Keratinolytic protease 생산균, Pseudomonas sp. KP-364의 분리 및 배양 (Isolation of Keratinolytic Protease Producing Microorganism and Its Cultivation Condition)

  • 경기도 일대의 가금류 공장부근 토양로부터 단백질 분해활성이 높은 균주를 분리한 후 이것들 가운데 Keratinolytic protease 생산성이 우수한 균주는 KP-364를 선별하여 효소생산을 위한 최적 배양조건을 검토하였다. 선별된 KP-364 는 형태학적 생화학적 특성을 종합한 결과 Pseudomonas sp 로 동정되었으며 편의상 Pseudomonas sp. KP-364로 명명하였다. 효소생산을 위한 최적 배지조성은 Keratin 0.1% glucose 2.0% soybeam meal 0.5% NaNO3 0.5% KCI 0.2%이었고 초기 pH 6.5 배양온도 $37^{\circ}C$, 48시간진탕 배양할때 효소생산이 가장 우수하였으며 이때의 효소의 역가는 1,270 U/ml/hr이었다.

  • 스웨덴 스톡홀름의 항구부근 토양시료로부터 분리한 Bacillus sp. SMMJ-2는 단백질 분해활성이 높은 균주로써 특히 keratinolytic protease 생산성이 우수한 균주이다. 효소생산을 위한 최적 배지조성은 0.7% $K_2HPO_4$, 0.2% $KH_2PO_4$, 1.0% fructose, 1.2% soybean meal(roasted), 그리고 0.01% $Na_2CO_3$ 이었으며 배양초기 pH는 7.0, 배양온도는 30$^{\circ}C$에서 200 rpm조건일 때 효소생산이 가장 우수하였으며 이때의 활성은 105 units/ml/min로 조사되었다.

Performance of a submerged membrane bioreactor for wastewater mimicking fish meal processing effluent

  • The objective of this work was to analyze organic matter removal, nitrification, biomass growth and membrane fouling in a submerged flat-sheet membrane bioreactor, fed with synthetic wastewater, of similar composition to the effluents generated in a fish meal industry. After biomass acclimatization with saline conditions of 12 gNaCl/L and COD/N ratio of 15 in the bioreactor, results showed that the organic matter removal was higher than 90%, for all organic loading rates (0.8, 1, 1.33 and $2gCOD/L{\cdot}d$) and nitrogen loading rates (0.053, 0.067, 0.089 and $0.133gN/L{\cdot}d$) tested during the study. However, nitrification was only carried out with the lowest OLR ($0.8gCOD/L{\cdot}d$) and NLR ($0.053gN/L{\cdot}d$). An excessive concentration of organic matter in the wastewater appears as a limiting factor to this process' operating conditions, where nitrification values of 65% were reached, including nitrogen assimilation to produce biomass. The analysis of membrane fouling showed that the bio-cake formation at the membrane surface is the most impacting mechanism responsible of this phenomenon and it was demonstrated that organic and nitrogen loading rates variations affected membrane fouling rate.

  • 가축 사료첨가용 probiotics용 균주를 얻기 위해 특히 내열성이 높아 가공사료에 적합한 Bacillus sp.를 위주로 하여 가축에게 유용한 효소인 phytase를 비롯하여 protease, cellulase, xylanase, amylase의 활성을 모두 나타내는 4-3 균주를 얻어 동정한 결과 Bacillus subtilis로 밝혀졌으며, 이를 B. subtilis 4-3으로 명명하였다. 본 균주를 원료사료에 순수배양하여 사료의 항영양인자인 phytic acid 분해율을 검토한 결과 대두박 및 쌀겨에 있어서는 phytic acid 분해율이 낮았으나, 밀기울의 경우 80.63%로 상대적으로 높은 phytic acid 분해율을 나타내었다. 원료 사료를 이용한 균주의 생산 조건은 대두박 1%(w/v)와 당밀 2%(w/v)가 가장 적합한 균주 생산을 위한 배지조성으로 검토되었다.

단일품목 동적 롯트량결정에 대한 이론적 고찰과 적용 (Single-prodect dynamic lot-sizing : review and extension)

  • In this study, We reviewed the solution methods (for the heuristic and optimization method) for the single-item dynamic lot-sizing problem, and improved the efficiency (speed and optimality) of the conventional heuristic method by utilizing the inventory decomposition property. The iventory decomposition property decomposes the given original problem into several independent subproblems without violating the optimality conditions. Then we solve each decomposed subproblems by using the conventional heuristics such as LTC, LUC, Silver-Meal etc. For testing the efficiency of the proposed decomposition method, we adopted the data sets given in Kaimann, Berry and Silver-Meal. The computational results show that the suggested problem solving framework results in some promising effects on the computation time and the degree of optimality.

돈혈의 적정 건조조건과 육계사료로서의 재활용 방안 (Optimum Drying Condition for Slaughter Porcine Blood and Its Utilization as Broiler Diets)

  • Optimum drying conditions to utilize porcine blood from slaughter house for blood meals, and the effects of blood meals on growth in broiler chicks were investigated. Moisture and protein con-tents of slaughter porcine blood were 79.8 and 16.4%, respectively. The protein contents of the flash dried blood meals at 80˚C were not different from those of the spray dried blood meals at 160 and 190˚C, but higher by 17% relative to those of the spray dried blood meals at 80 and 120˚C. Results from protein analysis by SDS-polyacrylamide electrophoresis showed that flash dried blood meals at 80˚C and spray dried blood meals at 160˚C were better than spray dried blood meals at 80, 120 and 190˚C in terms of protein quality. In Feeding Trial I with broiler chicks, body weights of chicks fed 2, 4 and 6% flash dried blood meal diets at 80˚C were increased at 35 days by 5.6, 7.9 and 4.0%, respectively, compared to control group(P<0.05). In Feeding Trial II, body weights of chicks fed 4 and 6% flash dried blood meal diets at 80˚C were increased at 42 days by 4.9 and 5.3%, respectively, compared to control group(P<0.05). Feed conversion ratios of chicks fed diets 4 and 6% flash dried blood meal diets at 80˚C were significantly improved at 42 days by 7.0 and 3.7%, respectively, compared to that of control group(P<0.05). The optimum drying condition of slaughter porcine blood seemed to be the flash drying method at 80˚C

위탁급식계약 개발전략수립을 위한 업체선정 요인분석 (Sales Strategic Planning through analyzing the factors affecting the foodservice management contract)

  • The purposes of this study were to compare, through a factor analysis, the perceived level of importance of 4 categories of features relating to food service management between salespeople and clients, to establish an effective marketing strategy for successful contracting. To obtain data on the perceived level of importance level of the factors affecting foodservice management contracts, questionnaires were developed using the Delphi technique, which were modified by apilot test. The questionnaires consisted of 4 categories and 19 items on the factors affecting foodservice management contracts, with the importance level of these factors measured on a 5 point-Likert type scale. Between March 12 and April 13 2003, the self-administrative questionnaires were mailed to the 60 salespeople and 280 clients. A total of 50 clients (25%) and 48 salespeople(77%) responded to the questionnaires. As a result, forprivate contracts and in competitive biding, the differences of the perceived importance level between the salespeople and clients of the 3 categories (the appropriateness of foodservice operation plan, sales ability, the conditions and costs of the contract) were significant. For the 5 items relating to private contracts, Field trip, Menu Management Plan, Sanitation and Safety Management, Cost per meal and Food Cost per meal, both the salespeople and clients perceived high levels of importance for all these items. For competitive biding, both the salespeople and clients perceived high levels of importance for the 6 item the Foodservice operation supportive system, Field trip, Menu Management Plan, Renewal plans for interior and environment, Cost per meal and Food Cost per meal.

사료 공급횟수가 어린 강도다리(Platichthys stellatus)의 성장에 미치는 영향 (Effect of Feeding Frequency on the Growth of Juvenile Starry Flounder Platichthys stellatus)

  • Effects of feeding frequency on the growth performance of juvenile starry flounder Platichthys stellatus were investigated. One thousand fifty fishes were distributed into 15 of 500-L flow-through tanks. Triplicate groups of were fed experimental diet with five feeding frequency (one meal every two days, one meal a day, two meals a day, three meals a day and four meals a day) for 9 weeks. Weight gain and specific growth rate (SGR) of fish fed two meals a day were higher than those of fish fed one meal 2 days and one meal a day, but not different from those of fish fed three meals a day and four meals a day. Feed consumption of fish fed two meals a day was not different from that of fish fed three meals a day and four meals a day. Feed efficiency (FE) and protein efficiency ratio (PER) of fish fed two meals a day were higher than those of fish fed all other diets, except for three meals a day. These results suggest that the optimal feeding frequency is two meals a day for the optimum growth performance of juvenile starry flounder under the experimental conditions.

Effects of Expander Conditioning of Complex Nursery Diets on Growth Performance of Weanling Pigs

  • Three experiments were conducted to determine the effects of conditioning a complex (20% whey, 10% lactose, 4% plasma protein, 4% wheat gluten and 2% blood meal) diet on growth performance of weanling pigs. In Exp. 1,180 pigs (average initial BW of 6.4 kg) were fed the experimental diet (1.7% lysine) during a 7-d growth assay. Treatments were a meal control (M), standard (ST), and expander (EX) conditioned mash or pellets. Rate and efficiency of gain were decreased by 39% and 21% (p<0.005) respectively, for pigs fed EX diets compared to those fed the ST diet. In Exp. 2,196 pigs (average initial BW of 6.5 kg) were used to determine the effects of EX operating conditions on nutritional value of a pelleted complex diet. When steam conditioning temperature (prior to expanding) was $54^{\circ}C$, increasing cone pressure of the EX from 0 to 7 to 14 kg/cm2 resulted in linear decreases in rate of gain of weaned pigs (p<0.006), suggesting heat damage of the diet. Increasing conditioning temperature (i.e., adding steam) of the diets from 46 to 54 to $63^{\circ}C$ (cone pressure at $12kg/cm^2$) resulted in improved rate of gain (p<0.04) of the pigs. However, none of the pigs fed expanded diets compared favorably to the pigs fed the conditioned $(54^{\circ}C)$ pellets processed with no cone pressure. In Exp. 3,168 pigs (average initial BW of 6.6 kg) were used to determine the effects of expanding the various components of the diet. Treatments were M and ST pellets as controls, EX-corn, EX-corn soybean-meal, EX corn-soybean meal-oil, and EX-complete diet. Efficiency of gain was increased by 13% with EX portions of the diet compared to the mash control, but there was a marked decrease in performance when the complete diet was expanded (p<0.001). Expanded corn-soybean meal-oil supported the greatest ADG with a 19% increase compared to the average of the EX corn and EX corn-soybean meal treatments (p<0.005). In conclusion, our results suggest no benefit from expanding complete phase-I diets.

버섯폐배지로부터 섬유소분해력이 높은 중온성 균의 분리 및 균주생산을 위한 배지조건의 최적화 (Isolation and Identification of High Cellulolytic Bacteria from Spent Mushroom Substrate and Determination of Optimal Medium Conditions for the Growth)

  • 본 연구는 톱밥주원료 버섯폐배지의 효과적 사료화를 위한 균주개발을 목적으로 버섯폐배지로부터 섬유소분해력이 높은 중온성 균주의 분리 동정하고 균주생산을 위한 배지의 최적화 조건 도출을 위하여 실시하였다. Xylanase와 CMCase의 활력이 높으면서 amylase와 pretense의 활력이 낮아 선발된 균은 201-3, 206-3번이며, 동정결과 201-3은 Enteerobacter ludwigii로 동정되어 Ent. ludwigii KU201-3으로, 206-3은 Bacillus cereus로 동정되어 B. cereus KU206-3으로 명명하였다. 균주생산을 위한 적정 액상배양조건은 Ent. ludwigii KU201-3 질소원으로 soybean meal 1%, 탄소원으로 sucrose 3%가, B. cereus KU206-3은 질소원으로 soybean meal 3%, 탄소원으로 molasses 1%인 것으로 나타났다.
