  • The purpose this research is to develop QCM (quartz crystal microbalance) biosensor for the determination of haptoglobin. Haptoglobin is an acute-phase protein with a hemoglobin-binding activity and has a potential to be used as a biomarker for infection or cancer. Haptoglobin level in milk has been used for the diagnosis of cow mastitis. In this study, anti-bovine haptoglobin antibody or bovine hemoglobin was chemically immobilized on the surface of the QCM, and the resulting sensor chips were tested for their response to samples containing haptoglobin at different concentrations. Concentration dependent frequency change was observed with both of the sensor chips. Especially, the sensor chip containing anti-bovine haptoglobin antibody showed sufficient sensitivity in the concentrations typically observed in the cows with mastitis.

Capsular Polysaccharide Serotypes Among Staphylococcus aureus Isolates from Cases of Bovine Mastitis and Dogs (젖소 유방염과 개에서 분리된 황색포도상구균에 대한 Capsular Polysaccharide형의 동정)

  • 본 연구는 임상형 및 준임상형 유방염에 이환된 젖소의 유즙에서 분리된 24주의 황색포도상구균에 대한 capsular polysaccharide(CP)형을 확인하고, 1% 토끼 혈청이 함유된 serum-soft agar 배지에서 집락의 모양을 관찰하였다. 또한 동물에 따라 우세한 CP형의 차이가 있는지를 비교하고자 개에서 분리된 13주에 대하여 동일한 실험을 수행하여 다음과 같은 결과를 얻었다. 집락모양을 관찰한 결과 젖소 유래 24주 중 16주(66.7%)는 diffuse, 5주(20.8%)는 compact, 나머지 3주(12.5%)는 분류가 불가능(indeterminate)하였다. CP형 확인결과 9주(37.5%)는 type 5, 2주(8.3%)는 type 8, 13주(54.2%)는 분류가 불가능(non-typeable)하였다. 한편, 개에서 분리된 균주 중 1주(type 8)를 제외한 12주(92.3%)는 type 5로 분류되었으며, 13주 중 8주(61.5%)가 diffuse형의 집락을 보였다. 본 실험에 사용한 균주의 수가 충분하지 못한 문제가 있지만 동물에 따라 우세한 CP형이 다를 수 있으며 분류 불가능한 균주가 상대적으로 높은 비율을 차지하였는데 이는 새로운 CP형의 분류형이 필요함을 시사하는 것으로 판단된다. 또한 type 5와 type 8만을 포함한 유방염 백신은 한계가 있을 것으로 사료된다.

Efficacy of Bovine Staphylococcal Mastitis Vaccine Composed of Alpha toxin, Capsular Polysaccharide and Fibronectin Binding Protein in Lactating Cows and Heifers (비유우와 처녀우에서 황색포도구균의 alpha toxin, capsular polysaccharide와 fibronectin binding protein으로 구성된 유방염 백신의 효능)

  • capsular polysaccharide(CPS), alpha toxin과 재조합 fibronectin binding protein (FnBP)으로 구성된 유방염 백신을 개발하여 야외효능실험을 수행하였다. 이 subunit 백신은 비유우 23두와 처녀우 20두를 대상으로 10개월동안 각각 수행되었다. 비유우는 매 3주간격으로 2회 백신접종을 하였으며 대조군은 PBS를 상유방림조절 주위에 피하주사하였다. 처녀우를 대상으로 한 실험에서는 분만 8주전부터 시작하여 매 3주간격으로 2회 비유우 백신접종부위와 동일하게 접종하였다. 접종후 비유우에서는 황색포도상구균에 의한 총유선내 감염율이 대조균에 비해 유의성있게 감소하였으며 (p<0.001) 처녀우에서도 총유선내 감염이 백신 접종균(1.6%)이 대조군(11.7%)에 비해 다음 비유기동안 유의성있게 낮았다. (p<0.001). 비유기동안 백신접종한 젖소의 체세포수는 변화가 없었으며 처녀우에서는 추가접종후 백신접종군에서 대조군의 체세포수에 비해 낮았지만 통계학적으로 유의성은 없었다(p>0.05). 본 실험결과 황색포도구균에 대한 유방염 아단위 백신은 비유우와 처녀우에서 체세포수를 증가시키지 않고 총 유선내 감염율을 낮추어주었다. 하지만 본 실험은 1군데의 목장을 대상으로 하였기 때문에 향후 대규모 목장을 대상으로 하는 유방염 백신의 야외효능실험이 심도있게 이루어져야 한다고 생각된다.

Characterization of Bacteriocin Produced by Lactobacillus bulgaricus

  • The antimicrobial substance produced by Lactobacillus bulgaricus was inactivated by protease. It showed inhibitory activity against Staphylococcus aureus ATCC6538, Streptococcus agalactiae ATCC14364, some Gram-positive and Gram-negative bacteria, and characteristics of a bacteriocin. The optimal temperature and culture time for the production of bacteriocin were $30^{\circ}C$ and 10 h, respectively, in the culture of L. bulgaricus. The bacteriocin production started in the exponential phase and reached a maximum at the early stationary phase. Using Staph. aureus ATCC6538 and Strep. agalactiae ATCC14364, known as common bovine mastitis pathogens, as indicator strains for determination of the bacteriocin activity, the antimicrobial activity of the bacteriocin was found to be stable in acidic and neutral pH's (2- 7) even at lOOT, whereas it was lost at high pH (10- 11) and $100^{\circ}C$. The mode of action for the antimicrobial activity was bacteriocidal, and the molecular weight determined by SDS-PAGE and overlay method was 14 kDa.

Study of plasma β-carotene concentration in dairy cows (젖소의 혈장 베타카로틴 농도 조사)

  • This study was carried out to determine the effects of ${\beta}-carotene$ on the control of mastitis in dairy cows during the dry period. The relationship between the levels of plasma ${\beta}-carotene$ and the status of udder health in Holstein dairy cows were investigated. Blood samples were collected from 117 cows to compare the levels of plasma ${\beta}-carotene$ in lactating cows. The levels of plasma ${\beta}-carotene$ were $1.82{\mu}g/ml$ in healthy cows(n = 65) and $1.12{\mu}g/ml$ in mastitic cows(n = 52), respectively(p < 0.01). In the experiment to compare the level of plasma ${\beta}-carotene$ in the cows at different stages of lactation, the plasma ${\beta}-carotene$ levels were $1.73{\mu}g/ml$ in lactating cows(n = 22), $1.29{\mu}g/ml$ in nonlactating cows(n = 35) and $0.43{\mu}g/ml$ in cows after calving(n = 16)(p < 0.05).

Detection and Identification of $\beta$-lactamase, Enterotoxin and Other Exotoxins Genes of Staphylococcus aureus by PCR

  • Staphylococcus aureus is a major pathogen for cattle, causing various forms of subclinical and clinical mastitis and could be a causative agent of food poisoning, it produces various superantigenic exotoxins which have a great public health significance. A total of 72 S. aureus clinical isolates from dairy farms located in Kyunggi Province Korea were examined for the species identification by biochemical method, and for the detection of $\beta$-lactamase, enterotoxin and other exotoxins genes by PCR. The results of species identification by biochemical method agreed with those of PCR done with species specific primer STA-AU. $\beta$-lactamase is an enzyme closely associated with the resistance to antibiotic penicillin, which is an important means of treatment of mastitis, all the isolates were positive for the presence of genes encoding $\beta$-lactamase, which were reproduced in penicillin susceptibility disc assay. Six types of toxin genes, Staphylococcal enterotoxin (SE)A, SEB, SEC, SEE, toxic shock syndrome toxin (TSST-1) and exfoliative toxin A (ET A) were detected in 72 isolates by PCR associated genotypic method in this study, none of the isolates carried the genes for enterotoxin D (SED) and exfoliative toxin B (ETB). The occurrence rate of exotoxin genes rated as 12.5%, and the precision of the PCR identification results has been confirmed using the reference strains.

Milk Quality and Antimicrobial Resistance against Mastitis Pathogens after Changing from a Conventional to an Experimentally Organic Dairy Farm

  • The present study was to investigate the effect of the transition from conventional to organic dairy farming on the antimicrobial resistant pattern of pathogens in milk. A farm with tie-stall management, with an average herd size of 20 milking cows, was selected based on the owner' willingness to accept, for at least 6 months, the highly restricted protocol developed in this study. Comparisons of bacterial isolates and antimicrobial susceptibilities before changing to an organic farm system (BEFORE) and for 6 months after (AFTER) operating the experimental organic farm system were performed by Fisher's Exact Chi-square tests. Significant levels were defined at p<0.05. During the AFTER period, average frequency of antibiotic treatment was decreased from more than 3 cases/month to less than 1 case/month during which the antibiotic use was authorized only by the veterinarian. In total, 92 and 70 quarter milk samples from 24 and 18 cows during BEFORE and AFTER, respectively, were included in the study. Overall, isolates ranged from a non-resistant level for cephazolin to a very high resistant level to streptomycin (64.71% to 95.45%). Percentages of antimicrobial resistant isolates during BEFORE were significantly higher than during AFTER for ampicillin (43.48% and 5.88%, respectively) and streptomycin (95.45% and 64.71%, respectively). In conclusion, percentages of antimicrobial resistant isolates were decreased after 6 months of operating as an organic farm system.

Anti-inflammatory Effects of Gagamtunong-san (가감투농산(加減透膿散)의 항염작용에 대한 실험적 연구)

  • The purpose of this research was to investigate the anti-inflammatory effects of Gagamtunong-san(GTNS) which has been medicated the patient such as mastitis. The results were as follows. The cytotoxicity on mouse lung fibroblast Cells(mLFC) was not served at all concentration of GTNS. GTNS in RAW264.7 cell inhibited $IL-1{\beta}$, IL-6, $TNF-{\alpha}$, COX-2 and NOS-II mRNA genes expression in a concentration-dependent manner. Specially GTNS inhibited NOS-II production very significantly at 100 ${\mu}g/ml$. GTNS inhibited NO production significantly in a concentration-dependent manner. GTNS inhibited ROS production in a concentration-dependent manner. GTNS inhibited $IL-1{\beta}$, IL-6 and $TNF-{\alpha}$ production significantly in serum of acute anti-inflammation-induced mice. GTNS increased the survival rate from the 3rd day on LPS-induced lethal endotoxemia. These results suggest that Gagamtunong-san(GTNS) can be useful in treating a lot of women mammary diseases caused by inflammation such as acute and chronic mastitis.

Isolation and Drug Sensitivity of Gram-Negative Rods from Bovine Udder Infections (젖소 유방염(乳房炎)으로부터 Gram 음성간균(陰性桿菌)의 분리(分離) 및 약제감수성(藥劑感受性)에 관(關)한 연구(硏究))

  • One hundred and eighteen cultures of Gram-negative rods isolated from cases of clinical bovine mastitis during lactation were examined for distribution of specific types, and activity of several antimicrobial agents to the isolates was determined by two-fold tube dilution method employing sterile whole milk as fluid medium. Of the isolates, 59.2% were Escherichia coli. Most of the remaining isolates were Klebsiella pneumoniae and Enterobacter aerogenes. Most Gram-negative rods(89.8%) were isolated from acute local and chronic mastitis. The cases of peracute systemic form with a marked symptoms of toxemia were associated with Escherichia coli and Klebsiella pneumoniae. The minimal lethal concentrations (MLC) of gentamicin and oxolinic acid in sterile whole milk were 16-128 times higher than the MLC obtained in trypticase soy broth (TSB), while the MLC of ampicillin and tetracycline in milk increased 2-4 times compared with TSB. Of the drugs tested, gentamicin was the most active antibiotics with MLC of $100{\mu}g/ml$ in sterile whole milk against all of Gram-negative rods isolated from bovine udder infections.
