Selection of a New Calanthe discolor Lindle Cultivar 'Saegdong' for color variation by natural population (자생새우란 화색변이주 "색동" 선발)

  • These studies were carried out to develop native Calanthe in Korea. Calanthe native to southern islands in korea has beautiful flowers with various color and sweet fragrance, and it has been reported to have very good ornamental value. Concerning the classified 57 lines had surveyed their characteristics during the three years, and confirmed which that had manifestation stably. And then, a line was developed and given a name of horticultural cultivar to ‘Saegdong’. The major characteristics of the selected line, ‘Saegdong’, were as follows. In its color of flower, sepal was reddish orange, petal was yellow and lip was yellow. ‘Saegdong’ had a bended-inner blooming type.

Intraplant Variations of Sesquiterpene Lactone Content in Lettuce Genetic Resources Grown in Two Cultivation Seasons

  • Inflorescence, stem, and leaf samples of lettuce grown in a greenhouse in spring and autumn seasons were assayed for sesquiterpene lactones (SLs) content by high performance liquid chromatography. The concentrations of SLs were significantly higher in the inflorescences followed by upper leaf and stem compared to the other plant parts in most of the samples. SLs content (sum of lactucin and lactucopicrin) in various tissues of lettuce cultivated in spring season varied from 5.7 to 22.5 fold ranging from $27.4{\mu}g/g$ dry weight (DW) in the upper stem (cultivar "PI 176588") as the lowest to as high as $2,292.0{\mu}g/g$ DW in the inflorescence (cultivar "709849-1"). During autumn cultivation, the concentration of SLs varied from 2.0 to 14.4 fold ranging from as low of $32.4{\mu}g/g$ DW in the lower stem (cultivar "PI176588") to as high of $838.0{\mu}g/g$ DW in the upper leaf (cultivar "Dambaesangchu"). Higher lactucin (1.2 to 5.6 fold) and lactucopicrin (1.1 to 3.9 fold) concentration was observed during spring compared to autumn cultivation in most of the samples. SLs content in various organs of lettuce increases from the basal plant part going upwards. As lactucin and lactucopicrin are the major SLs which affects the sensory property of lettuce, their quantitative variation in the lettuce cultivars is useful for breeding new varieties with better consumer acceptance.

Breeding of Medium Standard Rose, 'Honey Lemon' with White Color and Good Vase Life (절화수명이 긴 백색 중형 스탠다드 장미 'Honey Lemon' 육성)

  • A new standard rose cv. 'Honey Lemon' was bred by the cross between white standard cv. 'Escimo' and light pink standard cv. 'Medeo' at the National Institute of Horticultural Research Institute. The cross was made in 1999 and 'Honey Lemon' was finally selected in 2003 after investigating characteristics for three years from 2001 to 2003. 'Honey Lemon', a white standard cultivar grows vigorously and has good flower shape. The major characteristics of this cultivar are $248.0stems/m^2/year$ in yield, 53.0 cm in length of cut flower, 8.0 cm in flower diameter, 17.0 in petal number, and 16.9 days in vase life. This cultivar can be propagated by both cutting and grafting. The consumer's preference of this cultivar is relatively higher than that of control cv., 'Tineke'. (Registration number: 1151).

Studies on the Agronomic Characteristics in the Winter Primary Octaploid Triticale (추파형 Primary Octaploid Triticale의 농업형질에 따른 연구)

  • The experiment was dealt with an investigations on the agronomic potential of primary octraploid triticale in comparion with Chokwang, a major common wheat, local rye cultivar and new triticale selection Suweon #1 and Suweon #2. This octaploid triticale was originated from the cross of Jukoku #81 x Local rye cultivar. The results obtained were summarized as follows; Cold tolerance of the P-Tcl was better than those of the wheat cultivar Chokwang, triticale varieties Suweon #1 and Suweon #2, and comparable to local rye. Culm length of P-Tel was 113cm that was intermediate between the wheat and rye, and its culm thickness was thiner than the checks except the rye. Lateness of heading and maturing date of the P-Tel seemed to be over-dominant. However, flowering date after heading was 3 to 7 days earlier than the Suweon #1 and Suweon #2. Yield potential of the P-Tel was poor in comparision with the triticale cultivar Suweon #1 and Suweon #2 which was attributed to the low fertility and less number of the heads per squ are meter. If it was . improved the low fertility and less number of the valid tillers through the cross with the good secondary triticale it seemed to be an useful material for triticale breeding, expecially for improving cold tolerant winter triticale varieties in Korea.

Effect of Small Paper Bag Coverage at Young Fruit Stage on the Reduction of Fruit Damage Incidence and Quality Deterioration during 'Hwasan' Pear Cultivar (유과기 소봉지 괘대가 '화산배' 병해 및 품질에 미치는 영향)

  • Different timings of coverage by a small paper bag at the young fruit stage were tested in order to evaluate the effect on the protection of fruit damage occurred by disease or pest, and quality deterioration during organic cultivation of 'Hwasan' (Pyrus pyrifolia Nakai) pear cultivar. The occurrence of four major moth populations and two major diseases in pears were monitored from April to September 2015. Among major moths and diseases, Grapholita molesta and pear rust were found often in the organic pear orchard, and they were observed frequently in May. Coverage by a small paper bag at the young fruit stage resulted in better fruit quality and a higher proportion of commercial fruits compared to a coverage timing of conventional paper bag. Interestingly, the fastest coverage timing of a small paper bag at the young fruit stage (when it was covered in April) showed the lowest occurrence of damaged fruits, which indicates that it successfully protected fruits from the damage caused by moths and diseases during April and May. Moreover, this timing of small paper bag coverage did not negatively affect the major qualitative and quantitative characteristics of fruits. These results suggest that early coverage by a small paper bag at the young fruit stage can be considered an efficient management strategy for organic cultivation of 'Hwasan' pear cultivar.

Development of dry milling suitable rice cultivar to invigorate rice processing products

  • Rice consumption has been continuously decreasing as the eating habits of Koreans have become westernized and diversified. The per capita annual rice consumption in Korea has dropped sharply from 136.4 kg in 1970 to 61.9 kg in 2016. The Korean government, therefore, has been trying to promote rice consumption by invigorating the processed food industry using rice flour. To facilitate the market for processed rice foods, it is essential to develop proper milling technology in terms of flour particle size and damaged starch content to produce high quality rice flour at competitive cost. Dry milling and wet milling are the two major processes used to produce rice flour. Although the dry milling process is relatively simple with a lower production cost, damaged starch content increases because of the high grain hardness of rice. In wet milling, the quality of rice flour is improved by reducing flour particle size as well as damaged starch content through soaking procedures. However, the production costs are high because of the additional expenses associated with the disposal of waste water, sterilization and drying of the wet flour. Recently developed technologies such as jet milling and cryogenic milling also require expensive investment and production. Therefore, developing new rice cultivars with dry milling adaptability as well as good processing properties is an important goal of rice breeding in Korea. 'Suweon 542' is a floury endosperm mutant line derived from sodium azide treatment on a high-yield, early maturing, and non-glutinous japonica rice cultivar, 'Namil'. Compared with the wild type, after dry milling process, the grain hardness of 'Suweon 542' was significantly lower because of its round and loosely packed starch granules. Also, the flour of 'Suweon 542' had significantly smaller particles and less damaged starch than 'Namil' and other rice cultivars and its particle size distribution was similar to a commercial wheat cultivar. Recently, through collaborations with nine universities and food companies, a total of 21 kinds of processed prototypes, using the dry milling flour of 'Suweon 542', were evaluated. In the production of major rice processing products, there was no significant quality difference between the flours prepared by wet milling and dry milling. Although the amount of water added to the dough was slightly increased, it was confirmed that the recipe applying the wet flour could be used without significant change. To efficiently transfer the floury endosperm characteristics of 'Suweon 542' to other commercial rice cultivars, it is essential to develop DNA marker tightly linked to the target gene. Association analysis using 70 genome-wide SSR markers and 94 F2 plants derived from 'Suweon 542'/'Milyang 23' showed that markers on chromosome 5 explained a large portion of the variation in floury grains percentage (FGP). Further analysis with an increased number of SSR markers revealed that the floury endosperm of 'Suweon 542' was directed by a major recessive locus, flo7(t), located in the 19.33-19.86 Mbp region of chromosome 5, with RM18639 explaining 92.2% of FGP variation in the F2 population. Through further physical mapping, a co-segregate and co-dominant DNA marker with the locus, flo7(t) was successfully developed, by which, thereby, breeding efficiency of rice cultivars having proper dry milling adaptability with high yield potential or useful functional materials would be improved. 'Suweon 542' maintained the early maturity of the wild type, Namil, which can be used in rice-wheat double cropping systems in Korea not only for improved arable land but also for sharing flour production facilities. In addition to the high susceptibility against major rice diseases, nevertheless, another possible drawback of 'Suweon 542' is the high rate of viviparous under prolonged rainfall during the harvesting season. To overcome susceptibility and vivipary of 'Suweon 542', the progeny lines, derived from the crosses 'Suweon 542' and 'Jopyeong', an early maturing rice cultivar with multiple resistance against rice blast, bacterial blight, and rice strip virus, and 'Heugjinju', a anthocyanin pigment containing black rice cultivar, were intensively evaluated. As the outputs, three dry milling suitable rice elite lines, 'Jeonju614', 'Jeonju615', and 'Jeonju616' were developed.

  • 4, 8, 13-Duvatriene-1, 3-viols have been identified in the cuticular lipid of fresh Sohyang (aromatic cultivar) tobacco leaves. Their $\alpha, \beta$-isomers were directly separated by SE-54 glass capillary column and identified by mass spectrometry. Gas chromatographic analysis revealed that the duvatrienediols are major components in the culticular lipid of Nicotiana Tabacum.

  • Whole genome sequence of Abeliophyllum distichum Nakai (Oleaceae) cultivar Ok Hwang 1 Ho, which is Korean endemic species, was recently sequenced to understand its characteristics. Acteoside is one of major useful compounds presenting various activities, and its several proposed biochemical pathways were reviewed and integrated to make precise biochemical pathway. Utilizing MetaPre-AITM which was developed for predicting secondary metabolites based on whole genome with the precise biochemical pathway of acteoside and the InfoBoss Pathway Database, we successfully rescued all enzymes involved in this pathway from the genome sequences, presenting that A. distichum cultivar Ok Hwang 1 Ho may produce acteoside. High-performance liquid chromatography result displayed that callus of A. distichum cultivar Ok Hwang 1 Ho contained acteoside as well as isoacteoside which may be derived from acteoside. Taken together, we successfully showed that MetaPre-AITM can predict secondary metabolite from plant whole genomes. In addition, this method will be efficient to predict secondary metabolites of many plant species because DNA can be analyzed more stability than chemical compounds.

Evaluation of Antithrombosis and Antioxidant Activities of the Ethanol Extract of Different Parts of Hibiscus cannabinus L. cv. 'Jangdae' (케나프 장대 품종의 부위별 에탄올 추출물의 항혈전 및 항산화 활성)

  • Kenaf (Hibiscus cannabinus L.), one of the four major fiber crops, is attracting attention for its efficient CO2-absorbing ability and versatility for producing daily supplies, including textiles. In Korea, a new cultivar 'Jangdae' was established in 2013. The ease of cultivation and seed gathering of 'Jangdae' has led to its nationwide cultivation. However, evaluation of the bioactivities of the different parts of kenaf, and especially the 'Jangdae' cultivar, remains rudimentary. In this study, the antithrombosis and antioxidant activities of extracts prepared from different parts of the 'Jangdae' cultivar were evaluated by determining their effects on blood clot formation. Extracts prepared from seeds (HC-SD), seedpods (HC-SP), leaves (HC-L), stems (HC-S), and roots (HC-R) of the 'Jangdae' cultivar strongly inhibited blood clot formation. In particular, the HC-SD, HC-SP, and HC-S extracts showed strong inhibition against the coagulation factors prothrombin, and thrombin. The HC-SP extract showed strong antioxidant activities, such as scavenging ability against DPPH anion, ABTS cation, nitrite, and reducing power. Since blood clot formation is closely related to oxidative stress, the HC-SP extract could be developed as a novel anticoagulation and antioxidant resource. This is the first report of the antithrombosis activities of different parts of H. cannabinus L. cv. 'Jangdae'.
