Changes of Antioxidant Activities on Cultured Ginseng with Mushroom Mycelia During Cultivation (배양기간에 따른 버섯균사체 인삼배양물의 항산화활성)

  • This study was conducted to investigate the antioxidant activities of the cultured ginseng with mushroom mycelia (Phellinus linteus (PL), Ganoderma lucidum (GL), and Hericium erinaceum (HE)) during cultivation periods of 10, 20, 30, 40, and 50 days. The lyophilized powder from the cultured ginseng with mycelia was extracted with 80% ethanol, and then evaluated for antioxidant activities. Total phenolic contents ranged from 149.63 to 205.91 mg/g, and the highest value was 80% EtOH extract from the cultured ginseng with GL at 30 days. The highest antioxidant activity ($IC_{50}$) for DPPH was 1.16 mg/mL in the cultured ginseng with HE at 40 days, and total antioxidant activity for ABTS was the highest value of 4.03 mg AA eq/g in PL cultivation at 30 days. $\alpha$-Glucosidase inhibitory activity was the highest value of 92.51% in EtOH extract from the cultured ginseng with PL at 50 days, and tyrosinase inhibitory activity was highest value of 13.21% in GL cultivation at 40 days. These results suggest that mushroom mycelium cultivation period for enhancement of antioxidant activity might be 40 days.

Phylogenetic Relationships between the Genus Inonotus and its Related Genera Based on the Nucleotide Sequences of Internal Transcribed Spacers (ITS 염기서열에 기초한 차가버섯과 근연속간 유연관계분석)

  • In this study the ITS1, ITS2 and 5.8S ribosomal DNA sequences from 29 strains of the Genus Inonotus and its related genera were compared with 31 strains obtained from GenBank database. Using the neighbor-joining (NJ) method and most parsimonious analysis the phylogenetic tree was constructed. The hymenochaetales formed no monophyletic group and several non-hymenochaetales appeared as intermingled with the Hymenochaetales. Strains 6, 46, 49, 50, 53, 55 showed no certain affinities within the Hymenochaetales, whereas Inonotus sp. (51) was closely related to Phellinus baumii, and Inonotus sp. (52), and Inonotus glomeratus (10) was related to Phellinus linteus, and Fomes fomentarius (30) was related to Ganoderma lucidum. Inonotus sp. and Phellinus sp. formed no monophyletic groups and a subdivision in the following genera is accepted: Inonotus sp. Phellinus baumii, Phellinus linteus, Phellinus igniarius, Phellinus pini, Hericium erinaceum, Ganoderma lucidum and Sparassis sp. were confirmed and separated genera. The taxonomic status of Inonotus remained uncertain. Eight new combinations are proposed.

버섯의 항암효과 -영자 균사체 다당류의 항종양 활성-

  • 영지의 약리효과는 triterpenoid계의 저분자와 polysaccharide인 고분자로 대별할 수 있다. 저분자 물질은 항염증작용, 항알러지작용, 간암세포 성장 억제작용, 혈압 강하작용, 혈소판 응집저해작용 등이 있으며, 고분자 물질은 혈압강하작용, 정혈작용, 고지혈증 개선작용, 혈당 강하작용, 면역 증강작용 및 항종양작용 등이 있는 것으로 알려졌다. 현재까지의 영지에 관한 연구는 주로 자실체에 대한 연구가 주류를 이루고 있으며 이들의 산지, 환경조건, 채집시기 및 실험자들에 따라 약리 효과 등에 많은 차이가 있다. 이들에 대한 효과가 영지의 대표적 약리효과라고 언급하기엔 부족한 점이 많으므로 약리효과가 우수하고 일관성이 있는 영지 유래 다당류를 얻기 위한 방법으로서는 균사체의 대량배양법 및 분리방법이 확립 되어야만 한다. 본 연구에서는 새로운 항암활성 다당류의 개발을 위해 국내 자생 영지를 채집하여 검색 및 계통적 분류를 행하였으며 분리된 균주의 최적 액체배양 조건, 다당류 분리 조건, 다당류의 물리화학적 특성 및 약리활성 등에 관한 연구를 실시하였다. 그 중 약리활성이 우수한 G- lucidum IY009 균주를 선발 하였다. 구조적 특징과 종양 면역 활성과의 관계 규명을 위해, G- lucidum IY009 배양균사체로부터 다당류를 T-AS, U-AS, M-AS, S 및 H로 분획 하였다. 추출조건에 따른 다당류 GLG의 수율은 열수 추출 분획이 2.98g/l로 가장 높았으며, U-AS및 T-AS에서 각각 2.32g/l, 2.07g/l이었다. GLG의 특성을 조사하기 위하여 수용성과 비수용성으로 분리한 결과 수용성 분획은 비수용성 분획보다 높은 당 함량을 나타냈으며, T-AS는 70.3%의 당과 7.8%의 단백질로 구성 되었다. GLG 대부분의 분획들은 60~93%의 glucose로 구성된 다당류 이었으며, 주로 $\beta$-glucose로 구성된 다당류 이었다. 아미노산은 Asp 및 Glu의 산성 아미노산과 Ala, Leu 등의 함량이 높게 나타났으며, 비알칼리 추출물에서 Ser과 Thr의 함량이 높게 나타났다. 다당류 T-AS는 평균 분자량이 2,000 kD와 12kD에서 주 peak를 나타냈으며, 수용성 분획의 평균 분자량은 12kD이고 비수용성 분획은 36~2,000 kD의 평균 분자량 분포를 갖는 것으로 나타났다. IR과 NMR 분석 결과 890 cm-1에서 흡수 peak를 나타내어 $\beta$-(1,3)0glucan과 $\beta$-(1,6)-glucan의 구조를 갖는 다당류로 확인 되었다. T-AS 분획은 C:H:O:N의 함량비가 38.9:5.7:49.6:1.84%이며, 이 물질의 융점은 163 $^{\circ}C$로 연한 갈색을 나타낸다. 분리된 GLG의 항암활성 기전 규명을 위해, in vivo 항암실험, 항보체 활성능, 항체 생성능, serum protein 분비능, 대식세포의 탐식능과 활성능 및 세포간 물질 분비 등의 상관관계를 조사하였다. 다당류 GLG 분획물들 가운데 항보체의 활성이 높았던 분획은 sarcome 180에 대한 항암 활성이 높게 나타났다. 다당류 T-AS의 보체 활성화 기작은 classical과 alternative complement pathway의 양 경로를 통해 활성화 되었다. T-AS 분획은 mouse내의 특정 혈청단백을 증가시켰으며, 항체 생성능의 증가가 관찰되어 effect T 세포의 활성화가 나타나고 있음을 알 수 있었다. T-AS는 생체내 투여시에 대식세포의 탐식능이 증진되었으며, 대식세포 기능 저해제에 의한 대식세포의 기능 저해 현상이 회복되었다. 이와 같은 결과들로부터, T-AS의 항암 활성은 활성화된 보체 성분 및 당 수용체들이 존재하는 대식세포의 개입을 시사한다.

Changes of Saponin and β-Glucan Content on the Cultured Ginseng with Mushroom Mycelia (버섯균사체로 배양된 인삼 Saponin과 β-Glucan 함량 변화)

  • This study investigated the changes of saponin and $\beta$-glucan content on the cultured ginseng with mushroom mycelia of Phellinus linteus (PL), Ganoderma lucidum (GL), and Hericium erinaceum (HE). Cultured ginsengs with mushroom mycelia were extracted with 80% ethanol, fractionated with n-buthanol, and analysed for ginsenosides by high performance liquid chromatography (HPLC). Crude saponin content of raw ginseng was 4.11% (d.b) but cultured ginseng with mushroom mycelia of PL, GL, and HE were increased to 6.74, 6.77 and 6.23% (d.b), respectively. Ginsenoside-Rd, among the 12 ginsenosides which were available for analysis, was remarkably increased to 13.61, 24.26, and 32.69 mg/g, respectively (raw ginseng: 0.80 mg/g). The $\beta$-glucan content of cultured ginseng with mushroom mycelia of PL, GL, and HE were decreased to 8.85, 5.51 and 5.46% rather than mushroom mycelia of 29.14, 19.44, and 23.39% (d.b), respectively.

Study on the anti-allergy activity of mushrooms in IgE-sensitized RBL-2H3 cells (IgE로 감작된 RBL-2H3 세포에서 버섯자원의 항알레르기 활성 탐색)

  • For evaluating the anti-allergy activity of mushrooms, forty two mushroom extracts were studied their inhibitory activities on the IgE-mediated degranulation in rat basophilic leukemia cell line (RBL-2H3 cell). Effects of mushroom extracts on the release of interleukin-4 (IL-4) and beta-hexosaminidase, and the cell viability of the IgE-sensitized were measured. From the analysis, five mushroom extracts such as the water extract from Flammulina velutipes (Curt.:Fr.) Sing showed the suppressive activities on IL-4 release as 20%. Eight extracts including the water extract of Ganoserma lucidum showed the suppressive activities on ${\beta}$-hexosaminidase release as 20%. Almost all of the extracts stimulated the proliferation of RBL-2H3 cell. The water extracts of Flammulina velutipes and Phellinus linteus were examined against the inhibitory activity in the production of IL-4 and ${\beta}$-hexosaminidase. Additionally, the extracts from Ganoserma lucidum, Isaria japonica, Phellinus linteus and Pleurotus ostreatus inhibited dose-dependently on ${\beta}$-hexosaminidase production. In conclusion, the result suggests that the mushrooms with the potent inhibitory efficacies on the degranulation of the mast cell would be candidate resources for the anti-allergy resources, and thus need to study for their utilization.

Antitumor effect of Ganoderma lucidum : Cytotoxicity and Tumor Growth Delayt(1) (영지버섯의 항암효과 :세포독성과 종양의 성장억제에 미치는 영향(1))

  • Purpose :. To investigate the effect of aqueous extract of Ganoderma lucidum(G.I.) on the surival of tumor cells in vitro and on the growth of tumors in vivo. Materials and Methods : Dried G.I. was made into powder, extracted with distilled water, filtered and diluted from a maximum concentration of 100 mg/ml in sequence. The cytotoxicity of G.I, in vitro was evaluated from its ability to reduce the clonogenicity of SCK tumor cells. For the tumor growth delay study, about $2{\times}10^5$$ of SCK tumor cells were subcutaneously inoculated in the legs of A/J mice. The first experimental group of mice were injected i.p. with 0.2ml of 250 mg/kg of G.I. from the first day after tomor inoculation for 10 days. The second experimental group of mice were injected i.p. with 0.2ml of 250 mg/kg of G.I. either once a day for 10 days or twice a day for 5 days beginining from the 7th day after tumor inoculation Results : 1. Cytotoxicity in vitro;survival fraction, as judged from the curve, at G.I. concentration of 0.5, 1,5, 10, 25, 50 and 100 mg/ml were 1.0, $0.74{\pm}0.03$, $0.18{\pm}0.03$, $0.15{\pm}0.02$, $0.006{\pm}0.002$, 0.015 and 0.0015, respectively. 2. Tumor growth delay in vivo; a) the time required for the mean tumor volume to grow to $1,000mm^3$ was 11 days in the control group and 14 days in the experimental group. b) the time required for tumor volume to increase 4 times was 11 days in the control group while it was 10.5 and 12 days in the groups injected with G.I. once a day and twice a day from the 7th day after tumor inoculation respectively. Conclusion : Aqueous extracts of G.I. showed a marked cytotoxicity on the SCK mammary cells in vitro. Tumor growth delay was statistically signiricant when G.I. in-jection was started soon after tumor inoculation, but it was not significant when injection was started after the tumors were firmly established.

Preliminary Screening of Some Jeju Island Native Plants for Whitening and Antioxidant Activity (제주도 자생식물들에 대한 미백 및 항산화 효능 탐색)

  • In this study, we investigated the whitening and anti-oxidant activity of 37 Jeju island native plants. The active ingredients of the plants were prepared by methanol extraction. Whitening activity of plant extracts was examined from the inhibitory effect of tyrosinase and the inhibition of melanin synthesis of the B16-F1 cell line. Their anti-oxidant activity was measured by electron donating ability of DPPH (1,1-diphenyl-2-picrylhydrazyl) and ROS (reactive oxygen species) scavenging activity in V79-4 lung fibroblast cells using DCF-DA (dichlorofluorescin diacetate). Cytotoxicity of the extracts on cell s based experiments was investigated by MTT (3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide) assay. Also, the toxicity test using a rabbit and the human skin patch test were carried out for examining the safety of the extracts which showed the high whitening activity. It is interesting that the extracts of Lespedeza cuneata, Ligustrum lucidum (stem), Morus bombycis (stem) and Prunella vulgaris var. lilacina showed both potent whitening and anti-oxidant activities.

Thermophile mushroom cultivation in Cambodia: Spawn production and development of a new substrate, acacia tree sawdust

  • To minimize cultivation costs, prevent insect-pest infestation, and improve the production efficiency of thermophilic mushrooms, plant substrates obtained from local areas in Cambodia were used for production of both spawn and mushrooms. In this experiment, different sawdusts different organic wastes and grain ingredients and analyzed for improvement of spawn-production efficiency. Four thermophilic mushroom species, Pleurotus sajor-caju (oyster mushroom, Sambok), Ganoderma lucidum (deer horn shaped), Auricularia auricula (ear mushroom), and Lentinula edodes (shiitake), were used to identify efficient new substrates for spawn and mushroom production. Although the mycelia in the rubber tree sawdust medium showed a slightly slower growth rate (10.9 cm/15 days) than mycelia grown in grains (11.2 cm/15 days in rice seeds), rubber tree sawdust appeared to be an adequate replacement for grain spawn substrates. Th findings indicate that rubber tree sawdust, sugarcane bagasse, and acacia tree sawdust supplemented with rice bran and calcium carbonate could be new alternative the substrates for. Although sugarcane bagasse and rubber tree sawdust showed similarly high biological efficiencies (BE) of 60% and 60.8%, respectively, acacia tree sawdust exhibited relatively a low biological efficiency of 22.4%. However, it is expected that acacia sawdust has potential for the mushroom cultivation when supplemented with currently used sawdust substrates in Cambodia, because of its relatively low price. The price of the sawdust (20 kg sawdust= 6500 Riel or 1.6 USD) currently used was 6.5 times higher than the price of acacia sawdust (201000 Riel or 0.25 USD). Therefore, utilization for acacia sawdust for mushroom cultivation could become feasible as it would reduce by producing costs of mushrooms in rural areas of Cambodia.

Interspecific Protoplast Fusion of Ganoderma applanatum and Ganoderma lucidum and Fruit Body Formation of the Fusants (잔나비 걸상버섯과 영지(靈芝)의 종간원형질체(種間原形質體) 융합(融合)과 자실체형성(子實體形成)에 관한 연구(硏究))

  • Interspecific fusion products were obtained from fusion of Ganoderma applanatum and Ganoderma luridum. Frequency of fusion was 0.77-1.38%. Fusion products were selected by the comparision of morphology and color of colony. Fusion Products had chracteristics of two parental strains and generally grew faster than the parents. Some fusants were segregated on GCM. Fusion products were confirmed by mycelial morphology and electrophoretics pattern of esterase isozyme from mycelium. Most of fusion products were lack in clamp connection but those fusion products, that were segregated produced mycelium with and without clamp connection. Some of the fusion products produced fruit body on sawdust medium.

The Effects of Extracts Mixture Drink from Inonotus Obliquus, Phellinus Linteus and Ganoderma Lucidum on Hematopoietic Stem Cells and Lymphocyte Subset of Blood in Human (차가버섯, 상황버섯 및 영지버섯 복합추출물 복용이 인체의 혈중 조혈모세포와 면역세포에 미치는 영향)

  • This study was performed to investigate the effect of extract mixture(IPGE) drink from Inonotus Obliquus, Phellinus Linteus and Ganoderma Lucidum on hematopoietic stem cells and lymphocyte subset[lymphocyte, $CD4^+T$ cell, $CD8^+T$ cell, Natural Killer(NK) cells] of blood in 37 participants who were healthy and about $40{\sim}70$ years old. They were divided into two groups; extract mixture drink administration group(n=27) and placebo administration group(n=12). They were given the test drink daily for 4 weeks. Blood was obtained from the subjects every two week in the beginning of administration day to evaluate the $CD34^+$ hematopoietic stem cells and immune cells. As results, $CD34^+$ hematopoietic stem cells were significantly increased after taking IPGE drink for 4 weeks compared to that before taking the drink (p<0.001). There was no significant changes in number of lymphocytes, $CD4^+T$ cells, $CD8^+T$ cells, NK cells and in the ratio of $CD4^+/CD8^+$ cell after taking the test drink. From these results, it was suggested that IPGE have a good health effect by promoting the proliferation of the hematopoietic stem cells.
