Development of Universal Primers for Phylogenetic Analysis and Species-specific Band Identification in the Genus Actinidia (다래나무속 식물의 분류 및 계통 특이밴드 탐색을 위한 범용 프라이머 개발)

  • To develop universal primers for phylogenetic analysis and species-specific marker for breeding program of kiwifruit, eighteens primers were designed from kiwifruit genome-specific repeat sequences. Seven species including twenty two varieties collected from native eastern Asia were examined using 18 to 22 mer kiwifruit target(KT) primers. among eighteen primers, we selected seven primers for phylogenetic relationship. The genus Actinidia was divided into two large groups; group I,A. arguta, A. melanandra, A. kolomikta, and A. marcrosperma, characterized by the non-hair in fruits and loaves or a few pubescences only in young stage, which belongs to the section Leiocarpae, and group II, A. chinensis, A. deliciosa, and A. eriantha, characterized by a lot of hairs only in young fruit stage and with a lot of hairs or fuzzes in leaves and branches, which belongs to the section Stellatae. Group II especially belongs to the series Perfectae of the section Stellatae and was divided into two subgroups; subgroup I containing A. chinensis and A. deliciosa, and subgroup II containing A. eriantha. In contrast, the two species, A. chinensis and A. deliciosa, which are known to have common parents, were divided into two independent subgroups with 80% of a similarity value. On the other hand, we selected KT6F for variety specific bands, KT12E primers for 'Hayward' and 'Tomuri'. KT7F or KT12F primers were useful for analysis of inheritance pattern in kiwifruit cross-breeding. We suggest that these primers will be a powerful tool for elucidating phylogenetic relationship and selection of novelty kiwifruit in a breeding program.

  • The purpose of this study was to evaluate the methods of identifying zygotic seedlings of crosses between 'Morita unshiu' (Citrus. unshiu Marc.) and 'Ponkan' (C. reticulata Blanco). In order to investigate the frequency and position of zygotic seedlings and to determine the efficiency of zygotic seedling identification, random amplified polymorphic DNA (RAPD) and sequence-related amplified polymorphism (SRAP) were performed using UBC (9, 27, 229, 230, and 254) primers and F4/R27, F7/R14, F12/R10, and F44/R62 primer sets, respectively. A total of 37 putative zygotic seedlings out of 55 individuals were selected by RAPD and SRAP. The F7/R14 primer pair showed a screening ability of 45.5% (25/55), whereas the primer UBC27 revealed the highest efficiency of zygotic seedling identification (50.9%, 28/55). When both UBC27 and F7/R14 were properly used for selection of hybridized seedlings of 'Morita unshiu' (C. unshiu Marc.) and 'Ponkan' (C. reticulata Blanco), screening efficiency was increased to 60% (33/55) for putative zygotic seedlings. Thus, it is possible to select putative hybrid zygotic seedlings in an accurate and effective manner by RAPD and SRAP.

  • Elephant food(Amorphophallus konjac K.) have been utilized its tubers in workedmaterials for a health and diet food. The author supposed that it was increased the area of cultivation and demand. This experiments were conducted to select the proper covering material during winter in order to increase yield of tubers and decrease input by 2 year's continuous cultivation, also to verified ability of seed germination and to measured efficiency of photosynthesis of plant. The proper covering materials for wintering were rice straw and rice hull. These materials were covered at 5 cm thick and at field was promoted according to emergence appearing after winter. The yields were 5,790kg /10a at 4,730kg /10a, respectively. Yield increase was 120% and 80% than that of control. The seeds collected at August 22 were germinated about 84 percent, and it was not necessary to treatment of low temperature or germination-accelerated chemicals. The widest leaf area was ranged $1,218-1,438cm^2$ at October 20 and was varied. The efficiency of photosynthesis was highest at 65-95 days after leaf emergence. The line of broad leaf and high photosynthetic efficiency per unit area was greater compare with yield. Therefore, it was supposed that these characteristics will use a marker for selection for high-yielding lines.

  • This article is to analyse the economic efficiency of capital market, which plays a role of resource allocation in terms of financial claims such as stock and bond. It provides various contributions to the welfare theoretical aspects of modern capital market theory. The key feature that distinguishes the theory described here from traditional welfare theory is the presence of uncertainty. Securities has time dimensions and the state and outcome of the future are really uncertain. This problem resulting from this uncertainty can be solved by complete market, but it has a weak power to explain real stock market. Capital Market is faced with the uncertainity because it is a kind of incomplete market. Individuals and firms in capital market made their consumption-investment decision by their own criteria, i. e. the maximization of expected utility form intertemporal consumption and the maximization of the market value of firm. We noted that allocative decisions that had to be made in the economy could be naturally subdivided into two groups. One set of decisions concerned the allocation of first-period resources among consumption $C_i$, investment in risky firms $I_j$, and riskless investment M. The other decisions concern the distribution among individuals of income available in the second period $Y_i(\theta)$. Corresponing to this grouping, the theoretical analysis of efficiency has also been dichotomized. The optimality of the distribution of output in the second period is distributive efficiency" and the optimality of the allocation of first-period resources is 'the efficiency of investment'. We have found in the distributive efficiency that the conditions for attainability is the same as the conditions for market optimality. The necessary and sufficient conditions for attainability or market optimality is that (1) all utility functions are such that -$\frac{{U_i}^'(Y_i)}{{U_i}^"(Y_i)}={\mu}_i+{\lambda}Y_i$-linear risk tolerance function where the coefficients ${\mu}_i$ and $\lambda$ are independent of $Y_i$, and (2) there are homogeneous expectations, i. e. ${\Large f}_i(\theta)={\Large f}(\theta)$ for every i. On the other hand, the efficiency of investment has disagreement about optimal investment level. The investment level for market rule will not generally lead to Pareto-optimal allocation of investment. This suboptimality is caused by (1)the difference of Diamond's decomposable production function and mean-variance valuation model and (2) the selection of exelusive investment or competitive investment. In conclusion, this article has made an analysis of conditions and processes of Pareto-optimal allocation of resources in capital marker and tried to connect with significant issues in modern finance.

Monitoring of Bifenazate Resistant Two-spotted Spider Mite, Tetranychus urticae Using Molecular Detection Method (분자학적 진단방법을 이용한 bifenazate 저항성 점박이응애 모니터링)

  • In 2010, two-spotted spider mite, Tetranychus urticae was collected from the rose greenhouse and apple orchards in Cheongju (CJ), Chungju (CUJ)-1, CUJ-2, Kangjin (KJ), Yesan (YS), and Yeongju (YJ). Among them, KJ and YS strain showed high resistance to bifenazate of 964.5- and 1l30-fold, respectively. The other strains showed low resistance to bifenazate. By analyzing the mitochondrial cytochrome b (cytb) sequence, G126S point mutation was detected in KJ and YS strain. Thus, G126S point mutation in the mitochondrial cytb was available molecular detection marker for selection of bifenazate resistant T. urticae. Two molecular detection methods, quantitative sequencing (QS) and PCR amplification of specific alleles (PASA) were well detected specific G126S point mutation. Therefore, these methods can be used to monitor the resistance allele in field population of T. urticae and bifenazate resistance management strategy.

Analysis of Quantitative Trait Loci for Yield Component Traits in Soybean Using Recombinant Inbred Lines (콩에서 수량구성요인과 관련된 양적형질유전자좌의 분석)

  • Seed and pod numbers are the main yield components in soybean. Selection for increased yield potential is main goal of plant breeding. The objective of this study was to identify quantitative trait loci(QTLs) that control pod number per plant, seed number per plant and pod in soybean. The 117 $F_{2:10}$ recombinant inbred lines(RILs) developed from a cross of 'Keunolkong' and 'Shinpaldalkong' were used. Two independent QTLs for pod number per plant were identified from linkage group(LG) F and L. Two QTLs for seed number per plant were located on LG F and L. Seed number per pod was related with three QTLs located on LG D1a, D1b and F. Pod and seed number per plant have two common QTLs on LG F and L.

A New Rice Variety Developed from an Interspecific Cross, "Hwaweon 2" (종간교잡 유래 중생 다수성 벼 품종 "화원 2호")

  • "Hwaweon 2" was developed from an interspecific cross between Hwaseongbyeo and a wild species, Oryza rufipogon L. (IRGC 105491) based on marker-aided selection. The recurrent parent "Hwaseongbyeo" is a high grain quality cultivar with medium-maturity. Hwaweon 2 is nearly isogenic to Hwaseongbyeo except a small O. rufipogon introgression on chromosome 9. This segment was associated with genes controlling a number of traits including grain weight, heading date, culm length, and spikelets per panicle. The preliminary and replicated yield trial was conducted at Chungnam National University in 2004, 2005 and 2006. The local adaptability test was carried out by the National Seed Management Office (NSMO) in 2007 and 2008. This cultivar was registered to NSMO with a cultivar designated as "Hwaweon 2". This cultivar averaged 98cm in culm length and has a medium to late growth duration. This variety is resistant to stripe virus as the recurrent parent. Milled rice of "Hwaweon 2" is translucent and the grain quality traits are comparable to those of the recurrent parent. The yield potential of "Hwaweon 2" in grain is about 7.68 MT/ha at the ordinary fertilizer level about 14% higher than that of Hwaseongbyeo due to increase in grain weight and spikelets per panicle.

Analysis of Genetic Variation Among Popcorn Inbred Lines by SSR Markers (튀김용 옥수수 자식계통들에 대한 유전적 변이성)

  • Knowledge of genetic diversity and genetic relationships among inbred lines gives a significant impact on the selection of parental lines for hybrid maize varieties. Genetic diversity and genetic relationships among 86 popcorn inbred lines were analyzed using 50 SSR markers distributed over the whole genome. A total of 256 alleles were identified at all the SSR loci with an average of 5.1 and a range between two and sixteen per locus. The gene diversity values varied from 0.21 to 0.831 with an average of 0.579. The cluster tree generated using the described SSR markers recognized three major groups at 35.8% genetic similarity. Groups I, II, III respectively included 40, 39 and 7 inbred lines. The present study indicates that the SSR markers chosen for this analysis are effective for the assessment of genetic diversity and genetic relationships among 86 popcorn inbred lines in Korea.

Mapping Grain Weight QTL using Near Isogenic Lines from an Interspecific Cross (벼 종간잡종 유래 근동질 유전자계통 이용 종자중 관여 유전자 분석)

  • In previous studies, we reported QTLs for grain weight (GW), qGW3 and for spikelets per panicle (SPP), qSPP3 linked to RM60 on chromosome 3 using advanced backcross lines derived from a cross between Oryza sativa ssp. Indica cv. Milyang 23 and O. glaberrima. The O. glaberrima alleles at this locus increased GW and spikelets per panicle in the Milyang 23 background. To further confirm and narrow down the position of the QTLs on chromosome 3, substitution mapping was performed using five lines containing the target O. glaberrima segment on chromosome 3. The size and position of the O. glaberrima segment on chromosome 3 were different in each line. These lines possessed 3-10 non-target O. glaberrima introgressions in the Milyang 23 background. These five lines were evaluated for seven agronomic traits including 1,000 grain weight and spikelets per panicle and also genotyped with seven SSR markers. Four lines were informative in delimiting the position of QTLs, qGW3 and qSPP3. Two lines with the O. glaberrima segment flanked by SSR markers, RM60 and RM523 displayed significantly higher values than Milyang 23 in GW and SPP whereas two lines without that O. glaberrima segment displayed no difference in GW and SPP compared to Milyang 23. The result indicates that two QTL, qGW3 and qSPP3 are located in the interval between RM60 and RM523 which are 1.2-Mb apart. Introgression lines having QTLs, qGW3 and qSPP3 would be useful materials not only to indentify the relationship between these two yield QTLs, but also to develop high yielding variety via marker-aided selection technology.

Analysis of Genetic Characteristic of Jujube (Ziziphus jujuba Mill.) Cultivated in Korea Revealed by ISSR Markers (ISSR 표지를 이용한 국내 재배 대추나무의 유전특성 분석)

  • Chinese jujube (Ziziphus jujuba Mill.), a member of family Rhamnaceae, is an economically important species. It has high nutritional value in fruits, and its medicinal properties have led to extensive use in traditional oriental medicine. In this study, we investigated the genetic characteristics of 270 jujube germplasms collected from the six major jujube fruit production areas in Korea including Boeun and Gyeongsan, in order to understand the current situation on cultivated varieties of domestic jujube and to obtain useful data for selection of superior germplasms and establishing their breeding plans. The results of ISSR marker analysis revealed that the Shannon's information index of the 270 germplasms was 0.107 and the genetic similarity was as high as 0.935 showing the dominance of a specific cultivar or germplasm, and almost 67% (180) of them were identified at the same genotype with 'Bokjo'. The domestic commercial jujube populations could be characterized by very low genetic diversity and thus may be vulnerable to external disturbances. In addition, it seems that most of the jujube fruits in domestic market were largely produced through parthenocarpy.
