한삼덩굴에 대한 최근 국내외 연구 동향 분석 - 의약학 연구를 중심으로 - (Analysis on Recent Studies Trends of Humulus japonicus - Focusing on Research of Medical Sciences -)

  • Objective The purpose of this study was to provide information on the international trends in Humulus japonicus by analyzing recently reported papers and theses. Methods Papers on the medicinal in vivo and in vitro activities of Humulus japonicus were reviewed and analyzed. Data on the publication year, research and experimental methods, and subjects were extracted and summarized. Results Twenty-eight papers with recent experimental studies and clinical trials related to Humulus japonicus were included. The following results were obtained from this study. 1. An average of 2.8 papers were published annually from 2014 to 2023. 2. There were 27 experimental studies and one clinical trials during the study period. 3. Among the studies, there were 13 in vivo, seven in vitro, and seven with both types of experiments. 4. Based on the subject area, studies on neuropsychiatric activity were the most numerous, totaling six reports. Conclusions Humulus japonicus has been studied for various activities, including neuropsychiatric, anti-inflammatory, liver function-related, digestive system-related, antibacterial, anti-obesity, anti-aging, and antioxidant effects. However, more clinical research is needed to further explore its medicinal uses.

중금속류가 취절편의 Amylase 분비에 미치는 영향 (Effect of Heavy Metals on the Secretion of Amylase in Rat Pancreatic Fragments)

  • Heavy metals which are present as trace elements in human body have been known to modify various enzymatic reaction. These metals can be essential or non-essential. Zinc, copper and calcium are essential in maintaining some biological processes, whereas non-essential metals such as cadmium, lead and mercury produce accumulatve toxic effect. Cadmium accumulated in pancreas can cause toxicity and damage of pancreatic cells, thereby influencing CHO metabolism. Lead compounds are known to produce toxic effects on the kidney, digestive system and brain fellowed by inhibition of activity of ${\rho}-aminolevulinic$ acid and biosynthesis of hemoproteins and cytochrome. Evidence has been accumulated that zinc not only acts as a cofactor in enzyme reaction but also prevents toxic effect induced by heavy metal such as copper and cadmium. To demonstrate the effect of heavy metals on pancreatic secretion, part of uncinate pancreas was taken and incubated in Krebs-Ringer bicarbonate buffer with heavy metals used. Additional treatment with CCK-OP was performed when needed. After incubation during different period of time, medium was analyzed for amylase activity using Bernfeld's method. The present study was attempted in order to elucidate the effect of several kinds of heavy metal on exocrine pancreatic secretion in vitro. The results obtained are as follows: 1) CCK-OP stimulated significantly amylase release from pancreatic fragments in vitro. 2) CCK-OP response of amylase release from pancreatic fragments was inhibited by treatmant with cadmium, especially high doses of cadmium. 3) CCK-OP response of amylase release from pancreatic fragments was inhibited when pretreated with $10^{-4}M$ copper chloride. 4) Lead chloride at the concentration of $10^{-3}M\;and\;10^{4}M$ stimulated the basal amylase release in vitro but CCK-OP response did not augment by lead chloride. 5) Zine chloride did not affect amylase release from pancreatic fragment in vitro. From the results mentioned above, it is suggested that CCK-OP response was inhibited it the amylase release from pancreatic fragments pretreated with cadmium and copper chloride.

Multilayer Coating with Red Ginseng Dietary Fiber Improves Intestinal Adhesion and Proliferation of Probiotics in Human Intestinal Epithelial Models

  • To exert their beneficial effects, it is essential for the commensal bacteria of probiotic supplements to be sufficiently protected as they pass through the low pH environment of the stomach, and effectively colonize the intestinal epithelium downstream. Here, we investigated the effect of a multilayer coating containing red ginseng dietary fiber, on the acid tolerance, and the adhesion and proliferation capacities of three Lactobacillus strains (Limosilactobacillus reuteri KGC1901, Lacticaseibacillus casei KGC1201, Limosilactobacillus fermentum KGC1601) isolated from Panax ginseng, using HT-29 cells, mucin-coated plates, and human pluripotent stem cell-derived intestinal epithelial cells as in vitro models of human gut physiology. We observed that the multilayer-coated strains displayed improved survival rates after passage through gastric juice, as well as high adhesion and proliferation capacities within the various gut epithelial systems tested, compared to their uncoated counterparts. Our findings demonstrated that the multilayer coat effectively protected commensal microbiota and led to improved adhesion and colonization of intestinal epithelial cells, and consequently to higher probiotic efficacy.

Antioxidant and Immunoenhancement Activities of Ginger (Zingiber officinale Roscoe) Extracts and Compounds in In Vitro and In Vivo Mouse and Human System

  • Ginger (Zingiber officinale Roscoe) is traditionally used as appetite enhancer, improver of the digestive system, antithusive, anti-cold, antipyretic, analgesic, and antiinflammation. In vitro evaluation using human lymphocyte cultures showed almost similar indication with those in in vivo mouse study, NK cell lysing activity was improved significantly. Proliferation activity of B and T cells, and CD3$^{+}$ and CD3$^{+}$CD4$^{+}$T cell subset were better observed using oleoresin or gingerol and shogaol fractions. Although there were higher activities in gingerol, the improvement was almost equal to that by oleoresin. Shogaol did not show better improvement except at higher concentration. It could be concluded that treatment with single bioactive compound, such as gingerol, did not show significant effects compared to oleoresin, the crude extract. In human study, involving healthy male adult, the improvement of NK cell lysing activity was again demonstrated and even more apparent. The mechanism involved in the protection seemed to be through the antioxidant activity of gingerol. However, other mechanism underlying the improvement of NK cell lysing activity must be involved since this improvement seemed to be specifically toward NK cell activity. Since NK cells ave specific for the elimination of virus-infected cell and mutated cells, this positive effect on the immune system are very interesting. This work has also scientifically proved that the traditional beliefs that ginger had preventive effects on common cold appeared to be reasonable.

군리탕가감방(君理湯加減方)이 항종양(抗腫瘍) 면역반응(免疫反應)과 항암제로 유발(誘發)한 부작용(副作用)에 미치는 영향(影響) (Effects of Gunleetang Gagambang Extract on Antitumoral Immunological Response and the Side Effect Induced by Antitumoral Agents)

  • Even though appropriate immune response is necessary for the survival of the individual, excessive or insufficient immune Response might cause autoimmune or allergic disease. So the immune response must be controlled to the degree that is beneficial for the well being of the individual. This study was undertaken to know the effects of Gunleetang Gagambang on the immune system of the mouse. Gunleetang Gagambang has been used for cure of tumor as a traditional medicine without any experimental evidence to support the rational basis for its clinical use. This study was carried out to evaluate the possible therapeutic or antitumoral effects of Gunleetang Gagambang extract against tumor, and to carry out some mechanisms responsible for its effect. Some kinds of tumor were induced by the typical application of 3-methylcholanthrene(MCA) or by the implantation(s.c) of malignant tumor cells such as leukemia cells(3LL cells) or sarcoma cells(S180 cells). Treatment of the Gunleetang Gagambang on water-extract(dailly 1mg/mouse, i. p.) was continued for 7 days prior to tumor induction and after that the treatment was lasted for 20 days. Against squamous cell carcinoma induced by MCA, Gunleetang Gagambang decreased not only the frequency of tumor production but also the number and the weight of tumors per tumor bearing mice(TBM). Gunleetang Gagambang on also significantly suppressed the development of 3LL cell and S180 cell-implanted tumors in occurrence-frequency and their size. and some developed tumors were regressed by the continuous treatment of Gunleetang Gagambang extract into TBM. In vitro, treatment of Gunleetang Gagambang extract had no effect on the growth of some kinds of cell line such as FsaII, A431 strain but significantly inhibited the proliferation of 3LL, S180 cells and augmented the DNA synthesis of mitogen-activated lymphocytes. Gunleetang Gagambang also stimulated the migrative ability of leukocyte, the MIF and IL-2 production of T lymphocytes, but not IL 6 production of B cells. Gunleetang Gagambang administration to mice enhanced NK cells activities. These results demonstrated that Gunleetang Gagambang extract exhibited a significant prophylactic benefits against tumors and its antitumor activity was manifested depending on the type of tumor cells. And these results also suggested that effect of Gunleetang Gagambang might be chiefly due to nonspecitie enhancement of NK cell activities and cell-mediated immune responses.

SMA를 이용한 캡슐 내시경의 이동메커니즘 설계 및 제작 (Design and fabrication of the Locomotive Mechanism for Capsule Endoscopes Using Shape Memory Alloys (SMA))

  • Newly commercialized wireless capsule endoscope has many advantages compared to conventional push-type endoscopes. However, it is moved by the peristaltic waves. Therefore, it can not diagnose desired zones actively. In this paper, a locomotive mechanism for wireless capsule endoscope is proposed to increase the efficiency of endoscopy. We designed and fabricated a prototype using SMA springs and bio-mimetic clamping device. The hollow space in the prototype is allocated for further system integration of a camera module, a RF module and a battery. And the sequential control scheme is employed to improve the efficiency of its locomotion. To validate the performance of the locomotive mechanism, experiments on a silicone rubber pad and in vitro tests are carried out. The results of the experiments indicate that proposed mechanism is effective in harsh environments such as digestive organs of a human.

Cytochrome P450 유도제에 노출시킨 명주조개 (Coelomactra antiquata) 약물대사효소계의 in vitro 반응 (In vitro Response of the MPO System of the Clam, Coelomactra antiquata with Exposure to Cytochrome P450 Inducers)

  • 강원 북부 연안에 많은 명주조개 (Coelomactra antiquata)의 중장선으로 미크로좀을 만들고, cytochrome P45O (CYP)의 유도제로 알려진 phenobarbital (PB), 3-methylcholanthrene (MC) 및 $\beta$-naphthoflavone (BNF)을 DMSO에 녹여 함께 30$^{\circ}C$의 수조 상에서 in vitro로 배양하면서 약물대사효소계의 CYP 함량과 7-othoxyresorufin-O-deethylase (EROD) 활성의 변화를 경시적으로 조사하였다. 그리고 비교를 위한 sham구는 미크로좀에 DMSO만을 첨가하여 같은 조건으로 배양하였다. 그 결과, sham구의 CYP 함량은 배양 6시간이 지나서는 처음 수준의 $91\%$로 줄었으며, 0.1 mM과 0.2mM, 0.4mM 및 1.0 mM BNF 배양구는 배양 4$\~$6시간 후에 최대값이 되었는데 각각 처음 보다 $116\%$, $114\%$, $126\%$$132\%$로 되었다. PB와의 배양에서는 0.1 mM과 0.4 mM 및 1.0 mM 배양구는 배양 4$\~$6시간 후에 최대 값이 되었고 각각 처음의 $112\%$, $114\%$$114\%$로 되었으며, MC와의 배양에서도 0.1 mM과 0.4 mM 및 1.0 mM 배양구는 배양 4시간 후에 최대값이 되어 각각 $116\%$, $130\%$$132\%$로 증가하였다. 그리고 EROD 활성의 변화도 sham구는 배양 7시간 후에도 변화가 거의 없었으나, BNF의 0.1 mM과 0.4 mM 및 0.8 mM 배양구는 배양 7시간 후에 각각 처음의 $120\%$, $123\%$$115\%$로 되었고, PB의 0.1 mM과 1.0 mM 배양구는 배양 3시간 후에 최대로 되어 각각 $121\%$$136\%$로 증가하였으며, MC와의 배양에서는 0.1 mM과 0.4 mM 및 0.8 mM 배양구는 배양 5시간 후에 최대값이 되어 각각 $204\%$, $198\%$$191\%$로 증가하였다. 이처럼 BNF나 PB, MC는 명주조개에서도 미크로좀의 CYP 함량과 EROD 활성을 증가시키는 유도효과를 나타내었으며, 특히 MC에 의한 유도효과가 가장 강했고 BNF, PB의 순이었고, 대체로 약물의 농도가 높을수록 유도 효과도 큰 경향을 보였다.

자가미세유화를 통한 아토르바스타틴 칼슘의 난용성 개선 (Improvement of Solubility of Atorvastatin Calcium Using Self-Microemulsion Drug Delivery System(SMEDDS))

  • SMEDDS is mixture of oils, surfactants, and cosurfactants, which are emulsified in aqueous media under conditions of gentle agitation and digestive motility that would be encountered in the gastro-intestinal(GI) tract. The main purpose of this work is to prepare self-microemulsifying drug delivery system(SMEDDS) for oral bioavailability enhancement of a poorly water soluble drug, atorvastatin calcium. Solubility of atorvastatin calcium was determined in various vehicles. Pseudo-ternary phase diagrams were constructed to identity the efficient self-emulsification region and particle size distributions of the resultant micro emulsions were determined using a laser diffraction sizer. Optimized formulations for in vitro dissolution and bioavailability assessment were $Capryol^{(R)}$ 90(50%), Tetraglycol(16%), and $Cremophor^{(R)}$ EL(32%). The release rate of atorvastatin from SMEDDS was significantly higher than the conventional tablet ($Lipitor^{(R)}$), 2-fold. Our studies illustrated the potential use of SMEDDS for the delivery of hydrophobic compounds, such as atorvastatin calcium by the oral route.

  • Kefir exhibits antimicrobial activity in vitro against gram-positive and gram negative bacteria, as well as some fungi. The ability of LAB to inhibit the growth of closely related bacteria is well known. This inhibition of pathogenic and spoilage microbes may be due to the production of organic acids, hydrogen peroxide, acetaldehyde, diacetyl, carbon dioxide, or bacteriocins. Lactobacilli are the major contributors to acid production and, hence, a determining factor in the flavor development in kefir. Lactic acid, proteolytic activity, and acetaldehyde are the essential flavor compounds in kefir. Both acid and bacteriocins contribute to the antimicrobial activity of kefir and kefir grains. Kefir is rich in proteins, calcium, vitamin $B_{12}$, niacin, and folic acid. Many studies have investigated the benefits of consuming kefir and have shown that it is a natural probiotic, which when consumed regularly, can help relieve intestinal disorders, promote bowel movement, reduce flatulence, and improve the overall health of the digestive system. Tibetan kefir, which is different from traditional kefir, is produced in China. It has been reported to exhibit antimicrobial activity that is nearly identical to that of traditional kefir. Kefir production is considered a rapidly growing food industry in China.

혈관 긴장성 조절에 미치는 반하백출천마탕 (Effects of BanhabackchulChunma-tang on the Vascular Relaxation and its mechanism)

  • Hypertension is not only a well-established cardiovascular risk factor but also increase the risk of atherosclerosis. Most studies conducted to investigate the effectiveness of treatment for cardiovascular disease such as hypertension have focused primarily on conventional drug and physiotherapeutic treatments. BanhabackchulChunma-tang(半夏白朮天麻湯:BCT) is popular herbal medicine used in clinic for the treatment of various symptoms of drulatory disorders and weakness of digestive system, including anorexa and nausea with vertigo, severe headache, vomiting and so on. However, the mechanisms underlying its efficacy are unknown. This study investigated the effects of BCT as an alternative medication on the contraction induced by phenylephrine and KCI in rat thoratic aorta. BCT revealed siginificant relaxation on phenylephrine-induced arterial contraction, but revealed noncompetitive effect on concentration responses of phenylephrine-induced contraction. Treatment of N-L/sup ω/ -argine methyl ester(L-NAME) and methylene blue(MB)(10/sup -5/M) reduced the relaxation of BCT. BCT also increased in vitro NO production. It suggest that the relaxation effect of BBT is related with NO pathway, becausse the effect of L-NAME and MB are due to inhibition of NO synthesis from endothelial cells. These results indicate that BCT would be effective in hypertension treatment and its mechanism of relaxtion on arterial contraction is likely to be related with NO production, blocking of α-receptor and signal transduction after receptor activation.
