suitable substrate formulation for bottle and bag cultivation of Pleurotus pulmonarius (산느타리(Pleurotus pulmonarius) 병·봉지재배에 적합한 배지조성 연구)

  • Pleurotus pulmonarius usually grows on the dead wood of deciduous trees from mid summer to early fall. Cultivation of this mushroom is recently increasing due to its good flavour and fragrance. This study was conducted to find out the favorable substrate formulation in the bottle and bag cultivation of P. pulmonarius. The V formulation (10 : 50 : 20 : 20 mixing ratio of poplar sawdust, cottonseed hull, cottonseed meal and beet pulp) showed the fastest mycelial growth in the test tube column medium and highest yields in both bottle cultivation and bag cultivation. Those chemical conditions were pH 5.52, 29.07% organic matter contents, 1.12% total nitrogen, 25.95 OM/N ratio, 0.16% CaO, 0.51% $K_2O$, 0.17 MgO and 0.26 $P_2O_5$.

Composition analysis of raw material constituting the medium for mushroom cultivation (버섯재배용 배지재료의 성분분석)

  • The contents of raw materials which are components of mixed substrate for mushroom cultivation were analyzed to optimize the composition. The pure protein(amino acid) level of soybean meal was the highest, 44.02% followed by those of soybean curd residue(31.5%) and cotton seeds meal(30.6%). The non protein nitrogen(NPN) contents in crude protein of main nitrogen materials were 2.4% for soybean meal and 5.6% for dried soybean curd residue, while those of wheat bran and rice bran used as the carbon source were relatively higher, 17.6% compared to that of nitrogen supplying media. Crude protein content per price was 6.0 for rapeseed meal, indicating that it is high crude protein content per price. Nitrogen-free extract(NFE) considering as an ingredient for mycelial growth were high in alphacorn(72.9%) and wheat bran B(57.2%). Acid detergent fiber(ADF) was high in corncob, 51.88%, its use for cultivation of brown rot fungi including Lentinus lepideus should pay attention because the fungi lack complete lignin degradation activity.

Isolation and Identification of α-Glucosidase Inhibitory Compounds, Hyperoside, and Isoquercetin from Eleutherococcus senticosus Leaves (가시오갈피(Eleutherococcus senticosus) 잎으로부터 α-Glucosidase의 저해 활성 물질, Hyperoside와 Isoquercetin의 분리 및 구조·동정)

  • In the present investigation, the anti-diabetic potential of 80% ethanol extract of Eleutherococcus senticosus leaves (EEES) was examined based on ${\alpha}$-glucosidase inhibitory activities. EEES was sequentially fractionated with n-hexane, chloroform, ethyl acetate (EtAOc), n-butanol, and $H_2O$. Of the various fractions, EtAOc fraction effectively inhibited ${\alpha}$-glucosidase activity by 68.05%. Therefore, EtAOc fraction was selected for further isolation and identification studies. EtAOc fraction was separated by medium pressure liquid chromatography with silica and ODS gel to yield eight fractions (EAA~EAH). Based on the results of ${\alpha}$-glucosidase inhibitory activity, EAH fraction was re-chromatographed to yielded four more fractions (EAHA~EAHD). Of these, EAHC fraction showed higher ${\alpha}$-glucosidase inhibitory activity of 93.60%. EAHC fraction was re-chromatographed and yielded EAHCA and EAHCB fractions. Further, identification and chemical structures of these two fractions were analyzed using $^1H$-NMR, $^{13}C$-NMR, and mass spectra data. Based on the results of the spectral data, the isolated compounds were identified as hyperoside and isoquercetin. Results of the present study indicate that the isolated compounds, hyperoside, and isoquercetin from leaves of E. senticosus could be used for the development of new anti-diabetic drugs.
