Sequence Analysis of iap Gene PCR Products using Listeria monocytogenes Serotypes

  • The polymerase chain reaction (PCR) amplification technique was used for comparison of Listeria monocytogenes serotypes. PCR primers for the fragment of invasion-associated protein (iap) gene were highly specific for all the serotypes of L. monocytogenes. Other Listeria spp., such as Listeria ivanovii and Listeria innocua were not produced the PCR fragments by above primer set. The nucleotide sequences of PCR products showed high homologies in comparison of all the isolated serotypes except unknown type II-2. The deduced amino acid sequences of the PCR products also showed similar to one another. The various region of the PCR products, called a Thr-Asn repeat region was presented. All of isolated L. monocytogenes serotypes possessed 16 to 20 Thr-Asn repeats.

  • The 2-step competition graph of D has the same vertex set as D and an edge between vertices x and y if and only if there exist (x, z)-walk of length 2 and (y, z)-walk of length 2 for some vertex z in D. The 2-step competition number of a graph G is the smallest number k such that G together with k isolated vertices is the 2-step competition graph of an acyclic digraph. Cho, et al. showed that the 2-step competition number of a path of length at least two is two. In this paper, we characterize all the minimal acyclic digraphs whose 2-step competition graphs are paths of length n with two isolated vertices and construct all such digraphs.

  • Reverse transcription and polymerase chain reaction (RT-PCR) techiniques were used to identification and differentiation of cucumber mosaic virus isolated from Forsythia koreana (CMV-Fk). RT-PCT used by two set of 20-mer primers one was CMV-common primers and another was CMV subgroup I-specific primers designed in a conserved region of the 3' end of CMV RNA3, amplified about 490 bp and 200 bp DNA fragments from CMV-Fk, respectively. CMV could be detected by RT-PCR at a dilution as low as 10-4 in forsythia crude sap extracts. Restriction enzyme analysis of RT-PCR products using EcoRI and MspI showed that CMV-Fk belonged to CMV subgroup I. But, analysis of RNA fingerprinting by arbitrarily primed polymerase chain reaction (RAP-PCR) showed heterogeneity of RNA3 between CMV-Fk and CMV-Y as a member of subgroup I.

  • An Infectious hematopoietic necrosis virus strain (IHNV-RTK) was isolated from cultured rainbow trout at Kumi and Jechun area in Korea during 2000 and 2001. In the RT-PCR amplification with the specific primer set designed from IHNV G protein region, a 540 bp PCR product was amplified from the RTK strain. The RTK strain showed higher sequence homology with the published IHNV G protein genes (RB-76, LR-73, Col-85, and Carson-89)

Supose a group G acts smoothly on .SIGMA. with the fixed point set .SIGMA.$^{G}$ consists of two isolated fixed points p and q. In this case, tangent spaces $T_{p}$ .SIGMA. and $T_{q}$ .SIGMA. at isolated fixed points, as isotropy representations of G are called Smith equivalent. Moreover .SIGMA. is called a supporting homotopy sphere of Smith equivalent representations $T_{p}$ .SIGMA. and $T_{q}$ .SIGMA..   • Let G be a compact abelian Lie group, and M an odd-dimensional closed smooth G-manifold. If the fixed point set $M^G\neq\emptyset$ and dim $M^G=0$, then G has a subgroup H with $G/H{\cong}\mathbb{Z}_2$, the cyclic group of order 2. The tangential representation $\tau_x$(M) of G at $x{\in}M^G$ is also regarded as a representation of H by restricted action. We show that the number of fixed points is even, and that the tangential representations at fixed points are pairwise isomorphic as representations of H.

  • The competition graph of a digraph D is a graph which has the same vertex set as D and has an edge between x and y if and only if there exists a vertex v in D such that (x, v) and (y, v) are arcs of D. For any graph G, G together with sufficiently many isolated vertices is the competition graph of some acyclic digraph. The competition number k(G) of a graph G is defined to be the smallest number of such isolated vertices. In general, it is hard to compute the competition number k(G) for a graph G and it has been one of important research problems in the study of competition graphs. In this paper, we compute the competition numbers of Hamming graphs with diameter at most three.

Performance assessment of buildings isolated with S-FBI system under near-fault earthquakes

  • This study investigates the optimum design parameters of a superelastic friction base isolator (S-FBI) system through a multi-objective genetic algorithm to improve the performance of isolated buildings against near-fault earthquakes. The S-FBI system consists of a flat steel-PTFE sliding bearing and superelastic NiTi shape memory alloy (SMA) cables. Sliding bearing limits the transfer of shear across the isolation interface and provides damping from sliding friction. SMA cables provide restoring force capability to the isolation system together with additional damping characteristics. A three-story building is modeled with S-FBI isolation system. Multiple-objective numerical optimization that simultaneously minimizes isolation-level displacements and superstructure response is carried out with a genetic algorithm in order to optimize S-FBI system. Nonlinear time history analyses of the building with optimal S-FBI system are performed. A set of 20 near-fault ground motion records are used in numerical simulations. Results show that S-FBI system successfully control response of the buildings against near-fault earthquakes without sacrificing in isolation efficacy and producing large isolation-level deformations.

  • Because of resonance between natural frequency of the second floor base for IPB(Isolated Phase Bus) duct supports in a power plant and operation frequency of the turbine, there was high amplitude vibration on IPB duct. To reduce vibration of IPB duct, Firstly it was set a FEM model to seek the mode shape for the concrete structure. Secondly, it was carried out dynamic analysis for the FEM model. Lastly, because the natural frequency of the concrete structure could not be changed, it was changed supports position for the IPB duct near to beams. It resulted in reducing vibration of IPB duct.

  • Turnip mosaic virus)TuMV-Ca) was isolated from a Chinese cabbage showing severe mosaic and black necrotic spots symptoms in Korea. The virus was identified as a strain of TuMV by its host range test, particle morphology, serology, double stranded RNA analysis. For detection of the virus, reverse transcription and polymerase chain reaction(RT-PCR) was performed with a set of 18-mer TuMV-specific primers to amplify a 876 bp DNA fragment The virus was rapidly detected from total nucleic acids of virus infected tissues as well as native viral RNA of purified virion particles by RT-PCR. Detection limit of the viral RNA by RT-PCR was 10 fg.
