싸리나무 줄기 추출물의 추출 용매에 따른 항산화활성과 Nitric Oxide 생성 억제 활성 (Antioxidative and Nitric Oxide Production Inhibitory Activities of Lespedeza bicolor Stem Extracts Depending on Solvents)

  • In this study, we investigated on antioxidative activity and nitric oxide production inhibitory activity of various solvent extracts of Lespedeza bicolor. The total polyphenol content of the methanol extract was 192.6 mg/g and flavonoid content of the acetone extract was 40.6 mg/g, as the highest content. In DPPH radical scavenging ability, $SC_{50}$ values of the ethanol and methanol extract were exhibited $0.69mg/m{\ell}$ and $0.89mg/m{\ell}$, respectively. However, in nitric oxide(NO) scavenging ability, $SC_{50}$ values of the acetone was exhibited $0.72mg/m{\ell}$ as the highest activity. Moreover, the acetone extract showed strong NO production inhibitory effect in lipopolysaccharide(LPS)-stimulated Raw 264.7 cell. In the cytotoxicity measurement by MTT assay, the extracts were exhibited Raw 264.7 cell viabilities of 92.57~129.04% as nontoxic result in concentration of $65{\sim}650{\mu}g/m{\ell}$. As a result, the acetone extract of L. bicolor could be applicable to functional materials for anti-inflammatory related fields.

황기, 백출 및 오가피의 항산화성 및 미백효과에 관한 연구 (Antioxidative Properties and Whitening Effects of the Astragali Radix, Atractylodis Rhizoma Alba and Acanthopanacis Cortex)

  • In an attempt to find natural sources of antioxidants and whitening agents, Comparisons of the antioxidative and tyrosinase inhibitory activities of various ethanol extracts of Astragali Radix, Atractylodis Rhizoma Alba and Acanthopanacis Cortex were carried out. Comparison of the four ethanol extracts revealed that, Astragali Radix had the highest electron-donating ability(72.5%) and the highest SOD-like ability(26.1%). The xanthine oxidase experiment exhibited a hindrance effect of 88.5% in Atractylodis Rhizoma Alba, 81.1% in Acanthopanacis Cortex, 75.8% in Astragali Radix. A tyrosinase inhibitory activity assay was conducted to evaluate the whitening effects of the extracts, The tyrosinase inhibitory activity was 42.1% in the Acanthopanacis Cortex, 37.2% in the Atractylodis Rhizoma Alba, 6.0% in the Astragali Radix. Based on these results, we suggest that the ethanol extracts of Astragali Radix, Atractylodis Rhizoma Alba and Acanthopanacis Cortex can be used as food and cosmetic ingredients.

산벚나무 수피 및 코르크층 추출물의 생리활성 비교 (Comparison of the Physiological Activity of Extracts of Bark and Cork Layer from Prunus sargentii)

  • In this study, we investigated on antioxidative activity, antibacterial activity and tyrosinase inhibitory activity in 75% ethanol extracts of bark and cork layer from Prunus sargentii. The total polyphenol content of cork and bark extract was found to be 217.4 mg/g and 184.3 mg/g. And flavonoid content was found to be 31.9 mg/g and 58.6 mg/g respectively. In DPPH radical scavenging ability, cork extract was exhibited stronger scavenging ability than bark extract. Moreover, cork extract was showed higher inhibitory activity than bark extract in tyrosinase inhibitory activity. In antibacterial activity, bark extract was showed higher growth inhibition effect than cork extract against tested bacterial strains. In measurement of cytotoxicity by MTT assay, bark and cork extract showed fine cell viabilities(95.7~120.9%) against RAW 264.7 cell.

토사자, 복분자 및 작약의 항산화성 및 미백효과에 관한 연구 (Antioxidative Properties and Whitening Effects of the Cuscutae Semen, Rubi Fructus and Paeoniae Radix)

  • In an attempt to find natural sources of antioxidants and whitening agents, comparisons of the antioxidative and tyrosinase inhibitory activities of various ethanol extracts of Cuscutae Semen, Rubi Fructus and Paeoniae Radix were carried out. Comparison of the three ethanol extracts revealed that, Paeoniae Radix had the highest electron-donating ability(79.3%),; however, Rubi Fructus had the highest SOD-like ability(31.1%). The xanthine oxidase experiment exhibited a hindrance effect of 74.3% in Cuscutae Semen, 80.4% in Rubi Fructus and 60.8% in Paeoniae Radix. A tyrosinase inhibitory activity assay was conducted to evaluate the whitening effects of the extracts, The tyrosinase inhibitory activity was 20.1% in the Cuscutae Semen, 54.2% in the Rubi Fructus, 56.3% in the Paeoniae Radix. Based on these results, we suggest that the ethanol extracts of Cuscutae Semen, Rubi Fructus and Paeoniae Radix can be used as food and cosmetic ingredients.

청대(靑黛)와 생지황(生地黃)의 항염 및 항산화 효과에 대한 실험적 연구 (The Experimental Study on Anti-inflammation and Anti-oxidation of Indigo Naturalis and Rehmanniae Radix)

  • Objectives : This experiment study was performed to investigate the anti-inflammation and anti-oxidant effects of Indigo Naturalis(IN) and Rehmanniae Radix(RR) which were herbs for clearing heat. Methods : Anti-inflammation and anti-oxidation effects of IN and RR that were measured by the inhibitory ability of Nitric oxide(NO) production and the scavenging for 1,1-diphenyl-2-picrylhydrazyl(DPPH) radical. Results : 1. IN and RR groups were not showed cell toxicity. 2. In the inhibitory ability of NO production, IN groups were better than RR groups, but there were no statistical significances among the groups. 3. In the scavenging for DPPH radical, IN groups were better than RR groups, but there were no statistical significances among the groups. Conclusions : These results showed that the IN group was better than the RR group in treating skin eruptions, especially those such as erythema, purple spot, papule, pustules which appear in acute inflammation.

Antioxidative Effects and Anticancer Activities of Puer Tea Extract

  • Puer tea is a traditional beverage originating from Yunnan area of China. We have analyzed 11 different commercial tea brands provided by Daboo Culture and Art Center. This study was carried out to evaluate the contents of polyphenols, antibacterial activity, antioxidantive ability and physiological activities of extracts from Puer tea. The electron donating ability was ranged from 57.26~99.16% and SOD-like activity was ranged from 1.4~10.4%. The inhibitory effect on the growth of cancer cell lines was examined by MIT assay. The Puer tea extract exhibited the greatest inhibitory effect at the concentration of 2% for all cancer cells tested.

수원 5호 메밀의 유리라디칼 소거 및 Xanthine oxidase 활성 저해 (Free radical Scavenging Activities and Inhibitory Effects on Xanthine oxidase of Buckwheat (Suwon No. 5))

  • To examine the free radical scavenging activities from buckwheat, Suwon No. 5 was extracted with various solvents. The seed of Suwon No. 5 had a high contents of carbohydrate, but the vegetable of that had a 31.4% of protein, 28.6% of ash and 25.7% carbohydrate as moisture free basis. Phenols contents of vegetable extracts had higher than those of seed extracts. Acetone extract of vegetable showed the greatest electron donating ability. Butanol and acetone extracts of vegetable showed high inhibitory effect on lipid peroxidation. Acetone extract of vegetable has also excellent activity in the superoxide radical scavenging activity by xanthine/xanthine oxidase-cytochrome c reduction system. The inhibitory effects of extracts on xanthine oxidase were measured. Acetone extract had the strongest inhibitory effect on xanthine oxidase and $IC_{50}$ was 2.2$\mu\textrm{g}$.

Inhibitory Lignans against NFAT Transcription Factor from Acanthopanax koreanum

  • Three lignans isolated from the roots of A. koreanum (Araliaceae), namely eleutheroside E(1), tortoside A(2), and hemiariensin(4), were evaluated for their ability to inhibit NFAT transcription factor. Of these compounds, compound 4, possessing a diarylbutane skeleton, exhibited potent inhibitory activity against NFAT transcription factor (($IC_{50}$ : 36.3${\pm}2.5{\mu}\textrm{M}$). However, the activities of 1 (($IC_{50}$:>500 11M) and 2 (($IC_{50}$: 136.1 ${\pm}9.4\mu\textrm{M}$), which possess bisaryldioxabicy-clooctane skeletons, were lower. As the lignan derivatives of the same skeletons, hinokinin (5) and (-)-yatein (6) with diarylbutane skeletons and(+)-syringaresinol (3) with a bisaryldioxabicy-clooctane skeleton were also studied for their inhibitory effects on NFAT transcription factor.

Protective Effect of Kaempferol on Cultured Neuroglial Cells Damaged by Induction of Ischemia-like Condition

  • This study was performed to evaluate the cytotoxicity induced by ischemia-like condition (ILC) in cultured neuroglial cells (C6 glioma cells). The protective effect of kaempferol (KAE), flavonoid against the cytotoxicity induced by ILC induction was assessed. In addition, antioxidative effects of KAE were done by colorimetric assays. Cell viability and the antioxidative effects such as DPPH-radical scavenging activity, superoxide dismutase (SOD)-like activity and inhibitory activity of lipid peroxidation (LP) were analyzed. ILC induction decreased cell viability in a dose-dependent manner, and the $XTT_{90}$ value (low cytotoxicity value) and $XTT_{50}$ value (high cytotoxicity value) were determined during ILC induction for 15 and 40 minutes, respectively. The butylated hydroxytoluene (BHT) antioxidant significantly increased cell viability damaged by the ILC-induced cytotoxicity. In the protective effect of KAE on ILC-induced cytotoxicity, KAE protected the ILC-induced cytotoxicity by the significant increase of cell viability, and also it showed DPPH-radical scavenging ability, SOD-like ability and inhibitory ability of LP. From these results, it is suggested that ILC induction showed cytotoxicity in these cultures and the oxidative stress is involved in the ILC-induced cytotoxicity. While, KAE prevented ILC-induced cytotoxicity by antioxidative effects. In conclusion, natural products like KAE may be a putative therapeutic agent for the treatment of disease associated with oxidative stress such as ischemia.

Inhibitiory Activity of Lactic Acid Bacteria against Hazardous Microbes

  • One hundred of lactic cultures were evaluated for their ability to inhibit hazardous microbes, such as Salmonella enteritidis, Salmonella typhimurium, Escherichia coli, Listeria monocytogenes, and Bacillus cereus by agar well diffusion method. None of them showed inhibitory halo against S. enteritidis, while 27 strains showed inhibitory activity against S. typhimurium, 6 against E. coli, 9 against ampicillin resistant E. coli, 31 against L. monocytogens, 10 against B. cereus. pH of the culture does not explain for the inhibitory activity except against B. cereus. A neutralized culture from corn silage showed highest inhibitory activity against S. typhimurium, and the size of inhibitory halo was same as 10 ug/mL of ampicillin. The culture was identified to be Lactobacillus buchneri on the basis of biochemical characteristics and utilization of substrates. Using the culture as probiotics could be expected to reduce antibiotics for animal feeding.
