흰쥐의 간손상(肝損傷)에 대한 가미유간탕(加味愈肝湯)의 간(肝) 보호효과(保護效果) (Protective Effects of Gami Yugan-tang on Liver Damage in Rats)

  • In order to investigate the protective effects of Gami Yugan-tang on liver damage in rats induced by $CCl_{4}$ and d-galactosamine, the serum transaminase(ALT & AST), alkaline phosphatase(ALP), lactic dehydrogenase(LDH), superoxide dismutase(SOD), catalase, glutathione S-transferase(GST), glutathione peroxidase(GPX) for enzyme activities, and lipid peroxidation were measured. All animals were divided into 4 groups: normal group (untreated), control group (treated with 0.9% saline solution), sample I group (treated with 740mg/kg Gami Yugan-tang), and sample II group (treated with 1,480mg/kg Gami Yugan-tang). The results were as follows : 1. The results of liver damage in rats induced by $CCl_4$ : The protective effects of ALT were displayed in sample I and sample II, and AST, ALP, LDH, SOD, catalase, GST, GPX, and lipid peroxidation were noted in sample II group. It showed slight necrosis of hepatic cell and pathologic changes, for example, inflammatory cells infiltration were improved in sample II group compared to the control group. 2. The results of liver damage in rats induced by d-galactosamine : The inhibitory effects of AST, ALT, LDH, and ALP activities were noted in both sample I and sample II groups. The findings from this experiment suggests that Gami Yugan-tang has protective effects against liver damage in rats induced by $CCl_{4}$ and d-galactosamine.

  • The methanol extracts of five medicinal plant materials selected were tested for antihepatotoxic activity. Protective effects on the liver defect caused by $CCl_4$ can be proved by liver enzyme activities of serum GOT, GPT, LDH and ALP. The curative effects of these materials against $CCl_4-induced$ liver damage in albino rats were compared with those of control groups. It was shown that the extracts of Hepatica asiatica and Stellaria media showed antihepatotoxic effect on $CCl_4-induced$ liver damage; however, the extract of Gleditschia officinalis had no effect.

Protective Effect of ACTIValoe N-931 Complex, a Mixture of Aloe vera and Silybum marianum, on Experimental Acute Liver Injury

  • The aim of this study was to investigate the hepatoprotective effect of $ACTIValoe^{(R)}$ N-931 complex, a mixture of Aloe vera and Silybum marianum, against acute liver injuries. Acute liver damages were induced by intraperitoneal injection of galactosamine (GalN, 700 mg/kg), naphthylisothiocyanate (ANIT, 40 mg/kg) and ethionine (500 mg/kg). $ACTIValoe^{(R)}$ N-931 (85, 170 and 340) was administered orally 48 h, 24 h, 2 h before and 6 h after the injection of hepatotoxins. At 24 h after GalN treatment the levels of serum aminotransferases and hepatic lipid peroxidation were significantly elevated, whereas hepatic glutathione, serum triglyceride (TG) and total cholesterol were decreased. These changes were attenuated by $ACTIValoe^{(R)}$ N-931 complex. The serum aminotransferase activities and total bilirubin significantly increased at 48 h after ANIT treatment, but were attenuated by $ACTIValoe^{(R)}$ N-931 complex. The bile flow was lower after ANIT treatment, which was restored by $ACTIValoe^{(R)}$ N-931 complex. $ACTIValoe^{(R)}$ N-931 complex reduced the ethionine-induced elevated hepatic TG contents. Histopathological analysis revealed that signs of liver injury were prominent at 24 h as result of ethionine injection, demonstrated by extensive areas of fatty change and microvesicular steatosis were observed around cells. These changes were attenuated by $ACTIValoe^{(R)}$ N-931 complex. Our results suggest that the $ACTIValoe^{(R)}$ N-931 complex has a protective effect on acute liver injury.

오리나무 추출물(AI-1367)의 간질환 동물모델에서의 간 보호효과 (Hepatoprotecive Effects of Alnus japonica Extract on Experimental Liver Injury Models)

  • The protective effect of AI-1367 (Alnus japonica extract) on liver injury was investigated. Primary rat hepatocyte intoxication was induced by tert-butyl hydroperoxide (tBH), carbon tetrachloride ($CCl_4$), or D-glactosamine (D-GalN). Liver injury was induced by $CCl_4$, D-GalN or MCD (methionine choline deficient)-diet in mouse. The cellular leakage of lactate dehyrogenase and cell viability followed by the treatment of hepatotoxicants were significantly improved by AI-1367 treatment at a concentration range of 5~50 ${\mu}g/ml$ for tBH, 5~50 ${\mu}g/ml$ for D-GalN, and 5~100 ${\mu}g/ml$ for $CCl_4$, respectively. Treatment with AI-1367 (20, 10, 5 mg/kg, p.o.) on liver injury induced by subcutaneous injection of $CCl_4$ or D-GalN reduced significantly the levels of aspartate transaminase and alanine transaminase in serum. Histological observations revealed that fatty acid changes, hepatocyte necrosis and inflammatory cell infiltration in $CCl_4$ (D-GalN)-induced liver injury was improved by administration of AI-1367. AI-1367 treatment (10, 5, 2.5 mg/kg, p.o.) also significantly recovered the body weight change and serum levels of aspartate transaminase, alanine transaminase and triglyceride in liver injury induced by MCD diet. From these results, AI-1367 shows protective effects against tBH, $CCl_4$, D-GalN, or MCD diet-induced hepatotoxicity in vitro or in vivo.

Anti-oxidative Effect of a Protein from Cajanus indicus L against Acetaminophen-induced Hepato-nephro Toxicity

  • Overdoses of acetaminophen cause hepato-renal oxidative stress. The present study was undertaken to investigate the protective effect of a 43 kDa protein isolated from the herb Cajanus indicus, against acetaminophen-induced hepatic and renal toxicity. Male albino mice were treated with the protein for 4 days (intraperitoneally, 2 mg/kg body wt) prior or post to oral administration of acetaminophen (300 mg/kg body wt) for 2 days. Levels of different marker enzymes (namely, glutamate pyruvate transaminase and alkaline phosphatase), creatinine and blood urea nitrogen were measured in the experimental sera. Intracellular reactive oxygen species production and total antioxidant activity were also determined from acetaminophen and protein treated hepatocytes. Indices of different antioxidant enzymes (namely, superoxide dismutase, catalase, glutathione-S-transferase) as well as lipid peroxidation end-products and glutathione were determined in both liver and kidney homogenates. In addition, Cytochrome P450 activity was also measured from liver microsomes. Finally, histopathological studies were performed from liver sections of control, acetaminophen-treated and protein pre- and post-treated (along with acetaminophen) mice. Administration of acetaminophen increased all the serum markers and creatinine levels in mice sera along with the enhancement of hepatic and renal lipid peroxidation. Besides, application of acetaminophen to hepatocytes increased reactive oxygen species production and reduced the total antioxidant activity of the treated hepatocytes. It also reduced the levels of antioxidant enzymes and cellular reserves of glutathione in liver and kidney. In addition, acetaminophen enhanced the cytochrome P450 activity of liver microsomes. Treatment with the protein significantly reversed these changes to almost normal. Apart from these, histopathological changes also revealed the protective nature of the protein against acetaminophen induced necrotic damage of the liver tissues. Results suggest that the protein protects hepatic and renal tissues against oxidative damages and could be used as an effective protector against acetaminophen induced hepato-nephrotoxicity.

Acetaminophen 유도 간 손상에 대한 조릿대 애엽 추출물의 보호 효과 (Protective Effects of Sasa borealis Bamboo Browse Extract on Acetaminophen-induced Liver Damage in Mouse Model)

  • Acetaminophen (N-acety1-p-aminophenol, paracetamol) is widely used as an over-the-counter analgesic and antipyretic drug. Intake of a over dose of acetaminophen may result in severe hepatic necrosis. In this study, we investigated the liver damage in mice using single dose (300 mg/kg) of acetaminophen and the possible protective effects of administration (50-200 mg/kg body weight) of SB-Ex on acetaminophen-induced liver damage in mice. The alanine aminotransferase (ALT), and aspartate aminotransferase (AST) activities were determined in the plasma of mice. The effect of SB-Ex on lipid peroxidation product thiobarbituric reacting substances (TBARS) and some antioxidant enzymes superoxide dismutase (SOD), catalase, d-aminolevulinate dehydratase (${\sigma}$-ALA-D) activities, and gluthathione peroxidase (GPx), were also evaluated in the mouse liver homogenate. Acetaminophen caused liver damage as evident by statistically significant increased in plasma activities of AST and ALT. There were general statistically significant losses in the activities of SOD, catalase, ${\sigma}$-ALA-D, and GPx and an increase in TBARS in the liver of acetaminophen-treated group compared with the control group. However, SB-Ex was able to counteract these effects. These results suggest that SB-Ex can act as hepatoprotectives against acetaminophen toxicity and is a good candidate for further evaluation as an effective chemotherapeutic agent.

후코이단이 쥐의 급성 알콜성 간손상에 미치는 영향 (Protective Effects of Fucoidan against Acute Alcohol-induced Liver Injury in Rats)

  • 본 연구는 해조류에 존재하는 황산기를 함유하고 있는 후코이단의 급성 알코올 간 손상 모델에서의 간 보호능을 관찰하였다. 실험 결과 후코이단은 급성으로 알코올을 섭취시킨 쥐의 체중감소를 억제시켰으며, 알코올 섭취를 통해 나타날 수 있는 중성지질, 콜레스테롤, LDL-콜레스테롤 상승을 유의적으로 감소시킨 것으로 나타났다. 또한 간 기능을 대표하는 혈중의 GOT는 유의적인 차이는 없었으나, GPT는 유의적으로 후코이단 경구투여에 의해 감소되었다. 또한 후코이단은 지방이 비대해 지는 현상과 간세포의 지방축척을 감소시킨 것으로 관찰되었다. 따라서 이러한 결과로 보아 후코이단은 급성 알코올 섭취에 의한 간손상을 완화시켜 줄 수 있는 식품소재로 활용가능하다고 사료된다.

산수유의 유리자유기에 의한 간손상 보호효과 (Study on the Protective Effect of Corni Fructus against Free Radical Mediated Liver Damage)

  • Carbon tetrachloride ($CCl_4$)-induced liver injury depends on a toxic agent that has to be metabolized by the liver NAPDH-cytochrome P450 enzyme system to a highly reactive intermediate. Alternations in the activity of cytochrome P450 enzymes affect the susceptibility to hepatic injury from $CCl_4$. In this study, we evaluated the potential protective activity of the traditional Korean medicinal herb, Corni fructus (CF), against an experimental model of hepatotoxicity induced by $CCl_4$. The CF exhibited a hepatoprotective activity against $CCl_4-induced$ liver damage in Sprague-Dawley (SD) rats, as measured by GOT, GPT, ALP and histological observation. The CF also showed significant decrease of malodialdehyde (MDA) and increase of glutathion (GSH), catalase activity in rat liver homogenate. In addition, the expression of CYP2E1, as measured by reverse transcriptase-polymerase chain reaction (RT-PCR) and Western blot analysis, was significantly decreased in the liver of CF treated SD rats. But $CCl_4$ and CF has no significant effect on 1A1 and 3A1 isoform of cytochrome P450. Based on these findings, it is suggested that hepatoprotective effects of CF possibly related to antioxidative effects and regulation of CYP2E1 expression.

G009의 간 보호작용에 관한 연구 (Effects of G009 on Chemical-Induced Liver Damage in Rats)

  • The present study was performed to determine the protective effect of G009 on liver damage induced bv ethanol $CCl_4$ and thioacetamide in rats. In acute fatty liver animal model induced by ethanol, triglyceride accumulation was markedly decreased to the normal control level by 25 mg/kg G009 treatment. In addition, G009 significantly reduced serum ALT and AST levels in $CCl_4$-induced acute hepatitis animals. Treatment of G009 to the acute hepatitis rats induced by thioacetamide resulted in a dose dependent reduction of serum ALT level as well as AST level up to the normal control level. These protective effects of G009 were confirmed by histological examinations of the liver. These results suggested that G009 could be effective for the protection from the liver damage induced by ethanol, $CCl_4$and thioacetamide.

  • Five liglans, sauchinone (1), di-0-methyltetrahydrofuriguaiacin B (2), manassantin A (3), manassantin B (4) and saucerneol B (5), have been isolated from the MeOH extract of Saururus chinesis roots. The evaluation for protective effect of compounds 1-5 against tacrine-induced cytotoxicity in human liver-derived Hep G2 cells was conducted. Compounds 1,2, and 5 showed significant protective effects with the EC$_{50}$ values of74.2${\pm}$0.9, 111.3${\pm}$0.8,64.3${\pm}$0.8 ${\mu}$M, respectively. Silybin, one of the well-known hepatoprotective agents, used as a positive control, and also showed protective effect with an EC$_{50}$ value of 86.2${\pm}$0.5 ${\mu}$M.
