포도 송이가지에서 레스베라트롤의 분리 정제 (Isolation and Purification of Resveratrol from a Grape Twig)

  • 포도즙 착즙 후 폐기되는 포도송이가지를 재활용하여 항산화 물질로 잘 알려진 resveratrol을 80% 에탄올로 초음파 추출기를 이용하여 60분 동안 추출한 후 실리카겔 컬럼을 적용시켜 99% 이상의 순도로 정제하였다. 정제된 resveratrol의 안정성은 4$^{\circ}C$에서는 반감기가 90일이었으며, 25$^{\circ}C$에서는 60일로 조사되었다. 또한 고순도 resveratrol 추출물을 적용하여 기존의 식품에 포함된 양의 2$\sim$10배 정도 강화시킨 포도즙의 관능평가를 수행한 결과 전반적인 기호도가 높았으며, 색, 향 및 당도는 일반 포도즙에 비해 유의적인 차이가 없었다. 앞으로 포도착즙의 부산물인 포도송이가지에서 추출된 resveratrol은 건강보조식품, 기능성 향장품 소재 등으로 활용가치가 높을 것으로 사료된다.

  • The isolation, morphological study and growth characteristics of the algae were investigated from Lake Chuam. The isolated algae were applied the Agal Growth Potential test. The method of isolation and purification of the algae were used to Agar plating(AP), nutrient enrichment(NE), dilution(DI) and micro capillary technique(MC). Total isolated algae were 21 species. They were composed of Cyanophyceae, Dinophyceae, Bacillariophyceae, Euglenophyceae and Chlorophyceae. The numbers of algal strain by isolation technique were highest in dilution(21 species), and those of the rests were showed in order of NE > MC > AP. The sizes of isolated Selenastrum and Scenedesmus were $1.8\pm 1.4 \mu m$, $3.3\pm 0.9 \mu m$ in diameter and $6.4\pm 2.3 \mu m$, $13.6\pm 1.9 \mu m$ in length respectively. The morphology of isolated algae and NIES-collection strain was very similar each other, but the size was smaller isolated algae than that of NIES-collection. The optimum culture condition of isolated Selenastrum and Scenedesmus was about 30$\circ$C(25$\circ$C-35$\circ$C) in temperature and the maximum growth was appeared between 7,000 lux and 8,000 lux in the light intensity. The comparison of $\mu$(specific growth rate) on the concentration of nutrients such as nitrate and phosphate, isolated Selenastrum was appeared maximum it at 1.0 mg $NO_3-N/l$ but NIES-collection strain was showed 95% of maximum it at same nitrate concentration. Maximum g of isolated algae and NIES-collection strain in Scenedesmus onto nitrate concentration were very similar with the result of selenastrum. The specific growth rates of isolated algae and NIES-collection strain on the gradient concentration of phosphate were showed 0.72/day and 0.70/day at 0.02 mg $PO_4-P/l$ in Selenastrum but those of Scenedesmus were appeared 0.61/day and 0.57/day at same concentration $PO_4-P$.

  • Glycolipids produced by Pseudomonas aeruginosa YPJ-80 were characterized by chromatographic and spectorscopic techniques as a mixture of two rhamnolipids. For recovery of glycolipids from the culture broth, various isolation methods including ultrafiltration, adsorption and solvent extraction were compared. Ultrafiltration showed the best results in terms of glycolipids recovery. Further purification for spectroscopic analysis was carried out by adsorption chromatography and preparative thin layer chromatography. From the spectroscopic analysis, such as IR spectroscopy. FAB-MS, 1H-NMR and 13C-NMR and hydrolysis analysis, the glycolipids were identified as L-${\alpha}$-rhamnopyranosyl-${\beta}$-hydroxydecanoly-${\beta}$-hydroxydecanoate and 2-O-${\alpha}$-L-rhamnopyranosyl-${\alpha}$-L-rhamnopyranosyl-${\beta}$-hydroxydecanoyl-${\beta}$-hydroxydecanoate. Monorhamnolipid and dirhamnolipid lowered the surface tension of water to 28.1 mN/M and 29.3 mN/m, respectively.

Aspergillus속 균주가 생산하는 Phytase의 분리 정제 및 특성 (Isolation, Purification and Characterization of Phytase from Asperfillus sp.)

  • To extract insoluble proteins and to improve funtional properties of abolished proteins, an phytase producing Aspergillus sp. SM-15 was isolated from soil. The enzyme was purified and its enzymological characteristics were investigated. Phytase production reached to maximum when the wheat bran medium containing 1% mannose, 1% yeast extract, 1% (NH4)2HPO4 and 0.2% calcium chloride was cultured for 4 days. Phytase was purified 17.1 fold and specific activity was 244.32unit/mg by a sequencial process of ammonium sulfate fraction, ion exchange chromatography and gel filtrations Pruified enzyme was confirmed as a single band by the polyacrylamide gel electro-phoresis. The molecular weight of phytase was estimated to be 46,000. The optimum pH and temperature for the phytase activity were 5.5 and 5$0^{\circ}C$. The enzyme is stable in pH 4.5~5.5, 6$0^{\circ}C$. The activity of purified enzyme was inhibited by Hg2+ whereas activited by Pb2+ and Fe2+. The activity of phytase was inhibited by the treatment with iodine. The result indicate the possible involvement of histidine at active site. Km and Vmax of the puridied phytase were 37.037mM/L and 159.87umol/min, respectively.

도시철도 역사개선에 관한연구 (A study on the subway-station improvement of metropolitan subway)

  • Subway station is installed with equipment of deficient ventilation and air purification in need of routine activity for users because projectors didn't have recognition of the importance of prevention equipment on disasters in subway stations. So if the accident in Deagu is occurred again, we can't prevent it. And it is urgently necessary to improve antique subway station for the safety and service to citizens because users were increased far more. Improvement of subway station has to be modernized the backward and antique subway station and maintained and developed by the center of traffic to give the social contribution with management of social facilities. And subway station has to be made conveniently so that everyone may use the station with one-stop system including sales, business, service equipment. This study will show the improvement plan of problem in underground space and situation of metropolitan-train history to improve metropolitan-train station .we will study a traffic line and equipment relocation of underground space considering safety by choosing the improved station and problems of the underground station in seoul. And then we will study the improvement plan of air purification and sound isolation to upgrade service in citizens.

  • 누에의 성충특이체액단백질(adult specific protein : ASP)의 생리 생화학적 특성 및 성충화 발육에 따른 기능을 구명하기 위한 연구의 일환으로 ASP의 분리·정제 및 분자적 특성을 조사하였다. ASP는 Sephadex G-100 gel 여과 및 CM52 ion exchange chromatography에 의해 분리·정제되었으며, 정제된 단백질의 순도는 native-PAGE 및 면역전기영동에 의해 확인되었다. ASP는 분자량 19.5kDa 및 17.5kDa으로 추정되는 2종류의 subunits으로 구성되어 있고 또한 당 및 지질을 포함하지 않는 단순단백질임이 밝혀졌다.

  • 천잠의 알에서 난황 주단백질인 vitellin을 분리·정제하고 가잠 및 상잠의 vitellin과 첨잠 nitellin과의 면역학적 특성을 비교하였으며 천잠의 배자발생에 따른 난내 vitellin의 변동을 조사하였다. 1. 전잠의 체액 vitellogenin은 유충기의 토사말기부터 검출되었다. 2. 점잠의 vitellinm은 가잠 및 상잠의 vitellin과 저기영동상의 이동도는 같았으나 면역학적인 특성에 있어서는 비동질성이었다. 3. 천잠 vitellin은 함량은 전체 가용성 난단백질의 46.0%였으며, 배자발생에 따른 vitellin의 변동은 산난후 8일까지는 같은 수준이었나, 10일 이후부터는 감소되는 경향을 보이다가 5개월된 난과 17개월된 난에서는 상당히 미량만이 검출되었다.

Meroparamycin Production by Newly Isolated Streptomyces sp. Strain MAR01: Taxonomy, Fermentation, Purification and Structural Elucidation

  • Twelve actinomycete strains were isolated from Egyptian soil. The isolated actinomycete strains were then screened with regard to their potential to generate antibiotics. The most potent of the producer strains was selected and identified. The cultural and physiological characteristics of the strain identified. the strain as a member of the genus Streptomyces. The nucleotide sequence of the 16S rRNA gene (1.5kb) of the most potent strain evidenced a 99% similarity with Streptomyces spp. and S. aureofaciens 16S rRNA genes, and the isolated strain was ultimately identified as Streptomyces sp. MAR01. The extraction of the fermentation broth of this strain resulted in the isolation of one major compound, which was active in vitro against gram-positive, gram-negative representatives and Candida albicans. The chemical structure of this bioactive compound was elucidated based on the spectroscopic data obtained from the application of MS, IR, UV, $^1H$ NMR, $^{13}C$ NMR, and elemental analysis techniques. Via comparison to the reference data in the relevant literature and in the database search, this antibiotic, which had a molecular formula of $C_{19}H_{29}NO_2$ and a molecular weight of 303.44, was determined to differ from those produced by this genus as well as the available known antibiotics. Therefore, this antibiotic was designated Meroparamycin.

철도 정비창의 폐기물과 혼합된 중금속 오염토 분리에 관한 연구 (A Study on Isolation of Mixed Heavy Metal-Contaminated Soil and the Waste in Railroad Workshop)

  • 본 연구에서는 철도정비창 부지 내에 폐기물 및 중금속오염 구간에서 채취한 토양을 대상으로 하였다. 그리고 효율적인 정화공정 설계를 위하여 고농도 오염구간, 저농도 오염구간, 폐주물사 함유 시료를 대상으로 입도분포 및 입도 분포 오염농도 분석을 실시하였다. 하지만 폐콘크리트, 폐목재 등의 건설폐기물, 폐주물사, 소각재 등이 부지 전반에 걸쳐 매립되어 있어 일반토양 오염과 다른 양상을 보이고 있었다. 따라서 일반적인 중금속정화기술로는 오염원이 감소하지 않아 혼합된 폐기물 중에 자성을 띠는 성분을 자력선별을 적용하여 실험한 결과 중금속 오염도는 감소하는 것으로 나타났다.

Purification of Pig Muscle Stem Cells Using Magnetic-Activated Cell Sorting (MACS) Based on the Expression of Cluster of Differentiation 29 (CD29)

  • The muscle stem cells of domestic animals are of interest to researchers in the food and biotechnology industries for the production of cultured meat. For producing cultured meat, it is crucial for muscle stem cells to be efficiently isolated and stably maintained in vitro on a large scale. In the present study, we aimed to optimize the method for the enrichment of pig muscle stem cells using a magnetic-activated cell sorting (MACS) system. Pig muscle stem cells were collected from the biceps femoris muscles of 14 d-old pigs of three breeds [Landrace×Yorkshire×Duroc (LYD), Berkshire, and Korean native pigs] and cultured in skeletal muscle growth medium-2 (SkGM-2) supplemented with epidermal growth factor (EGF), dexamethasone, and a p38 inhibitor (SB203580). Approximately 30% of total cultured cells were nonmyogenic cells in the absence of purification in our system, as determined by immunostaining for cluster of differentiation 56 (CD56) and CD29, which are known markers of muscle stem cells. Interestingly, following MACS isolation using the CD29 antibody, the proportion of CD56+/CD29+ muscle stem cells was significantly increased (91.5±2.40%), and the proportion of CD56 single-positive nonmyogenic cells was dramatically decreased. Furthermore, we verified that this method worked well for purifying muscle stem cells in the three pig breeds. Accordingly, we found that CD29 is a valuable candidate among the various marker genes for the isolation of pig muscle stem cells and developed a simple sorting method based on a single antibody to this protein.
