아토피피부염이 유발된 마우스에 대한 3종 가감방풍통성산(활석 제외)의 효과 비교 (Effects of Three Types of GagamBangpungtongseong-san(Except Talcum) on the Atopic Dermatitis in Mice)

  • Objectives : This study was to examine the effects of 3 types of BTS which were excluded talcum only or replaced talcum to Lonicera japonicae Flos or Kochiae Fructus on the DNCB-induced atopic dermatitis in mice. Methods : In this study, Balb/c mice were divided into five groups: normal, control, GBT(BTS except talcum), GBTG(GBT added Lonicera japonicae Flos), and GBTJ(GBT added Kochiae Fructus). And the effects on atopic dermatitis were evaluated by weight change, ear's thickness and weight, thickness of dorsal skin, severity scale of dorsal skin, histopathologic findings of dorsal skin by H&E and toluidine blue stain, proliferation of splenocyte and thymocyte in vitro, proliferation of splenocyte in vivo, IL-4, TNF-α, IgE in serum. Results : There were no significantly changes in body weight and effect of ear's weight in GBT, GBTG, and GBTJ group. The thickness of ear of GBT and GBTJ group showed significant decrease. And the thickness of dorsal skin of GBTJ group significantly decreased compared to the control, GBT, and GBTG group. All the treated groups significantly decreased in severity scale, histopathologically reduced epidermal thickness, and mast cell infiltration. In vitro, all the treated groups increased in the proliferation rates of splenocyte. However, in vivo study, it showed a falling tendency and GBT group significantly decreased compared to control, GBTG, and GBTJ group. In vitro study, GBTG group significantly decreased in the proliferation rates of thymocyte. There was no IgE contents chnage in GBT, GBTG, and GBTJ groups but IL-4 and TNF-α contents were significantly decreased. Conclusions : GBT, GBTG, and GBTJ are expected to improve symptoms of atopic dermatitis and further studies are needed for development of BTS's transformation.

금은화(金銀花) 열수 추출물이 피부 미백(美白)에 미치는 영향 (Effects of Hot Water Extracts from Lonicera Japonica Flos Extracts on whitening using B16F10 cell lines)

  • Objectives : This study was designed to investigate effects of Lonicera Flos Extracts(LFE) on whitening using B16F10 cell lines. Methods : In this experiment, we observed effects of LFE on cell viability, inhibitory effects of melanin synthesis, inhibitory effect on tyrosinase, tyrosinase activity, superoxide dismutase(SOD)-like activity and mRNA expression. Results : In results, more than $2000{\mu}g/ml$ of LFE treated group showed lowered cell viability rates significantly compared to albutin treated group. More than $2000{\mu}g/ml$ of LFE treated groups were lower levels of melanin synthesis. Inhibitory effects of melanin production showed in $1000{\mu}g/ml$ of LFE treated group. $1,000{\mu}g/ml$ of LFE treated group significantly suppressed tyrosinase activities in vitro. LFE and albutin treated group significantly decreased tyrosinase activity compared to non treated group. SOD-like activity of LFE treated group was lower than vitamin C treated group but increased depending on concentration. $500{\mu}g/ml$ of LFE treated group and $1,000{\mu}g/ml$ of LFE treated group was significantly increased. Tyrosinase mRNA expression of ${\alpha}$-MSH and LFE $250{\mu}g/ml$ treated group significantly decreased compared to ${\alpha}$-MSH treated group. Conclusions : These results suggest that LFE can inhibit melanin synthesis through inhibitory action on tyrosinase activity. And LFE suppressed tyrosinase activities B16F10 cells significantly. So I suggest LFE can apply to whitening.

초임계유체 $CO_2$ 및 Co-solvent 첨가에 따른 금은화(Lonicera fles)의 Flavonoid류 추출특성 (Extraction Characteristics of Flavonoids from Lonicera flos by Supercritical Fluid Carbon Dioxide ($SF-CO_2$) with Co-solvent)

  • 순수 methanol을 cd-solvent로 첨가하였을 때 소량의 수율을 얻을 수 있었던 반면 methanol에 물을 첨가하여 수용액을 조제한 후 각각 첨가하였을 때 co-solvent의 극성도 증가에 비례하여 수율이 현저하게 증가하였다. 60% methanol을 첨가하였을 때 수율이 가장 높았으며 순수 methanol 첨가구와 비교하여 luteolin이 5.9배, quercetin이 4.1배, apigenin이 7.8배 가량 증가하였다. Co-solvent에 citric acid를 농도별로 첨가한 결과 첨가량 1%까지는 추출수율이 농도 비례적으로 증가하는 경향을 나타내어 대조구에 비해 대략 luteolin이 14.64mg/100g, quercetin이 10.17mg/100g, apigenin이 2.4mg/100g의 유의적인 증가율을 보였으나 그 이상의 농도에서는 소량 감소하는 경향을 보였다. 한편 산을 첨가한 모든 조건에서 대조구보다 높은 수율을 얻을 수 있었다. 추출수율이 가장 우수하였던 citric acid가 1% 첨가된 60% aqueous methanol을 co-solvent로 초임계상태에 첨가하였을 때 공정압력이 100bar에서 200ber로 증가함에 따라 추출수율이 비례적으로 증가하였으나 250bar 이상에서는 감소하는 경향을 나타내었다. 온도에 따라서는 $40^{\circ}C$에서 $50^{\circ}C$까지는 증가하는 경향을 보이다 이상의 온도에서는 오히려 감소하였다. 추출 시간의 증가와 더불어 추출 수율은 유의적으로 증가하다가 60분일 때 최대값을 보였으며 90분 이상의 추출시간에서는 추출수율이 거의 일정해졌다. $CO_{2}$ 대비 cosolvent를 15(0.3mL/min)% 첨가하였을 때 최대 추출수율을 보였으며 25% 이상의 첨가구에서는 첨가량은 증가하였으나 수율은 감소하는 경향을 보였다. 것으로 나타났다. 그러므로 감마선 조사 및 저온저장($10^{\circ}C$)은 김밥재료 뿐만 아니라 김밥의 미생물 제어에 효과적인 것으로 확인되었다.와 비례하는 경향을 나타내었다. 또한 FA-swelling mica의 중금속 이온의 선택성은 Pb>Cu>Cd$\geq$Zn 순으로 나타났다.지 않았다.l years and a new type of transfer crane has been developed. 금은화(金銀花)추출물의 마우스 단회 경구투여 독성실험 (Mouse Single Oral Dose Toxicity Test of Lonicerae Flos Aqueous Extracts)

  • Objectives : The object of this study was to obtain accurate information (single oral dose toxicity) of Lonicerae Flos (LF; Dried flower bud parts of Lonicera japonica Thunb (Caprifoliaceae)), which has traditionally been used in Korean medicine for treating various inflammatory diseases. Methods : In order to observe the 50% lethal dose (LD 50), approximate lethal dosage (ALD) and target organs, test articles were once orally administered to female and male ICR mice at dose levels of 2,000, 1,000, 500 and 0 (control) mg/kg (body weight.). The mortality and changes on body weight, clinical signs and gross observation were monitored for 14 days after single oral treatment of LF aqueous extracts with organ weights and histopathological observations of 12 types of principle organs. Results : 1. After single oral treatment of LF aqueous extracts, we could not find any mortality and toxicological evidences up to 2,000 mg/kg treated group, the limited dosages in rodents at body and organ weights, clinical signs, gross and histopathological observations. 2. Slight diarrhea was detected in most mice treated with 2,000 mg/kg of LF aqueous extracts and male mice of LF aqueous extracts 1,000 mg/kg within 2 days after end of treatment, respectively. Conclusion : The results obtained in this study suggest that the LD 50 and ALD of LF aqueous extracts in both female and male mice after single oral treatment were considered as over 2,000 mg/kg because no mortalities were detected up to 2000 mg/kg, the highest dose recommended by KFDA and OECD. However, we also observed the possibility of digestive disorders like diarrhea when over 1,000 mg/kg of LF aqueous extracts were administered in the present study.

Protective Effect of Lonicerae Flos Aqueous Extracts on a Pressure Overload-induced Heart Failure Model

  • Objectives: Lonicerae flos (LF), a dried flower part of Lonicera japonica Thunb., has been widely used in Korean medicine as anti-inflammatory and antioxidative agent. The purpose of this study was to determine the cardioprotective effects of LF, through potential antioxidant effects, on the pressure overload (PO)-induced heart failure (HF) in C57BL/6 mice after transverse aortic constriction (TAC) surgery. Methods: Resveratrol (10 mg/kg body weight) or LF (125, 250 or 500 mg/kg body weight) was orally administered, once daily for 14 days, starting 14 days after TAC surgery. Changes in the mortality, body weights, heart weights, histopathology of the heart, and antioxidant defense systems of the heart were analyzed. Results: Marked and noticeable increases of heart weights, mortalities, and hypertrophic, focal, and lytic fibrotic histological changes in the LVs were observed, with destruction of heart antioxidant defense systems after surgery. However, HF signs, induced by TAC surgery through PO, and destruction of heart antioxidant defense systems were significantly and dose-dependently inhibited by 14 days of maintained oral treatment with LF 500, 250 or 125 mg/kg. Treatment with 250 mg/kg LF was comparable to treatment with 10 mg/kg resveratrol. Conclusions: The results in this study suggest that oral administration of LF favorably relieves PO-induced HF following TAC, through increase of heart antioxidant defense systems. The overall effects of 250 mg/kg LF were similar to those of 10 mg/kg resveratrol. More detailed mechanistic studies should be conducted in the future, with screening of the biologically active compounds in LF.

인동 추출물의 성장호르몬 유발 효과 (Induction of Growth Hormone Release by the Extracts of Lonicera japonica $T_{HUNB.}$)

  • Lonicerae Flos (LF) has been used as an anticancer, anti-viral, and anti-inflammatory agent in traditional herbal medicine. In this study, induction of rat growth hormone (rGH) by addition of methanol (MeOH) extract of LF of Lonicerae (L.) Folium or several constituents of L. Folium were carried out in the pituitary cell culture system. Induced rGH level by addition of 70% MeOH extract of LF was increased to $732.65{\pm}105.64%$ of control (n=18, p<0.01), however, the other sequential fractions were not significantly different from the control. Ochnaflavone, a constituent of L. Folium, induced rGH level in the cell culture to $329.73{\pm}160.00%$ of control (n=6, p<0.01). An I..v. injection of the MeOH extract of LF did not increase plasma rGH level in anesthetized rats. Unfortunately, the MeOH extract of LF induced prolactin and LH release about 7 and 5 fold of the control, respectively (p<0.05, each). In conclusions, 70% MeOH extract of LF exerted induction of rGH release in rat pituitary cell culture. Further studies to investigate mechanisms of the inducded rGH by LF are in progress.

  • objectives: We investigated the moisturizing and cooling effects of the cosmetic product with Scrophularia Buergeriana, Poria Cocos, Lonicera Japonica, Portulaca Oleracea and Ginko Biloba extract on human skin by using non-invasive instrument. Methods: We measured physiological effects after volunteers(female, n=5) applied the cosmetic product containing the extract of oriental herbs. The forearm skin moisturization was measured with $Corneometer^{\circledR}$ CM825 after 15min, 3hr, and 6hr. The forearm skin surface roughness was with Scanning electron microscopy after 15min, and 6hr. The face skin temperatures cooling and microcirculation effects were measured with ThermaCAMTM PM525 and LDl-Periscan PlM II after 15min, and 30min. Results: The forearm skin moisturization was imcreased 108% after 15min. The forearm skin surface roughness was decreased significantly after 15min. The face skin temperatures were cooling down to $30.7^{\circ}C$. The face skin microcirculation effect was normalized time-dependently. Conclusions: We concluded that cosmetic product containing the extract of Scrophularia Buergeriana, Poria Cocos, Lonicera Japonica, Portulaca Oleracea and Ginko Biloba had a moisturization effect and improving the surface roughness, temperature and microcirculation on the human skin.

HaCaT Cell에서 황기 에탄올 추출물의 Aquaporin-3 발현 효과 (Ethanol Extract of Astragalus membranaceus Bunge Induces Aquaporin-3 Expression in HaCaT Cells)

  • Astragalus membranaceus Bunge is used in herbal medicine in Eastern Asian countries including Korea. In this study, we assessed the effects of A. membranaceus extract (AM) on the aquaporin-3 (AQP3) protein expression in HaCaT cells. AM did not affect viability of HaCaT cells. AQP3 expression and cell migration seem to be maximal at $100{\mu}g/mL$ concentration. Epidermal growth factor receptor (EGFR) kinase inhibitor, PD153035, blocked AM-induced AQP3 expression and cell migration. In addition, an 80% ethanol extracts of herbal prescription, SinhyoTakleesan (ST), which is composed of A. membranaceus, Angelicae gigantis, Glycyrrhiza glabra Linne, and Lonicera japonica Flos also induced AQP3 expression at $20{\mu}g/mL$ in HaCaT cells. Collectively, these results suggest that AM induce AQP3 expression via EGFR pathway.

문헌 고찰을 통한 비만 치료 한약의 장내미생물 변화 연구 (A Study on Changes in the Gut Microbiome of Herbal Medicines for Treating Obesity Using Literature Review)

  • Objective This study identified useful herbal medicines and prescriptions for obesity treatment by analyzing herbal medicines and prescriptions that showed meaningful results in weight loss by changing the gut microbiota. Methods Using PubMed, we selected and studied 23 papers showing meaningful results on weight loss through changes in the gut microbiota using herbal medicines. Result Of the 23 selected papers, 17 were of studies using herbal medicines, and 6 of studies using prescriptions. Pomegranate peel, Flos Lonicera, Rehmannia glutinosa, Rhein, Coix seed, Platycodon grandiflorus, mulberry leaves, Poria cocos, asperuloside, Bupleuri radix, Astragalus polysaccharides, Ephedra sinica, Ganoderma lucidum, Hirsutella sinensis, Caulis spatholobi, aconite, and Bletilla striata were used as herbal medicine. Linggui zhugan-tang, Bofutsushosan, Shenling baizhu powder, Chowiseungcheng-tang, Daesiho-tang, and Yijin-tang were used as prescription. Conclusion Seventeen herbal medicines and six prescriptions associated with meaningful results in weight loss through changes in the gut microbiota, suggest the possibility of treatment and prevention of obesity through herbal medicine.

Geumeunyoungji-tang Inhibits Serum IgE Level in Mouse Dermatitis Induced by 2,4-dinitrofluorobenzene

  • Lonicerae Flos containing formulation, Geumeumyoungji-tang (GYT), is an herbal prescription prepared using 5 different herbal drugs, namely, Lonicerae Flos, Ganoderma, Lactucae Herba, Xanthii Fructus, and Smilacis Rhizoma. This study was focused on the investigation of the pharmacological effects of GYT on allergic reactions. As the first step of the study, GYT was administered BALB/c mice which were sensitized by 2,4-dinitrofluorobenzene (DNFB). As the result GYT ameliorated dermatitis provoked by DNFB. The serum IgE level of the DNFB sensitized-mouse was significantly decreased when GYT was administered. In order to confirm the moderating effect of this prescription on allergic reaction, GYT was pretreated to human mast cells (HMC-1) before they were stimulated by phorbol-12-myristate 13-acetate plus calcium ionophore A23187 (PMACI). GYT suppressed secretion of inflammatory cytokines, interleukin (IL)-6 and IL-8, from HMC-1. Additionally, pretreatment of GYT showed regulating effect on COX-2 expression. Collectively, these findings provide insights into the pharmacological actions of GYT as a potential agent for treatment of allergic dermatitis.
