Production and Characterization of a Recombinant Antibody Neutralizing Botulinum Neurotoxin A (보툴리눔 신경독소 A를 중화하는 재조합 항체의 제조와 특성 분석)

  • Botulinum neurotoxin (BoNT/A) is a neurotoxin that selectively attacks the peripheral cholinergic nerve endings. It is produced by Gram -positive, endospore-forming strict anaerobic bacteria, Clostridium botulinum. Since BoNT/A could be a biothreat agent, as well as a contaminator of food and water supplies, the development of sensitive assays for toxin detection and potent antitoxin for the treatment of intoxication is necessary. In this study, for the purpose of producing monoclonal antibodies (mAbs) that are capable of neutralizing Botulinum neurotoxin type A (BoNT/A), scFv (single-chain variable domain fragment) libraries from the rabbit antisera against BoNT/A was fused to a human IgG. The resulting recombinant scFvIgG antibody protein was expressed in stable cell lines and was purified using a protein A affinity chromatography. The efficacy of scFvIgG mAb was confirmed by ELISA and was evaluated for the neutralization of BoNT/A in vivo. Such an in vivo toxin neutralization assay was performed using mice. Although scFvIgG antibody proteins (10 ug) failed to fully protect the mice challenged with BoNT/A (100,000 $LD_{50}$), it significantly prolonged the survival time. These results suggest that scFvIgG mAb may be capable of neutralizing BoNT/A single-chain variable domain fragment.

A Prostate Segmentation of TRUS Image using Average Shape Model and SIFT Features (평균 형상 모델과 SIFT 특징을 이용한 TRUS 영상의 전립선 분할)

  • Prostate cancer is one of the most frequent cancers in men and is a major cause of mortality in the most of countries. In many diagnostic and treatment procedures for prostate disease, transrectal ultrasound(TRUS) images are being used because the cost is low. But, accurate detection of prostate boundaries is a challenging and difficult task due to weak prostate boundaries, speckle noises and the short range of gray levels. This paper proposes a method for automatic prostate segmentation in TRUS images using its average shape model and invariant features. This approach consists of 4 steps. First, it detects the probe position and the two straight lines connected to the probe using edge distribution. Next, it acquires 3 prostate patches which are in the middle of average model. The patches will be used to compare the features of prostate and nonprostate. Next, it compares and classifies which blocks are similar to 3 representative patches. Last, the boundaries from prior classification and the rough boundaries from first step are used to determine the segmentation. A number of experiments are conducted to validate this method and results showed that this new approach extracted the prostate boundary with less than 7.78% relative to boundary provided manually by experts.

Multi-view Image Generation from Stereoscopic Image Features and the Occlusion Region Extraction (가려짐 영역 검출 및 스테레오 영상 내의 특징들을 이용한 다시점 영상 생성)

  • In this paper, we propose a novel algorithm that generates multi-view images by using various image features obtained from the given stereoscopic images. In the proposed algorithm, we first create an intensity gradient saliency map from the given stereo images. And then we calculate a block-based optical flow that represents the relative movement(disparity) of each block with certain size between left and right images. And we also obtain the disparities of feature points that are extracted by SIFT(scale-invariant We then create a disparity saliency map by combining these extracted disparity features. Disparity saliency map is refined through the occlusion detection and removal of false disparities. Thirdly, we extract straight line segments in order to minimize the distortion of straight lines during the image warping. Finally, we generate multi-view images by grid mesh-based image warping algorithm. Extracted image features are used as constraints during grid mesh-based image warping. The experimental results show that the proposed algorithm performs better than the conventional DIBR algorithm in terms of visual quality.

A Study on the Fabrication of the Sensor Module for the Detection of Resistive Leakage Current (Igr) in Real Time and Its Reliability Evaluation (실시간 Igr 검출을 위한 센서 모듈의 제작 및 신뢰성 평가에 관한 연구)

  • The purpose of this study is to fabricate a sensor module to detect the resistive leakage current (Igr) in real time that occurs to low voltage electric lines and to verify its reliability. In the case of the developed sensor module, wires are inserted into the zero current transformer (ZCT) and current transformer (CT) in advance and then the branch line is connected to the circuit breaker. The measurement result of the resistance of the distribution panel equipped with the developed sensor module shows that the resistance is $0.151m{\Omega}$ between the R and R phases, $0.169m{\Omega}$ between the S and S phases, and $0.178m{\Omega}$ between the T and T phases, respectively. The insulation resistance measured at AC 500 V and 1,000 V is $0.08m{\Omega}$ between the R, S, T and N phases, respectively. Then, the insulation resistance measured at DC 500 V is $83.3G{\Omega}$ between the R, S, T and G terminal, respectively. In addition, the applied withstanding voltage is AC 220 V/380 V/440 V and it was found that characteristics between all phases are good. This study measured the standby power by installing the developed sensor module at the rear of the MCCB and switching the circuit breaker on sequentially. The standby power is 1.350 W when one circuit breaker is turned on, 1.690 W when 2 circuit breakers are turned on, and 4.371 W when 10 circuit breakers are turned on. This study also verified the reliability of the standby power of the distribution panel equipped with the developed sensor module using the Minitab Program (Minitab PGM). Since the analysis shows the statistical average of 1.34627 in the reliable range of normal distribution, standard deviation of 0.001874, AD of 0.554, and P value of 0.140, it is found that the distribution panel equipped with the developed sensor module has high reliability.

Antimicrobial and Immunological activities of Vinca minor Extracts (빈카 마이너 추출물의 항균 및 면역활성 연구)

  • Vinca alkaloid compounds including vincamine from Vinca minor L. have been extracted by ethanol and hot water extraction methods. Antimicrobial properties of those extracts have been investigated against dandruff causing microorganism Malassezia furfur, yeast, Gram positive and negative bacteria. Vincamine standard and ethanol extract showed no sign of antimicrobial activity, whereas hot-water extract had the activity against M. furfur. Furthermore hot water extract had antimicrobial activity against Gram positive Bacillus sp. and Gram negative Escherichia coli. Cytotoxic properties of those extracts have also been investigated with HaCaT cell (human keratinocyte), HT-29 cell (human colorectal adenocarcinoma cell) and Raw cell, showing no significant cytotoxic effects. We also measured the ROS using dichlorofluorescein diacetate (DCFDA), a popular fluorescence-based probe for reactive oxygen species detection. The result showed the increasement of reactive oxygen species formation (20%) in HaCaT and HT-29 cell lines.

Characteristics of Thermoluminescence and Electron Spin Resonance and Organoleptic Quality of Irradiated Raisin and Dried Banana During Storage (건포도와 건바나나의 감마선 조사와 저장기간에 따른 열발광 및 전자스핀공명 특성과 관능적 품질)

  • The characteristics of thermoluminescence (TL) and electron spin resonance (ESR) and organoleptic qualities of gamma-irradiated raisin and dried banana were investigated during storage at 4$^{\circ}C$ for 6 months. The minerals separated from non-irradiated raisins showed TL glow curve (TL$_1$) with very low intensity around 200~30$0^{\circ}C$, while the irradiated samples at 1 kGy or more showed glow curves with higher intensity around 18$0^{\circ}C$, with linear increase by irradiation dose ($R^2$=0.9684), which made it possible to identify irradiated samples during 6 months. Moreover, TL ratios (TL$_1$/TL$_2$) through the reirradiation step at 1 kGy enhanced confidence in the identification of irradiated raisins. The ESR signals of multicomponent lines resulted from crystalline sugar radicals were shown in irradiated banana, identifying irradiated samples. The ESR signal intensity was dependent on irradiation doses ($R^2$=0.8977) and the signals were stable enough to be detected by 6th month after storage. Considering tile marketability of irradiated dried fruits during 6 months at low temperature TL and ESR analyses were shown suitable for the identification of irradiated raisins and dried banana, respectively.

  • The IPCC 5th Assessment Report (Climate Change 2013: The Physical Science Basis) was accepted at the 36th Session of the IPCC on 26 September 2013 in Stockholm, Sweden. It consists of the full scientific and technical assessment undertaken by Working Group I. This comprehensive assessment of the physical aspects of climate change puts a focus on those elements that are relevant to understand past, document current, and project future of climate change. The assessment builds on the IPCC Fourth Assessment Report and the recent Special Report on Managing the Risk of Extreme Events and Disasters to Advance Climate Change Adaptation. The assessment covers the current knowledge of various processes within, and interactions among, climate system components, which determine the sensitivity and response of the system to changes in forcing, and they quantify the link between the changes in atmospheric constituents, and hence radiative forcing, and the consequent detection and attribution of climate change. Projections of changes in all climate system components are based on model simulations forced by a new set of scenarios. The report also provides a comprehensive assessment of past and future sea level change in a dedicated chapter. The primary purpose of this Technical Summary is to provide the link between the complete assessment of the multiple lines of independent evidence presented in the main report and the highly condensed summary prepared as Policy makers Summary. The Technical Summary thus serves as a starting point for those readers who seek the full information on more specific topics covered by this assessment. Warming of the climate system is unequivocal, and since the 1950s, many of the observed changes are unprecedented over decades to millennia. The atmosphere and ocean have warmed, the amounts of snow and ice have diminished, sea level has risen, and the concentrations of greenhouse gases have increased. Total radiative forcing is positive, and has led to an uptake of energy by the climate system. The largest contribution to total radiative forcing is caused by the increase in the atmospheric concentration of $CO_2$ since 1750. Human influence on the climate system is clear. This is evident from the increasing greenhouse gas concentrations in the atmosphere, positive radiative forcing, observed warming, and understanding of the climate system. Continued emissions of greenhouse gases will cause further warming and changes in all components of the climate system. Limiting climate change will require substantial and sustained reductions of greenhouse gas emissions. The in-depth review for past, present and future of climate change is carried out on the basis of the IPCC 5th Assessment Report.

A Gene-based dCAPS Marker for Selecting old-gold-crimson (ogc) Fruit Color Mutation in Tomato (토마토 과색 돌연변이 유전자(old-gold-crimson) 선발을 위한 dCAPS 분자표지 개발)

  • The old-gold-crimson ($og^c$) fruit color mutation produces deep red tomato fruit with high lycopene content. age is a null mutation allele of lycopene-${\beta}$-cyclase (Crt-b) gene (B locus) that converts lycopene to ${\beta}$-carotene in the cartenoid biosynthesis pathway in tomato. Breeding of high lycopene tomato cultivars can be accelerated by marker-assisted selection (MAS) for introgression of $og^c$ allele by using a gene-based DNA marker. In order to develop a marker, single nucleotide deletion of adenine(A) with. in a poly-A repeat that has been known to be responsible for frame-shift mutation of $og^c$ was confirmed by resequencing mutant allele and wild-type allele at B locus of several tomato lines. For allele discrimination and detection of $og^c$, derived CAPS (dCAPS) approach was used by designing a primer that artificially introduced restriction enzyme recognition site of Hin fI in PCR products from $og^c$ allele. This dCAPS marker is co-dominant gene-based PCR marker that can be efficiently used for MAS breeding program aiming the development of high lycopene tomato.

Development of a River Maintenance Management Technology Related with National River Management Data (국가하천관리자료와 연계한 하천유지관리 기술개발)

  • This study has developed a technology for river basin including the management of the data related with riverbed and the analysis of the riverbed maintenance based on the high-resolution imagery data and LiDAR (Light Detection and Raging) in order to enhance the utilization of river management by using RIMGIS(River Information Management GIS) and to acquire the advanced operation for river management. Using the detailed river topographical map specially designed in the form of LiDAR or high-resolution images, riverbed data and river bank channel information that are dynamically changed were informationized and established in the RIMGIS DB. At this stage, a monitoring techniques that is established in the river management system associated with RIMGIS and minimized the impact of riverbed maintenance (fluctuations) has been studied. In addition, functions and data structure of RIMGIS containing the information of geography and management of the river have been supplemented to make an improvement of the real-time management of the river. Furthermore, a technology that is capable of supplementing RIMGIS has been developed, making it feasible to maintain the river in use of structural method including an structural scheme of cross-section of the river by providing the information of riverbed and cross-section of the river. This is considered to solve an issue of insufficient data on accuracy and based on a lack of information of the river based on the two-dimensional lines, making it feasible to (steadily) improve the function of RIMGIS and to operate management tasks. Therefore, it is highly expected to enhance aforementioned technology of the river information management as a great foundation that maximizes the utilization of the river management to support RIMGIS for the development of national river management data.

Extraction of Renal Glomeruli Region using Genetic Algorithm (유전적 알고리듬을 이용한 신장 사구체 영역의 추출)

  • Extraction of glomeruli region plays a very important role for diagnosing nephritis automatically. However, it is not easy to extract glomeruli region correctly because the difference between glomeruli region and other region is not obvious, simultaneously unevennesses that is brought in the sampling process and in the imaging process. In this study, a new method for extracting renal glomeruli region using genetic algorithm is proposed. The first, low and high resolution images are obtained by using Laplacian-Gaussian filter with ${\sigma}=2.1$ and ${\sigma}=1.8$, then, binary images by setting the threshold value to zero are obtained. And then border edge is detected from low resolution images, the border of glomeruli is expressed by a closed B-splines' curve line. The parameters that decide the closed curve line with this low resolution image prevent the noises and the border lines from breaking off in the middle by searching using genetic algorithm. Next, in order to obtain more precise border edges of glomeruli, the number of node points is increased and corrected in order from eight to sixteen and thirty two from high resolution images. Finally, the validity of this proposed method is shown to be effective by applying to the real images.
