Effect of Paddy Drying by Solar Energy Concentration Blast-Grain Circulation Dryer (태양열집열송풍(太陽熱集熱送風), 곡물순환식(穀物循環式) 건조기(乾燥機)의 벼 건조효과(乾燥效果))

  • A 2.5 ton scale of solar energy concentration blast-grain circulation dryer (SECD) was developed in order to shorten the drying time without damaged paddy. Comparative experiments were carried out on performance, drying efficiency, consistency in moisture content, milling recovery, grade of milled rice, and energy requirement and cost against all that of in-bin drying and storage (IBDS) method. The experiments were performed using mixture of several rice varieties of Tongil type(Japonica-Indica breeding type) under the autumn weather in Korea. The circulating air temperature inside SECD was $4{\sim}5^{\circ}C$ higher than that of IBDS. The moisture content of the paddy during the drying period in SECD was uniform while substantially varied in upper, middle or bottom layer in IBDS. By SECD, 24% initial moisture content of paddy was reduced to 15% after only 3 days of drying as compared to 14 days at IBDS. The percentage of cracked kernels in upper, middle and bottom layers in IBDS was 6, 6 and 12%, respectively, whereas 7% in all layers in SECD. Both types of dryers did not significantly affect the milling recovery of dried paddy and grade of milled rice. Energy requirement of SECD(28.8Kw/2.5ton) for paddy drying was much less than that of IBDS(108Kw/2.5ton).

Characteristics of a planar Bi-Sb multijunction thermal converter with Pt-heater (백금 히터가 내장된 평면형 Bi-Sb 다중접합 열전변환기의 특성)

  • A planar Bi-Sb multijunction thermal converter with high thermal sensitivity and small ac-dc transfer error has been fabricated by preparing the bifilar thin film Pt-heater and the hot junctions of thin film Bi-Sb thermopile on the $Si_{3}N_{4}/SiO_{2}/Si_{3}N_{4}$-diaphragm, which functions as a thermal isolation layer, and the cold junctions on the dielectric membrane supported with the Si-substrate, which acts as a heat sink, and its ac-dc transfer characteristics were investigated with the fast reversed dc method. The respective thermal sensitivities of the converter with single bifilar heater were about 10.1 mV/mW and 14.8 mV/mW in the air and vacuum, and those of the converter with dual bifilar heater were about 5.1 mV/mW and 7.6 mV/mW, and about 5.3 mV/mW and 7.8 mV/mW in the air and vacuum for the inputs of inside and outside heaters, indicating that the thermal sensitivities in the vacuum, where there is rarely thermal loss caused by gas, are higher than those in the air. The ac-dc voltage and current transfer difference ranges of the converter with single bifilar heater were about ${\pm}1.80\;ppm$ and ${\pm}0.58\;ppm$, and those of the converter with dual bifilar heater were about ${\pm}0.63\;ppm$ and ${\pm}0.25\;ppm$, and about ${\pm}0.53\;ppm$ and ${\pm}0.27\;ppm$, respectively, for the inputs of inside and outside heaters, in the frequency range below 10 kHz and in the air.

A Reliable Protocol for transfection of mature primary hippocampal neurons using a neuron-glia co-culture system (신경세포-신경교세포 공동배양을 이용한 성숙한 해마신경세포의 효율적인 형질전환 방법)

  • DNA transfection is a powerful tool for studying gene functions. The $Ca^{2+}$-phosphate precipitation remains one of the most popular and cost-effective transfection techniques. Mature neurons are more resistant to transfection than young ones and most other cell types, and easy to die if microenvironment changes. Here, we report a transfection protocol for mature neurons. The critical modifications are inclusion of glial cells in culture and careful control of $Ca^{2+}$-phosphate precipitation under microscope. Cerebral glial cells were grown until ${\sim}70-80%$ confluence in DMEM/10% horse serum, which was thereafter replaced with serum-free Neurobasal/Ara-C, and 319 hippocampal neurons were plated onto the glial layer Formation of fine $DNA/Ca^{2+}$-phosphate precipitates was induced using Clontech $CalPhos^{TM}$ Mammalian Transfection Kit, and the size ($0.5-1\;{\mu}m$ in diameter) and density(about 10 particles/$100\;{\mu}m^2$) were carefully controlled by the time of incubation in the medium. This modified protocol can be reliably applied for transfection of mature neurons that are maintained longer than two weeks in vitro, resulting in 10-15 healthy transfected neurons per a well of 24-well plates. The efficacy of the protocol was verified by punctate expression of $pEGFP-CaMKII{\alpha}$, a synaptic protein, and diffuse expression of pDsRed2. Our protocol provides a reliable method for transfection of mature neurons in vitro.

Management Guideline and Avifauca of Odaesan area in Odaesan National Park (오대산 국립공원 오대산 지역의 야생조류상 및 관리 방안)

  • This study was conducted to investigate the avifauna and to suggest the management discipline for the protection of bird community in Odaesan National Park. Field survey was carried over 2 main trails by line transect method from mid June to early December in 1996. The 1st section included the area from Woljongas to Sangwonsa 7km distance. The 2nd section survey started from Sangwonsa via Bukdaesa and the summit of Odaesan to Sangwonsa 9.8 km distance. The study results were as follows ; The observed birds belonged to 9 orders 22families 52species, they also had Black Woodpecker(Dryocopus martius)designated as natural monument No. 242, Chinese Sparrow Hawk(Accipiter soloensis) and Kestrel(Falco tinnunculus) as natural munumet No. 323, Scops Owl(Out scops)and Korean Wood Owl(Strix aluco) as No. 324. These birds also were classified into 25 species for residents, 16 species for summer visitors, 8 species for passage migrants, 3 species for winter visitors, respectionely. The 2nd section showed a high species richness and individuals in every season, however, had a difference in species composition with 1st section. Nesting guild of breeding bird community used highly in order of bush, hole, and canopy as a nest resources. It is suggested that high bush-nesting guild had a deep relationship with bush layer located in the ecotone of 1st section and that located in the high elevated zone in 2nd section. Hole-nesting guild such as Black Woodpecker(Dryocopus martius), Gray-headed Woodpecker(Picus canus) and Great Spotted Woodpecker(Dendrocopos major) were surveyed only in 2nd section, so it could be attribute to the small fragmentation and the growing of high diameter at breast height(D.B.H) tree in 2nd section. It is urgent that the management of camping ground and people for the conservation of brook in 1nd section, trail protection for the prevention from trail enlargement in 2nd section for the bird protection. Artificial food in snowy winter will provide the good breeding condition with the residents and migrants. And, the endeavor to lessen the habitat fragmentation will be beneficial to the birds who have a large home range such as Black Woodpecker(Dryocopus martius) and Korean Wood Owl(Strix aluco). For the control of Domestic Dove(Columba livia) populations, it could be recommended that the elimination of their nesting resources by net.

Establishment of the Appropriate Risk Standard through the Risk Assessment of Accident Scenario (사고시나리오별 위험도 산정을 통한 적정 위험도 기준 설정)

  • An off-site consequence analysis is used to calculate the risks when hazardous chemicals that is being used on-site has been exposed off-site; the biggest factor that impacts the risk is the risks of accident scenarios. This study seeks to calculate risks according to accident scenarios by applying OGP/LOPA risk calculating methods for similar facilities, calculate risk reduction ratio by inspecting applicable IPL for incidents, and propose an appropriate risk standard for different risk calculating methods. Considering all applicable IPL when estimating the safety improvement of accident scenarios, the risk of OGP is 8.05E-04 and the risk of LOPA is 1.00E-04, According to the case of IPL, the risk is 1.34E-02. The optimal risk level for accident scenarios using LOPA was $10^{-2}$, but the appropriate risk criteria for accident scenarios in foreign similar studies were $10^{-3}{\sim}10^{-4}$, the risk of a scenario can be determined at an unacceptable level. When OGP is applied, it is analyzed as acceptable level, but in case of applying LOPA, all applicable IPL should be applied in order to satisfy the acceptable risk level. Compared to OGP, the risk is high when LOPA is applied. Therefore, the acceptable risk level should be set differently for each risk method.

  • We have fabricated mixed-ionic conducting membranes, L $a_{0.6}$S $r_{0.4}$ $Co_{0.2}$F $e_{0.8}$ $O_{3-}$$\delta$/ and L $a_{0.7}$S $r_{0.3}$G $a_{0.6}$F $e_{0.4}$ $O_{3-}$$\delta$/ by the solid state method. Ceramic membranes consisted of perovskite-type structures and exhibited high relative density, >95%. Especially, dense L $a_{0.6}$S $r_{0.4}$Co $O_{3-}$$\delta$/ layer was coated on the L $a_{0.7}$S $r_{0.3}$G $a_{0.6}$F $e_{0.4}$ $O_{3-}$$\delta$/ membranes by using screen printing technique in order to improve oxygen ion flux. We measured oxygen ion flux on uncoated L $a_{0.6}$S $r_{0.4}$ $Co_{0.2}$F $e_{0.8}$ $O_{3-}$$\delta$/, uncoated L $a_{0.7}$S $r_{0.3}$G $a_{0.6}$F $e_{0.4}$ $O_{3-}$$\delta$/, and coated L $a_{0.7}$S $r_{0.3}$G $a_{0.6}$F $e_{0.4}$ $O_{3-}$$\delta$/ membranes. The L $a_{0.6}$S $r_{0.4}$ $Co_{0.2}$F $e_{0.8}$ $O_{3-}$$\delta$/ membranes showed the highest flux, 0.26 mL/min.$\textrm{cm}^2$ at 90$0^{\circ}C$, after steady state had been reached. The oxygen flux of coated L $a_{0.7}$S $r_{0.3}$G $a_{0.6}$F $e_{0.4}$ $O_{3-}$$\delta$/ membranes showed higher value, 0.19 mL/min.$\textrm{cm}^2$ at 95$0^{\circ}C$. This flux was as much as 2 or 3 times higher than those of uncoated L $a_{0.7}$S $r_{0.3}$G $a_{0.6}$F $e_{0.4}$ $O_{3-}$$\delta$/ membranes. 3-$\delta$/ membranes.X> 3-$\delta$/ membranes.membranes.

  • We fabricated a Si nano floating gate memory with Schottky barrier tunneling transistor structure. The device was consisted of Schottky barriers of Er-silicide at source/drain and Si nanoclusters in the gate stack formed by LPCVD-digital gas feeding method. Transistor operations due to the Schottky barrier tunneling were observed under small gate bias < 2V. The nonvolatile memory properties were investigated by measuring the threshold voltage shift along the gate bias voltage and time. We obtained the 10/50 mseconds for write/erase times and the memory window of $\sim5V$ under ${\pm}20\;V$ write/erase voltages. However, the memory window decreased to 0.4V after 104seconds, which was attributed to the Er-related defects in the tunneling oxide layer. Good write/erase endurance was maintained until $10^3$ write/erase times. However, the threshold voltages moved upward, and the memory window became small after more write/erase operations. Defects in the LPCVD control oxide were discussed for the endurance results. The experimental results point to the possibility of a Si nano floating gate memory with Schottky barrier tunneling transistor structure for Si nanoscale nonvolatile memory device.

Development of Dermal Transduction Epidermal Growth Factor (EGF) Using A Skin Penetrating Functional Peptide (피부투과 기능성 펩타이드를 이용한 경피투과성 상피세포성장인자의 개발)

  • The epidermal growth factor (EGF) has a intrinsic function of inducing growth and proliferation of cells through interacting with cell membrane receptors in human epidermis and dermis layer. These functions of EGF are used as a main ingredient for wound healing medicines and anti-aging cosmetics. As a cosmetic ingredient, the EGF has a problem in exhibiting its natural efficacy due to the lack of the ability to penetrate through the stratum corneum, which is known as the skin barrier. In this study, a recombinant human epidermal growth factor ($MTD_{151}-EGF$) fused with the macromolecule transduction domain $(MTD)_{151}$ with the skin penetration ability was developed to improve the skin penetration efficiency of the EGF. Expression of $MTD_{151}-EGF$ was performed in E. coli transformed with a vector encoding the $MTD_{151}-EGF$ gene and then purified. The purified $MTD_{151}-EGF$ was evaluated using cell proliferation assay, cytotoxicity test and skin penetration test by franz diffusion cell assay and artificial skin. Cell proliferation activity of $MTD_{151}-EGF$ purified to high purity of 99% or above was equivalent to the EGF or better, and cytotoxicity was not observed. In addition, the $MTD_{151}-EGF$ showed an excellent penetration efficiency compared to the EGF in the skin penetration test with EGF and $MTD_{151}-EGF$ labeled by FITC in an artificial skin penetration model. Based on the quantitative analysis of the penetrating substance using franz diffusion cell assay, the amount of penetration was about 16 times more than that of EGF. These results can be regarded as an effective alternative to improve the existing physical transdermal penetration method related to the use of various active ingredients for cosmetics.

  • Sangjogam, located in Goseong, Gyeongsangnam-do, was designated as Natural Monument #411, because of its diverse geological heritage, such as fossils, ripple marks, dykes, and columnar joints. In the area, Byeongpungbawi, with its beautiful columnar joints vertical to the bedding plane of the underlying sedimentary rocks and spectacular coastal view, was named after its overall shape reminiscent of a huge folding screen. The purpose of this study was to investigate the formation process of the columnar joints using the anisotropy of magnetic susceptibility (AMS) method. AMS measurements showed that the k1 and k3 values representative of directions of the long and short axes of a magnetic particle at each point strongly clustered, and the oblate magnetic foliation structure in Byeongpungbawi developed during sill-type intrusion rather than lava flow. In summary, Byeongpungbawi was produced by sill-type intrusion along the bedding plane of the underlying sedimentary layer, and the subsequent formation of columnar joints was accompanied by the cooling and contraction of intruding rhyolite magma. This study potentially provides a basic research tool in understanding the formation mechanism of columnar joints which are widely distributed in southern Korea.

  • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70$\pm$1.32mmHg/min)compared to CF dialyzers(4.32$\pm$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The strip loader and cassette were made to obtain more accurate reflectance parameters. The purpose of this study is to identify major geometrical differences between Korean femurs 3nd others that we believe belong to Caucasians so that we would be able to get insights into the femoral component design that fits Asians including Koreans. We utilized computerized tomography (CT) images of femurs extracted from cadavers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm. In this paper, a new method to detect nonlinear propagation characteristics of the internal vibration in the tissue for the low frequency mechanical vibration by using bispectral analysis is proposed. In the method, low frequency vibration of f0( = 100Hz) is applied on the surface of the object, and the waveform of the internal vibration x (t) is measured from Doppler frequency modulation of silmultaneously transmitted probing ultrasonic waves. Then, the bispectra of the signal x (t) at the frequencies (f0, f0) and (f0, 2f0) are calculated to estimate the nonlinear propagation characteristics as their magnitude ratio, w here since bispectrum is free from the gaussian additive noise we can get the value with high S/N.
