Target Recognition Triggered Split DNAzyme based Colorimetric Assay for Direct and Sensitive Methicillin-Resistance Analysis of Staphylococcus aureus

  • The accurate and rapid detection of methicillin-resistant Staphylococcus aureus (MRSA) holds significant clinical importance. This work presents a new method for detecting methicillin-resistant Staphylococcus aureus (S. aureus) in clinical samples. The method uses an aptamer-based colorimetric assay that combines a recognizing probe to identify the target and split DNAzyme to amplify the signal, resulting in a highly sensitive and direct analysis of methicillin-resistance. The identification of the PBP2a protein on the membrane of S. aureus in clinical samples leads to the allosterism of the recognizing probe, and thus provides a template for the proximity ligation of split DNAzyme. The proximity ligation of split DNAzyme forms an intact DNAzyme to identify the loop section in the L probe and generates a nicking site to release the loop sequence ("3" and "4" fragments). The "3" and "4" fragments forms an intact sequence to induce the catalytic hairpin assembly, exposing the G-rich section. The released the G-rich sequence of LR probe induces the formation of G-quadruplex-hemin DNAzyme as a colorimetric signal readout. The absorption intensity demonstrated a strong linear association with the logarithm of the S. aureus concentration across a wide range of 5 orders of magnitude dynamic range under the optimized experimental parameters. The limit of detection was calculated to be 23 CFU/ml and the method showed high selectivity for MRSA.

  • The determination of DNA hybridization can apply the molecular biology research, clinic diagnostics, bioengineering, environment monitoring, food science and other application area. So, The determination of hybridization is very important for the improvement of DNA detection system. In this study, we report the characterization of the DNA hybridization by the electricalchemical methods. A new electrochemical biosensor is described for voltammetric detection of gene sequence related to probe oligonucleotide of bacterium Escherichia coli O157:H7. The biosensor involves the immobilization of a 18-mer probe oligonucleotide, which is complemetary to a specific gene sequence related to Escherichia coli O157:H7 on a gold electrode through specific adsorption. The probe oligonucleotide was used to determine the amount of target oligonucleotide in solution using mitoxantrone(MTX) as the electrochemical indicators. The cathodic peak currents $(I_{peak})$ of MTX were linearly related to the concentration of the target oligonucleotide sequence in the range $1[{\mu}M]{\sim}0.1[nM]$. The detection limit of this approach was 0.01[nM]. In addition, these indicators were capable of selectivity discriminating against various mismatching condition.

Distance Measurement Using a Single Camera with a Rotating Mirror

  • A new distance measurement method with the use of a single camera and a rotating mirror is presented. A camera in front of a rotating mirror acquires a sequence of reflected images, from which distance information is extracted. The distance measurement is based on the idea that the corresponding pixel of an object point at a longer distance moves at a higher speed in a sequence of images in this type of system setting. Distance measurement based on such pixel movement is investigated. Like many other image-based techniques, this presented technique requires matching corresponding points in two images. To alleviate such difficulty, two kinds of techniques of image tracking through the sequence of images and the utilization of multiple sets of image frames are described. Precision improvement is possible and is one attractive merit. The presented approach with a rotating mirror is especially suitable for such multiple measurements. The imprecision caused by the physical limit could be improved through making several measurements and taking an average. In this paper, mathematics necessary for implementing the technique is derived and presented. Also, the error sensitivities of related parameters are analyzed. Experimental results using the real camera-mirror setup are reported.

  • Stepwise efficiency improvement in data envelopment analysis (DEA)-based benchmarking is a realistic and effective method by which inefficient decision making units (DMUs) can choose benchmarks in a stepwise manner and, thereby, effect gradual performance improvement. Most of the previous research relevant to stepwise efficiency improvement has focused primarily on how to stratify DMUs into multiple layers and how to select immediate benchmark targets in leading levels for lagging-level DMUs. It can be said that the sequence of benchmark targets was constructed in a myopic way, which can limit its effectiveness. To address this issue, this paper proposes an optimization approach to the construction of a sequence of benchmarks in DEA-based benchmarking, wherein two optimization criteria are employed : similarity of input-output use patterns, and proximity of input-output use levels between DMUs. To illustrate the proposed method, we applied it to the benchmarking of 23 national universities in South Korea.

THE LUMINOSITY FUNCTION AND INITIAL MASS FUNCTION FOR THE PLEIADES CLUSTER

  • In the best observed Pleiades cluster, the luminosity function(LF) and mass function(MF) for main sequence(MS) stars extended to $Mv{\approx}15.5(V{\approx}21)$ are very similar to the initial luminosity function(ILF) and initial mass function(IMF) for field stars in the solar neighborhood showing a bump at log $m{\simeq}-0.05$ and a dip at log $m{\simeq}-0.12$. This dip is equivalent to the Wielen dip appearing in the LF for the field stars. The occurence of these bump and dip is independent of adopted mass-luminosity relation(MLR) . and their characteristics could be explained by a time-dependent bimodal IMF. The model with this IMF gives a total cluster mass of $\~700M_\bigodot,\;\~25$ brown dwarfs and $\~3$ white dwarfs if the upper mass limit of progenitor of white dwarf is greater than $4.5M_\bigodot$. The cluster age on the basis of LF for brightest stars is given by $\~8\times10^7yr$ and all stars in the cluster lie along the single age sequence in the C-M diagram without showing a large dispersion from the sequence.

  • We prove the uniqueness of solutions for the boundary value problem of certain nonlinear elliptic operators in the setting: Given any continuous function f on the p-harmonic boundary of a complete Riemannian manifold, there exists a unique solution of certain nonlinear elliptic operators, which is a limit of a sequence of solutions of the operators with finite energy in the sense of supremum norm, on the manifold taking the same boundary value at each p-harmonic boundary as that of f.

  • In this paper, motivated by [1] and [7] we give a sequential definition of conditional Wiener integral and then use this definition to evaluate conditional Wiener integral of several functions on C [0, T]. The sequential definition is defined as the limit of a sequence of finite dimensional Lebesgue integrals. Thus the evaluation of conditional Wiener integrals involves no integrals in function space [cf, 5].
