Development of an Environmental Friend Pellet Coating Oil for Replacement of Antibiotics for Ruminant Aninmal (천연물질을 이용한 반추동물 항생제 대체용 친환경 펠렛코팅오일의 개발)

  • This study investigated the effect of coated feed using mixed oil on rumen fermentation characteristics. Two experiments were conducted based on materials that were mixed. First, cashew nut and soybean oils were mixed with white mineral oil. And second, different plant extracts were mixed with white mineral oil. At first experiment, inclusion levels of mixed oil on diet (0.03%, 0.1%, 0.5%) were applied as variables. A coated diet was fermented with rumen inoculum according to in vitro rumen fermentation and its parameters were investigated. In the result of first experiment, no negative effects on rumen pH were found. Significantly decreased dry matter digestibility was detected at 0.5% treatment (P<0.05). Total gas productions in control and 0.03% were significantly greater than those of others (P<0.05). Significantly reduced methane productions were found in all treatments compared to the control (P<0.05). Inclusion of mixed oil did not affect on ammonia production. Total volatile fatty acid production was also not influenced by coating with mixed oils. Rumen fermentation parameters were greatly changed according to introduced plant extracts at second experiment. The significantly lowest and greatest ammonia productions were found at treatments with Ixeris dentata and Plantago asiatica, respectively (P<0.05). The significantly greatest acetate and propionate productions were detected at treatments with Crucuma longa and Zizyphus vulgaris 1, respectively (P<0.05). All treatments, except Chrysanthemum idicum, Euyale ferox seed, Moringa leaf and fruit and Zizyphus vulgaris 1, showed significantly increased total volatile fatty acid production compared to the control (P<0.05). Only Paeonia lactiflora showed significantly lesser gas production than the control (P<0.05). In methane production, Ceramium, Zizyphus, Paeonia, Agrimonia, Torilis, Mugwort, Foeniculum, Euphorbia, Taraxacum, Artemisia, Momordica, Curcuma and Moringa reduced methane significantly compared to the control (P<0.05).

  • We have studied the mycelia growth of four soil-borne fungal pathogens under light qualities and two lighting types (continuous and intermittent) provided by light-emitting diodes (LEDs). As a result, each mycelia growth on Phytophthora cactorum KACC40166, Athelia rolfsii KACC40170, and Helicobasidium mompa KACC40836 strain showed the similar growth rates within 10% or less difference among treatments compared to dark control, regardless of lighting types. However, the mycelia growth on Rosellinia necatrix KACC40168 strain was significantly suppressed by blue, blue+green and blue+red LED as well as fluorescent lamp compared to a dark control, in common with lighting types. The melanin pigment on R. necatrix KACC40168 strain showed relatively to induce more strongly under green LED and fluorescent lamp, whereas no induction under red LED and a control, regardless of lighting types. Thus, the hypha width on R. necatrix KACC40168 was significantly thinned by blue and blue+green LED compared to a control, in common with lighting types.

  • Rice bran is by-product from the dehulling process of rice, an important staple food in Korea. Various studies have been reported immune-enhancing effects of rice bran cultured with Lentinus edodes. In particular black rice bran that contains anthocyanin has the beneficial effects on immune system. SuperC3GHi, a recently released black colored rice cultivar, has higher contents of cyanidine-3-glucoside (C3G), a strong antioxidant than those of currently cultivated black rice cultivars. In this study, we investigate the beneficial effects of fermented SuperC3GHi rice bran (C3G-F) with Lentinus edodes. Using in vivo and in vitro model for inflammation and allgergy, anti-inflammatory and anti-allergic effects of C3G-F were determined. C3G-F induced tumor suppressive effect on Sarcoma-180 cell and B16 melanoma cell injected to mice. These results suggest that the capacity of C3G-F seems to act as a potent immunomodulator and with the absence of notable side-effects, C3G-F could be used as a novel functional food ingredient having possible therapeutic effects against immunological disorders.

  • This study aims to investigate the effects of hydrogen peroxide and grapefruit seed extract used as a chemical and natural disinfectants on human conjunctival cells in vitro. The main component of grapefruit seed extract is a narigin. It is one of the flavonoid types in citrus fruits and f1avonoids are widely recognized as naturally occurring(삭제) antioxidants. Cytotoxicity was determined by mitochondrial activity(MTT assay) and DNA damage was analyzed by measuring Comet assay. In LDH assay, 5% of grapefruits seed extract has been observed as a material is giving recovery effect of damaged cultured conjuctival cells by hydrogen peroxide. And also, each of concentrations has been treated simultaneously with same amounts and cytotoxicity of hydrogen peroxide and grapefruit seed extract have been estimated by LDH leakage assay after 24 hours. In conclusion, H2O2-induced cytotoxicity, apoptosis were Significantly prevented by grapefruit seed extract. It is a main component of bioflavonoids that we can simply take it as food. The present results suggest that grapefruit seed extract is a useful disinfectanct having antioxidant and antiapoptopic activity as a natural product.

The Effect of the Addition and Removal of Various Cryoprotectants on the Nuclear Maturation and ATP Content of Immature Porcine Oocytes

  • This study was undertaken to investigate the influence of exposure and removal of four different cryoprotectants (CPAs) on the ATP content of cumulus cell-enclosed (COs) and cumulus cell-denuded (DOs) immature porcine oocytes. The in vitro nuclear maturation of the COs, exposed to and removed from the CPAs was also assessed. Both COs and DOs were exposed to 1.5 M concentrations of each CPA (ethylene glycol (EG); propylene glycol (PG); dimethyl-sulfoxide (DMSO); and glycerol (G)) for durations of 5, 15, and 30 minutes at room temperature ($23.5{\pm}1.5^{\circ}C$), and immersed in physiological saline supplemented with 20% (v/v) fetal bovine serum for 5 minutes ($39^{\circ}C$) to remove each CPA. Before, during and after exposure to each CPA, the ATP content of both the COs and the DOs was measured. After removal from each CPA an aliquot of the COs was matured for 44${\pm}$2 h, and their nuclear maturation rates were measured up to the metaphase stage of the second meiotic division (the M-II stage). The maturation rates up to the M-II stage were not significantly different between all the groups that were exposed to each CPA for 5 minutes. For 15 and 30 minute exposures, the maturation rates of the COs exposed to PG, DMSO and EG were almost the same as those of the control groups; however, the rates of G group exposed for 15 and 30 minutes were significantly lower (p<0.05) than the control group. These groups were also found to have a decrease in the ATP content of COs and DOs during and after exposure for the same periods (p<0.05). In addition, although the ATP contents of the COs after exposure to EG for any period were the same as the controls, the ATP content of the DOs exposed to EG for any period were significantly lower (p<0.05) than those of the controls. When the ATP content of the COs and DOs of each CPA were compared, the DOs exposed to PG were found to have a significantly greater level (p<0.05) than DOs exposed to G for any duration. In addition, the ATP content of DOs exposed to PG for 30 min and removal was also higher (p<0.05) than when exposed to DMSO for the same period. These findings indicate that PG may be a useful CPA for the cryopreservation of immature porcine oocytes.

  • Local extravasation during intravenous administration of adriamycin (doxorubicin HCl) can cause severe skin ulceration and necrosis. To investigate the mechanism of adriamycin-induced skin toxicity, effects of adriamycin on reactive oxygen radical metabolism using cultured skin cells of fetal rat. Adriamycin produced significant release of lactic dehydrogenase from cultured skin cell preparations dose- and time-dependently. The production of superoxide anion in sonicated suspensions of cultured skin cells was significantly increased by adriamycin under the presence of NADPH and NADH. The drug also stimulated malondialdehyde (MDA) production, an index of lipid peroxidation, in NADPH- and NADH-supported cell preparations. The increased production of MDA was significantly inhibited by oxygen radical scavengers (superoxide dismutase, catalase, thiourea) and antioxidants (butylated hydroxytoluene, ${\alpha}-tocopherol$). Treatment of cultured skin cells with 1, 3,-bis (2-chloroethyl)-1-nitrosourea (BCNU), an inhibitor of glutathione reductase, enhanced the lipid peroxidation induced by adriamycin. The present study suggests that lipid peroxidation which is resulted from the stimulated production of reactive oxygen radical causes cellular damage in adriamycin-treated skin cells of rat.

Effect of White and Red Ginseng Extracts on the Immunological Activities in Lymphocytes Isolated from Sasang Constitution Blood Cells (백삼 및 홍삼 추출물의 사상체질별 면역세포 활성효과)

  • The effects of white ginseng and red ginseng ethanol extracts on the proliferation, nitric oxide (NO) production and tumor necrosis factor-$\alpha$ (TNF-$\alpha$) in human Band T lymphocytes isolated from blood cell of sasang constitution were examined in vitro. White ginseng extract known as medicine suitable for soeumin constitution showed the highest immune activities such as proliferation, NO production and TNF-$\alpha$ in soeumin constitution, and followed by taeeumin and soyangin, respectively (P<0.05). However, there were immunologically insignificant differences among sasang constitution lymphocytes treated by red ginseng ethanol extract. Both white and red ginseng ethanol extracts showed significantly higher immunological activities than LPS-induced sasang constitution lymphocytes on NO production and TNF-$\alpha$ release (P<0.05). The different immunological activities of white and red ginseng extracts in sasang constitution lymphocytes might be due to the component changes during the processing of ginseng. Thus, the findings of components suitable for all constitutions from red ginseng extract could be applied as a new constitution marker.

  • The cells associated with normal defense mechanism in inflammation release free oxygen radicals, hydroxy radicals, and various protease, all of which can damage the surrounding cells(fibroblasts) and matrix molecules(collagen). The objective of this study was to evaluate the effects of "scavenger" enzyme, superoxide dismutase(SOD). to periodontal ligament (PDL) cells. Human PDL cells were cultured from the teeth extracted for non-periodontal reason. Cultured PDL cells in vitro were treated with SOD and LPS according to dosage and culture times. Cellular activity was exaimed by Microtitration(MTT) assay. The quantitative expression of cellular proliferation by proliferating cell nuclear antigen(PCNA), collagen type I and fibronectin by indirect immunocytochemically stain in PDL cells were done. The results were as follows: 1. As only SOD treated group at 2 and 3 days, PDL cell activity was significantly increased at more than 150U(P<0.05). 2. When LPS(0.5, $5{\mu}g/m{\ell}$) and SOD(more than 150U) were added together, it was significantly increased than LPS only treated and control groups at 2 days(P<0.05). 3. When LPS($5{\mu}g/m{\ell}$) and SOD(150, 300U) were added together, PCNA index was significantly increased than LPS only treated and control groups at 2 and 3 days(P<0.05). 4. When LPS($5{\mu}g/m{\ell}$) and SOD(150U) were added together, collagen type I was significantly increased than LPS only treated and control groups at 3 days(P<0.05). 5.When LPS($5{\mu}g/m{\ell}$) and SOD(300U) were added together, fibronectin was significantly increased than LPS only treated and control groups at 3 days(P<0.05). On the above the results, the SOD in association with collagen type I, fibonectin, and PCNA may afford biological protection to oxy-radicals that were typically liberated during normal inflammatory response. Thus, the exogenous application of SOD may be effective in sthe treatment of the localized breakdown associated with chronic periodontal disease.

EFFECTS OF POLYPHOSPHATE MIXED IN ACRYLIC RESIN ON THE ATTACHMENT AND GROWTH OF ORAL BACTERIA (Acrylic Resin에 혼합된 Polyphosphate가 미생물의 부착 및 성장에 미치는 영향)

  • The purpose of this present study was to develop a new way of self-curing acrylic resin, using commercially available polyphosphate, Calgon, which is known to be antimicrobial and safe. For the study, polyphosphate(polyP) was blended with acrylic powder and devided into four groups as follows: no polyP(control), 1% polyP, 2% polyP, and 3% polyP. For the experiment, Streptococcus mutans GS5, Streptococcus sobrinus 6715, Streptococcus gordonii G9B and Challis, Porphyromonas gingivalis 2561, and Candida albicans ATCC 90027 were used. Resin specimens in each group were tested in vitro for the purpose of investigating the effect of polyP on the microbial attachment, growth and hydrophobicity of the resin surface. The results were as follows. 1. PolyP added to the acrylic resin decreased attachment of S. mutans GS5, S. sobrinus 6715, S. gordonii G9B. The greater binding inhibition was found in acrylic resin polymerized with polyP at higher concentrations. 2. The addition of polyP to acrylic resin failed to significantly affect the growth of the tested microorganisms. 3. The addition of polyP to acrylic resin seemed to reduce hydrophobicity of the acrylic resin. PolyP in acrylic resin does not seem to exert a direct antibacterial activity, but rather inhibit attachment of oral bacteria, especially mutans streptococci to saliva-coated acrylic resin. The acrylic resin reduces attachment of streptococci may be due to the decreased hydrophobicity caused by polyP added to the resin. PolyP may be included to acrylic resin to inhibit dental caries which often occurs when removable acrylic resin appliance is placed.

Biological Evaluation of the Methanolic Extract of Eriobotrya japonica and Its Irradiation Effect (비파 메탄올 추출물의 생리활성 및 방사선 조사 효과)

  • Eriobotrya japonica has been used as a folk medicine for treatment of skin diseases, inflammation, coughing, phlegm, and ulcers in Korea and other Asian countries. In a search for possible bioactive agents from natural sources, we found that the methanolic extracts from various parts of E. japonica showed moderate antioxidative and antimicrobial activities in several in vitro bioassay systems. Additionally, the respective parts of E. japonica were irradiated at 20 kGy to investigate the effects of irradiation. Gamma irradiation of E. japonica extracts removed the deep greenish color without affecting its natural biological activities such as its antioxidative and antimicrobial properties. Based on these findings, the methanolic extracts of this plant source may be not affected by gamma irradiation as its bioactive constituents may be insensitive to this irradiation. Moreover, the methanolic extract of E. japonica may serve as a good natural resource for beneficial functions in food and other related industries.
