통일계 벼의 Brassinosteroid활성물질 검색 (Investigation of Brassinosteroid substances in the shoots of hybrid type korean rices)

  • The production of bassinosteroid-like substances of two hybrid types of Korean rice, Jangseongbyeo, Taebackbyeo were investigated. The shoots at the maximum tillering stage were extracted and purified by solvent fractionation, silica gel adsorption chromatography Sephadex LH-20 chromatography, charcoal adsorption chromatography, Bondesil chromatography and HPLC of reverse phase, successively. Biological activities of each purification step were monitored by the rice lamina inclination test. Higher activities against the rice lamina inclination test in the each purification step showed that the shoots of two cultivars biosynthesize brassinosteroids. Two cultivars also showed a similar distribution of biological activities of endogenous brassinosteroids detected by HPLC.

Pseudorabies virus를 이용한 신장, 신수 및 경문에서 투사되는 중추신경계내 표지영역에 관한 연구 (Central Localization of the Neurons Projecting to the Kidney, UB23 and GB25 Using the Pseudorabies Virus)

  • Objective : To investigate the relation to the organs, shu points and mu points. The labeled common locations of the spinal cord and brain were observed following injection of pseudorabies virus(PRV) into the the kidney, UB23 and GB25. Methods : After survival times of 96 hours following injection of PRV, The fifteen rats were perfused, and their spinal cord and brain were frozen sectioned($30{\mu}m$). These sections were stained by PRV immunohistochemical staining method, and observed with light microscope. Results : In spinal cord, PRV labeled neurons projecting to the kidney, BL23 and GB25 were founded in cervical, thoracic, lumbar and sacral spinal segments. Dense labeled areas of cervical segments were overlap in lateral cervical n. and lamina III-V area. Thoracic segments were overlap in lateral spinal n., intermediolateral n. and lamina V-X areas. Lumbar segments were overlap in lamina I-V areas. Sacral segments were overlap in lamina IV, V and X areas. In brain, PRV labeled areas projecting to the kidney, UB23 and GB25 were overlap in the A1 noradrenalin cells/C1 adrenalin cells/caudoventrolateral reticular n./rostroventrolaterai n., raphe obscurus n,, raphe pallidus n., raphe magnus n., gigantocellular reticular n., locus coeruleus, subcoeruleus n., A5 cell group and paraventricular hypothalamic n.. Conclusions : This results suggest that PRV labeled overlap areas of projecting to the kidney may be correlated to shu and mu points related to the kidney. These morphological results provide that organs-shu(transport) and mu(alarm) points interrelationship may be related to the central autonomic pathways.

  • Cyclosporin A(CsA) is now widely used to treat organ transplant recipients. But CsA has various short-and long-term side effects. Especially, gingival hyperplasia is not easy to resolve since its nature is still unknown. This study discusses the pathogenesis of CsA-induced gingival hyperplasia on the basis of data obtained from light and electron microscopic studies of biopsis from patients on CsA treatment after kidney transplantation. Light microscopically, the multilayered squamous epithelium showed an irregular surface of parakeratosis and deep invaginations in the subepithelial tissue. At lamina propria, we observed bundles of irregularly arranged collagen fiber, some fibroblasts, numerous capillary vessels and a large diffuse infiltration of plasma cells. Ultrastructurally, many fibroblasts, collagen fibers, collagen fibrils were present in lamina propria. On the basis of the data collected, we propose that the morphological features of the dimensional increase in gingival tissue associated with CsA treatment in kidney transplant patients may be considered proliferative fibroblasts, collagen fibers, collagen fibrils in lamina propria.

자궁과 삼음교에서 투사하는 충추신경계내 신경원의 표지영역에 관한 연구 (Central Localization of Neurons Projecting to the Uterus and Sanyinjiao(Sp6))

  • This experimental studies was to investigate location of labeled neurons in CNS following injection of pseudorabies virus(PRV), Bartha strain, into the uterus and Sanyinjiao(Sp6) of rats. After survival times of 4-5 days following the injection of PRV, the rats were perfused, and their brain and spinal cord were frozen sectioned($30\mu\textrm{m}$). These sections were stained by PRV immunohistochemical staining methods, and observed with light microscope. The results were as follows: 1. In the spinal cord, overlap areas of PRV labeled neurons projecting to uterus and Sp6 were observed in lamina VII, IX and X areas of cervical segments. In thoracic segments, overlap areas were observed in lamina IV, VII, X and intermediolateral n.. In lumbar segments, overlap area of PRV labeled neurons were observed in lamina I, V-VII, IX, X and intermediolateral n.. In sacral segments, overlap areas of PRY labeled neurons were observed in lamina N, V, VII, X and sacral parasympathetic n.. 2. In the brain, overlap areas of PR V labeled neurons projecting to the uterus and Sp6 were observed in lateral paragigantocellular n., rostroventrolateral reticular n., raphe obscurus n., raphe pallidus n., raphe magnus n., locus coeruleus n., Barrington's n., A5 cell group, central gray n., paraventricular hypothalamic n. and arcuate n. This results suggest that overlap areas of PRV labeled neurons of the spinal cord projecting to the uterus and Sp6 might be the first-order neurons related to the viscera-somatic sensory and sympathetic preganglionic neurons. PRV labeled neurons of the brain may be the second and third-order neurons response to the movement of smooth muscle of uterus. These PRV labeled neurons may be central autonomic center related to the integration and modulation of reflex control linked to the sensory and motor system monitoring the internal environment. These overlap areas of spinal cord and brainmay be related to autonomic centers related to regulation of uterus.

특별직교이방성 복합적층판 이론을 응용한 포스트텐션된 슬래브 교량의 해석 (Analysis of Post-tensioned Slab Bridge by Means of Specially Orthotropic Laminates Theory)

  • 포스트텐션된 슬래브 교량은 특별직교이방성 복합적층판 이론으로 해석할 수 있다. 슬래브 교량의 해석에 있어 단면의 기하학적, 물리적 특성이 중립축을 중심으로 휨-연계 강성 $B_{ij}=0$ 이고, $D_{16}=D_{26}=0$ 임을 고려해야 한다. 각각의 횡방향철근과 수직방향철근은 하나의 lamina로 간주하고, 재료상수는 각각의 lamina의 혼합법칙에 의해서 계산되어진다. 단순지지된 슬래브 교량은 등분포하중과 축하중을 받고 있다. 본 논문에서는 유한차분법과 보 이론에 의해서 해석하였다. 그 결과 보 이론에 의한 해석 값이 판 이론에 의한 해석 값에 근접함을 알 수 있었다. 본 논문에서 얻은 연구 결과를 이용하여 가까운 장래에 학부정도의 실력을 가진 기술자가 포스트텐션된 슬래브 교량의 해석에 있어서 유용하게 사용 할 수 있다.

해저터널용 복합신소재 배수복합관 부재의 정적거동에 관한 실험적 연구 (An experimental study on the static behavior of advanced composite materials drainage pipe member for an undersea tunnel)

  • 해저터널용 복합신소재 배수관구조를 설계하기 위해서는 복합신소재 구조부재의 적층형태별 역학적 성질을 결정하는 것이 필수적이다. 복합신소재는 일반적으로 등방성 재료와 달리 치수효과가 매우 큰 것으로 알려져 있다. 본 연구에서는 복합신소재 부재의 적층형태별 인장시험을 상온($20^{\circ}C$)과 해수온도($0^{\circ}C$)에서 각각 수행하였다. 또한, 이론적 해석방법인 혼합물의 법칙과 탄성해법을 적용하여 재료의 역학적 성질을 추정하고 시험결과와 비교를 하였다. 해저터널 복합신소재 배수관 구조부재를 설계할 때 사용되는 역학적 성질의 값은 상온에서 얻어진 값을 보정하여 적용하여야 된다. 이러한 자료는 향후 해저터널용 복합신소재 배수관구조의 설계의 기초자료로 제공하고자 하였다.

Evaluation of Bonding Strength of Larch Cross-Laminated Timber

  • The delamination along the annual ring on the cross-section of laminae and the bonding strength according to the tangential angle between laminae were evaluated for the production of 3-ply cross-laminated timber (CLT) using domestic larch. Since there is no standard for CLT in Korea, the production and test of specimens for bonding strength followed the standard procedure of "Structural glued laminated timber" (KS F 3021). The standard specifies to exclude any measurement from the cracks of timbers resulted from drying or knots during delamination test of the glued laminated timbers. However, the failure of cross-sectional tissues along the annual rings was observed near the glue-line of all specimens during the delamination test. Because this phenomenon can generate defects in the CLT that may be exposed to various temperatures and relative humidities after the actual construction, the delamination percentage was measured by including this wood failure. As a result, the delamination percentage of the CLT which had been combined in such a way that the annual rings of outer lamina were directed inward was the lowest, which was around 13%, regardless of the annual ring direction of the middle lamina. On the other hand, the delamination percentage of the CLT which had been combined in such a way that the annual rings of outer lamina were directed outward was the highest, which was around 26%. Furthermore, end-split occurred in the outer lamina during the drying process of the boiling delamination test, which affected the delamination percentage. Therefore, the soaking delamination test was found to be more appropriate for evaluating the delamination strength of CLT. The block shear strength of larch CLT was $3.9{\pm}0.9$ MPa on average, which was 46% lower than the block shear strength requirement (7.1 MPa) of the standard, but satisfied the criteria of the block shear strength (3.5 MPa) of the European Standard (prEN 16351: 2013).

A Computed Tomography-Based Anatomic Comparison of Three Different Types of C7 Posterior Fixation Techniques : Pedicle, Intralaminar, and Lateral Mass Screws

  • Objective : The intralaminar screw (ILS) fixation technique offers an alternative to pedicle screw (PS) and lateral mass screw (LMS) fixation in the C7 spine. Although cadaveric studies have described the anatomy of the pedicles, laminae, and lateral masses at C7, 3-dimensional computed tomography (CT) imaging is the modality of choice for pre-surgical planning. In this study, the goal was to determine the anatomical parameter and optimal screw trajectory for ILS placement at C7, and to compare this information to PS and LMS placement in the C7 spine as determined by CT evaluation. Methods : A total of 120 patients (60 men and 60 women) with an average age of $51.7{\pm}13.6$ years were selected by retrospective review of a trauma registry database over a 2-year period. Patients were included in the study if they were older than 15 years of age, had standardized axial bone-window CT imaging at C7, and had no evidence of spinal trauma. For each lamina and pedicle, width (outer cortical and inner cancellous), maximal screw length, and optimal screw trajectory were measured, and the maximal screw length of the lateral mass were measured using m-view 5.4 software. Statistical analysis was performed using Student's t-test. Results : At C7, the maximal PS length was significantly greater than the ILS and LMS length (PS, $33.9{\pm}3.1$ mm; ILS, $30.8{\pm}3.1$ mm; LMS, $10.6{\pm}1.3$; p<0.01). When the outer cortical and inner cancellous width was compared between the pedicle and lamina, the mean pedicle outer cortical width at C7 was wider than the lamina by an average of 0.6 mm (pedicle, $6.8{\pm}1.2$ mm; lamina, $6.2{\pm}1.2$ mm; p<0.01). At C7, 95.8% of the laminae measured accepted a 4.0-mm screw with a 1.0 mm of clearance, compared with 99.2% of pedicle. Of the laminae measured, 99.2% accepted a 3.5-mm screw with a 1.0 mm clearance, compared with 100% of the pedicle. When the outer cortical and inner cancellous height was compared between pedicle and lamina, the mean lamina outer cortical height at C7 was wider than the pedicle by an average of 9.9 mm (lamina, $18.6{\pm}2.0$ mm; pedicle, $8.7{\pm}1.3$ mm; p<0.01). The ideal screw trajectory at C7 was also measured ($47.8{\pm}4.8^{\circ}$ for ILS and $35.1{\pm}8.1^{\circ}$ for PS). Conclusion : Although pedicle screw fixation is the most ideal instrumentation method for C7 fixation with respect to length and cortical diameter, anatomical aspect of C7 lamina is affordable to place screw. Therefore, the C7 intralaminar screw could be an alternative fixation technique with few anatomic limitations in the cases when C7 pedicle screw fixation is not favorable. However, anatomical variations in the length and width must be considered when placing an intralaminar or pedicle screw at C7.

흰쥐 삼차신경감각핵에서 $P2X_3$와 TRPV1의 공존에 관한 연구 (Coexpression of $P2X_3$ with TRPV1 in the Rat Trigeminal Sensory Nuclei)

  • 삼차신경계에서 $P2X_3$와 TRPV1 면역양성 일차들신경섬유는 통각정보의 전달에 중요한 역할을 한다. 본 연구에서는 삼차신경절 및 삼차신경꼬리핵에서 $P2X_3$와 TRPV1 면역양성 신경세포의 형태학적 특성 및 투사양식을 이해하기 위하여, 흰쥐 삼차신경절 및 삼차신경꼬리핵에서 $P2X_3$와 TRPV1에 대한 항체를 사용하여 형광면역염색법 및 형태계측학적인 기법을 시행하여 다음과 같은 결과를 얻었다. $P2X_3$ 면역양성 신경세포중 77.4%의 신경세포에서 (1,401/1,810) TRPV1이 동시에 발현되었으며, TRPV1 면역양성 신경세포중 51.9% (1,401/2,698)의 신경세포에서 $P2X_3$와 공존을 보였다. $P2X_3$와 TRPV1에 동시에 면역양성반응을 보이는 신경세포는 중간크기의 신경세포에서 주로 관찰되었으며, $P2X_3$ 혹은 TRPV1 면역양성 신경세포중 아주 작거나 큰 신경세포에서는 공존하지 않았다. 삼차신경꼬리핵에서 $P2X_3$ 면역양성 신경섬유 및 신경종말들은 제1층과 제2층에 분포하는데 주로 제2층의 안쪽부위에서 밀도가 높게 관찰되었으며, TRPV1 면역양성 신경섬유 및 신경종말들은 제1층과 제2층의 바깥쪽에서 밀도가 높게 관찰되었다. $P2X_3$와 TRPV1이 공존하는 신경섬유 및 신경종말들은 제2층의 안쪽과 바깥쪽의 경계부위에서 관찰되었다. 이러한 연구결과는 $P2X_3$와 TRPV1을 동시에 발현하는 신경세포는 구강안면영역에서 통각정보의 처리에 독특한 역할을 수행할 것이라는 것을 시사한다.

Alport 증후군에서 사구체 기저막의 형태학적 변화와 사구체 상피세포의 구조 및 임상지표와의 관계 (Relationship of Glomerular Basement Membrane Alterations to Epithelial Cell Structure and Clinical Parameters in Alport Syndrome)

  • 목 적 : Alport증후군 환자의 사구체 기저막의 형태학적 변화 및 사구체 상피세포의 구조 변화에 따른 임상지표간의 상관 관계를 알아보고자 하였다. 방 법 : Alport 증후군으로 확진된 15명의 환자를 대상으로 하였다. 사구체 기저막을 형태학적 변화에 따라 1) 정상(C1), 2) minor alteration: widening of lamina rara interna or externa without lamina densa changes (C2), 3)nonspecific splitting of lamina densa (C3), 4) basket-weave pattern of lamina densa splitting (C4)의 4가지 범주로 나누고 각 범주에 해당하는 사구체 기저막의 비율을(%) 측정하였다. 또한 각 범주의 사구체 기저막에서 사구체 기저막 $10\;{\mu}m$에 대한 상피세포의 세과여극(slit pore)의 밀도를 구하였고, 나이에 따른 사구체 기저막의 형태학적 변화를 살펴보았다. 결 과 : 요 중 단백질 배설량과 정상 기저막 소견(C1) 사이는 역비례 관계를 보였고(P<0.05), 요 중 단백질 배설량과 비정상 사구체 기저막 소견(C2+C3+C4) 사이는 정비례 관계를 보였다(P<0.05). 비정상적 기저막 소견을 보이는 각 범주와 단백뇨나 크레아티닌 청소율과는 의미 있는 상관관계가 없었다. 크레아티닌 청소율과 정상 사구체 기저막(C1)의 비율과의 관계는 통계적인 의미가 없었고(P=0.085), 비정상 사구체 기저막(C2+C3+C4)의 비율과도 통계적으로 의미가 없었다(P=0.080). 신생검 당시의 나이와 사구체 기저막의 형태학적 변화도 상관관계가 없었다. 결 론 : Alport 증후군 환자에서 사구체 기저막의 변화는 단백뇨와 관련성이 있지만 사구체 상피세포 구조와 사구체여과율과는 관련성이 없었다.
