  • The efficient production of glucose isomerase (G. I0.) produced form Arthrobacter sp. L-3 was studied. The optimum culture time of the enzyme was 40hr. The maximum enzyme activity was found at glucose concentration 1%. G. I. activity did not affect inoculum size. The glucose isomerase activity was strongly influenced by the addition of glucose.

김치에서의 활성 TMV 검출 (Detection of tobacco mosaic virus from "Kimchi")

  • Tobacco mosaic virus (TMV) was detected from Kimchi by biological and serological assay 5. Kimchi samples three month after cooked were collected, and were inoculated on N. tabacum var. Burley 21 and NC 95. Out of 33 samples, 6 showed typical symptoms induced by TMV, local necrotic lesions on Burley 21 and mosaic on NC 95. All saps from tobacco leaves showed the mosaic symptom reacted positively against TMV antiserum by agar gel double diffusion test. Based on the results, the Kimchi is considered as one of the important inoculum sources in Korea.

세균성마름병에 대한 담배의 저항성검정 방법 (A Screening Method on Resistance of Tobacco Plants to Bacterial Wilt)

  • Three kinds of inoculation methods, capillary, root cutting and dipping were compared for an efficient way to screening the resistant tobacco variety against bacterial wilt, Ralstonia solanacearum. The pricking a capillary tube contained the pathogenic bacterial suspension(10$^{7}$ cfu/$m\ell$) to an axillary bud of each tobacco plant showed different resistance well between varieties. The less period was required in inoculating work and in disease development for the inoculation method used with capillary tube than for two other inoculation methods tested also.

  • Soybean Whetzelinia rot caused by Whetzelinia sclerotiorum was observed in Jinju area. The diseased soybean plants showed withering and sudden collapse under field conditions. Diseased parts exhibited numerous black, irregularly-shaped scleratia embedded in dense white cottonly mycelium on tissue and in the pith of diseased stems. A sclerotium in the moist sand produced several apothecia under laboratary condition. The primary inoculum was supposed to originate from overwintered sclerotia of soil and soybean debris.

Optimization of Mass cultivation Media for the Production of Biomass and Natural Colourants from Two Marine Cyanobacteria by a Mathematical Design of Experiments

  • Optimization of chemicals in the large scale sea water medium and inoculum for biomass and natural colourants production in the marine cyanobacteria, Phomidium tenue BDU 46241 (phycoerythrin producer) and P.valderianum BDU 30501 (phycocyanin producer) was carried out by experiments in L8 orthogonal array. Mathematical analysis revealed the significance of these factors. The factor(s) that critically control the yield varied with the organism and the end-product further, the desirable level of these factors between the normal and a higher level tested was identified and improved media were evolved. In both cyanobacteria, higher level of $K_2$$HPO_4$, $NaNO_3$ and inoculum with normal level of ferric ammonium citrate was found to be desirable for biomass production and additionally, higher level of $MgSO_4$ for pigment production. The level of other factors varied with the organism and the end-product. Confirmation experiments showed that the clues obtained based on mathematical experimentation are valid. In P.tenue, the medium optimized for biomass production increased the yield of biomass by 495% and the medium optimized for phycoerythrin production increased the yield of biomass by 408% with 30% increase in phycoerythrin content of the biomass. Similarly in P.valderianum, the medium optimized for biomass production increased the yield of biomass by 224% and the medium optimized for phycocyanin production increased the yield of biomass by 143% with 44% increase in phycocyanin content of the biomass.

분무경재배에 의한 arbuscular 균근균 증식에서 양액내 인산농도의 영향 (Effect of P Levels in Nutrient solution on the Propagation of Arvuscular Mycorrhizal Funfi in Aeroponics)

  • 본 연구는 분무경 방식을 이용한 arbuscular 균근균 접종원의 대량배양증식을 위하여 기주식물의 종류와 분무양액의 P 농도 등에 따른 균근감염율 및 감염양상 변화 등을 비교하였다. 기주식물 선발에서는 고구마와 토마토가 수단그라스와 질경이에 비해 우수하였다. 또한 인산농도가 5 또는 $10{\mu}M$ 보다 $20{\mu}M$의 수준으로 높아질수록 뿌리의 생성량이 많아진 반면, 뿌리의 균근감염은 인산농도가 낮을수록 높고, 하부 뿌리로 갈수록 arbuscular 균근균 감염율이 낮아지는 경향을 보였으며 감염율은 18.6~26.0% 범위였다. 16주후의 포자형성을 조사한 결과 신선중 1 g 당 500개의 포자가 형성되었고, 분무생육상당 생산된 포자는 총830.479개였다.

반응표면분석법을 통한 Arthrobacter sp.의 amylase 생산 최적화 (Optimization of Physical Factor for amylase Production by Arthrobacter sp. by Response Surface Methodology)

  • 본 연구에서는 극지 연구소로부터 분양 받은 Arthrobacter sp. PAMC 27388 균주에서 생산되는 아밀라아제(amylase)를 물리적 요인(physical factor)들의 변화를 통하여 생산배지 최적화를 수행하였다. 한천 배지 상에서 lugol solution을 이용한 클린환의 확인을 통하여 아밀라아제가 생산됨을 확인하였으며, 16S rDNA를 이용하여 동정한 결과 Arthrobacter sp. 임을 확인할 수 있었다. 최적화 이전의 아밀라아제 생산량은 1.66 mU/L로 확인되었다. 최적화 결과, 2.49 mL의 접종부피, pH 6.85, 42.87 mL의 배지 부피의 조건에서 가장 많은 양의 아밀라아제가 생산될 것으로 예상되었으며, 생산량은 2.84 mU/L로 예상되었다. 확인 실험을 통하여 최적화 이전과 비교하여 생산량이 약 150% 증가한 2.50 mU/L의 아밀라아제가 생산됨을 확인할 수 있었다.

Antagonistic Bacillus species as a biological control of ginseng root rot caused by Fusarium cf. incarnatum

  • Background: This study aimed to develop a biocontrol system for ginseng root rot caused by Fusarium cf. incarnatum. Methods: In total, 392 bacteria isolated from ginseng roots and various soils were screened for their antifungal activity against the fungal pathogen, and a bacterial isolate (B2-5) was selected as a promising candidate for the biocontrol because of the strong antagonistic activity of the bacterial cell suspension and culture filtrate against pathogen. Results: The bacterial isolate B2-5 displayed an enhanced inhibitory activity against the pathogen mycelial growth with a temperature increase to $25^{\circ}C$, produced no pectinase (related to root rotting) an no critical rot symptoms at low [$10^6$ colony-forming units (CFU)/mL] and high ($10^8CFU/mL$) inoculum concentrations. In pot experiments, pretreatment with the bacterial isolate in the presumed optimal time for disease control reduced disease severity significantly with a higher control efficacy at an inoculum concentration of $10^6CFU/mL$ than at $10^8CFU/mL$. The establishment and colonization ability of the bacterial isolates on the ginseng rhizosphere appeared to be higher when both the bacterial isolate and the pathogen were coinoculated than when the bacterial isolate was inoculated alone, suggesting its target-oriented biocontrol activity against the pathogen. Scanning electron microscopy showed that the pathogen hyphae were twisted and shriveled by the bacterial treatment, which may be a symptom of direct damage by antifungal substances. Conclusion: All of these results suggest that the bacterial isolate has good potential as a microbial agent for the biocontrol of the ginseng root rot caused by F. cf. incarnatum.

Effect of Dispersed and Proximate Inoculation Methods of Glomus etunicatum on Root Colonization of Sorghum-Sudangrass Hybrid

  • Information on the effective application method of arbuscular mycorrhizal fungi (AMF) inoculum is still inadequate. This work was performed to assess two AMF inoculation methods (dispersed and proximate) on root colonization of sorghum-sudangrass hybrid (Sorghum bicolor L.). In dispersed inoculation method, spores were inoculated in 2 kg pots of soil in which 5 day-old seedlings were transplanted and maintained for 50 days. In the proximate inoculation method, spores were first introduced in 500 mL pots where seeds were sown. After 10 days, the seedlings with the 500 mL soil were transferred to 2 kg pots without disturbing the contents. After 50 days of growth, root colonization and arbuscule abundance significantly increased (over 100%) in proximate method of inoculation. Moreover, sorghum-sudangrass hybrid had higher shoot growth (182.5 cm) and Glomalin related soil protein (GRSP) production in proximate method. Nutrient accumulation, particularly total nitrogen (82.61 mg $plant^{-1}$), was also found to be higher in proximate method of inoculation. Our results demonstrate that the proximate method of inoculation may improve the early stage mycorrhizal symbiosis and inoculum performance in Saemangeum reclaimed soil.
