Effect of Dried Powder and Juice of Aster Scaber on Lipid Metabolism and Antioxidative Capacity in Rats (참취의 건분 및 녹즙이 흰쥐의 지방대사와 항산화능에 미치는 영향)

  • This study was performed to investigate the effects of dried powder and juice of Aster scaber on lipid metabolism and antioxidative capacity in rats. Twenty one male Sprague-Dawley rats weighing 170.9$\pm$15.1g were blocked into 3 groups according to body weight and raised for 4 weeks with diets containing 5%(w/w) dried powder and juice from the same amount of Aster scaber. Food intake, body weight gain, food efficiency ratio and organ weights were not different among all the experimental groups. On lipid metabolism, both dried powder and juice supplementation decreased plasma total lipid, triglyceride(TG), total cholesterol and liver cholesterol levels by increasing fecal lipid excretions. Plasma thiobarbituric acid reactive substance(TBARS) concentrations were significantly decreased in dried powder and juice groups, whereas liver TBARS concentrations were not affected by experimental diets. Red blood cell(RBC) superoxide dismutase(SOD) activities were significantly increased by dried powder and juice supplementation, while activities of other antioxidative enzymes in RBC and liver were decreased or showed no changes. In conclusion, both Aster scaber dried powder and juice had the effects on lowering lipid levels by increasing fecal lipid excretions, and inhibiting of plasma lipid peroxidation in animals and the effects of juice were higher than that of dried powder.   • Objectives : To investigate the effects of acupuncture at Gansoo (B-18) and Chungwan (CV-12) on lipid metabolism in rats fed peroxidized lipid diet. Methods : Effects of acupuncture at Gansoo and Chungwan on plasma and liver lipid composition and antioxidative capacity were investigated in rats fed peroxidized lipids. Results : Although the level of plasma total cholesterol and triglyceride showed a tendency to decrease in the acupuncture group, the plasma HDL-cholesterol concentration showed no significant difference. The level of liver total cholesterol and triglyceride showed no significant difference in all treatment groups. Thiobarbituric acid( TBARS ) values in plasma and liver showed a tendency to decrease in the acupuncture groups. The plasma GOT and GPT activities showed low values in the acupuncture groups. The liver glutathione peroxidase, superoxide dismutase and catalase activity showed high values in the acupuncture groups. Conclusions : The results suggest that acupuncture at Gansoo (B-18) and Chungwan (CV-12) may have an influence on lipid metabolism via enhancing antioxidative capacity.

Antioxidant Characteristics and Phytoremediation Potential of 27 Taxa of Roadside Trees at Industrial Complex Area (공단지역에서 생육하는 가로수 27종의 항산화특성과 잠재적 환경정화능력)

  • In order to screen for the best species for mitigating air pollutants by plants at an industrial complex area, we investigated antioxidant capacity, lipid peroxidation and nitrogen content in the leaves of 27 taxa of woody plants that are mostly utilized as roadside trees. Among 27 taxa, the highest value of antioxidant capacity was given by Cedrus deodara (91.4%) and the lowest one was by Firmiana simplex (56.9%). At lipid peroxidation level, little malondialdehyde (MDA) was observed in Lagerstroemia indica and Ginkgo biloba, but Platanus occidentalis, Castanoposis cuspidata var, sieboldii, Machilus thunbergii and Juniperus chinensis showed high MDA content. Antioxidant capacity of the deciduous woody plants was not significantly different in comparison with that of the evergreen ones. But MDA content of the deciduous woody plants was lower than that of the evergreen ones. The 27 taxa of woody plants appeared to be classified into four types: those of high antioxidant capacity and low lipid peroxidation, those of high antioxidant capacity and high lipid peroxidation, those of low antioxidant capacity and low lipid peroxidation, and those of low antioxidant capacity and high lipid peroxidation. The taxa included in these types are 7 (first type), 6 (second one), 8 (third one) and 6 (fourth one) taxa. first or second type species which have a high antioxidant capacity represented low nitrogen content in their leaves. However, third or forth type species which have low antioxidant capacity showed high nitrogen content in their leaves. Metasequoia glyptostroboides, Platycarya strobilacra and P. occidentalis which belong to the first or second type had extraordinarily high antioxidant capacity and high nitrogen content. Thus, three species are considered to be good phytoremediators for an industrial complex area.

  • Solid Lipid Nanoparticle(SLN), one of the colloidal carrier systems, has many advantages such as good biocompatibility, low toxicity and stability. In this paper, the effects of drug lipophilicity and surfactant on the drug loading capacity, particle size and drug release profile were examined. SLNs were prepared by homogenization of melted lipid dispersed in an aqueous surfactant solution. Ketoprofen, ibuprofen and pranoprofen were used as model drugs and tweens and poloxamers were tested for the effect of surfactant. Mean particle size of prepared SLNs was ranged from 100 to 150nm. The drug loading capacity was improved with the most lipophilic drug and low concentration of surfactant. Particle size and polydispersity of SLNs were changed according to the used lipid and surfactant. The rates of drug release were controlled by the loading drug and surfactant concentration. SLN system with effective drug loading efficiency and proper particle size for the intravenous or oral formulation can be prepared by selecting optimum drug and surfactant.

Effects of Adsorption Condition on Fat-binding Characteristics of Chitosan (흡착조건이 키토산의 지방질 흡착 특성에 미치는 영향)

  • To study the lipid adsorption characteristic of chitosans with different molecular weights and the degrees of deacetylation, in vitro test and near-infrared (NIR) spectroscopic analysis have been performed for the measurement of lipid adsorption characteristics of chitosan. The degrees of deacetylation in chitosans were $70{\%},\;85{\%}\;and\;92{\%}$ at different deacetylation times (1 hr, 2 hrs, 3 hrs), respectively. The molecular weight of each chitosan was controlled by enzymatic hydrolysis, and then the molecular weight of the chitosan was 4 kDa. The bulk density, water holding capacity and fat binding capacity of each chitosan powder were $96.2-504.0{\%},\;374.4-1217.9{\%},\;and\;307.0-659.3{\%}$, respectively. The higher molecular weight of chitosan was exhibited the lower bulk density and the higher water and fat binding capacities. Bindinf capacities of chitosan powders to bile salts, cholesterol and linoleic acid were $41.2-63.3{\%},\;40.8-67.4{\%},\;42.6-72.6{\%}$, respectively. In NIR spectrum of lipid adsorbed chitosan the occurrence static eletronical binding between chitosan and lipid was identified by NIR spectrum peak induced from combination of carboxylic group in lipid and amino group in chitosan. In conclusion, the higher degree of deacetylation and molecular weight of chitosan showed the higher lipid binding capacity and the lipid adsorption of chitosan were occurred by combination of carboxylic group in lipids and amino group in chitosan.

Effect of Chestnut and Acorn on Lipid Metabolism, Antioxidative Capacity and Antithrombptic Capacity in Rats (밤과 도토리의 과육 및 내피가 흰쥐의 지방대사, 항산화능 및 항혈전능에 미치는 영향)

  • This study wa performed to investigate the effect of dired powder of chestnut and acorn on lipid metabolism, antioxidative capacity and antithrombotic effect in rats. Fifty-four male Sprague-Dawley rats weighing 199$\pm$17g were blocked into nine groups according to their body weight. Rats were raised with diets containing only flesh or flesh with inner skin of 5% and 10% dried nut powders for four weeks. Food intake, body weight gain, food efficiency ratio and organ weight were no different among the experimental groups. The plasma and liver lipid levels of all the nut diet groups were lover than those of the control group. The nut diets showed hypolipidenic effect in the plasma and liver. Plasma and liver thiobarbituric acid reactive substance (TBARS) concentrations were significantly decreased in all the nut diet groups. The plasma TBARS levels of the inner skin groups were significantly different from the control group dose-dependently. Superoxide dismutase(SOD) activity was significantly different among the experimental groups, and all the nut groups showed higher activity than the control group. There were significant differences in SOD activity between the chestnut and acorn groups and the chestnut groups showed higher erythrocyte SOD activity and the acorn groups showed higher liver SOD activity than the other groups. Whereas catalase and GSH-Px activities in the erythrocyte and liver of both nut groups showed a tendency to increase, they were not significantly different among the experimental groups. The bleeding time and whole blood clotting time tended to be extended by feeding both types of nut but they were not significantly different among the experimental groups. Production of TX $B_2$ and PG $F_{1{\alpha}}$ was no different among the experimental groups. These results suggest that chestnut and acorn diets have the effect of lowering plasma and liver lipid levels, inhibiting lipid peroxide formation and increasing antioxidative enzymes activity. Thus, it is plausible that chestnut and acorn could be recommended in the treatment and prevention of cardiovascular diseases.

Effect of Lecithin Intake on Lipid Metabolism and Antioxidative Capacity in Rats Fed High Fat Diet (레시틴섭취가 고지방 식이를 섭취한 흰쥐의 지방대사와 항산화능에 미치는 영향)

  • This study was performed to investigate the effect of lecithin on lipid metabolism and antixidative capacity in 9-week-old rats. Forty-five male Sprague-Dawley rats weighing 249.8 g were blocked into three groups according to their body weight and raised for 8 weeks with experimental diets containing 1% (LM) or 5% lecithin (LH) and control (C) diet. Plasma and liver total lipids, triglyceride, total cholesterol and plasma HDL-cholesterol concenterations, and fecal total lipids, triglyceride, total cholesterol and bile acid excretions were measured. Malondialdehyde (MDA) levels in plasma, liver, superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GSH-Px) activities in red blood cell and liver, xanthine oxidase (XO) activities in plasma and liver, and total antioxidant status (TAS) in plasma were also measured. Effect of lecithin intake on antioxidative capacity was not significantly different among all the groups. Plasma total lipids, triglyceride and total cholesterol levels were lower in lecithin groups compared to control group, and these three lipid levels of lecithin groups were lowered dose-dependently as dietary lecithin level increased. But liver total lipids, triglyceride and total cholesterol levels were not different among all the groups. Also fecal total lipids, triglyceride and total cholesterol excretions were highest in high lecithin groups compared to two other groups. Thus it is plausible that lecithin intake decreases plasma lipid levels through increasing fecal lipid excretions, and may be beneficial for treatment and prevention of hyperlipidemia, but has no effect on antioxidative capacity.

Effects of a Rubus coreanus Miquel supplement on plasma antioxidant capacity in healthy Korean men

  • Korean raspberry, Rubus coreanus Miquel (RCM), contains high concentrations of phenolic compounds, which prevent oxidative stress. To determine the effect of RCM on antioxidant capacity in humans, we assessed in vivo lipid oxidation and antioxidant enzyme activities from plasma in 15 healthy men. The subjects ingested 30 g of freeze-dried RCM daily for 4 weeks. Blood was taken at baseline and at the end of the study to determine blood lipid profiles, fasting plasma glucose, liver function, lipid peroxidation, and antioxidant enzyme activities. RCM supplementation had no effect on blood lipid or fasting plasma glucose concentrations but decreased alkaline phosphatase activity. RCM supplementation increased glutathione peroxidase activities (P<0.05) but had no effect on lipid peroxidation. These results suggest that short-term RCM supplementation may offer health benefits by enhancing antioxidant capacity in a healthy population.

Effect of Korean Native Plant Diet on Lipid Metabolism, Antioxidative Capacity and Cadmium Detoxification in Rats (한국 고유의 산채류 첨가 식이가 흰쥐의 지방 대사 및 항산화능과 Cadmium 제독에 미치는 영향)

  • This study was performed to investigate the effects of Korean native plant diets on lipid and cadmium (Cd) metabolisms and also antioxidative enzyme activities in rats. Seventy male Sprague-Dawley rats weighing 210.3$\pm$2.7g were blocked into ten groups according to body weight. Rats were raised for four weeks with diets containing 0 or 0.04%(w/w) cadmium chloride and 5%(w/w) plant powder-Ssook(Artemisia princeps var. orientalis HARA), Chamchwi(Aster scaber THUNB), Gomchwi(Lingularia fischeri TUBCZ). or Solbeerum (Portulaca oleracea LINNE). Food intake, weight gain, food efficiency ratio, and weights of liver, kidney, spleen and epididymal fat were significantly lower in Cd-exposed groups, food efficiency ratio, and weights of liver, kidney, spleen and epididymal fat were significanlty lower in Cd-exposed groups. Plasma total lipid level, liver total lipid, cholesterol and triglycerde concentrations, and fecal total lipid, cholesterol and triglyceride excretions were decreased by Cd administration. Total lipid and triglyceride concentration in plasma and liver were lower in Chamchwi groups together with increased fecal total lipid and triglyceride excretions. Cholesterol concentration in plasma and liver were lower in Gomchwi groups with increased fecal cholesterol excretions. Activities of red-blood-cell superoxide dismutase(SOD), and liver catalase, SOD and glutathione peroxidase (GSH-px) were decreased significantly by Cd administration. Thiobarbituric acid reactive substance(TBARS) concentrations of plasma and liver were increased by Cd administration. Thiobarbituric acid reactive substance(TBARS) concentration of plasma and liver were increased by Cd administration. Administration of plant diets decreased plasma and liver catalase, SOD and GSH-px activities in Cd-free diet groups and increased them in Cd-administered groups. Plasma and liver TBARS concentrations were decreased in animals fed plant diets, with Chamchwi showing the most effective antioxidative capacity. The concentration of Cd accumulated in blood and tissue decreased in Cd-exposed groups fed plant diets. Liver Cd concentration were lower in the Chamchwi and Gomchwi groups, and kidney Cd levels were lower in the Ssook and Chamchwi groups. Among them, Chamchwi was the most effective in Cd detoxification. In conclusion, Chamchwi and Gomchwi were effective in lowering tissue lipid levels, with Chamchwi having the greatest antioxidative and detoxifying effects.

The Effect of Spirulina on Lipid Metabolism, Antioxidant Gapacity and Immune Function in Korean Elderlies (스피루리나 복용이 노인의 혈중 지질 농도, 항산화능 및 면역능에 미치는 영향)

  • This intervention study was performed to assess the effect of spirulina on lipid metabolism, antioxidant capacity, and immune function in elderly Koreans. The subjects were 6 male and 6 female people between the ages of 60 and 75, who were given spirulina supplements of 7.5 g/day for 24 weeks. Dietary intake, anthropometric measurements and biochemical assessment for plasma lipid levels, antioxidant status and immune function were measured before and throughout the intervention period. Before entering the study, the subjects were in relatively good health. Their nutrient intake was satisfactory, and anthropometric indices and plasma nutrient levels were within the normal range. Spirulina supplementation for 24 weeks did not affect dietary intake and anthropometric parameters. However, considerable changes were observed in blood lipid profiles, antioxidant capacity, and immune indices. The plasma concentrations of triglycerides, total- and LDL-cholesterol decreased from 4 weeks of the supplementation period. The antioxidant capacity improved, as shown in increasing TAS and decreasing TBARS after supplementation. Improved immune function was also observed as the PBL lymphocyte proliferation rate and plasma C3 levels increased. The above effects of spirulina supplementation did not differ between mild hypercholesterolemic (cholesterol $\geq$ 200mg/d1) and normochole-sterolemic (cholesterol < 200 mg/dl) subjects. This study provided evidence that spirulina could be used as dietary supplementation in nutritionally vulnerable groups to improve nutritional and health status and to prevent chronic disease such as hyperlipidemia or oxidation-prone diseases. further studies in this area with various population groups are warranted.
