In vitro propagation of a rare and endangered species, Echinosophora koreensis Nakai, by axillary bud culture (희귀 및 멸종위기 수종 개느삼의 액아배양을 통한 기내번식)

  • An efficient micropropagation was established by using axillary bud explants from two-year-old tree(Echinosphorea koreensis Nakai), which has been known as a rare and endangered species. Among various basal media tested, DKW medium was shown to be the best for axillary shoot elongation. The addition of both BA and TDZ to the medium induced 6 to 10 shoots per explant during eight weeks of culture, without showing any abnormal morphology at the shoot proliferation stage. However, high concentration of TDZ(>0.05 mg/L) appeared to cause hyperhydration on either leaf or shoot at the later developmental stage. Approximately 20% of shoots produced roots by the addition of 1.0 mg/L NAA but not by IBA($0.2{\sim}1.0$ mg/L). Ex vitro micro-cuttings were better source for root induction; up to 58.6% of the micro-cuttings rooted when 100 mg/L IBA was applied to the soil(vermiculite). More than 90% of plantlets with roots were successfully acclimatized and grew normally in the field. Therefore, we suggest that this endangered tree species can be effectively micropropagated by axillary bud culture system developed in this study.

Effect of Surfactant Concentrations on Anti-waterdrop, Environment and Lettuce(Lactuca sativa L.) Growth in Polyethylene Film Greenhouse (계면활성제 처리 농도가 무가온 플라스틱온실의 방적성, 환경 및 상추 생육에 미치는 영향)

  • In polyethylene film house treated with surfactants(SF316+FB0800), waterdrops attached on film surface were absorbed to filter paper of 9 cm in diameter and measured 1.21 mL in control, 0.15 mL in 1% and 0.07 mL in 2% on December 10 at 10:00, 2002. However, there was no clear difference between surfactant concentrations 1% and 2%, And the most waterdrop was measured at 10:00 in daytime. As the air temperature was higher in greenhouse, waterdrop was sweled. So, the least waterdrop was measured at 14:00. In greenhouse covered with surfactants concentrated 1% and 2%, transmittances of solar radiation were 9.3% and 12.9% higher than control, respectively. In air temperature and relative humidity, there were no significant difference in all greenhouse. in forenoon, the air temperature and relative surfactant treated film greenhouse tended 1~2$^{\circ}C$ warmer than that of control. However, sil temperatures of surfactant treated film greenhouse tended 3~4$^{\circ}C$ warmer than that of control. During winter, the lettuce growth in surfactant treated film greenhouse was faster than that of control. However, there was no difference between surfactant concentrations of 1% and 2%.

  • It is generally known that handoff delay degrades the QoS by packet drop, packet delay and jitter. Moreover. handoff highly effects on QoS in beyond 3G system because not only micro cells for achieving high capacity and fast moving nodes induce the frequent handoff but also hard handoff must be carried out in OFDM system. Therefore, study on a handoff algorithm for guaranteeing QoS is required. Related works on handoff for beyond 3G system mainly focused on reducing the L3 handoff delay or packet loss. That is, these schemes try to compensate L2 delay rather directly eliminate it. However, remained 1.2 delay degrades QoS, especially delay-sensitive realtime traffic. Therefore, we proposed the seamless handoff algorithmwhich can minimize the L2 handoff delay.

  • Fibrinolytic enzyme (FE-2) was purified from the fruiting bodies of Tricholoma saponaceum using DEAE-Cellulose chromatography and Mono-S column chromatography, The enzyme has a molecular weight of 18.23 kDa and include Zn$^{2+}$ ion as found by ICP/MS. The N-terminal amino acid sequence of the enzyme was A-L-Y-V-G-X-S-P-X-Q-Q-S-L-L-V It has a pH optimum at pH 7.5, suggested that FE-2 was a neutral pretense. The activity of FE-2 was highly inhibited by EDTA and 1,10-phenanthroline, indicating that the enzyme is a metalloprotease. The activity of FE-2 was increased by $Mg^{2+}$, Zn$^{2+}$, Fe$^{2+}$, and Co$^{2+}$, but the enzyme activity was totally inhibited by Hg$^{2+}$. No inhibition was found with PMSF, E-64, pepstatin and 2-mercaptoethanol. The enzyme hydrolyzed both $A\alpha$ and B$\beta$ chains of human fibrinogen. The $\gamma$ chain was resistant to hydrolysis by FE-2.

Effect of Supplying Volume and Frequency of Nutrient Solution on Growth and Fruit Quality of Blueberry (블루베리 양액재배시 공급량 및 공급횟수가 수체생육 및 과실품질에 미치는 영향)

  • In this study, the effect of supplying volume and frequency of a nutrient solution consisted with $NO_3-N$ 4.6, $NH_4-N$ 3.4, $PO_4-P$ 3, K 3, Ca 4.6 and Mg $2.2mmol{\cdot}L^{-1}$ on growth and fruit quality of 'Duke' blueberry was investigated. Three years old 'Duke' blueberry bushes cultivated in containers ($60{\times}80{\times}40cm$) filled with 130L peat moss and 40L pearlite (v/v) were selected for the experiment. The growth containers were mulched with sawdust. Two different volumes (4L and 8L) of nutrient solution was tested at three different supplying frequencies (one time, two times, and three times) per week and the drainage quality of nutrient solution and fruit quality of 'Duke' blueberry was evaluated. The optimal drainage rate for the vegetable cultivation is known to be 20-30%. The results revealed that the average drainage rates of 27% and 29% for the nutrient solution supplied in 'Duke' blueberry growth medium at 4L, 2 times/7 days and 4L, 3 times/7days, respectively. The highest shoot diameter (4.2mm) and shoot length (31cm) of 'Duke' blueberry was recorded with the 8L of nutrient solution supplied at 3 times per 7 days. According to the analysis of inorganic components in the drainage of nutrient solution, there was a tendency of absorbing nitrogen at the early stage of growth. The supplying volume and frequency of nutrient solution was not significantly affected on 'Duke' blueberry fruit weight, soluble solids content, and titratable acidity. The highest yield per bush (2.7kg) was recorded for the nutrient solution supplied with 4L at three times per 7 days, while the 4L nutrient solution supplied at one time per 7 days resulted the lowest yield of 1.4kg per bush. Consequently, the tested nutrient solution can be applied for the 'Duke' blueberry bushes with the volume of 4L at three times per week for the better crop growth.

Effect of Soil Texture on Rice Growth and Paddy Soil Percolation under Lysimeter Condition (라이시미터 조건에서 토성이 벼의 생육 및 논토양의 지하삼투수량에 미치는 영향)

  • The lysimeter experiment was conducted to investigate the temporal changes of irrigation requirement, soil water percolation and rice root distribution during rice growing period under different soil texture that were sandy loam, clay loam and clay paddy soil in 1999 and 2000. The irrigation requirement in the first year was 3,306 l/$m^2$ in clay loam, 2,650 l/$m^2$ in sandy loam and 2,002 l/$m^2$ in clay soil. However, the highest irrigation requirement was 5,281 l/$m^2$ in sandy loam and the next was 4,984 l/$m^2$ in clay loam and 3,968 l/$m^2$ in clay soil in the second year, Soil water percolation in the first year was 2,141 l/$m^2$ in clay loam, 1,228 l/$m^2$ in Sandy loam and 862 l/$m^2$ in clay soil. However, in the second year, the highest water percolation of 4,448 l/$m^2$ was measured in sandy loam, and was followed by 3,833 l/$m^2$ in clay loam and 2,925 l/$m^2$ in clay soil. Distribution ratio of rice roots measured in 0-10cm of soil depth was 56.0% in sandy loam, 61.4% in clay loam and 72.1% in clay soil, respectively. It was interpreted that the greater water percolation measured in the second year was caused mainly by the large amount of rice root growth. Therefore, it was concluded that the soil water percolation in rice paddy soil was affected greatly not only by soil texture but also the growth of rice root.

  • Production of crude glycerol from biodiesel industry is expected to exceed the commercial demand for purified glycerol in the near future. This study aimed to evaluate the feasibility of co-digestion of crude glycerol with swine manure. Crude glycerol up to 13.8 g/L was regarded as a good co-substrate for swine manure digester. It improved methane production and productivity by 90% and 120%, respectively. Methane yield of crude glycerol at the condition was estimated to be 232 mL/g. However, it inhibited methanogenic activity at above 27.5 g/L. Optimum concentration of crude glycerol for co-digestion with swine manure would be near to 13.8 g/L.

Antioxidant Activities of Methanol Extracts from Prunella vulgaris (하고초 메탄올 추출물의 항산화 활성)

  • The aim of the present study was to investigate the antioxidant activities of methanol extracts from whole plant, flower stalk and stem of Hagocho (Prunella vulgaris). Content of total phenolic compound was the highest in flower stalk (77.1 mg/100 g) and those of others were below 54.0 mg/100 g. Flavonoid contents was the highest in stem (36.1 mg/100 g) compared to other samples. Electron donating ability of Prunella vulgaris was activated at over 70% in all samples at $500{\mu}g/mL$ concentration, especially, the activity was the highest (92.1%) in flower stalk extracts. Reducing power showed similar tendency to electron donating ability, which was significantly higher flower stalk ($0.3{\sim}1.9$), whole plant ($0.2{\sim}1.6$) and stem ($0.2{\sim}1.5$). Hydroxyl radical was scavenged over 80% in $100{\mu}g/mL$ concentration and was not significantly different between parts. Antioxidant activity in $\beta$-carotene-linoleic acid system was $47.5{\sim}84.6%$ when $1,000{\mu}g/mL$ methanol extracts was added to reaction mixtures, and flower stalk showed the highest activity. Ability of ABTs cation decolorization from Prunella vulgaris was activated over 50% in all samples when $250{\mu}g/mL$ of methanol extracts was added to reaction mixtures and $500{\mu}g/mL$ were the most suitable concentration for its activation. Nitric oxide scavenging activity was lower under 20%, but its activity was significantly higher in flower stalk than other parts. The results indicate that flower stalk from Prunella vulgaris has potent antioxidant activities.

Antioxidative Effects of Ethanol Extracts from Rhus Verniciflua Stokes (RVS) on Mouse Whole Brain Cells (옻나무 에탄올 추출물의 쥐 뇌세포에 대한 항산화효과)

  • To measure antioxidative activities, the various extracts from RVS (Rhus Verniciflua Stokes) were tried out with either DPPH or thiocyanate method. Also we used the GO (Glucose Oxidase) 20 mU/mL hydroxyl radical system in mouse whole brain cell culture. Chloroform, n-hexane or ethanol were used as extract solutions which had different polarity respectively. In DPPH and thiocyanate method, the antioxidative activities of the crude ethanol extracts were stronger than other extracts. The crude ethanol extracts were fractionated 5 peaks by glass column. Among of them, antioxidative activity of peak II $(P_{II})$ was shown stronger than other fractions, a little for peak III $(P_{III})$ and peak IV $(P_{IV})$, and none for peak I $(P_I)$ and Peak V $(P_V)$. In the antioxidative effects of crude ethanol extracts (30 mg/mL), cell viabilities were evaluated $1\;{\mu}L\;(297\;{\mu}g/mL)$, $2\;{\mu}L\;(588\;{\mu}g/mL)$ of crude ethanol extracts 59%, 68% respectively. $10\;{\mu}L\;(2,727\;{\mu}g/mL)$ addition of crude ethanol extracts had 95% cell viabilities, 0.01% significant, comparing control. In addition, the compounds related to antioxidative effect of crude ethanol extract might be glycoproteins by means of SDS-PAGE. Comparison to antioxidative effects between several antioxidants (ascorbic acid, ${\alpha}-tocopherol$, catalase) $273\;{\mu}L/mL$ addition of crude ethanol extracts corresponds to $1\;{\mu}g/mL$ catalase in antioxidative effects.

The association between serum IGF-1 and neonatal growth and disease in a NICU (신생아집중치료실 입원아의 혈청 IGF-1과 성장 및 질병 사이의 관련성)

  • Purpose : The objective of this study was to establish the serum IGF-1 level in newborn infants, and investigate its association with growth and diseases. Methods : In a retrospective study, serum IGF-1 levels were measured for newborn infants admitted to NICU at Kyungpook University Hospital from March 2007 to July 2007. Birth data, disease history, and hospital course were obtained from medical records. Results : Of 52 blood samples obtained at birth, serum IGF-l levels in 30 preterm infants ($31.6{\pm}27.3$ ng/mL) were lower than in 22 full-term infants ($53.4{\pm}40.0$ ng/mL; P<0.05). In sick full-term infants, serum IGF-1 levels ($46.0{\pm}40.2$ ng/mL) were lower than in healthy full-term infants ($64.1{\pm}39.5$ ng/mL; P<0.05). In preterm infants, there were no differences in IGF-1 levels between healthy ($33.2{\pm}23.3$ ng/mL) and sick infants ($30.6{\pm}30.4$ ng/mL); however, IGF-1 levels in both sick and healthy preterm infants were lower than in healthy full-term infants. Among infants admitted after 8 days of life, serum IGF-1 levels were higher in infants who gained weight ($70.8{\pm}36.2$ ng/mL) than in infants who lost weight ($13.3{\pm}19.9$ ng/mL; P<0.01); however IGF-1 levels showed no difference between gender or method of delivery. Conclusion : The study showed lower IGF-l levels in preterm infants than in full-term infants. Additionally, the IGF-l level in infants with weight loss was lower than in infants with weight gain. These results indicate that serum IGF-1 is associated with gestational age and postnatal growth.
