Cytotoxicity and Antioxidative Activity of Artemisiae Argi Folium Alcoholic Extracts and Their Fractions (애엽(艾葉) 에탄올 및 메탄올 추출물과 용매별 분획의 세포독성과 항산화활성)

  • This experiment was carried out to obtain the cytotoxicity and antioxidative activity of Artemisiae Argi Folium. The total polyphenol contents in the ethyl acetate fraction of the ethanol extract and the methanol extract were 430.27mg/g and 427.84mg/g, respectively. In DPPH radical scavenging ability, $SC_{50}$ values of the ethyl acetate fraction of the ethanol extract and the methanol extract were 32.64 ${\mu}g/m{\ell}$ and 27.70 ${\mu}g/m{\ell}$ as the same level of statistical with ascorbic acid. In the cytotoxicity measurement by MTT assay, the chloroform and hexane fraction, and each extract were exhibited higher cytotoxicity than the other fractions. In particular, the ethyl acetate fractions appeared high activity in DPPH radical scavenging ability were began to show cytotoxicity in 125 ${\mu}g/m{\ell}$. As a result, the ethyl acetate fraction of Artemisiae Argi Folium extract was the most highly active fraction in antioxidative activity. However, for the use of extracts and fractions from Artemisiae Argi Folium to related fields, the setting of appropriate concentration is required.

  • To improve the catalytic activity of platinum on polymer electrolyte fuel cell(PEFC), platinum was alloyed with cobalt and nickel at various temperature. By XRD, it was observed the crystal structure of alloy catalysts were the ordered face centered cubic(f.c.c) due to the superlattice line at $33^{\circ}$. As heat-treatment temperature was increased, the particle size of alloys also were increased and the crystalline lattice parameters were decreased. According to the results from mass activity, specific activity and Tafel slope measured by cell performance test and cyclic voltammogram, the catalyst activities of alloys are higher than that pure platinum.

Biological Activity of Fermented Gryllus bimaculatus extracts (발효 쌍별귀뚜라미 추출물의 생리활성 연구)

  • Gryllus bimaculatus extract (GbE) have reported that anti-inflammatory activity by suppression of pro-inflammatory cytokines. However, the effects of fermented Gryllus bimaculatus extract (FGbE) have not yet been investigated. In this study, we evaluated the anti-inflammatory and anti-wrinkle effect of the fermented Gryllus bimaculatus extracts using Bacillus subtilis (JB PMB-18) in RAW264.7 cells. Both GbE and FGbE exerted no cytotoxic effects until 1000 ㎍/mL concentration. FGbE decreased NO production and decreased iNOS and COX-2 mRNA levels in a concentration-dependent manner. In addition, the protein production of inflammatory cytokines TNF-α, IL-1β and IL-6 was effectively reduced compared to the GbE. Inhibitory activities of elastase and collagenase associated with skin wrinkle improvement were measured to be 45% and 69%, respectively, at a concentration of 500 ㎍/mL in FGbE. From these results, FGbE can be used as a health functional food and skin functional cosmetic materials for preventing inflammatory diseases because it has excellent anti-inflammatory and anti-wrinkle effects.

Ethanol Eluted Extract of Rhus verniciflua Stokes Showed both Antioxidant and Cytotoxic Effects on Mouse Thymocytes Depending on the Dose and Time of the Treatment

  • For a long time Rhus verniciflua Stokes (RVS) has traditionally been used as a herbal plant. It is known to contain various biological activities. Previously, a crude ethanol extract from RVS was reported to have antioxidant effects, and antiproliferative activities, on human cancer cell lines. In this report, we prepared a highly purified ethanol extract from RVS, which did not contain the urushiol derivatives, named REEE-1 ($\underline{R}$hus $\underline{e}$thanol $\underline{e}$luted $\underline{e}$xtract-1), to investigate the mechanisms of the scavenging activity of hydroxyl radicals using mouse thymocytes. The results from the deoxyribose, DNA nicking, and glucose/glucose oxidase enzyme assays showed that REEE-1 contained a strong scavenging activity of oxygen free radicals, especially of hydroxyl radicals. However, interestingly, REEE-1 also showed cytotoxicity against the thymocytes, although the effect was variable, depending on the concentrations and times of treatment. The REEE-1-mediated cytotoxicity against thymocytes, which has been used as one of the well-characterized models for apoptosis studies, was verified to be apoptotic. This was proven by the following: the appearance of DNA laddering, increases in DNA fragmentation, low fluorescence intensity in the nuclei after propidium iodide staining, and positive Annexin V staining of the cells. These results suggested that REEE-1 had both antioxidative activity and cytotoxicity against the thymocytes, although the effect of the cytotoxicity was variable, depending on the dose and time of the treatment.

  • The purpose of this study is to develop the programming language teaching materials for the gifted of information with using the game. There are many curricula for teaching the information and technology to the gifted of information. There are not differences between these programs and the general IT curricula. The gifted of information has some complains about his curriculum(58/60, 96.67%). So this study developed the alternative programming language teaching materials for the gifted of information. This study concluded the effects of alternative materials for teaching programming language depend on learners' academic achievement about programming language with game activity. This study also looks forward to being able to indicate new direction to develop the teaching materials for the gifted of information.

Prokaryotic Selectivity, Anti-endotoxic Activity and Protease Stability of Diastereomeric and Enantiomeric Analogs of Human Antimicrobial Peptide LL-37

  • LL-37 is the only antimicrobial peptide (AMP) of the human cathelicidin family. In addition to potent antimicrobial activity, LL-37 is known to have the potential to inhibit lipolysaccharide (LPS)-induced endotoxic effects. To provide the stability to proteolytic digestion and increase prokaryotic selectivity and/or anti-endotoxic activity of two Lys/Trp-substituted 19-meric antimicrobial peptides (a4-W1 and a4-W2) designed from IG-19 (residues 13-31 of LL-37), we synthesized the diastereomeric peptides (a4-W1-D and a4-W2-D) with D-amino acid substitution at positions 3, 7, 10, 13 and 17 of a4-W1 and a4-W2, respectively and the enantiomeric peptides (a4-W1-E and a4-W2-E) composed D-amino acids. The diastereomeric peptides exhibited the best prokaryotic selectivity and effective protease stability, but no or less anti-endotoxic activity. In contrast, the enantiomeric peptides had not only prokaryotic selectivity and anti-endotoxic activity but also protease stability. Our results suggest that the hydrophobicity and ${\alpha}$-helicity of the peptide is important for anti-endotoxic activity. In particular, the enantiomeric peptides showed potent anti-endotoxic and LPS-neutralizing activities comparable to that of LL-37. Taken together, both a4-W1-E and a4-W2-E holds promise as a template for the development of peptide antibiotics for the treatment of endotoxic shock and sepsis.

Larvicidal and Antifeeding Activities of Oriental Medicinal Plant Extracts against Plutella xylostella and Spodoptera litura (배추좀나방과 담배거세미나방 유충에 대한 한방식물체 추출물의 살충 및 섭식저해활성)

  • Methanol extracts from 45 species of oriental medicinal plants belonging to 17 families were subjected to a screening test for their larvicidal and antifeeding activities against two species of lepidopteran larvae, diamondback moth (Plutella xylostella L.) and tobacco cutworm (Spodoptera litura F.) by a leaf-dipping method. At a concentration of 8,000 ppm, methanol extract of Corydalis turrschaninovii roots only was found to have potent larvicidal activity against S. litura. At 5,000 ppm, strong antifeeding activity against both larvae was observed from the extracts of Clerodendron trichotomum, C. trichotomum var. esculentum, Inura helenium, Arctium lappa, Artemisia messerschmidtiana, Anthriscus sylvestris, Angelica dahurica, and C. turrschaninovii.

Inhibitory Effects of Quinizarin Isolated from Cassia tora Seeds Against Human Intestinal Bacteria and Aflatoxin $B_1$ Biotransformation

  • The growth-inhibitory activity of Cassia tora seed-derived materials against seven intestinal bacteria was examined in vitro, and compared with that of anthraquinone, anthraflavine, anthrarufin, and 1-hydroxyanthraquinone. The active constituent of C. tore seeds was characterized as quinizarin, using various spectroscopic analyses. The growth responses varied depending on the compound, dose, and bacterial strain tested. At 1 mg/disk, quinizarin exhibited a strong inhibition of Clostridium perfringens and moderate inhibition of Staphylococcus aureus without any adverse effects on the growth of Bifidobacterium adolescentis, B. bifidum, B. longum, and Lactobacillus casei. Furthermore, the isolate at 0.1 mg/disk showed moderate and no activity against C. perfringens and S. aureus. The structure-activity relationship revealed that anthrarufin, anthraflavine, and quinizarin moderately inhibited the growth of S. aureus. However. anthraquinone and 1-hydroxyanthraquinone did not inhibit the human intestinal bacteria tested. As for the morphological effect of 1 mg/disk quinizarin, most strains of C. perfringens were damaged and disappeared, indicating that the strong activity of quinizarin was morphologically exhibited against C. perfringens. The inhibitory effect on aflatoxin $B_1$ biotransformation by anthraquinones revealed that anthrarufin ($IC_50,\;11.49\mu\textrm{M}$) anthraflavine ($IC_50,\;26.94\mu\textrm{M}$), and quinizarin ($IC_50,\;4.12\mu\textrm{M}$), were potent inhibitors of aflatoxin ${B_1}-8,9-epoxide$ formation. However, anthraquinone and 1-hydroxyanthraquinone did not inhibit the mouse liver microsomal sample to convert aflatoxin $B_1$ to aflatoxin ${B_1}-8,9-epoxide$. These results indicate that the two hydroxyl groups on A ring of anthraquinones may be essential for inhibiting the formation of aflatoxin ${B_1}-8,9-epoxide$. Accordingly, as naturally occurring inhibitory agents, the C. tora seed-derived materials described could be useful as a preventive agent against diseases caused by harmful intestinal bacteria, such as clostridia, and as an inhibitory agent for the mouse liver microsomal conversion of aflatoxin $B_1$ to aflatoxin ${B_1}-8,9-epoxide$.

  • CS Unplugged education is emphasized as the component of the basic principles of Elementary SW education. This document produced by two other Timbell presents the contents in a variety of topics about computer science. One of the main components is the finite Automata, and this requires the development of educational materials for teaching our situation. So We'll present the finite Automata learning materials for elementary school classes. Learning model that we have presented is a process of self-directed and activity-based learning. For verification of this experiment was the validation of the expert group and was concluded that adequate through the analysis of the diagnostic tests.
