Diketopiperazines from Cordyceps militaris (동충하초의 Diketopiperazine 성분)

  • In a continuation of investigation on Cordyceps militaris, thirteen compounds were isolated from the $CH_2Cl_2$ and n-BuOH-soluble fraction of C. militaris. They were identified as twelve diketopiperazines such as cyclo($\small{L}$-Gly-$\small{L}$-Pro) (1), cyclo($\small{L}$-Ala-$\small{L}$-Pro) (2), cyclo($\small{L}$-Ser-$\small{L}$-Pro) (3), cyclo($\small{L}$-Val-$\small{L}$-Pro) (4), cyclo($\small{L}$-Thr-$\small{L}$-Pro) (5), cyclo($\small{L}$-Pro-$\small{L}$-Pro) (6), cyclo($\small{L}$-Thr-$\small{L}$-Leu) (7), cyclo($\small{L}$-Tyr-$\small{L}$-Ala) (8), cyclo($\small{L}$-Phe-$\small{L}$-Ser) (9), cyclo($\small{L}$-Phe-$\small{L}$-Pro) (10), cyclo($\small{L}$-Tyr-$\small{L}$-Pro) (11) and brevianamide F (13), and an amino acid, tryptophan (12). Their structures were identified on the basis of chemical evidences and spectroscopic analysis including 1D-NMR ($^1H$, $^{13}C$), 2D-NMR (HSQC, HMBC) and MS spectral data. Among the isolated compounds, compounds 1, 2, 6-11 are first reported from C. militaris.

  • The most effective cytokinin source for adventitious shoot regeneration of in vitro grown leaves from 'Gala' apple was BA with the concentration of 4.0 mg/L, while auxin source was IAA, IBA and NAA with the concentration of 0.1 mg/L, respectively. As the result of combinational treatment of BA and NAA, 6.0 or 8.0 mg/L BA with 0.5 mg/L NAA was effective for adventitious shoot regeneration from leaf tissues of 'Gala', 0.1 mg/L NAA + 8.0 mg/L BA and 1.0mg/L NAA + 8.0 mg/L BA for internode, and 0.1 mg/L NAA + 4.0 mg/L BA for petiole.

The Evidence for Pepsin-Catalyzed Transpeptidation (펩신촉매에 의한 Transpeptide의 생성)

  • Procine pepsin hydrolysis of hexapeptide L-S-pNF-Nle-A-OMe in the presence of dipeptide L-L generates a new peak on HPLC analysis of reaction mixtures that is not seen when enzyme is incubated with either peptide alone. The peaks can be detected spectroscopically at either 214 or 254 nm, the latter consistent with a new peptide containing the p-nitro-F residue. The data suggest acyl transpeptidation between E(L-S-pNF) and L-L to form L-S-pNF-L-L. Consistent with this inference are (1) the ability of L-L-NH$_{2}$ and inability of Boc-L-L to undergo a similar transpeptidation reaction, and (2) the data from electrospray mass spectrum. This synthesis requires that Nle-A-L-OMe be released before L-S-pNF, an order opposite to that proposed on the basis of product inhibition kinetics. Consistent with this inference are reciprocal solvent isotope effects ; normal isotope effects of 1.736$\pm$0.121 on the formation of Nle-A-L-OMe and 2.281$\pm$0.184 in the formation of L-S-pNF, coupled to an inverse isotope effects of 0.576$\pm$0.045 on the formation of L-S-pNF-L-L. Because transpeptidation precedes faster in D$_{2}$O, the isotopically-sensitive step must occur after release of Nle-A-L-OMe. Isotopically-enhanced transpeptidation is consistent with the Uni-Bi iso memchanism postulated on the basis of an isotope effects on Vmax but not on Vmax/Km$^{1)}$ and confirmed by isotope effects on the onset of inhibition by pepstatin$^{2)}$.

Enfluence of Herbal Extract on Lactic Acid Bacteria Growth and Cryoprotectants (생약 추출물이 유산균의 생육 및 동결건조 보호제로서의 효과)

  • Extract of Eleutherococcus senticosus, Lycium chinensis, Angelica acutiloba and Schizandra chinensis were investigated to determine whether its addition may inflict on growth of lactic acid bacteria, freeze dry and cell viability during the storage. In cultivation with herbal extract, all strains tested did not demonstrate a significant reduction in their cell population, particularly Lactobacillus. acidophilus, L. bulgaricus, L. paracasei. But Schizandra chinensis extract inhibited growth of several strains. Immediately after freeze-drying using the skim milk 10%(w/v), sucrose 5%(w/v) with herbal extract 2.5%(v/v), the percentage viability was about 81.7%, 63.8%, 73.2%, 78.1 % in L. acidophilus, L. bulgaricus, L. paracasei, L. casei respectively. The protective effect of herbal extract to cell damage from freeze-drying was weak in comparison with control. During accelerated storage of freeze-dried lactic acid bacteria, those survival rate decrease rapidly, reaching 8 -18% in one month. But addition of Eleutherococcus senticosus extract in freeze dry of L. acidophilus showed a positive activity in storage.

Varietal Difference in Plant Regeneration from Cotyledon Culture of Capsicum annuum L. (고추 자엽에서 식물체 재분화의 품종간 차이)

  • Effects of genotype and culture medium on plant regeneration from cotyledon segments of red pepper(Capsicum annuum L.) was investigated. Among combinations of IAA(0.25 and 0.50 mg/L) and zeatin(2.0 and 4.0 mg/L) added to MS medium, combination of 2.0 mg/L zeatin and 0.25 mg/L IAA was shown to be the best for shoot differentiation from cotyledon segments. Shoot regeneration from cotyledon explants took 9 to 25days, depending on genotypes and culture media. Early shooting was observed in Yeongyangjaelae, Putgochw, Karkovskij-A-35, Gris I-A-1 on MS medium containing 2.0 mg/L zeatin and 0.25 IAA mg/L. Percent of explants producing shoots, as also influenced by genotypes and culture media, were over 90% for 621, Yeongyangjaelae, Putgochw, Nikko jacksacgmulgochw, Ch-6-Num-216, and Kajenskij-A-35 when cultured on MS medum supplemented with 2.0 mg/L zeatin and 0.25 mg/L IAA and for Fresno chile, PI 169126, Kajenskij-A-35, jacksacgmulgochw, and PI 297438 on MS medium including 2.0 mg/L BA and 1.0 mg/L IAA.

The Effects of L1 Rhetorical Styles on L2 Writing Quality

  • This study is a small-scale replication study of Kubota (1998). Kubota discovered that L1 rhetoric was not the main cause of L2 writing problems. The conclusion of Kubota's study was in contrast to that of contrastive rhetoric research that claimed that the use of L1 rhetorical styles in L2 writing negatively affects the quality of L2 writing, in particular, at the organizational level. According to Kubota(1998), the use of Japanese rhetorical styles did not deteriorate the quality of English writing. By investigating Korean ESL learners, this replication study examined whether L1 and L2 writings have the same rhetorical styles and how L1 rhetorical styles affect the L2 writing quality. The study analyzed Korean and English persuasive writings written by 12 Korean ESL learners in terms of organization. The results show that Korean learners of English used different rhetorical styles in their L1 Korean writing and L2 English writing. However, there was a positive relationship between L1 Korean and L2 English organization scores.

Analysis of Latent Classes and Influencing Factors According to the Love Types of Korean Adults (한국 성인의 사랑유형 잠재집단 및 영향요인 분석)

  • This study was conducted to classify 601 Korean adults into latent classes according to their love types and identify the differences in depression and find variables that affect the latent classes classification. As a result of the latent class analysis, the latent group for love types of Korean adults were classified into the L-H (7.7%) group, which showed the highest level of all three factors of intimacy, passion, and commitment, and the L-MH (33.6%) group, which all three factors were higher than the average, the L-M (39.8%) group with the mean of all three factors, the L-ML (14.6%) group with all three factors lower than the mean, and the L-L (4.3%) group with the lowest all three factors. Also, as a result of ANOVA, the L-MH group was psychologically healthier and more adaptive than the L-ML group. As a result of multinomial logistic analysis, females were more likely to belong to L-M, L-ML and L-L groups than males. In addition, singles were more likely to belong to the L-M and L-ML groups than those who were married. Also, the higher the anxiety attachment level, the higher the likelihood of belonging to the L-M, L-ML, and L-L groups than the L-H and L-MH groups, the L-ML and L-L groups than the L-M groups, and the L-L group rather than the L-ML groups. However, age, neuroticism, and emotional regulation did not affect the classification of latent classes. This study is meaningful in that it identified the various latent classes for the love types of Korean adults more three-dimensionally and suggested the possibility of differential interventions according to the characteristics of each group.

A Study on the Integrated Management of XML and EDI Electronic Letters of Credit (EDI 방식과 XML 방식간 통합관리를 통한 전자신용장의 활성화 방안에 관한 연구)

  • Traditionally, a L/C (letter of credit) has been known as a relatively low risk method of payment. This is why L/C has been used in international trade. However, L/C has a number of weak points such as high cost, long processing time, and complicated documents. Using an electronic L/C is one way to solve those weak points. In Korea, there are two types of electronic L/C. One is the EDI (Electronic Data Interchange) based L/C and the other one is the XML (Extensible Markup Language) based L/C. The former, established in 1990's, is sent from banks to the beneficiary solely through VAN (Value Added Network, KTNET) operators. On the other hand the latter, started in 2005, is sent from banks to KFTC (Korea Financial Telecommunications & Clearings Institute) for management of the L/C balance, as well as to KTNET. So far, paper L/C and EDI based L/C have been used overwhelmingly instead of XML based L/C in spite of the aforesaid disadvantages. In this paper, the authors examined empirically why the users of electronic L/C were reluctant to use XML based L/C. The results are as follows. First, the users of paper L/C were more dissatisfied than the users of electronic L/C due to many factors such as cost, the time required, and information reuse. Second, the users who have more experience with XML based L/C wanted to adopt integrated management with EDI based L/C more than the users who had not experienced XML based L/C. Third, the users who had used more than one form of L/C wanted to adopt integrated management to EDI and XML based L/C more than the users who had only used one form of L/C. Therefore, the authority for electronic L/C should consider a change of the policy from the XML based electronic L/C oriented to integrated management of the various types of L/C.

  • As a possible biocompatible and biodegrable polymer skeleton for drug delivery system, block copolymers of L-lactic acid and L-glutamic acid with different composition were synthesized and characterized. Poly (L-lactide) was prepared by polymerization of L-lactide with zine oxide at $130^{\circ}C$ for 72 hrs. 3-Amino-l-propanol was introduced to poly (L-lactide) by an ester linkage in order to initiate polymerization. Polymerization of $\gamma-benzyl-L-glutamate-N-carboxyanhydride(\gamma-BLG-NCA)$ utiliizing the amino group of modified poly (L-lactide) as an initiator gave rise to the block copoly $(L-lactide-\gamma-benzyl-L-glutamate).$ The NMR study of resulting block copolymers showed that the composition of L-lactic acid and $\gamma-benzyl-L-glutamate$ in block copolymers was depended on the weight ratio of poly (L-lactide) and $\gamma-BLG-NCA.$ The thermal properties of the resulting block copolymers were determined by the differential scanning calorimetry and by the thermogravimetry.
