Inflammation is the immune system's response to infection and injury-related disorders, and is related to pro-inflammatory factors (NO, $PGE_2$, cytokines, etc.) produced by inflammatory cells. Atopic dermatitis (AD) is a representative inflammatory skin disease that is characterized by increasing serum levels of inflammatory chemokines, including macrophage-derived chemokine (MDC). Carpinus tschonoskii is a member of the genus Carpinus. We investigated the anti-inflammatory activity of C. tschonoskii by studying the effects of various solvent fractions prepared from its leaves on inflammatory mediators in HaCaT and RAW264.7 cells. We found that the chloroform fraction of C. tschonoskii inhibited MDC at both the protein and mRNA levels in HaCaT cells, acting via the inhibition of STAT1 in the IFN-${\gamma}$ signaling pathway. In addition, the chloroform fraction significantly suppressed the expression of inflammatory factors induced by lipopolysaccharide stimulation, except COX-2 and TNF-${\alpha}$. These results suggest that the chloroform fraction of C. tschonoskii leaves may include a component with potential anti-inflammatory activity.

Heracleum moellendorffii roots (HM-R) have been long treated for inflammatory diseases such as arthritis, backache and fever. However, an anti-inflammatory effect and the specific mechanism of HM-R were not yet clear. In this study, we for the first time explored the anti-inflammatory of HM-R. Results: HM-R dose-dependently blocked LPS-induced NO and PGE2 production. In addition, HM-R inhibited LPS-induced overexpression of iNOS, COX-2, $IL-1{\beta}$ and IL-6 in RAW264.7 cells. HM-R inhibited LPS-induced $NF-{\kappa}B$ signaling activation through blocking $I{\kappa}B-{\alpha}$ degradation and p65 nuclear accumulation. Furthermore, HM-R inhibited MAPK signaling activation by attenuating the phosphorylation of ERK1/2, p38 and JNK. HM-R increased nuclear accumulation of Nrf2 and HO-1 expression. However, NAC reduced the increased nuclear accumulation of Nrf2 and HO-1 expression by HM-R. In HPLC analysis, falcarinol was detected from HM-R as an anti-inflammatory compound. These results indicate that HM-R may exert anti-inflammatory activity by inhibiting $NF-{\kappa}B$ and MAPK signaling, and activating ROS/Nrf2/HO-1 signaling. From these findings, HM-R may have potential to be a candidate for the development of anti-inflammatory drugs.
Airborne fine dust (FD) particles smaller than 10 ㎛ in diameter (PM10) are one of the major causes of air pollution in East Asia, including Korea, and have become a major contributor to respiratory and skin problems. FD inordinately promotes the production of reactive oxygen species and inflammatory response in macrophages, leading to cell damage and death. Rosa rugosa, a deciduous shrub of the Rosa genus, has been used in traditional East Asian herbal medicine to treat various illnesses. The present study investigated the anti-inflammatory effects of R. rugosa organ extracts on PM10-stimulated RAW264.7 macrophages. Compared to non-treated RAW264.7 cells, treatment with 100 ㎍.ml-1 PM10 resulted in increased nitric oxide (NO) production, similar to lipopolysaccharide treatment. Additionally, 100 ㎍/ml stem extract reduced NO production by more than 45% compared to mock treatment. Furthermore, PM10-induced expression of interleukin (IL)-1β, IL-6, inducible NO synthase, and cyclooxygenase-2 was significantly reduced by stem extract treatment, indicating that the anti-inflammatory effect of the stem extract is mediated by the inhibition of pro-inflammatory mediators in PM10-stimulated RAW 264.7 cells. These results indicate that the R. rugosa stem could be considered a natural remedy with a protective effect against inflammatory responses induced by harmful airborne dust.

The chloroform extract of Croton roxburghii (Family: Euphorbiaceae) was evaluated for its antipyretic effects in Brewer's yeast induced hyperthermia in rats. The anti-inflammatory effect of the Croton roxburghii was also evaluated by using carrageenan, dextran, histamine, serotonin induced rat paw oedema and cotton pellet induced granuloma (chronic) models in rats. The chloroform extract of Croton roxburghii (CECR) exhibited significant anti-pyretic and anti-inflammatory effect at the dose 50, 100 and 200 mg/kg. Maximum inhibition (55.32%) was notedat the dose of 200 mg/kg after 3 h of drug treatment in carrageenan induced paw oedema, whereas the Indomethacin (standard drug) produced 61.33% of inhibition. The extract exhibited significant anti-inflammatory activity in dextran induced paw edema in a dose dependent manner. In the chronic model (cotton pellet induced granuloma) the CECR (200 mg/kg) and Indomethacin (10 mg/kg) showed decreased formation of granuloma tissue by 52.32% and 56.32% respectively. The extract also exhibited a significant antipyretic response in Brewer's yeast induced pyrexia in rats. Thus, the present study revealed that the CECR exhibited significant antipyretic and anti-inflammatory activity in the tested animal models.
${\beta}$-sitosterol glucoside exists in a variety of plants and have anti-tumor, anti-microbial, and immunomodulatory activities. Mast cells and eosinophils play important roles in a variety of inflammatory diseases, specifically asthma and atopic dermatitis. In the present study, we used lactose-${\beta}$-sitosterol (L-BS) and investigated the effect of L-BS on inflammatory responses of the human mast cell line, HMC-1 and the human eosinophilic leukemia cell line, EoL-1. In HMC-1 cells, L-BS significantly inhibited cell migration in response to stem cell factor without cytotoxicity. However, the mRNA expression of CC chemokine receptors (CCRs), including CCR1-5, were not altered after L-BS treatment in HMC-1 cells. LPS-induced IL-4 production was also suppressed by L-BS in a dose-dependent manner. In EoL-1 cells, the concentration ranging from 0.1 ${\mu}M$ to 10 ${\mu}M$ of L-BS had no cytotoxicity and had no effect on mRNA expression of major protein-mediators derived from activated eosinophils. However, 100 ${\mu}M$ of L-BS induced the apoptosis of EoL-1 cells in a time-dependent manner. This finding indicates the possibility of L-BS as a potential therapeutic molecule in inflammatory diseases and may contribute to the need to improve current therapeutic drugs.

Inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) are important inflammatory mediators implicated in pathogenesis of inflammation and certain types of human cancers. The present study was designed to determine whether Red Ginseng (RG) could modulate $I{\kappa}B$-kinase, iNOS and COX-2 gene expression and immune responses in RAW 264.7 macrophages stimulated with lipopolysaccharide (LPS). RG extract suppressed the expression of LPS-induced $I{\kappa}B$, iNOS, COX-2, and immune responses in a dose-dependent manner. It also showed an anti-inflammatory effect by inhibiting NF-${\kappa}B$ immune response induced by LPS treatment. Inhibitory effect of RG on LPS-induced inflammation was mediated by suppressed phosphorylation of ERK, JNK and p38 through the regulation of the mitogen-activated protein kinase (MAPK) pathway leading to a decreased production of NO, iNOS, COX-2 and NF-${\kappa}B$. The results implied the role of RG as an inflammation regulator and its possible application for curing inflammatory diseases.

Inflammation is a double-edge sword, functioning as a response against external stimuli, but excessive inflammation can lead to a harmful result. Inflammation is important in the development of vascular diseases, such as atherosclerosis and restenosis. Sohamhyung-tang(SHHT) is one of major herbal prescriptions from "Sanghanron", treating congested heat in thorax. It can be seen as an origin of several prescriptions for Hyungbi(胸痺, thoracic congestion), with its major component Trichosanthes semen. This study was done to determine whether SHHT reduces the production of nitrite, index of NO, and pro-inflammatory cytokines in Lipopolysaccharide(LPS)-induced RAW 264.7 cells. The results show that SHHT inhibited LPS-induced production of NO and PGE2, and iNOS and COX-2 expression, meaning SHHT has a significant anti-inflammatory effect. Based on these results, we suggest that SHTT can be used for prevention and treatment of diseases which starts from chronic inflammatory reactions.
To investigate the anti-inflammatory activity of natural products, we determined the anti-inflammatory activity of purified epigallocatechin (EGC) from Camellia sinensis leaves. In the present study, we found that EGC inhibited the production of proinflammatory mediators (IL-6, TNF-${\alpha}$, NO, and $PGE_2$) in lipopolysaccharide (LPS)-stimulated Raw 264.7 cells. Suppression of IL-6 seems to be at least partly attributable to the inhibitory effect of EGC. TNF-${\alpha}$ is a major cytokine produced by LPS-induced macrophages, and they have a wide variety of biological functions including regulation of inflammation. The inhibition of IL-6 and TNF-${\alpha}$ production by EGC may downregulate the acute-phase response to LPS, thereby reducing LPS-induced inflammation. In addition to IL-6 and TNF-${\alpha}$, EGC effectively reduced the production of other key inflammatory mediators, including NO and $PGE_2$. The inhibitory effect of EGC on NO and $PGE_2$ production was supported by the suppression of inducible nitric oxide synthase and COX-2 at protein levels. These results support the traditional use of EGC in the alleviation of various inflammation-associated diseases and suggest that EGC might be useful in the development of new functional foods for inflammatory diseases.

Inducible nitric oxide synthase (iNOS) and cyclooxygenase (COX)-2 are important inflammatory mediators that have been implicated in pathogenesis of inflammation and certain types of human cancers. The present study was designed in order to determine whether Wild ginseng (Panax ginseng C. A. Mayer) could modulate $I{\kappa}B$-kinase (IKK), iNOS and COX-2 gene expression and its immune responses in RAW 264.7 macrophages stimulated with lipopolysaccharide (LPS, 1 ${\mu}/m{\ell}$). Wild ginseng extract dose-dependantly (*0.5 - 2 ${\mu}/m{\ell}$) decreased the LPS-induced IKK, iNOS and COX-2 mRNA expression and its immune responses. Moreover, it inhibited unclear factor (NF)-${\kappa}B$ immune response by LPS. These data be likely to indicate that Wild ginseng may acts as inflammatory regulator and may be possible to develope a useful agent for inflammatory diseases.
As a part of ongoing research to elucidate and characterize antioxidant and anti-inflammatory nutraceuticals, solvent-partitioned fractions from Spergularia marina were tested for their ability to scavenge radicals and suppress inflammation. The results of the 2',7'-dichlorofluorescein diacetate assay indicate that solvent-partitioned fractions from S. marina scavenged intracellular radicals in $H_2O_2$-stimulated mouse macrophages. The tested fractions decreased the generation of nitric oxide (NO) and the expression of inflammation mediators, namely, inducible nitric oxide synthase (iNOS) and interleukin (IL)-6, by lipopolysaccharide (LPS)-induced mouse macrophages, indicating that S. marina decreases inflammation. Among all tested fractions [i.e., $H_2O$, n-buthanol (n-BuOH), 85% aqueous methanol (aq. MeOH), and n-hexane], the 85% aq. MeOH fraction showed the strongest antioxidant and anti-inflammatory response. The 85% aq. MeOH fraction scavenged 80% of the free radicals produced by $H_2O_2$-induced control cells. In addition, NO production was 98% lower in 85% aq. MeOH fraction-treated cells compared to LPS-induced control cells. The mRNA expression of iNOS and IL-6 was also suppressed in 85% aq. MeOH fraction-treated cells. The results of the current study suggest that the phenolic compound components of S. marina are responsible for its antioxidant and anti-inflammatory effects.
