Inflammation is the immune system's response to infection and injury-related disorders, and is related to pro-inflammatory factors (NO, $PGE_2$, cytokines, etc.) produced by inflammatory cells. Atopic dermatitis (AD) is a representative inflammatory skin disease that is characterized by increasing serum levels of inflammatory chemokines, including macrophage-derived chemokine (MDC). Carpinus tschonoskii is a member of the genus Carpinus. We investigated the anti-inflammatory activity of C. tschonoskii by studying the effects of various solvent fractions prepared from its leaves on inflammatory mediators in HaCaT and RAW264.7 cells. We found that the chloroform fraction of C. tschonoskii inhibited MDC at both the protein and mRNA levels in HaCaT cells, acting via the inhibition of STAT1 in the IFN-${\gamma}$ signaling pathway. In addition, the chloroform fraction significantly suppressed the expression of inflammatory factors induced by lipopolysaccharide stimulation, except COX-2 and TNF-${\alpha}$. These results suggest that the chloroform fraction of C. tschonoskii leaves may include a component with potential anti-inflammatory activity.
The present study describes the preliminary evaluation of the anti-inflammatory activities of Phellinus linteus (PL) and Phellinus linteus Grow in Germinated Brown Rice (BRPL). In order to effectively screen for anti-inflammatory agents, we first examined the extracts' inhibitory effects on the expression of pro-inflammatory cytokines activated with lipopolysaccharide. Moreover, we examined the inhibitory effects of the PL and BRPL extracts on pro-inflammatory factors such as NO, iNOS, $TNF-{\alpha}$ and $IFN-{\gamma}$ in murine macrophage RAW 264.7 cells. NO production and iNOS expression was significantly augmented in LPS treated cell, the production of NO and iNOS was greater in the BRPL than in the PL group. In addition, protein and mRNA levels of $TNF-{\alpha}$ and $IFN-{\gamma}$ in BRPL showed relatively more potent pro-inflammatory-activity inhibition compared to that of PL. These results suggest that BRPL may have significant effects on inflammatory factors, and may be a potential anti-inflammatory therapeutic materials.
Objectives The purpose of this study was to investigate the antioxidant, anti-inflammatory and cartilage regeneration effects of Euiiin-tang water extract (EIIT) in the treatment of monosodium iodoacetate (MIA)-induced osteoarthritis in rats. Methods Animal models were divided into five groups. The normal group didn't do any treatments causing osteoarthritis. The control group was orally administerd distilled water instead of the drug, the positive control group used indomethacin 5 mg/kg, the EIIT 100 group used EIIT 100 mg/kg and the EIIT 200 group used EIIT 200 mg/kg, and seven rats were placed per group. We administered drug to rats for 2 weeks and analyzed oxidative stress-related proteins in joint tissue. Inflammation mediators and inflammatory cytokines induced by the activity of inflammation-related proteins were analyzed. In addition, the expression of anti-inflammatory cytokines and collagen-related factors were analyzed, and H&E staining and Safranin-O staining were performed to see the effect on histopathological changes. Results 1) Oxidative stress-related proteins were significantly reduced. 2) Inflammationrelated proteins, inflammatory mediators and inflammatory cytokines were significantly reduced. 3) Anti-inflammatory cytokines were significantly increased. 4) Collagen proteolysis factors significantly decreased, and collagen degradation inhibitory factor was significantly increased. 5) EIIT administration significantly reduced cartilage degeneration and deformation in H&E staining, and reduced proteoglycan destruction in Safranin-O staining. Conclusions From the above experimental results, it judges that Euiiin-tang has antioxidant, anti-inflammatory, and cartilage regeneration effects on osteoarthritis in rats induced by MIA.
The present study was designed to assess the dynamic patterns of pro-inflammatory cytokines, including $IFN-{\gamma}$, $TNF-{\alpha}$, IL-4, IL-6, acute phase protein (${\alpha}1$-acid-glycoprotein, AGP), and an inflammation associated factor (adenosine deaminase; ADA) following experimental caprine coccidiosis. Ten kids aging from 2 to 4 months were infected orally with $5{\times}10^4$ sporulated oocysts and 10 animals served as controls. Blood samples were collected in both groups before infection and at days 3, 7, 14, 21, 28, and 35 post-infection (PI), and the levels of above-mentioned factors were measured. $IFN-{\gamma}$, $TNF-{\alpha}$, IL-4, IL-6, AGP, and ADA activities were significantly higher in infected animals from day 7 PI (P<0.05). In conclusion, the circulatory levels of most systemic inflammatory markers, including pro-inflammatory cytokines ($IFN-{\gamma}$, $TNF-{\alpha}$, IL-4, IL-6), AGP, and ADA increased significantly starting from day 3 to day 7 PI in caprine coccidiosis.
The excessive inflammatory response induced by myocardial infarction exacerbates heart injury and leads to the development of heart failure. Recent studies have confirmed the involvement of multiple transcription factors in the modulation of cardiovascular disease processes. However, the role of KLF9 in the inflammatory response induced by cardiovascular diseases including myocardial infarction remains unclear. Here, we found that the expression of KLF9 significantly increased during myocardial infarction. Besides, we also detected high expression of KLF9 in infiltrated macrophages after myocardial infarction. Our functional studies revealed that KLF9 deficiency prevented cardiac function and adverse cardiac remodeling. Furthermore, the downregulation of KLF9 inhibited the activation of NF-κB and MAPK signaling, leading to the suppression of inflammatory responses of macrophages triggered by myocardial infarction. Mechanistically, KLF9 was directly bound to the TLR2 promoter to enhance its expression, subsequently promoting the activation of inflammation-related signaling pathways. Our results suggested that KLF9 is a pro-inflammatory transcription factor in macrophages and targeting KLF9 may be a novel therapeutic strategy for ischemic heart disease.
Inflammatory bowel disease (IBD) is a chronic relapsing immune-mediated disease of the intestinal tract. Although its prevalence is reportedly lower in Asia than in Western countries, the rapid increase in the incidence of IBD has drawn attention to its etiology, including genetic susceptibility and environmental factors. Specifically, recent studies concerning dietary treatments and intestinal microbiota suggest that these factors may interact with the immune system, and the imbalance of this relationship may lead to immune dysregulation in IBD. Changes in diet or alterations in the composition of the intestinal microbiota may be associated with the increasing incidence of IBD in Asia. Here, we aim to review recent studies on the role of diet and intestinal microbiota in IBD pathogenesis and the results of the investigations performed to modulate these factors.
본 연구는 구실잣밤나무(C. cuspidate) 잎 추출물의 항염증 활성이 염증성 매개인자 생성 저해 및 염증성 사이토카인의 억제와 관련이 있을 것으로 예상되어짐에 따라, 구실잣밤나무를 대상으로 80% 에탄올를 가지고 추출한 후 추출물을 극성에 따라 순차적으로 용매분획을 실시하여, 구실잣밤나무 80% 에탄올 추출물 및 용매분획물들이 염증반응의 주체가 되는 대식세포 계열인 RAW 264.7 세포에서 LPS로 유도된 NO의 생성억제효과, 그리고 iNOS와 COX-2의 mRNA와 단백질 발현 억제효과 및 IL-6와 IL-$1{\beta}$와 같은 염증성 사이토카인 생성 억제효과 등을 알아보았다. 대식세포 계열인 RAW 264.7 세포에 LPS로 자극을 주고 구실잣밤나무 80% 에탄올 추출물 및 분획물을 처리하여 확인해본 결과, 헥산, 디클로로메탄 및 에틸아세테이트 분획물에서 NO의 생성억제 효과가 강하게 나타났으며, 헥산과 디클로로메탄 분획물에서는 iNOS와 COX-2 생성 억제 효과가 다른 분획물에 비해 강하게 나타났으며 염증성 사이토카인 생성 억제 효능도 80% 에탄올 추출물 및 분획물들이 다소 차이는 있었지만 헥산과 디클로로메탄 분획물에서 IL-6와 IL-$1{\beta}$에서 생성억제 효과가 나타났다. 이러한 결과는 구실잣밤나무 잎에서 유효성분 추출을 통한 항염증 물질의 연구 또는 예방하거나 치료할 수 있는 염증 억제 성분의 분리 및 그 작용기전 연구에 중요한 기초 자료가 될 것이라 사료된다. 또한 구실잣밤나무 추출물로부터 염증억제 성분을 도출하고자 활성분획인 디클로로 메탄 분획물에 대하여 활성성분의 분리가 진행 중이다.
여드름은 만성 염증 질환으로 주로 청소년기에 발생하는 것으로 알려져 있으나 대기오염, 약물 남용 등의 원인에 의해 아동기 및 성인기에도 나타날 수 있다고 알려져 있다. 여드름을 유발하는 정확한 원인은 밝혀져 있지 않으나 한 가지 원인보다 스트레스, 호르몬의 변화, 유전, 외부 환경 등 다양한 요인들이 복합적으로 작용한다고 여겨지고 있다. Propionibacterium acnes (P. acnes)는 여드름을 유발하는 균으로 모낭 내에 상주하여 피지의 중성지방을 분해하고 유리 지방산을 형성하여 모낭 내 염증을 유발한다. 따라서 피지의 생성 증가는 P. acnes의 생존에 좋은 영향을 주고 피부에서 염증반응을 유발하는 monocytic cell의 활성화와 염증성 사이토카인(pro-inflammatory cytokine)의 증가를 유발한다. 따라서 여드름 치료를 위해서는 P. acnes의 증식 억제 및 염증반응을 최소화하는 것이 중요하다. 본 논문에서는 유칼립투스(Eucalyptus globules) 추출물이 P. acnes에 의한 염증반응에 나타내는 효과를 확인하고자 하였다. 유칼립투스 추출물 처리는 P. acnes가 유도하는 염증 매개자로 알려진 $TNF-{\alpha}$, $IL-1{\beta}$, IL-2와 인플라마좀 복합체인 NLRP3의 유전자 발현을 효과적으로 감소시키는 것을 확인하였다. 뿐만 아니라 염증성 사이토카인의 유전자 발현에 중요하다고 알려진 전사인자(transcription factors) $NF-{\kappa}B$와 NFAT의 활성 역시 유칼립투스 추출물을 처리하였을 때 감소하는 것을 알 수 있었다. 따라서 본 논문을 통해 유칼립투스 추출물이 P. acnes에 의해 초래되는 여드름의 치료 보조제로 사용될 수 있으며, 천연 추출물의 사용이 항생제 장기 복용으로 인해 유발되는 항생제 내성을 해결하는 좋은 대안이 될 것이라고 예상할 수 있다.
본 연구는 자소자 열수추출물과 압력은 일정하게 유지한 상태로 온도를 조절하여 추출한 3종의 자소자 초임계추출물에 대해 항산화력, 항균활성, biofilm 형성 억제능과 염증매개물질(nitric oxide), 염증성 사이토카인(interleukin-6, interleukin-8)의 생성 억제능을 평가하였다. 자소자 열수추출물과 비교하였을 때 자소자 초임계추출물이 항산화 효과는 떨어졌으나, Staphylococcus aureus(S. aureus)에 대한 minimal inhibitory concentration(MIC)이 월등히 낮은 것으로 확인되었고, biofilm 형성 억제능 또한 더 우수하였다. 염증매개물질과 염증성 사이토카인의 생성도 자소자 초임계추출물이 자소자 열수추출물에 비해 현저히 감소시킬 수 있었다. 따라서 우리는 초임계추출물중 가장 우수한 항균작용과 biofilm 형성 억제, 염증인자 생성 억제 효능을 보인 45℃로 추출한 자소자 초임계추출물이 아토피 피부염 환자의 가려움증과 경표피수분손실을 방지하기 위한 천연 기능성 소재로 적합함을 시사한다.
The pathophysiology and fundamental etiologic mechanism of inflammatory bowel disease (IBD) is not well understood even though therapeutic regimens and drugs are rapidly evolutionary. IBD has complicated connections with genetic, immunologic, gut microbial, environmental, and nutritional factors. It is not clearly well known to the physicians how to feed, what nutrients are more helpful, and what food to be avoided. This review discusses the issues of growth and important nutritional concerns in the management of IBD in childhood.
