삼두해정탕가미방이 $CCl_4$ 및 d-Galactosamine에 依하여 誘發된 흰쥐의 肝損傷에 미치는 影響 (The effects of Samduhaejungtang-gamibang on liver injury of rats induced by $CCl_4$ and d-Galactosamine)

  • In order to investigate the protective effect of Samduhaejungtang-gamibang on the liver injury of rats induced by $CCl_4$ and d-galactosamine, the serum transaminase(GOT& GPT) alkaline phosphatase(ALP), lactic dehydrogenase(LDH) for enzyme activities and triglyceride, total bilirubin amounts for serum component were measured. All animals were divided into 4 groups, those were normal group(untreated), control group(treated with vehicle 0.9% Saline solution), sample Ⅰ group(1500mg/kg administrated), sample Ⅱ group(3000mg/kg administrated). The results were as follows: 1. The inhibitory effects of the serum GOT activities in rats induced by $CCl_4$ were noted in sample Ⅱ group(p<0.01). In serum GPT activities, sample Ⅱ group(p<0.05) only showed the inhibitory effects. 2. The inhibitory effects of the serum ALP activities in rats induced by $CCl_4$ were noted in sample Ⅱ group(p<0.01) 3. The inhibitory effects of the serum LPH activities in rats induced by $CCl_4$ were noted in sample Ⅱ group, but it is not recognized statistically. 4. The increases effects of the serum triglyceride content level in rats induced by $CCl_4$ were inhibited in both sample Ⅰ group(p<0.05) and sample Ⅱ group(p<0.01) 5. The increases effects of the serum total bilirubin content level in rats induced by $CCl_4$ were inhibited in sample Ⅱ group(p<0.05) 6. The inhibitory effects of the serum GOT, GPT activities in rats induced by d-galactosamine were noted in sample Ⅱ group (p<0.001), but sample Ⅰ group was not recognized. 7. The signiticantly inhibitory effects of in the serum LDH activities in rats induced by d-galactosamine were noted in both sample Ⅰ group(p<0.05) and sample Ⅱ group (p<0.001) 8. The increases of the serum ALP content level in rats induced by d-galactosamine were inhibited in sample Ⅱ group(p<0.05) 9. The increases of the serum total bilirubin content level in rats induced d-galactosamine were inhibited in sample Ⅱgroup(p<0.05) According to the above results, it is considered that Samduhaejungtang-gamibang has protective effect against liver injury in rats induced by $CCl_4$ and d-galactosamine. So it is required to study about the actions of mutual relation of medicines and patho-mechanism by experiment.

Effects of GABAB Receptor Antagonist on the Cardiovascular Response of Adenosine A1 and Adenosine A2 Receptor Agonist in the Spinal Cord of the Rats

  • Adenosine and GABA are known to be major inhitory neurotransmitters in the central nervous system and its receptors mediate various neurophamacological effects including cardiovascular modulatory effects. Inhibitory cardiovascular effects induced by intrathecal (i.t.) administration of adenosine $A_1$ receptor agonist and its modulation by cyclic AMP was suggested by our previous report. In this experiment, we examined the modulation of cardiovascular effects of adenosine $A_1$ receptor and adenosine $A_2$ receptor by $GABA_B$ receptors antagonist in the spinal cord. I.t. administration of 10 nmol of $N^6$-cyclohexyladenosine (CHA, an adenosine $A_1$ receptor agonist), I.t. administration of 2 nmol of 5'-(N-cyclopropyl)-carboxamidoadenosine (CPCA, an adenosine $A_2$ receptor agonist), pretreatment with 5-aminovaleric acid (a $GABA_B$ receptor antagonist, 50 nmol, i.t.) prior to administration of CHA and pretreatment with 5-aminovaleric acid (a $GABA_B$ receptor antagonist, 50 nmol, i.t.) prior to administration of CPCA were performed in anesthetized, artificially ventilated Sprague-Dawley rats. I.t. administration of 50 nmol of 5-aminovaleric acid significantly attenuated the inhibitory cardiovascular effects of CHA but did not attenuated the inhibitory cardiovascular effects of CPCA. It is suggested that cardiovascular responses of adenosine $A_1$ receptor is modulated by $GABA_B$ receptor and adenosine $A_2$ receptor is not modulated by $GABA_B$ receptor in the spinal cord.

황기건중탕합리중탕(黃耆建中湯合理中湯)이 항궤양(抗潰瘍) 및 위장관(胃腸管) 기능(機能)에 미치는 효과(效果)에 대(對)한 실험적(實驗的) 연구(硏究) (An experimental study of Wahngkigunchungtanghapleechungtang on the effects of gastric ulcer and gastro-intestinal function in rats and mice)

  • An experimental studies were done to investigate the spontaneous movements to the isolated ileum with the liquid extracts of Wahngkigunchunghapleechungtang. Then the effects of isolated ileum, pylorus-ligated and indomethacin-induced ulcer, gastric juice secretion, and transport abilities of the small and large intestine were studied. The following results have been obtained; 1. Wahngkigunchunghapleechungtang showed to have an inhibitory effects on the smooth muscle contraction of the isolated ileum by acetylcholine chloride and barium chloride. 2. Wahngkigunchunghapleechungtang showed to have an inhibitory effect on the contraction of the gastric fundus-strip by acetylcholine chloride and barium chloride. 3. The preventive effects were not recognized on the pylorus-ligated ulcer, by administration of Wahngkigunchunghapleechungtang. 4. The preventive effects were recognized on the indomethacin-induced ulcer by administration of Wahngkigunchunghapleechungtang. 5. The Inhibitory effects on the secretion gastric juice, free & total acidity, and pepsin output were recognized by administration of Wahngkigunchunghapleechungtang. 6. The transport abilities in the small intestine were not recognized by administration of Wahngkigunchunghapleechungtang. 7. The transport abilities in the large intestine were increased by administration of Wahngkigunchunghapleechungtang. According to the above results, it is considered that the experimental effects of Wahngkigunchunghapleechungtang has correspond to the oriental medical literatures.

대건중탕(大建中湯)의 항궤양(抗潰瘍) 및 위장관(胃腸管)에 미치는 효과(效果)에 관(關)한 실험적(實驗的) 연구(硏究) (An experimental study of Daegunchungtang on the gastric ulcer and the function of gastrointestinal tract in rats and mice)

  • This study was done to investigate the clinical effects of Daegunchungtang on the peptic ulcer and the function of gastointestinal tract. The liquid extracts of Daegunchungtang was administerd orally to the rats and mice. Then the effects of isolated ileum, pylorus-ligated and indomethacine-induced ulcer, gastric juice secretion, and transport abilities of the small and large intestine, were studied. The following results have been obtained : 1. Daegunchungtang showed to have an inhibitory effects on the smooth muscle contraction of the isolated ileum by acetylcholine chloride and barium chloride. 2. Inhibitory Effect of Corynoline Isolated from the Aerial Parts of Corydalis incisa on the Acetylcholinesterase

  • In the course of screening Korean natural products for acetylcholinesterase (AChE) inhibitory activity, it was found that a methanolic extract of the aerial parts of Corydalis incisa (Papaveraceae) showed significant inhibitory effects on AChE. Corynoline isolated from this plant inhibited AChE activity in a dose-dependent manner, and the $IC_{50}$ value of corynoline was $30.6{\;}{\mu}M$. The AChE inhibitory activity of corynoline was reversible and noncompetitive.

태음조위탕(太陰調胃湯)의 유탕억제효능(遺瘍抑制效能)에 관(關)한 연구(硏究) (A study on the ulcerous inhibitory effects of Taeumchowetang)

  • The experimental study used in this paper was done to examine the clinical effects of Taeumchowetang for the gastric ulcer and the function of gastrointestinal tract using rats and mice which were administered orally the water extraction from Taeumchowetang. Then, the counter-action of the water extraction on the isolated ileum and gastric fundus, the inhibitory effects of pylorus-ligated ulcer and indomethacin-induced ulcer, the associations with gastric juice secretion, total acidity, pepsin output, the transportability in the small and large intestine, were studied with administering acetylcholine chloride and barium chloride. In addition, it was investigated whether the central nervous system related to pain control and sleeping time was influenced by Taeumchowetang. The following results have been obtained; 1. As resulting from injection of acetylcholine chloride and barium chloride into the isolated ileum of rats and mice, Taeumchowetang led to have an inhibitory effect on the muscle contraction of the ileum. Then, acetylcholine chloride was measured as lower effect than barium chloride 2. For the inhibitory effect on contraction for the gastric fundus strip in rats by either acetylcholine chloride or barium chloride, the one showed low inhibitory effect, on the other hand the other showed density-dependent effect. 3. The water extraction was given on the pylorus ligated ulcer with using two different administration groups of 1,300mg/kg and 2,600mg/kg, each result was measured as 22.9% and 36.5% for an ulcerous inhibitory effect (p<0.05). 4. According to the two administration groups, the preventive effect was tabulated 18.1% and 29.3% for indomethacin-induced ulcer (p<0.05, P<0.01). 5. For associations with gastric juice secretion, total acidity, pepsin output in the administration group 2,600mg/kg, Taeumchowetang was recognized as having an inhibitory effects related to suppressive actions involving gastric juice secretion(p<0.05), and free acidity(p<0.01), but there was no significant association with total acidity and pepsin output. 6. To know the transportability in the intestine, BaSO4 solution was used. The transportability of the small intestine in the administration group 2,600mg/kg was 22.2% which was statistically significant compared with the large intestine's transportability(P<0.01). 7. In the administration group 1,300mg/kg and 2,600mg/kg, analgesic effect with against acetic acid was measured as being 16.8% and 24.4% which was shown as statistically siginificant(p<0.01). 8. No statistically significant association between Taeumchowetang and sleeping time was found in both 1,300mg/kg and 2,600mg/kg by administering phenobarbital-Na. According to the results of this study, Taeumchowetang has agreed with the effects of literature review. Further more in this study, Taeumchowetang also has preventive effects on pylorus-ligated ulcer. Hence, Teaumchowetang can be significant effect such as both anti ulcer agent and increasing gastric activity for the patients who suffer from gastric ulcer.

Inhibitory Effects of Paeonia suffruticosa Extracts on Maltase and Sucrase

  • This study was carried out to investigate inhibitory effect of extracts from root cortex of Paeonia suffruticosa on ${\alpha}-glucosidase$ (EC 3. 2. 1. 20) and postprandial hyperglycemia. Methanol extract and organic solvent (n-hexane, ethyl acetate, butanol, aqueous) fractions from the crude drug were determined for the inhibitory activities against maltase, sucrase and ${\alpha}-amylase$. The methanol extract from the crude drug strongly inhibited maltase (72%) and sucrase (76%) at the concentration of $100\;{\mu}g/ml$. Among the fractions examined, the ethyl acetate fraction from the natural plant drug showed potent inhibitory effects on maltase (85%) and sucrase (81%) at the concentration of $100\;{\mu}g/ml$. The ethyl acetate fraction from root cortex of Paeonia suffruticosa also exhibited significant reductions (21%) of blood glucose elevation in mice loaded with maltose.

종대황 스틸벤 유도체의 Cyclooxygenase 저해작용 (Inhibitory Effects of Stilbene Derivatives from Rheum undulatum on Cyclooxygenase Activity)

  • In order to clarify the anti-thrombosis activity of rhubarb, we investigated the effect of stilbene derivatives from rhizomes of Rheum undulatun on cyclooxygenase activity. Stilbene derivatives (desoxyrhapontigenin, rhapontigenin, piceatannol) exhibited the inhibitory effects on COX-1, and desoxyrhapontigenin showed inhibitory effect on COX-2. These inhibitory effect may partially contributed to anti-thrombosis activity of rhubarb.

수원 5호 메밀의 유리라디칼 소거 및 Xanthine oxidase 활성 저해 (Free radical Scavenging Activities and Inhibitory Effects on Xanthine oxidase of Buckwheat (Suwon No. 5))

  • To examine the free radical scavenging activities from buckwheat, Suwon No. 5 was extracted with various solvents. The seed of Suwon No. 5 had a high contents of carbohydrate, but the vegetable of that had a 31.4% of protein, 28.6% of ash and 25.7% carbohydrate as moisture free basis. Phenols contents of vegetable extracts had higher than those of seed extracts. Acetone extract of vegetable showed the greatest electron donating ability. Butanol and acetone extracts of vegetable showed high inhibitory effect on lipid peroxidation. Acetone extract of vegetable has also excellent activity in the superoxide radical scavenging activity by xanthine/xanthine oxidase-cytochrome c reduction system. The inhibitory effects of extracts on xanthine oxidase were measured. Acetone extract had the strongest inhibitory effect on xanthine oxidase and $IC_{50}$ was 2.2$\mu\textrm{g}$.

Tyrosinase and melanin biosynthesis inhibitory activities of crude drugs

  • Melanin biosynthesis inhibitors are useful not only for the materials used in cosmetics as skin-whitening agents but also for the remedy of hyperpigmentation. In order to find the new skin-whitening compounds from the natural products. screening of tyrosinase and melanin biosynthesis inhibitory activities in vitro has been carried out. The MeOH extracts and/or fractions of Polygoni multiflori Radix, Dalbergiae odoriferae Lignum. Solnani nigri Herba. Polygoni cuspidati Radix. POlygoni multiflori Ramulus. Salviae Radix showed tyrosinase inhibitory effects. Four methanolic extracts also showed melanin biosynthesis inhibitory effects in B-16 melanoma cell line.
