In the present study, in vitro multiple shoot induction was achieved from cotyledonary node and stem nodal explants of cv. NARI-6 of safflower (Carthamus tinctorius L.). Among various growth regulators tested, MS salts and B5 vitamins supplemented with BA (6-Benzy-laminopurine) $17.76\;{\mu}M$ and KN (Kinetin) $6.96\;{\mu}M$ phytohormonal combination was found to be the most effective in initiating numerous shoot buds after 30 days of culture than BA ($4.44-44.39\;{\mu}M$) or KN ($2.32-46.40\;{\mu}M$) alone in the medium. In addition, 0.8% (w/v) agar (Hi-media) and 3.0% sucrose (w/v) was the optimum level for the formation of adventitious shoots. Further results showed the maximum shoot elongation occurred on MS medium with BA ($8.88\;{\mu}M$) and $GA_3$($11.56\;{\mu}M$) combinations. Efficient rooting occurred on quarter strength MS medium with NAA $10.74\;{\mu}M$. The regenerated plantlets were acclimatized and successfully transferred to the field.
Experiments were conducted to find out the optimum cultural conditions for adventitious shoot regeneration from leaf segments of Gypsophila paniculata L. Thidiazuron (TDZ) was remarkably effective for the regeneration of leaf segment in Gypsophila paniculata compared with BA and kinetin. TDZ showed the highest rate of regeneration at $3.0mg{\cdot}L^{-1}$, while kinetin did not affect the regeneration. BA in the medium increased vitrification. Shoot formation efficiency was much higher on $0.3mg{\cdot}L^{-1}$ of IAA-containing media than NAA-containing media. Regeneration of leaf segments was induced with the agar concentrations of 1.0, 1.2 and 1.6%. Dark treatment at the initial stage of the culture increased the rate of regeneration up to 75%. The leaf explants from the 3rd subcultured stock plants after meristem culture, showed the highest adventitious shoot regeneration efficiency.
Winter bud explants from 15 individual angelica tree (Aralia elata) were cultured in vitro to find out optimal conditions for somatic embryo induction as well as plant regeneration. Calli are induced and grown on MS medium supplemented with 1.0 mg/L 2,4-D for 4 weeks and subcultured on a half-strength MS medium without phytohormones to induce somatic embryos. Inter-simple sequence repeat (I-SSR) markers were analyzed with total DNAs extracted from the trees. Genotype effects on somatic embryo induction were examined by cluster analysis. Callus induction rate varied from 58.5 to 100% among the genotypes. Somatic embryo induction rate also greatly varied from 0 to 100% among the genotypes. There was a significant difference in somatic embryo induction rate even among the individual trees that showed close genetic relationships each other. This suggested that somatic embryo induction rate in Aralia elata be influenced by a few major specific genes rather than whole genomic similarity among individual trees. Four individuals of Ulneong-7, Cheju-1, Shingu and China, which are recalcitrant to somatic embryo induction, turned out to have a close genetic relationship, suggesting that both physiological and genetic factors affect somatic embryo induction. The results suggest that genotype selection be the most important factor to achieve an efficient propagation, although cultural optimization through medium and explant manipulation may also play crucial roles in somatic embryogensis as well as plant regeneration of these species.
Immature flowers and lateral buds of Neoregeria carorinae cv. Tricolor were cultured for micropropagation and the collecting times of materials, growth regulators and theirs concentrations, and cultural methods on the formation of adventitious buds and growth were investigated in this experiment The formation rate was the highest in immature flowers collected at 4weeks after flower bud differentiation and in buds at 7weeks after flower differentiation of adventitious buds. MS medium supplemented with 1.0 mg/L BA and 0.5 mg/L BA was the most favorable for the formation of adventitious buds. Solid medium was more effective for the formation of adventitious buds than liquid one. MS medium with 1.0 mg/L NAA was the most suitable for the rooting of regenerated shoot. Liquid medium was effective for the rooting of regenerated shoot than solid one.
Kiwi (Actinidia deliciosa) is exotic plant and thus susceptible to cold climate in the middle part of Korean peninsular. Several hybrids have recently been developed to enhance cold tolerance by crossing them with domestic species (A. arguta), We have developed an efficient micropropagation technique for the hybrids using both axillary bud and callus culture systems. Shoot proliferation from axillary buds was possible on St medium supplemented with 0.2 mg/L Bh and 3.0 mg/L GA$_3$. In vivo cuttings of the proliferated shoots were more effective for root induction and subsequent survival than in vitro rooting. More than 95% of the plantlets were successfully transferred to field. Effective callus induction was achieved on MS or B$_{5}$ medium with 2,4-D or NAA. Although callus induction could be made from any combinations of media and auxins, shoot regeneration was observed only from the callus induced on medium containing NAA.A.
This study was carried out to establish the micropropagation system of Bupleurum latissimum Nakai that is a Korean native endangered species. Callus were induced from the leaf, petiole and floral bud and the percentage of callus formation was highest in the floral bud on the MS medium containing 2.0 mg ${\cdot}$$L^{-1}$ 2,4-D. Especially, callus induced from floral bud was formed 77.8% and the percentage of shoot formation was 42.6% on the MS medium containing 2.0 mg ${\cdot}$$L^{-1}$ 2,4-D plus 1.0 mg ${\cdot}$$L^{-1}$ TDZ. For simultaneously callus formation and shoot regeneration, 1/2 MS medium was more effective than MS medium. The percentage callus formation, shoot regeneration and rooting were 46.3%, 13.0%, 13.0% in 1/2 MS medium, respectively. Soot regeneration from callus was good in 1/2 MS medium supplemented with 2.0 mg ${\cdot}$$L^{-1}$ 2,4-D plus 1.0 mg ${\cdot}$$L^{-1}$ BA where percentage of shoot regeneration was 74.1 %, and the number of shoot per explant was 2.4. The percentage of rooting was lowest (57.8%) in control while it was highest (97.8%) in 1.5 mg ${\cdot}$$L^{-1}$ NAA. In acclimatization of regenerated plantlets, the percentage of survived plantlets was highest (86.1%), and plant height, root length and fresh weight were good in the soil for horticulture.
In order to investigate the effect of plant growth regulators on effective in vitro micropropagation, apical meristems of Pulsatilla cernua var. koreana were cultured on Murashige and Skoog's (MS) medium with 2,4-D, NAA, TDZ and BA. Media containing 2,4-D and kinetin, 2,4-D and TDZ, NAA and TDZ were not effective on callus induction. However, embryogenic or organogenic callus was obtained on media containing NAA and BA. Especially, on MS medium with 0.5mg/L NAA and 1.0mg/L BA was optimal for a high frequency (62%) of shoot or shoot bud obtained from callus. Callus proliferation, shoot multiplication and elongation were significantly increased by adding 10% coconut water on MS media with 0.5mg/L NAA and 1.0mg/L BA. Repeated subculturing of in vitro grown shoots resulted in propagation rate of 12.9 shoots per explant every 30 days. Root formation from the adventitious shoots was not easily achieved. However, roots were only produced through callus on MS medium with 2.0mg/L NAA alone or 0.5mg/L NAA and 1.0mg/L BA. These roots were used materials for callus and shoot production repetitively.
Multiple shoots were induced from in vitro shoot originated from winter bud of P. yedoensis from Jeju. Most explants grow in similar type among the five different media but affected by supplement of sucrose regardless of media. For mass-propagation various concentrations of BAP or $GA_3$ were treated in the medium respectively. BAP was very effective to produce multiple shoots and 3.5~9.5 shoots were formed on the explant. The shoots induced on the high levels of BAP have short internodes. No shoots were induced on the treatment of $GA_3$ but roots were induced on it. When $GA_3$ was supplemented with the medium containing BAP, multiple shoots were produced from the explants. The medium(WPM) containing with $0.5mg/{\ell}$ BAP and $4.0mg/{\ell}$$GA_3$ was most effective to produce multiple shoots. When the explants were cultured for 8 weeks, 39.5 shoots were developed in average.

The Polygonatum odoratum cv. Gungangbeaksea, bred in Gyeongsangnam-Do Agricutural Research & Extension Service, was cultured in vitro for micropropagate rapidly through the culture of rhizome explants ($5{\times}5mm$). The $7{\times}7mm$ explants of adventitious multi-bud clusters (AMC), obtained through the culture of rhizome explants (MS + 3.0 mg/L BA) were cultured on MS media with BA and TDZ. The shoot multiplication was favorable on the MS medium containing 3.0 mg/L TDZ with 2.8 in shoot number. But the formation of AMC was low in all media tested. The explants of AMC were cultured on MS media containing 1.0~5.0 mg/L TDZ and NAA to multiplicate AMC more. The formation of AMC was a little more stimulated on combined MS media of TDZ and NAA, than that with TDZ alone. The multiplication of shoots and AMC was favorable on MS media with 3.0 mg/L TDZ and 5.0 mg/L NAA, and 5.0 mg/L TDZ and 3.0 mg/L NAA. As the concentration of MS salts increased, the formation of AMC was decreased. But the formation of AMC was more stimulated, as the concentration of sucrose increased to 7%. Therefore, the multiplication of shoots and AMC was suitable on media containing 3.0~5.0 mg/L TDZ and NAA, and 7% of sucrose. The explants of AMC were rooted on media with 3.0 mg/L IBA, or 2.0 mg/L NAA with more than 80% in rooting ratio. The plantlets were treated at $5^{\circ}C$ for 8 weeks, and cultured ex vitro for 8 weeks. The survival ratio of plantlets were 100% in vermiculite, and the mixed soil with perlite 1 volumn and vermiculite 1 volumn.
This study was undertaken to know the possibility of in vitro propagation of Stevia through axillary bud culture and the results indicated that: (1) Addition of NAA (0.01-0.05 mg/l) alone on Murashige-Skoog basal medium promoted shoot differentiation and growth rate. And also additional of kinetin of 0.5-1.0 mg/1 alone showed the same trend as that of NAA: (2) Addition of both NAA (0.01-0.05 mg/l) and kinetin (0.5-1.0mg/l) to MS medium promoted better shoot formation. (3) Shoot differentiation and growth were better on the full salt strength of MS medium (1X MS) than that of half strength ( $\frac{1}{2}$MS), while their effects were reversed for root differentiation
